SLC52A1 is a neofunctionalized primate urate transporter enabling intestinal urate secretion.

Serum urate levels are high in hominoids because of the evolutionary loss of uricase, an enzyme involved in purine metabolism during evolution. However, the mechanism underlying uricase loss remains unclear. We report the involvement of the neofunctionalized solute carrier family 52 member A1 (SLC52A1) in the evolutionary loss of uricase. Synteny analysis revealed that SLC52A1 was duplicated from SLC52A2, which encodes a riboflavin transporter and is conserved among primates. Functional studies demonstrated the ability of primate SLC52A1 to transport urate as well as riboflavin and the mediati…

Tracking blaCTX-M transmission through transposable elements in uropathogenic and commensal E. coli.

AIM: To investigate the nucleotide sequences associated with transposable elements carrying blaCTX-M allelic variants as potential markers f … sal E. coli and UPEC displayed identical nucleotide sequences surrounding the blaCTX-M gene and the synteny was similar to those found in other parts of the world; however phylogenetic analysis indicated that the genetic environments in Ecuadorian isolates were unique. CONCLUSION: These findings suggest that the nucleotide sequences flanking the blaCTX-M …

Lineage-specific evolution of Methylthioalkylmalate synthases (MAMs) involved in glucosinolates biosynthesis.

Methylthioalkylmalate synthases (MAMs) encoded by MAM genes are central to the diversification of the glucosinolates, which are important secondary metabolites in Brassicaceae species. However, the evolutionary pathway of MAM genes is poorly understood. We analyzed the phylogenetic and synteny relationships of MAM genes from 13 sequenced Brassicaceae species. Based on these analyses, we propose that the syntenic loci of MAM genes, which underwent frequent tandem duplications, divided into two independent lineage-specific evolution routes and were driven by positive selection after the divergen…

The genomes of four tapeworm species reveal adaptations to parasitism.

Tapeworms (Cestoda) cause neglected diseases that can be fatal and are difficult to treat, owing to inefficient drugs. Here we present an analysis of tapeworm genome sequences using the human-infective species Echinococcus multilocularis, E. granulosus, Taenia solium and the laboratory model Hymenolepis microstoma as examples. The 115- to 141-megabase genomes offer insights into the evolution of parasitism. Synteny is maintained with distantly related blood flukes but we find extreme losses of genes and pathways that are ubiquitous in other animals, including 34 homeobox families and several d…

ADAM12-deficient zebrafish exhibit retardation in body growth at the juvenile stage without developmental defects.

ADAM (a disintegrin and metalloprotease) constitutes a family of multi-domain proteins that are involved in development, homeostasis, and disease. ADAM12 plays important roles in myogenesis and adipogenesis in mice; however, the precise physiological mechanisms are not known, and the function of this gene in other vertebrates has not been examined. In this study, we used a simple model vertebrate, the zebrafish, to investigate the functions of ADAM12 during development. Zebrafish adam12 is conserved with those of mammals in the synteny and the amino-acid sequence. We examined adam12 expression…

Molecular Cloning, Characterization, Tissue-Specific Transcription, and Nutritional Regulation of FADS Genes by Dietary Curcumin in Chicken, Gallus gallus.

In this study, five FADS genes-FADS1a, FADS1b, FADS1c, FADS2, and FADS6-were cloned and characterized in chickens. Their nutritional regulations by diets containing various levels (100, 200, 300, and 400 mg/kg) of curcumin ( Curcuma longa ) were also determined. We aimed to determine the functions of FADS genes in fatty acid (FA) metabolism in chickens. We identified two critical conserved regions in chicken FADS: DWXXGH and GEXA, likely conferring structural stability to the iron-binding center and modulating the electrostatic potential. The chicken FADS genes are orthologs of vertebrate FADS

Evolutionary and molecular characterization of fish specific decoy receptor 3 isoforms (TNFRSF6B.1 and 2) in rock bream (Oplegnathus fascatus).

TNFRSF6B, commonly referred to as decoy receptor 3, interacts with TNFSF6, TNFSF14, and TNFSF15, thereby imparting anti-apoptotic and anti-inflammatory properties. This study identifies two isoforms, TNFRSF6B.1 and TNFRSF6B.2, in rock bream (RB) and explores their evolutionary and molecular attributes. Phylogenetic analysis indicates that the teleost isoforms TNFRSF6B.1 and TNFRSF6B.2 have diverged from ancestral forms of TNFRSF6B and TNFRSF11B. Notably, the coelacanth TNFRSF6B is positioned phylogenetically closer to frogs, while TNFRSF6B.2 branches off from TNFRSF11B. Synteny analysis reveal…

Comparative physical genome mapping of malaria vectors Anopheles sinensis and Anopheles gambiae.

BACKGROUND: Anopheles sinensis is a dominant natural vector of Plasmodium vivax in China, Taiwan, Japan, and Korea. Recent genome sequencing … isons of 3955 orthologous genes between An. sinensis and Anopheles gambiae identified 361 conserved synteny blocks and 267 inversions fixed between these two lineages. A review of the ecology, genomics, and stress response of Listeria innocua and Listeria monocytogenes.

Listeria monocytogenes is a Gram-positive foodborne pathogen responsible for a severe disease occurring in immuno-compromised populations. Foodborne illness caused by L. monocytogenes is a serious public health concern because of the high associated mortality. Study of the closely related, but nonpathogenic Listeria innocua has accounted for a better understanding of the behavior of L. monocytogenes in environments beyond the laboratory. Traditionally, the ecological co-habitation, genomic synteny, and physiological similarity of the two species have supported use of L. innocua for predicting …

Subfunctionalization of duplicated zebrafish pax6 genes by cis-regulatory divergence.

Gene duplication is a major driver of evolutionary divergence. In most vertebrates a single PAX6 gene encodes a transcription factor require … s role-sharing between the co-orthologues. Meticulous mapping of isolated BACs identified perturbed synteny relationships around the duplicates. This highlights the functional conservation of pax6 downstream (3') control sequences, which in most vertebrates reside within the introns of a ubiquitously expressed neighbour gene, ELP4, whose pax6a-linked exon …
