Antimicrobial effect of two fluoride-releasing adhesive tapes on Streptococcus mutans biofilm.

Fluoride-releasing adhesive tapes have been developed as a new fluoride delivery agent. However, application as caries prevention agents remains underexplored. This study aimed at evaluating the antimicrobial activity of two fluoride-releasing adhesive tapes against S. mutans biofilm. Two polyvinyl alcohol (PVA) tapes were investigated: (i) a fluoride-PVA (F-PVA) tape, (ii) a pullulan incorporated F-PVA (PF-PVA) tape. S. mutan strains were cultured and treated with the tapes. Antimicrobial effects were evaluated using the agar diffusion test, field-emission scanning electron microscopy (FE-SEM

ActA-mediated PykF acetylation negatively regulates oxidative stress adaptability of Streptococcus mutans.

UNLABELLED: Dental caries is associated with microbial dysbiosis caused by the excessive proliferation of Streptococcus mutans in dental biofilms, where oxidative stress serves as the major stressor to microbial communities. The adaptability of S. mutans to oxidative stress is a prerequisite for its proliferation and even for exerting its virulence. Protein acetylation is a reversible and conserved regulatory mechanism enabling bacteria to rapidly respond to external environmental stressors. However, the functions of protein acetylation in regulating oxidative stress adaptability of S. mutans …

Antimicrobial Peptides Targeting Streptococcus mutans: Current Research on Design, Screening and Efficacy.

Antimicrobial peptides (AMPs) are small-molecule peptides that play a vital role in the nonspecific immune defense system of organisms. They mainly kill microorganisms by physically destroying the cell membrane and causing the leakage of contents. AMPs have attracted much attention as potential alternatives to antibiotics due to their low susceptibility to resistance. Streptococcus mutans (S. mutans) is one of the main causative agents of human dental caries. The design, screening, and efficacy evaluation of AMPs targeting S. mutans offer new possibilities for the prevention and treatment of o…

Antibacterial photodynamic therapy with curcumin and Curcuma xanthorrhiza extract against Streptococcus mutans.

BACKGROUND: Bacteria are becoming increasingly resistant to conventional antibacterial chemotherapy. This has prompted the application of antibacterial photodynamic therapy (aPDT) in bacteria-related diseases due to its excellent biocide effects. However, few studies have attempted to develop a novel photosensitizer based on natural components. The aim of the present study was to compare the aPDT effects of curcumin and Curcuma xanthorrhiza extract (CXE) against Streptococcus mutans. METHODS: A planktonic suspension containing an S. mutans strain was treated in three separate groups: aPDT wit…

Pulpitis induction in baboon primary teeth using carious dentine or Streptococcus mutans.

The objective of this equivalency study was to see if a colony of Streptococcus mutans placed into cavities in primary molar teeth produced pulpitis similar to an established pulpitis induction method using carious dentine. In two juvenile baboons (Papio ursinus), occlusal cavities were cut in all 16 primary molar teeth, followed by making a small pulpal exposure after which the cavity was swabbed with 37 per cent phosphoric acid. In one half of the teeth, fresh soft human carious dentine was placed over the pulpal exposure; in the remaining teeth the exposure was covered with a colony of Stre…

Structure insight into FtsZ function maintaining under acid stress of Streptococcus mutans.

Understanding the acid resistance mechanism of S. mutans is crucial for preventing dental caries. FtsZ is the core protein for cell division in bacteria that can polymerize into Z-rings and drive cytokinesis. Our previous study revealed that the FtsZ in S. mutans (SmFtsZ) has higher self-assembly and GTPase activity under acidic stress, which may be responsible for acid resistance and cariogenesis of S. mutans. However, the functional structure mechanism of SmFtsZ under low pH conditions is still unclear. Here, we further reported the crystal structure of S. mutans FtsZ, revealing a unique lat

Acid shock triggers virulence surge accompanied by bacterial elongation in streptococcus mutans.

Pediatric caries is a major global oral health burden, children are particularly susceptible due to immature oral defense mechanisms, underdeveloped immune responses, and frequent exposure to sugar-rich diets that promote cariogenic bacterial activity. Streptococcus mutans (S. mutans) is widely recognized as a primary cariogenic pathogen, responsible for key virulence factors such as acid production, acid tolerance and exopolysaccharide (EPS) synthesis. Given that S. mutans can adapt to acidic environments through mechanisms such as the acid tolerance response (ATR), we cultured S. mutans in a…

Polymethyl methacrylate microplastics affect oral microbiota diversity and Streptococcus mutans biofilm formation.

INTRODUCTION: Polymethyl methacrylate (PMMA)-a widely applied dental resin-based material-contributes to oral PMMA microplastics (PMMA-MPs) exposure through masticating. PMMA-MPs may facilitate pathogen adhesion, posing substantial risks to oral health. Dental caries represents the most prevalent chronic infectious oral disease, characterized by progressive lesions that may induce pain, tooth structure loss, and compromised masticatory efficiency. Streptococcus mutans have been widely identified as the primary etiological agents responsible for caries pathogenesis. This study aimed to investig…

Cell wall glycosyltransferase of Streptococcus mutans impacts its dissemination to murine organs.

Streptococcus mutans, a cariogenic bacterium in humans, is associated with systemic disorders. Its cariogenic factors include glucosyltransferases (GTFs) and the glycosyltransferase rhamnose-glucose polysaccharide I (RgpI), which is involved in cell wall synthesis. However, the potential roles of these enzymes in systemic disorders remain unclear. We constructed a luciferase-tagged S. mutans UA159 mutant strain that lacked rgpI to explore the involvement of this enzyme in the systemic pathogenicity of S. mutans. We also employed the luciferase-tagged S. mutans UA159 variant, which exhibited re…

Streptococcus mutans and Cariogenic Biofilms: Mechanisms, Disruption Strategies, and Future Therapeutic Directions.

Dental caries, a significant global health concern, is intricately linked to the development and persistence of cariogenic biofilms on tooth surfaces. These complex microbial ecosystems orchestrate a multifactorial process of decay, adhering to teeth, producing acids that erode enamel, and exhibiting resistance to traditional therapies. This review delves into the intricate mechanisms of cariogenic biofilm formation and pathogenesis, exploring adhesion, acid production, extracellular polymeric substance (EPS) mediated acid tolerance, and nutrient acquisition within the biofilm. We then explore

Trans-Cinnamaldehyde Inhibition of Pyruvate Dehydrogenase: Effects on Streptococcus mutans Carbohydrate Metabolism.

Dental caries is a chronic oral infectious disease, and Streptococcus mutans (S. mutans) plays an important role in the formation of dental caries. Trans-cinnamaldehyde (CA) exhibits broad-spectrum antibacterial activity; however, its target and mechanism of action of CA on S. mutans needs to be further explored. In this study, it was verified that CA could inhibit the growth and biofilm formation of S. mutans. Further proteomic analysis identified 33, 55, and 78 differentially expressed proteins (DEPs) in S. mutans treated with CA for 1, 2, and 4 h, respectively. Bioinformatics analysis showe…

Relationship between dental fluorosis, dental caries and salivary levels of Streptococcus mutans.

BACKGROUND AND OBJECTIVE: Worldwide, dental caries is an endemic infection and a significant public health problem. Fluoride reduces caries by helping to prevent demineralisation, by remineralising early carious lesions, and by decreasing the activity and growth of Streptococcus mutans (S. mutans). Fluoride intake above the optimal levels leads to a condition known as dental fluorosis, which causes hypo-mineralisation of the tooth. Some studies have revealed that the severity of fluorosis is directly proportional to caries, but some showed opposite results. Hence, with these contradicting resu…
