Surviving without oxygen involves major tissue specific changes in the proteome of crucian carp (Carassius carassius).

The crucian carp (Carassius carassius) can survive complete oxygen depletion (anoxia) for several months at low temperatures, making it an excellent model for studying molecular adaptations to anoxia. Still, little is known about how its global proteome responds to anoxia and reoxygenation. By applying mass spectrometry-based proteome analyses on brain, heart and liver tissue from crucian carp exposed to normoxia, five days anoxia, and reoxygenation, we found major changes in particularly cardiac and hepatic protein levels in response to anoxia and reoxygenation. These included tissue-specific

Postdiagnosis Loss of Skeletal Muscle, but Not Adipose Tissue, Is Associated with Shorter Survival of Patients with Advanced Pancreatic Cancer.

BACKGROUND: Pancreatic cancer is associated with development of cachexia, a wasting syndrome thought to limit survival. Few studies have longitudinally quantified peripheral tissues or identified biomarkers predictive of future tissue wasting. METHODS: Adipose and muscle tissue were measured by computed tomography (CT) at diagnosis and 50 to 120 days later in 164 patients with advanced pancreatic cancer. Tissue changes and survival were evaluated by Cox proportional hazards regression. Baseline levels of circulating markers were examined in relation to future tissue wasting. RESULTS: Compare

FSH supplementation to culture medium is beneficial for activation and survival of preantral follicles enclosed in equine ovarian tissue.

This study investigated the effect of adding different concentrations of bovine recombinant follicle-stimulating hormone on the IVC of equine preantral follicles enclosed in ovarian tissue fragments. Randomized ovarian fragments were fixed immediately (fresh noncultured control) or cultured for 1 or 7 days in α-MEM(+) supplemented with 0, 10, 50, and 100 ng/mL FSH and subsequently analyzed by classical histology. Culture media collected on Day 1 or Day 7 and were analyzed for steroids (estradiol and progesterone) and reactive oxygen species (ROS). After Day 1 and Day 7 of culture, 50-ng/mL FSH

Effects of mineral trioxide aggregate on cell survival, gene expression associated with mineralized tissues, and biomineralization of cementoblasts.

The purpose of this study was to investigate the effects of mineral trioxide aggregate (MTA) on survival, mineralization, and expression of mineralization-related genes of cementoblasts. Immortalized cementoblasts (OCCM) were maintained with Dulbecco modified Eagle medium containing 10% fetal bovine serum. Methyl-thiazol-diphenyl-tetrazolium experiments were performed at 24 and 72 hours to evaluate bioactive components released by MTA (0.002-20 mg/mL) on the cell survival of OCCM. Von Kossa staining was used to evaluate biomineralization of OCCM cells. Images of cementoblasts were taken on day

Access to gastric tissue promotes the survival of axotomized neurons in the dorsal motor nucleus of the vagus in neonatal rats.

Lesioning the vagus nerve in the neck (cervical vagotomy) results in a rapid and virtually complete loss of motoneurons in the dorsal motor nucleus of the vagus in neonatal rats. The present study sought to determine whether access to gastric target tissue will promote the survival of these motoneurons after axotomy. Quantitative analysis demonstrates that subdiaphragmatic vagotomy, which leaves the cut vagal axons in close proximity to their normal gastric targets, results in significantly less motoneuron loss than cervical vagotomy. Furthermore, the loss of motoneurons after cervical vagotom

The incorporation of acetate-1-C14 into cholesterol and fatty acids by surviving tissues of normal and scorbutic guinea pigs.

The synthesis of cholesterol and fatty acids from acetate-l-C(14) by the isolated liver, adrenal, and aorta of scorbutic and pair-fed control guinea pigs has been studied. It was found that ascorbic acid deficiency does not affect the rate of incorporation of C(14)-acetate into cholesterol and fatty acids by the tissues investigated, under our experimental conditions. The relatively high metabolic activity of the artery with regard to cholesterogenesis and lipogenesis was noted. The elevation of serum cholesterol and hexosamine in scurvy has been confirmed.

Association of adipose tissue activity in cervical regions with weight loss and prognostic survival in gastric cancer patients.

BACKGROUND: With the application of positron emission tomography-computed tomography (PET-CT) in recent years, studies have found that brown adipose tissue is not only present in infants and hibernating mammals but also persists in the neck regions of adults. Cancer-associated cachexia, a complex metabolic syndrome, is characterized by adipose tissue loss and heightened metabolic activity. Despite the profound impact of cachexia on patient outcomes, research into the metabolic dynamics of brown adipose tissue has been sparse. The objective of this study is to quantify the metabolic activity of

Tissue Memory Score captures proximal tubular identity loss and independently stratifies survival in clear cell renal cell carcinoma.

PURPOSE: In clear cell renal cell carcinoma (ccRCC), stage dominates prognosis, and grading of tubular dedifferentiation depends on observer judgment. We evaluated the Tissue Memory Score (TMS), a continuous measure of retained proximal tubular identity, for independent prognosis, biological validity, platform robustness, and replication. METHODS: TMS scores a tumor's proximity to the kidney centroid in a locked principal-component space of 2,694 healthy GTEx samples; higher values denote greater kidney identity. The prespecified cohort was TCGA-KIRC (n = 540); a rank-based variant was transf

Lymph node metastasis in high-risk soft tissue sarcoma: Predictors and survival in a population-based SEER study.

BACKGROUND: Soft tissue sarcoma comprises heterogeneous malignant mesenchymal tumours with variable metastatic behaviour. Although spread is usually haematogenous, epithelioid sarcoma, rhabdomyosarcoma, clear cell sarcoma, and angiosarcoma have recognised nodal metastatic potential. Population-level data on nodal metastasis in these high-nodal-risk subtypes remain limited. METHODS: This retrospective population-based cohort study used prospectively collected SEER-17 registry data. Adults aged ≥18 years with microscopically confirmed epithelioid sarcoma, rhabdomyosarcoma, clear cell sarcoma of

Tumor metabolic activity is associated with subcutaneous adipose tissue radiodensity and survival in non-small cell lung cancer.

BACKGROUND: Cachexia-associated body composition alterations and tumor metabolic activity are both associated with survival of cancer patients. Recently, subcutaneous adipose tissue properties have emerged as particularly prognostic body composition features. We hypothesized that tumors with higher metabolic activity instigate cachexia related peripheral metabolic alterations, and investigated whether tumor metabolic activity is associated with body composition and survival in patients with non-small-cell lung cancer (NSCLC), focusing on subcutaneous adipose tissue. METHODS: A retrospective a

Cancer Cell Acid Adaptation Gene Expression Response Is Correlated to Tumor-Specific Tissue Expression Profiles and Patient Survival.

The acidic pH of the tumor microenvironment plays a critical role in driving cancer development toward a more aggressive phenotype, but the underlying mechanisms are unclear. To this end, phenotypic and genotypic changes induced by adaptation of cancer cells to chronic acidosis have been studied. However, the generality of acid adaptation patterns across cell models and their correlation to the molecular phenotypes and aggressiveness of human cancers are essentially unknown. Here, we define an acid adaptation expression response shared across three cancer cell models, dominated by metabolic re

Stereotactic body radiotherapy for pulmonary metastases from soft-tissue sarcomas: excellent local lesion control and improved patient survival.

PURPOSE: Patients with pulmonary metastases (PM) from soft-tissue sarcomas (STS) have historically been treated with surgery and/or chemotherapy. Since 2001, we have treated PM with stereotactic body radiation therapy (SBRT). We postulated that SBRT for PM from STS would yield excellent local control (LC) and overall survival (OS). METHODS AND MATERIALS: Fifty-two patients with PM from STS, diagnosed between 1990 and 2006 at the University of Rochester, were retrospectively reviewed. Most patients received multimodality treatment comprising of surgery, chemotherapy, and/or radiation. SBRT use

Comparison of baseline characteristics and survival between patients with idiopathic and connective tissue disease-related pulmonary arterial hypertension.

BACKGROUND: Both idiopathic pulmonary arterial hypertension (IPAH) and pulmonary arterial hypertension (PAH) related to connective tissue diseases (CPAH) are classified in the group of PAH disorders. However, CPAH has a particularly worse prognosis than IPAH. Few studies have compared the clinical, functional and hemodynamic profiles of IPAH and CPAH. METHODS: We performed a retrospective cohort study of patients with IPAH or CPAH. Demographic characteristics, functional status (FE), pulmonary function test and hemodynamic values at the time of diagnosis were compared between the two etiologi

Five-gene model to predict survival in mantle-cell lymphoma using frozen or formalin-fixed, paraffin-embedded tissue.

PURPOSE: Despite the common underlying translocation t(11;14) involving cyclin D1 that is present in nearly all cases of mantle-cell lymphoma (MCL), the clinical course of the disease is highly variable. The aim of the present study was to develop a quantitative gene expression-based model to predict survival in newly diagnosed patients with MCL that involves a minimum number of genes and is applicable to fresh-frozen and formalin-fixed, paraffin-embedded (FFPE) tumor samples. PATIENTS AND METHODS: The expression of 33 genes with potential prognostic and pathogenetic impact in MCL was analyze
