Peripheral peptidergic fibers of the trigeminal nerve in the olfactory bulb of the rat.

Axons immunoreactive for calcitonin gene-related peptide (CGRP) and substance P are present in the olfactory nerve, although few, if any, olfactory receptor cells contain immunocytochemically detectable levels of these peptides. The possible trigeminal origin of these fibers was tested by performing unilateral stereotaxic lesions of the ophthalmic division of the trigeminal nerve, followed 2-25 days later by immunocytochemistry for CGRP and substance P. As reported previously, free nerve endings immunoreactive for both peptides were found transversing the nasal epithelium on the unlesioned sid

The Oral-Brain Axis: A Unified Framework Linking Trigeminal Sensorimotor Dysfunction, Chronic Stress, Neuroinflammation, and Neurodegeneration.

Neurodegenerative diseases such as Alzheimer's disease (AD) and Parkinson's disease (PD) develop over decades, yet their earliest pathogenic drivers remain poorly understood. Epidemiological and experimental animal studies suggest that disturbances in oral sensorimotor regulation, particularly within trigeminal proprioceptive pathways, may contribute to neural dysfunction long before clinical symptoms emerge. The mesencephalic trigeminal nucleus (MesV), the only primary sensory neuron population located entirely within the central nervous system (CNS), links oral proprioception with brainstem

Effect of dental malocclusion on cerebellar neuron activation via the dorsomedial part of the principal sensory trigeminal nucleus.

Occlusion has been proposed to play a role for body posture and balance, both of which are mediated mainly by the cerebellum. The dorsomedial part of the principal sensory trigeminal nucleus (Vpdm) has direct projection to the cerebellum. The experimental unilateral anterior crossbite (UAC) has an impact on the motor nuclei in the brain stem via trigeminal mesencephalic nucleus (Vme). The current aim was to explore whether UAC has an impact on Vpdm-cerebellum circuit. The inferior alveolar nerve was injected into cholera toxin B subunit (CTb), the cerebellum was injected into fluoro-gold (FG),

A horseradish peroxidase study of the central projections of primary trigeminal neurons innervating the hard palate in the cat.

The central projections of primary sensory neurons innervating the hard palate in the cat were studied by transganglionic transport of horseradish peroxidase conjugated to wheat germ agglutinin (WGA-HRP). Following injection of WGA-HRP in the incisive papilla terminal labeling was observed in all subdivisions of the sensory trigeminal nuclear complex. In the main sensory nucleus labeling was located in the dorsal part, especially in its most rostral portion. At the levels of nuclei oralis and interpolaris labeling was observed along the medial borders of the nuclei. In addition, at these level

A mechanism of carbamazepine-analgesia as shown by bradykinin-induced trigeminal pain.

Single neuronal responses induced by micro-application of bradykinin solution to the tooth pulp of the rabbit were recorded in the subnucleus reticularis dorsalis and trigeminal subnucleus caudalis and characterized by intermittent episodes of paroxysmal activities. Carbamazepine, the primary agent for trigeminal neuralgia, suppressed such a paroxysmal response, suggesting that an analgesic action of the drug results from the suppression of synaptic transmission at the subnucleus reticularis dorsalis and trigeminal subnucleus caudalis.

Trigeminal neuropathic pain alters responses in CNS circuits to mechanical (brush) and thermal (cold and heat) stimuli.

Functional magnetic resonance imaging was used to study patients with chronic neuropathic pain involving the maxillary region (V2) of the trigeminal nerve in patients with spontaneous pain and evoked pain to brush (allodynia). Patients underwent two functional scans (2-3 months apart) with mechanical and thermal stimuli applied to the affected region of V2 and to the mirror site in the unaffected contralateral V2 region, as well as bilaterally to the mandibular (V3) division. Patients were stimulated with brush, noxious cold, and noxious heat. Significant changes were observed in regions withi

7.0 Tesla MRI tractography in patients with trigeminal neuralgia.

7.0 Tesla (T) high-resolution diffusion tensor imaging (DTI) can supply information on changing microstructures in cranial nerves. We investigated DTI parameters and the feasibility of DTI criteria for diagnosing trigeminal neuralgia (TN). In this study, 14 patients (28 hemispheres) of mean age 49.0 years (range, 31-64) with TN underwent DTI using 7.0 TMRI. We compared fractional anisotropy (FA), axial diffusivity (AD), mean diffusivity (MD), and radial diffusivity (RD) of affected-side and unaffected-side trigeminal nerves using DTI. We examined associations between DTI parameters and clinica

Primary T-cell/histiocyte-rich B-cell lymphoma arising in the trigeminal ganglion in a patient with rheumatoid arthritis.

We report a cased of a 68-year-old man with primary T-cell/histiocyte-rich B-cell lymphoma (T/HRBCL) that arose in the trigeminal ganglion. He had a 30-year history of rheumatoid arthritis and presented with progressive left facial pain that had started 3 weeks earlier. Magnetic resonance imaging (MRI) revealed an enhanced mass in the trigeminal ganglion and swelling of the distal part of the trigeminal root. The tumor, subtotally resected via the left anterior petrosal approach, was composed of proliferating CD30- and CD79-positive atypical large polygonal cells with hyperchromatic single or

Quantitative histochemical and microchemical changes in the adult mouse central nervous system after section of the infraorbital and optic nerves.

The sensory projections from the whiskers of mice and other rodents synapse somatotopically in 3 subnuclei in the brainstem trigeminal complex, in the ventrobasal complex of the thalamus and in the somatosensory cortex. Deafferentation of the whiskers in adult animals results in qualitative and quantitative changes in activities of the metabolic enzymes in the somatosensory cortex (e.g. J. Neuro-sci., 1 (1981) 929-935). We determined the time course and extent of changes in the subcortical trigeminal centers of adult mice after deafferentation. The right infraorbital nerve was sectioned in mic

The role of sensory fiber demography in trigeminal and postherpetic neuralgias.

In this study, we systematically investigated fiber demography, based on function and distribution, from the periphery to their destinations in the various central (sub) nuclei in the trigeminal brainstem nuclear sensory complex. Conventional and novel compelling information is provided, demonstrating that the ratio and somatotopy of types A and C sensory fibers at the site of a lesion can elucidate important puzzles in TNP disorders. For instance, we explain how of a major shift in the fibers' direction and ratio at the level of the trigeminal root entry zone (REZ) influences the pathophysiol

Cortical projections of functionally identified thalamic trigeminovascular neurons: implications for migraine headache and its associated symptoms.

This study identifies massive axonal arbors of trigeminovascular (dura-sensitive) thalamic neurons in multiple cortical areas and proposes a novel framework for conceptualizing migraine headache and its associated symptoms. Individual dura-sensitive neurons identified and characterized electrophysiologically in first-order and higher-order relay thalamic nuclei were juxtacellularly filled with an anterograde tracer that labeled their cell bodies and processes. First-order neurons located in the ventral posteromedial nucleus projected mainly to trigeminal areas of primary (S1) as well as second

Dental malocclusion stimulates neuromuscular circuits associated with temporomandibular disorders.

Unilateral anterior crossbite (UAC) has been demonstrated to cause masseter hyperactivity via the periodontal trigeminal mesencephalic nucleus (Vme)-trigeminal motor nucleus circuit. Here, we studied activation of motor neurons of the facial nucleus (VII), hypoglossal nucleus (XII), nucleus ambiguus (Amb), and spinal nucleus of the accessory nerve (SNA) in rats with UAC via their similar connections with Vme. An anterograde tracer, biotinylated dextran amine (BDA), was injected into the Vme to identify the central axon terminals around the motor neurons of VII, XII, Amb, and SNA. The expressio

The expression of voltage-gated sodium channels in trigeminal nerve following chronic constriction injury in rats.

Objectives: Despite the etiology of trigeminal neuralgia has been verified by microvascular decompression as vascular compression of the trigeminal root, very few researches concerning its underlying pathogenesis has been reported in the literature. The present study focused on those voltage-gated sodium channels, which are the structural basis for generation of ectopic action potentials. Methods: The trigeminal neuralgia modeling was obtained with infraorbital nerve chronic constriction injury (ION-CCI) in rats. Two weeks postoperatively, the infraorbital nerve (TN), the trigeminal ganglion (

Activation of brain stem neurons by irritant chemical stimulation of the throat assessed by c-fos immunohistochemistry.

The method of c-fos immunohistochemistry was used to identify the brain stem distribution of neurons activated following irritant chemical stimulation of the laryngopharyngeal mucosa. In pentobarbital-anesthetized rats, either water (control), nicotine (600 mM, 1 ml) or capsaicin (330 microM, 1 ml) was applied to the pharynx via a cannula placed posterior to the soft palate. Following nicotine and capsaicin, there was a significant increase in fos-like immunoreactivity (FLI) compared with controls in the following areas: nucleus of the solitary tract from the level of the pyramidal decussation

Interactions of Whisking and Touch Signals in the Rat Brainstem.

Perception is an active process, requiring the integration of both proprioceptive and exteroceptive information. In the rat's vibrissal system, a classical model for active sensing, the relative contribution of the two information streams was previously studied at the peripheral, thalamic, and cortical levels. Contributions of brainstem neurons were only indirectly inferred for some trigeminal nuclei according to their thalamic projections. The current work addressed this knowledge gap by performing the first comparative study of the encoding of proprioceptive whisking and exteroceptive touch

A systematic study of brainstem motor nuclei in a mouse model of ALS, the effects of lithium.

Transgenic mice expressing the human superoxide dismutase 1 (SOD-1) mutant at position 93 (G93A) develop a phenotype resembling amyotrophic lateral sclerosis (ALS). In fact, G93A mice develop progressive motor deficits which finally lead to motor palsy and death. This is due to the progressive degeneration of motor neurons in the ventral horn of the spinal cord. Although a similar loss is reported for specific cranial motor nuclei, only a few studies so far investigated degeneration in a few brainstem nuclei. We recently reported that chronic lithium administration delays onset and duration of

Migrainous vertigo: development of a pathogenetic model and structured diagnostic interview.

PURPOSE OF REVIEW: Vestibular symptoms occur frequently in patients with migraine. This review refines recently proposed diagnostic criteria for migraine-related vestibular symptoms, and develops a pathophysiological model for the interface between migraine and the vestibular system. RECENT FINDINGS: The epidemiological link between migraine and vestibular symptoms and signs suggests shared pathogenetic mechanisms. Links between the vestibular nuclei, the trigeminal system, and thalamocortical processing centers provide the basis for the development of a pathophysiological model of migraine-r

Physiological identification of jaw-movement-related neurons in the trigeminal nucleus of cats.

Although neurons responsive to jaw movements have been identified in most parts of the trigeminal brainstem nuclei, little is known about how this information is relayed to the thalamus and ultimately to the cortex for kinesthetic functions and sensorimotor integration. The present extracellular recording experiments showed that a substantial amount of movement-related information is relayed to the thalamus through the caudal part of subnucleus interpolaris (Vi) in adult cats. Vertical jaw displacements, natural mechanical stimuli, and electrical stimulation of the masseter nerve were used to
