Distribution, development, and identity of retinal ganglion cells labeled in the Sert-Cre reporter mouse.

The mouse retina contains over 40 types of retinal ganglion cells (RGCs) that differ in morphology, function, or gene expression. RGCs also differ by whether their axons target the brain.s ipsilateral or contralateral hemisphere. Contralaterally projecting RGCs (contraRGCs) are widespread in mouse retina, whereas ipsilateral projecting RGCs (ipsiRGCs) are confined to the ventro-temporal (VT) crescent of retina. In this study, we employed the Sert-Cre transgenic line, which had been reported to selectively label ipsiRGCs, to study ipsiRGCs during development. Although the number of Cre-expressi

Pathways for Naturalistic Looking Behavior in Primate I: Behavioral Characteristics and Brainstem Circuits.

Organisms control their visual worlds by moving their eyes, heads, and bodies. This control of "gaze" or "looking" is key to survival and intelligence, but our investigation of the underlying neural mechanisms in natural conditions is hindered by technical limitations. Recent advances have enabled measurement of both brain and behavior in freely moving animals in complex environments, expanding on historical head-fixed laboratory investigations. We juxtapose looking behavior as traditionally measured in the laboratory against looking behavior in naturalistic conditions, finding that behavior c

DSCAM is differentially patterned along the optic axon pathway in the developing Xenopus visual system and guides axon termination at the target.

BACKGROUND: The Xenopus retinotectal circuit is organized topographically, where the dorsal-ventral axis of the retina maps respectively on to the ventral-dorsal axis of the tectum; axons from the nasal-temporal axis of the retina project respectively to the caudal-rostral axis of the tectum. Studies throughout the last two decades have shown that mechanisms involving molecular recognition of proper termination domains are at work guiding topographic organization. Such studies have shown that graded distribution of molecular cues is important for topographic mapping. However, the complement of

Causal Role of Neural Signals Transmitted From the Frontal Eye Field to the Superior Colliculus in Saccade Generation.

The frontal eye field (FEF) and superior colliculus (SC) are major and well-studied components of the oculomotor system. The FEF sends strong projections to the SC directly, and neurons in these brain regions transmit a variety of signals related to saccadic eye movements. Electrical microstimulation and pharmacological manipulation targeting the FEF or SC affect saccadic eye movements. These data suggest the causal contribution of each region to saccade generation. To understand how the brain generates behavior, however, it is critical not only to identify the structures and functions of indi

Overt responses during covert orienting.

A distributed network of cortical and subcortical brain areas controls our oculomotor behavior. This network includes the superior colliculus (SC), which coordinates an ancient visual grasp reflex via outputs that ramify widely within the brainstem and spinal cord, accessing saccadic and other premotor and autonomic circuits. In this Review, we discuss recent results correlating subliminal SC activity in the absence of saccades with diverse components of the visual grasp reflex, including neck and limb muscle recruitment, pupil dilation, and microsaccade propensity. Such subtle manifestations

Projections of the superior colliculus to the pulvinar in prosimian galagos (Otolemur garnettii) and VGLUT2 staining of the visual pulvinar.

An understanding of the organization of the pulvinar complex in prosimian primates has been somewhat elusive due to the lack of clear architectonic divisions. In the current study we reveal features of the organization of the pulvinar complex in galagos by examining superior colliculus (SC) projections to this structure and comparing them with staining patterns of the vesicular glutamate transporter, VGLUT2. Cholera toxin subunit β (CTB), Fluoro-ruby (FR), and wheat germ agglutinin conjugated with horseradish peroxidase (WGA-HRP) were placed in topographically different locations within the SC

Morphology of superior colliculus- and middle temporal area-projecting neurons in primate primary visual cortex.

Layers 5 and 6 of primate primary visual cortex (V1) harbor morphologically diverse cell groups that have corticocortical and corticosubcortical projections. Layer 6 middle temporal area (MT)-projecting neurons are particularly interesting, as they are the only deep-layer cortical neurons that provide both corticocortical feedforward inputs (to area MT) and corticosubcortical feedback projections (to superior colliculus [SC]) (Fries et al. [1985] Exp Brain Res 58:613-616). However, due to limitations in anatomical tracing techniques, little is known about the detailed morphologies of these cel

Area 17 lesions deactivate area MT in owl monkeys.

The middle temporal visual area, MT, is one of three major targets of the primary visual cortex, area 17, in primates. We assessed the contribution of area 17 connections to the responsiveness of area MT neurons to visual stimuli by first mapping the representation of the visual hemifield in MT of anesthetized owl monkeys with microelectrodes, ablating an electrophysiologically mapped part of area 17, and then immediately remapping MT. Before the lesions, neurons at recording sites throughout MT responded vigorously to moving slits of light and other visual stimuli. In addition, the relationsh

Movements resembling orientation or avoidance elicited by electrical stimulation of the superior colliculus in rats.

Some studies have reported that stimulation of the superior colliculus in rats produces orienting responses, as it does in a number of species. However, other studies have reported movements resembling avoidance and escape, which are not characteristic of collicular stimulation in other mammals. This apparent discrepancy was investigated by systematically recording the effects on head and body movements of electrical stimulation at a large number of sites throughout the superior colliculus (SC) and surrounding structures. It was found that the nature of the movements observed depended on the l

MR imaging of the excised human brainstem: a correlative neuroanatomic study.

The purpose of this study was to examine an excised human brainstem with high-resolution MR imaging. The MR images were correlated with myel … trinsic brainstem nuclei such as the olivary complex, pontine nuclei, red nucleus, and inferior and superior colliculi were identified. Functional neuroanatomy of the brainstem is reviewed. This neuroanatomic description and review of the complex MR anatomy of the brainstem should assist in the delineation of brainstem structures on MR imaging.

Leading the way: collicular contributions to blindsight.

In this issue, Sepe et al. (2026) show that after only 2 weeks' recovery from a unilateral lesion of primary visual cortex, macaques show evidence that they can make saccades towards targets in the contralesional visual field when target onset is cued. We propose that the superior colliculus is a strong candidate for the generation of these spared visual responses. We go on to suggest that measuring other components of the orienting response, namely activity in the neck and limb muscles during saccades or pointing movements, could provide confirmatory evidence for a collicular contribution to

Transcranial temporal interference stimulation precisely targets deep brain regions to regulate eye movements.

Transcranial temporal interference stimulation (tTIS) is a novel non-invasive neuromodulation technique with the potential to precisely target deep brain structures. This study explores the neural and behavioral effects of tTIS on the superior colliculus (SC), a region involved in eye movement control, in mice. Computational modeling revealed that tTIS delivers more focused stimulation to the SC than traditional transcranial alternating current stimulation. In vivo experiments, including Ca2+ signal recordings and eye movement tracking, showed that tTIS effectively modulates SC neural activity

Functional cell types in the mouse superior colliculus.

The superior colliculus (SC) represents a major visual processing station in the mammalian brain that receives input from many types of retinal ganglion cells (RGCs). How many parallel channels exist in the SC, and what information does each encode? Here, we recorded from mouse superficial SC neurons under a battery of visual stimuli including those used for classification of RGCs. An unsupervised clustering algorithm identified 24 functional types based on their visual responses. They fall into two groups: one that responds similarly to RGCs and another with more diverse and specialized stimu

Phylogenetic origins of early alterations in brain region proportions.

Adult galliform birds (e.g. chickens) exhibit a relatively small telencephalon and a proportionately large optic tectum compared with parrots and songbirds. We previously examined the embryonic origins of these adult species differences and found that the optic tectum is larger in quail than in parakeets and songbirds at early stages of development, prior to tectal neurogenesis onset. The aim of this study was to determine whether a proportionately large presumptive tectum is a primitive condition within birds or a derived feature of quail and other galliform birds. To this end, we examined em

Collicular dysfunction in attention deficit hyperactivity disorder.

Attention deficit hyperactivity disorder (ADHD) is a prevalent neurodevelopmental disorder characterised by (inter alia) an increase in distractibility. The current front-line pharmacotherapies for the treatment of ADHD, namely the psychostimulants methylphenidate and amphetamines, have clear abuse potential, hence there is a strong need to develop new drug treatments for this disorder. Central to this process is the identification of the pathophysiological changes which underlie ADHD. Given the heterogeneity of the disorder, multiple loci are probably involved, providing multiple potential th

Saccade initiation and the reliability of motor signals involved in the generation of saccadic eye movements.

We examined the trial-by-trial relationship between the metrics of saccadic eye movements and the activity of individual putative premotor neurons in the paramedian pontine reticular formation (PPRF) of rhesus monkeys. The region of the pons containing these excitatory burst neurons (EBNs) extends for several millimetres. Motoneurons innervating extraocular muscles integrate the output of hundreds or even thousands of these neurons. Accordingly, there was no reason to expect that relatively small variations in the activity of a single pontine neuron would be related to variations in saccade am

Morphology and connections of neurons in area 17 projecting to the extrastriate areas MT and 19DM and to the superior colliculus in the monkey Callithrix jacchus.

Neurons of area 17, the primary visual cortex, project to various anatomically and physiologically different extrastriate areas and subcortical regions. In the present investigation, we addressed the question of whether the efferent neurons in area 17 can contribute to functional diversity between these regions. We approached this question by analyzing the dendritic morphology of neurons in area 17 projecting to area MT, area 19DM, and the superior colliculus in the new world simian primate Callithrix jacchus, because dendritic morphology is an important factor in determining physiological pro

Histological examination of chick embryos with ocular hypoplasia.

From so-called "dead-in-pipped-eggs" provided by the Poultry Breeding Center of Gifu Prefecture, we obtained two chick embryos with hypoplastic eyes on external examination, and studied the visual system and cranial nerve nuclei innervating the extraocular muscles. The upper bills of both embryos deviated toward one side when observed with the naked eye. In histological sections from the most severely affected eyeball of each case primordial retinal tissue in the poorly developed orbit seemed to be at the stage of optic vesicle formation; it had not differentiated into individual layers. Saccade deficits after a unilateral lesion affecting the superior colliculus.

A 70 year old patient with a small haematoma largely restricted to the area of the right superior and inferior colliculi is reported. Eye movements were electro-oculographically recorded 17 and 80 days after the onset of the haematoma. At the first examination, latency of lateral reflexive visually-guided saccades was asymmetrical, both in the gap task (central fixation point switched off 200 ms before the onset of the lateral target) and in the overlap task (central fixation point remaining switched on). Furthermore, latency of leftward saccades in the overlap task was increased, and accuracy
