A crRNA/Cas12a complex-driven rapid and visual detection method for four porcine diarrhea viruses.

BACKGROUND: Porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (TGEV), porcine delta-coronavirus (PDCoV), and porcine rotavirus-A (PoRV) G9 are major swine pathogens primarily responsible for gastrointestinal diseases, particularly affecting lactating piglets and resulting in significant economic losses, especially in China. This study reports a novel CRISPR-based nucleic acid detection method that integrates the high specificity of huLbCas12a with the sensitivity of loop-mediated isothermal amplification (LAMP) technology. Central to this method, the crRNA/Cas12a comp…

Case-control study of pathogens involved in piglet diarrhea.

BACKGROUND: Diarrhea in piglets directly affects commercial swine production. The disease results from the interaction of pathogens with the host immune system and is also affected by management procedures. Several pathogenic agents such as Campylobacter spp., Clostridium perfringens, Escherichia coli, Salmonella spp., group A rotavirus (RV-A), coronaviruses (transmissible gastroenteritis virus; porcine epidemic diarrhea virus), as well as nematode and protozoan parasites, can be associated with disease cases. RESULTS: All bacterial, viral, protozoan, and parasitic agents here investigated, w…

Inactivation of surrogate coronaviruses on hard surfaces by health care germicides.

BACKGROUND: In the 2003 severe acute respiratory syndrome outbreak, finding viral nucleic acids on hospital surfaces suggested surfaces could play a role in spread in health care environments. Surface disinfection may interrupt transmission, but few data exist on the effectiveness of health care germicides against coronaviruses on surfaces. METHODS: The efficacy of health care germicides against 2 surrogate coronaviruses, mouse hepatitis virus (MHV) and transmissible gastroenteritis virus (TGEV), was tested using the quantitative carrier method on stainless steel surfaces. Germicides were o-p…

Recovery and Quantification of Norovirus in Air Samples from Experimentally Produced Aerosols.

Norovirus is the leading cause of acute gastroenteritis in humans across all age groups worldwide. Norovirus-infected patients can produce aerosolized droplets which play a role in gastroenteritis transmission. The study aimed to assess bioaerosol sampling in combination with a virus concentrating procedure to facilitate molecular detection of norovirus genogroup (G) II from experimentally contaminated aerosols. Using a nebulizer within an experimental chamber, aerosols of norovirus GII were generated at known concentrations. Air samples were then collected in both 5 mL and 20 mL water using t

Coronavirus NSP5 protease cleaves CCDC50 to evade antiviral autophagy.

Selective macroautophagy/autophagy is a critical component of innate antiviral defense, relying on selective autophagy receptors to recogniz … conserved cleavage site also targeted by NSP5 orthologs from porcine epidemic diarrhea virus/PEDV, transmissible gastroenteritis virus/TGEV, and SARS-CoV-2. This cleavage disrupts the interaction of CCDC50 with ubiquitin and MAP1LC3/LC3, thereby impairing autophagic degradation of the E protein. Collectively, these findings establish CCDC50 as a selective autophagy receptor with an …

Genetic evolution and epidemiological dynamics of porcine epidemic diarrhea virus in Guangxi, China, from 2020 to 2023.

Porcine epidemic diarrhea virus (PEDV) remains a major threat to the global swine industry, leading to severe economic losses due to high mortality and morbidity in piglets. Although significant advances have been made in understanding its genetic features, comprehensive studies on the genetic evolution and epidemiology of PEDV remain limited. In this study, we analyzed PEDV N gene sequences obtained from Guangxi during the period of 2020 to 2023, alongside co-infection patterns involving other enteric viruses, including Porcine Rotavirus (PoRV) and Transmissible Gastroenteritis Virus (TGEV). …

Engineering a TGEV S-trimer chimera with PEDV D0-NTD generates potent neutralizing antibodies against both viruses.

Porcine epidemic diarrhea virus (PEDV) and transmissible gastroenteritis virus (TGEV), two economically devastating swine enteric coronaviruses, persistently threaten global pork production via severe clinical morbidity. Vaccination is the most effective means to prevent morbidity and mortality caused by porcine enteric coronavirus infection. Hence, the development of an effective and safe vaccination strategy to protect against both PEDV and TGEV infections is urgently needed. Here, we designed and produced trimerized full-length PEDV and TGEV spike (S) proteins using an efficient mammalian e…

Adenovirus types 2, 8, and 37 associated with genital infections in patients attending a sexually transmitted disease clinic.

Adenoviruses (Ads) are an important cause of respiratory illness, conjunctivitis, and gastroenteritis, but they are seldom recognized as a potential cause of sexually transmitted disease. We performed virus cultures on approximately 7,000 patients attending a sexually transmitted disease clinic or other health department clinics for the evaluation of genital ulcers, urethritis, or conjunctivitis. Ads were isolated from genital or conjunctival specimens obtained from 23 (0.33%) patients. Among the 20 Ad-positive men, 15 (75%) had urethritis, 12 (60%) had conjunctivitis, and 10 (50%) had both. FIRST DETECTION OF CHICKEN ANEMIA VIRUS AND NOROVIRUS GENOGROUP II IN STOOL OF CHILDREN WITH ACUTE GASTROENTERITIS IN TAIWAN.

To date, there has been no report of co-infection of chicken anemia virus (CAV) with enteric virus in patients with acute gastroenteritis (AGE). CAV has been recently detected in various types of human samples including stool, indicating pathogenicity in gastrointestinal tract. Examination by PCR-based methods of CAV and norovivus genogroup II (NV GII) in stool of 110 children with AGE at a hospital in Taiwan revealed for the first time of co-infection in two cases. This is the first description of CAV infection in children with AGE in Taiwan. Systematic surveillance and evidence-based studies

Coinfection and nonrandom recombination drive the evolution of swine enteric coronaviruses.

Coinfection with multiple viruses is a common phenomenon in clinical settings and is a crucial driver of viral evolution. Although numerous studies have demonstrated viral recombination arising from coinfections of different strains of a specific species, the role of coinfections of different species or genera during viral evolution is rarely investigated. Here, we analyzed coinfections of and recombination events between four different swine enteric coronaviruses that infect the jejunum and ileum in pigs, including porcine epidemic diarrhea virus (PEDV), transmissible gastroenteritis virus (T…
