Expansion of inflammatory monocytes in periphery and infiltrated into thyroid tissue in Graves' disease.

Monocytes are important mediators of immune system and are reported to be altered in autoimmune disorders. Little is known about the pathological role of monocytes in Graves' disease (GD). Thus, we investigated monocytes in periphery and thyroid tissue in GD. Untreated GD patients were enrolled and followed up until remission. Monocytes were significantly increased and positively correlated with anti-thyrotropin receptor antibody (TRAb) in untreated GD (rcounts = 0.269, P < 0.001; rpercentage = 0.338, P < 0.001). Flow cytometry showed CD14++ CD16+ monocytes were increased and CD14++ CD16- mono

[Tissue expansion in children: Indications and management of complications. A 10-year experience].

INTRODUCTION: Tissue expansion is a plastic surgery technique which enables optimal quality and skin surface reconstruction in case of soft tissue defect. Despite the good results obtained, both from a functional and aesthetic point of view, there is a high rate of complications whose management seems to be essential to ensure satisfactory results. PATIENTS AND METHODS: We retrospectively reviewed the medical files of 45 children treated in our department between 2002 and 2012: 73 expanders were placed during 51 protocols. RESULTS: The studied protocols gathered a large variety of situations…

Pms2 suppresses large expansions of the (GAA·TTC)n sequence in neuronal tissues.

Expanded trinucleotide repeat sequences are the cause of several inherited neurodegenerative diseases. Disease pathogenesis is correlated with several features of somatic instability of these sequences, including further large expansions in postmitotic tissues. The presence of somatic expansions in postmitotic tissues is consistent with DNA repair being a major determinant of somatic instability. Indeed, proteins in the mismatch repair (MMR) pathway are required for instability of the expanded (CAG·CTG)(n) sequence, likely via recognition of intrastrand hairpins by MutSβ. It is not clear if or

Tactile Sensitivity After Ear Reconstruction with Tissue Expansion: A Semmes-Weinstein Monofilament Study.

BACKGROUND: Protective sensation is crucial to avoid injury in the reconstructed auricle. This study aimed to objectively quantify cutaneous sensitivity following autologous ear reconstruction using tissue expansion for unilateral microtia compared to the sensation of healthy auricles. METHODS: In this retrospective study, participants who underwent autologous auricular reconstruction with tissue expansion and were at least 6 months post-operation were included. From January 2025 to June 2025, standardized sensory testing with Semmes-Weinstein monofilaments (SWM) was performed at 12 predefine…

MYCT1-IFITM2/3 interaction links endothelial endolysosomal trafficking to white adipose tissue expansion.

Vertebrates rely on a network of blood vessels to meet organ demands for oxygen and nutrients. While endothelial cells are known to transport excess nutrients to white adipose tissue (WAT) for energy storage, how their metabolic state impacts this process remains unclear. Here, we identify MYCT1 as a conserved, pan-endothelial protein essential for WAT expansion. Endothelial-specific MYCT1 deletion limited WAT expansion independently of angiogenesis, adipogenesis, or systemic metabolic parameters. Mechanistically, MYCT1 interacted with the transmembrane endolysosomal proteins IFITM2/3 to restr

The influence of perivascular tissue on lateral thermal expansion during bipolar vessel sealing.

BACKGROUND: Lateral heat propagation has been an unavoidable effect of bipolar sealing with the risk of damage to surrounding structures. It is presently unknown whether leaving the perivascular tissue in situ may be advantageous in the sense of an isolation effect. MATERIAL AND METHODS: Two groups were formed from ex vivo carotid specimens. Group A (n = 10) consisted of carotid artery with the perivascular connective tissue in place (mean preparation diameter: 10.57 ± 0.16 mm) and group B (n = 10) of skeletonized carotids (mean vessel diameter: 5.21 ± 0.12 mm). All specimens were fixed on a

The Treatment of Complex Extremity Wounds Using External Tissue Expansion: A Case Series.

SUMMARY: The goal of wound reconstruction is the approximation of soft tissue and re-establishment of an acceptable appearance with minimal risk of complications. For large wound closure in the extremities, skin graft and flap reconstruction are common treatments but are associated with a variety of complications. Comparatively, tissue expansion can provide the opportunity to reconstruct large wounds with native, durable, and sensate tissue without significant donor site morbidity. External tissue expansion is less invasive and avoids complications associated with internal expansion. The autho…

A Brain-Melanocortin-Vagus Axis Mediates Adipose Tissue Expansion Independently of Energy Intake.

The melanocortin system is a brain circuit that influences energy balance by regulating energy intake and expenditure. In addition, the brain-melanocortin system controls adipose tissue metabolism to optimize fuel mobilization and storage. Specifically, increased brain-melanocortin signaling or negative energy balance promotes lipid mobilization by increasing sympathetic nervous system input to adipose tissue. In contrast, calorie-independent mechanisms favoring energy storage are less understood. Here, we demonstrate that reduction of brain-melanocortin signaling actively promotes fat mass ga

Confocal super-resolution imaging of the glomerular filtration barrier enabled by tissue expansion.

The glomerular filtration barrier, has historically only been spatially resolved using electron microscopy due to the nanometer-scale dimensions of these structures. Recently, it was shown that the nanoscale distribution of proteins in the slit diaphragm can be resolved by fluorescence based stimulated emission depletion microscopy, in combination with optical clearing. Fluorescence microscopy has advantages over electron microscopy in terms of multiplex imaging of different epitopes, and also the amount of volumetric data that can be extracted from thicker samples. However, stimulated emissio

Tissue expansion in management of failed phallic reconstruction: initial report of clinical series.

OBJECTIVES: To report the first significant experience with the use of tissue expanders in phallic reconstruction. Patients who have undergone multiple failed surgical procedures for repair of hypospadias or epispadias often have minimal residual skin available for additional reconstructive efforts. Although a variety of graft options exist, local skin expansion can provide pigment and texture-matched skin for reconstructive procedures. METHODS: We reviewed our experience with tissue expansion in patients after multiple failed phallic reconstructive efforts. A total of 18 males (mean age 13.2…

Tissue expansion mass spectrometry imaging (TEMI) for high-spatial-resolution multiomics molecular mapping.

High-spatial-resolution in situ mapping of biomolecules within tissue reveals critical insights into the complex molecular landscape and spatial organization of biological systems. Mass spectrometry imaging (MSI) is a powerful tool for spatially resolved molecular analysis of biological samples, with ongoing demand for improved spatial resolution. Tissue expansion combined with MSI (TEMI) is a recently developed approach that enables multiomics molecular mapping across various biological tissues with significantly improved spatial resolution. Unlike conventional methods that depend on instrume…

CD153/CD30 signaling promotes age-dependent tertiary lymphoid tissue expansion and kidney injury.

Tertiary lymphoid tissues (TLTs) facilitate local T and B cell interactions in chronically inflamed organs. However, the cells and molecular pathways that govern TLT formation are poorly defined. Here, we identified TNF superfamily CD153/CD30 signaling between 2 unique age-dependent lymphocyte subpopulations, CD153+PD-1+CD4+ senescence-associated T (SAT) cells and CD30+T-bet+ age-associated B cells (ABCs), as a driver for TLT expansion. SAT cells, which produced ABC-inducing factors IL-21 and IFN-γ, and ABCs progressively accumulated within TLTs in aged kidneys after injury. Notably, in kidney

Copy number expansion of the STX17 duplication in melanoma tissue from Grey horses.

BACKGROUND: Greying with age in horses is an autosomal dominant trait, associated with loss of hair pigmentation, melanoma and vitiligo-like depigmentation. We recently identified a 4.6 kb duplication in STX17 to be associated with the phenotype. The aims of this study were to investigate if the duplication in Grey horses shows copy number variation and to exclude that any other polymorphism is uniquely associated with the Grey mutation. RESULTS: We found little evidence for copy number expansion of the duplicated sequence in blood DNA from Grey horses. In contrast, clear evidence for copy nu

Combining scalp tissue expansion with porous polyethylene total ear reconstruction in burned patients.

Burned ear reconstruction remains one of the most difficult areas of plastic surgery. A superior result is needed to justify the donor site morbidity when reconstructing with a costochondral graft. Recently, more studies have evaluated porous polyethylene ear reconstruction in microtia and in burns. A total loss of the pinna from burns is rare and often associated with significant alopecia. Tissue expansion is an excellent means of reconstructing burned scalp alopecia. Deeply burned skin is often so densely scarred that a costochondral graft would produce a nondescript scarred mass with little…

Severely resorbed mandible: predictable reconstruction with soft tissue matrix expansion (tent pole) grafts.

PURPOSE: Attempts to reconstruct the severely resorbed mandible (<or=6 mm) have historically been unsuccessful and often been complicated by fracture, fistulas, and neurosensory loss. In the past, bone grafts to these severely resorbed mandibles have rapidly resorbed due to a soft tissue volume deficiency that contracts about the graft. MATERIAL AND METHODS: A novel surgical approach using dental implants as "tent poles," along with bone grafting, is described, and the long-term follow-up of 64 cases is presented. RESULTS: The results revealed that all 64 grafts maintained 15 mm of bone heig

Morphological analysis of descending tracts in mouse spinal cord using tissue clearing, tissue expansion and tiling light sheet microscopy techniques.

Descending tracts carry motor signals from the brain to spinal cord. However, few previous studies show the full view of the long tracts fro … synaptic connections between the tracts and motor neurons in the spinal cord through hydrogel-based tissue expansion, and found several monosynaptic connections between the medullary reticular formation ventral part axons and spinal motor neurons. Changing the Donor Site Selection Concept of Facial Skin Expansion from Pure Healthy Tissue to Defect and Healthy Tissue Combination.

UNLABELLED: Facial defect reconstruction is a challenge for plastic surgeons due to unique esthetic and functional properties of the region. Facial tissue expansion provides an ideal reconstruction resource. However, the donor site is limited in the facial region. Thus, a cost-effective expansion management is crucial for an efficient reconstruction. In this article, the evolution of our donor site preference for tissue expansion from pure healthy tissue to a defect-healthy tissue combination is presented. Fifteen patients underwent skin reconstruction with local tissue expansion for facial an…

Systems mapping of intra-donor, cross-tissue T cell clonal expansion and tissue adaptation in the gut-liver-blood axis.

The human gut-liver axis is a critical immunological interface, yet factors that shape T cell adaptation and clonal expansion across tissues remain unclear. We performed integrated single-cell RNA and T cell receptor sequencing on T cells from matched colon epithelium, lamina propria, liver, and blood of the same donors, enabling clonal tracking and tissue-specific transcriptional profiling without inter-individual confounders. Colonic intraepithelial lymphocytes were largely clonally distinct from lamina propria T cells. Tissue-resident T cells in colon vs. liver displayed marked transcriptio
