LOXL3-related foveal hypoplasia in siblings.

PURPOSE: To report foveal hypoplasia and high myopia in siblings who harbored biallelic pathogenic variants in the gene LOXL3 (lysyl oxidase-like 3; biallelic) and highlight LOXL3 as a candidate gene for this phenotype. METHOD: Retrospective case series. RESULTS: Two siblings, a 12-year-old boy and his 2-year-old sister, had foveal hypoplasia in the setting of high myopia. Whole exome sequencing revealed both to be homozygous for NM_032603.5: LOXL3 (lysyl oxidase-like 3) c.1036C>T; p.Arg346Trp and NM_000315.4: PTH (parathyroid hormone) c.128 G>A; p.Gly43Glu. The LOXL3 variant has previously …

Xeroderma Pigmentosam: Case Report of Siblings.

AIM: This case report reveals how xeroderma pigmentosum (XP) influences the dental procedure and the required modification. It illustrates t … nd chemotherapeutic agents are commonly followed. CASE DESCRIPTION: Hereby, we report a case of XP siblings and their dental management. CONCLUSION: XP, a rare genetic disorder, can go undetected due to a lack of knowledge related to this condition. Pediatric dentists should make an effort to emphasize parental and patient anticipatory education for the …

Two siblings with triple A syndrome.

Triple A syndrome is a rare autosomal recessive disorder presenting as adrenal insufficiency, achalasia and alacrima, often accompanied by neurological complications. We present the cases of two siblings, a girl (patient 1) and a boy (patient 2) in their early adolescence, born from a consanguineous marriage. At the age of 4, patient 1 developed progressive dysphagia when consuming both solid and liquid foods, while patient 2 began displaying abnormal gait by 2 years. Both siblings were confirmed to have alacrima and achalasia. Endocrinological assessments unveiled adrenal insufficiency and re…

Clinical variability of BBS1 across siblings.

Bardet-Biedl syndrome (BBS), an autosomal recessive ciliopathy with pleiotropic effects, manifests as a spectrum of anomalies involving multiple genes and affects fewer than 3,000 individuals in the USA. Due to its rarity and phenotypic variability, early diagnosis of BBS poses a significant challenge. Therefore, we aim to shed light on the intrafamilial phenotypic variation of BBS resulting from a BBS1 variant by delineating the clinical presentation in two siblings.

Severe Congenital Lead Poisoning in Siblings.

Congenital lead toxicity is rare. Lead freely crosses the placenta, therefore, placing the developing fetus at a higher risk for lead toxicity. This can lead to adverse consequences, such as spontaneous abortion, low birth weight, low IQ, and neurodevelopmental impairment. We present a rare case of siblings born to a mother with pica behaviors. Her venous lead level was 42 μg/dL at the time of disclosure. The repeat venous level at delivery 2 weeks later was 61 μg/dL. The infant's venous level shortly after birth was 89 μg/dL. The neonate was transferred to the NICU, where he received 9 doses …

Pycnodysostosis: Clinical Insights From Two Siblings.

Pycnodysostosis is a rare autosomal recessive bone disorder caused by mutations in the cathepsin K (CTSK) gene, characterized by increased bone density, short stature, and skeletal fragility. This study reports on two siblings from a consanguineous marriage, observed at the Mohammed VI University Hospital in Oujda, Morocco. Both patients presented with typical symptoms, including craniofacial dysmorphism and skeletal abnormalities. Radiographic findings confirmed increased bone density and acro-osteolysis. The cases highlight the importance of early and accurate diagnosis, comprehensive manage…

Sedaghatian spondylometaphyseal dysplasia in two siblings.

Sedaghatian type spondylometaphyseal dysplasia (SSMD) is a rare skeletal dysplasia with only 24 reported cases to date. Despite the limited literature available, evidence suggests this is a multi-system disorder, with neurological and cardiovascular abnormalities reported in addition to the skeletal features. We report a new family with two affected siblings and detailed phenotypic description of the affected proband. Diagnosis in the neonatal period led to retrospective genetic diagnosis of a previous affected pregnancy that was terminated due to severe ventriculomegaly. We suggest that a dia…

Congenital ocular toxoplasmosis in consecutive siblings.

Toxoplasma gondii infection can cause ocular manifestations after acquired and congenital disease. We report two cases of symptomatic congenital toxoplasmosis with ocular involvement in non-twin siblings, with a 2-year interval between pregnancies. Vertical transmission of toxoplasmosis in successive pregnancies, which was once considered impossible, is now found to be plausible even in immunocompetent subjects.

Epilepsy with myoclonic absences in siblings.

BACKGROUND: Epilepsy with myoclonic absences (EMAs) is a distinct form of childhood epilepsy characterized by a peculiar seizure type that identifies this condition. PURPOSE: To describe the clinical, electroencephalographic features, treatment strategies and outcome in this first case series of two siblings with normal intelligence presenting with EMAs. MATERIALS AND METHODS: Both siblings underwent video-polygraphic investigations (simultaneous recording of electroencephalogram [EEG] and electromyogram [EMG] from deltoids), high-resolution magnetic resonance imaging (MRI), karyotyping, neu…

Testicular microlithiasis in siblings: clinical implications.

Testicular microlithiasis is a relatively uncommon condition in children. It is characterized by the presence of microcalcifications within the testicular parenchyma. Although it is a benign finding, underlying diseases and other conditions must be ruled out because testicular microlithiasis has been found in association with both benign and malignant lesions in the testes and other tissues. We present two brothers with testicular microlithiasis, and highlight the prevalence, natural history, associated malignant conditions and follow-up recommendations of children diagnosed with testicular mi

Mowat-Wilson syndrome affecting 3 siblings.

We herein report 3 cases of Mowat-Wilson syndrome, characterized by distinct facial features, severe psychomotor retardation, and epilepsy, recurring in 3 siblings from the same parents. The proband was a 15-month-old boy, the youngest of 3 children (2 elder sisters), who was referred to our hospital for the treatment of severe seizures. The clinical features and course of these 3 siblings were compatible with those of previously reported Mowat-Wilson syndrome patients, and all siblings had the same E87X nonsense mutation in ZFHX1B, whereas their mother did not show the mutation. Because Mowat…

Lipoid proteinosis: report of three siblings.

Lipoid proteinosis is a very rare genodermatosis characterized by infiltration of hyaline material into the skin, oral cavity, larynx and internal organs. It usually presents in infancy with hoarseness. Although about 300 cases have been reported in the literature, the occurrence of the disease is rare in siblings. In this report we introduce three siblings with this disease.

Persistent pupillary membranes in 3 siblings.

We report 3 siblings from a nonconsanguineous white family with bilateral persistent pupillary membranes with dissimilar forms of severity. Two of the patients required no treatment; 1 was treated surgically for occlusion of the pupillary axis and moderate visual acuity decrease. Anterior segment examination was otherwise normal in the siblings. Although persistent pupillary membranes are usually sporadic, cases in which they are associated with other anterior chamber abnormalities might be inherited autosomal dominant. To our knowledge, this is the first report of a familial isolated persiste…
