USP14 regulates DNA damage repair by targeting RNF168-dependent ubiquitination.

UNLABELLED: Recent reports have made important revelations, uncovering direct regulation of DNA damage response (DDR)-associated proteins and chromatin ubiquitination (Ubn) by macroautophagy/autophagy. Here, we report a previously unexplored connection between autophagy and DDR, via a deubiquitnase (DUB), USP14. Loss of autophagy in prostate cancer cells led to unrepaired DNA double-strand breaks (DSBs) as indicated by persistent ionizing radiation (IR)-induced foci (IRIF) formation for γH2AFX, and decreased protein levels and IRIF formation for RNF168, an E3-ubiquitin ligase essential for chr

The Emerging Role of Non-traditional Ubiquitination in Oncogenic Pathways.

The addition of ubiquitin to a target protein has long been implicated in the process of degradation and is the primary mediator of protein turnover in the cell. Recently, however, many non-proteolytic functions of ubiquitination have emerged as key regulators of cellular homeostasis. In this review, we will describe the various non-traditional functions of ubiquitination, with particular focus on how they can be used as signaling entities in cancer formation and progression. Elaboration of this topic can lead to a better understanding of oncogenic mechanisms, as well as the discovery of novel

PI3K/AKT activation induces PTEN ubiquitination and destabilization accelerating tumourigenesis.

The activity of the phosphatase and tensin homologue (PTEN) is known to be suppressed via post-translational modification. However, the mechanism and physiological significance by which post-translational modifications lead to PTEN suppression remain unclear. Here we demonstrate that PTEN destabilization is induced by EGFR- or oncogenic PI3K mutation-mediated AKT activation in cervical cancer. EGFR/PI3K/AKT-mediated ubiquitination and degradation of PTEN are dependent on the MKRN1 E3 ligase. These processes require the stabilization of MKRN1 via AKT-mediated phosphorylation. In cervical cancer

Beta3 integrin-mediated ubiquitination activates survival signaling during myocardial hypertrophy.

Identifying the molecular mechanisms activated in compensatory hypertrophy and absent during decompensation will provide molecular targets for prevention of heart failure. We have previously shown enhanced ubiquitination (Ub) during the early growth period of pressure overload (PO) hypertrophy near intercalated discs of cardiomyocytes, where integrins are important for mechanotransduction. In this study, we tested the role of integrins upstream of Ub, whether enhanced Ub contributes to survival signaling in early PO, and if loss of this mechanism could lead to decreased ventricular function. Biological meaning of ubiquitination and DNA fragmentation in human spermatozoa.

The ubiquitin-proteasome is an ubiquitous system mainly devoted to protein degradation. The presence of ubiquitinated proteins in male gametes suggests a role for this system also in reproduction. Available evidence indicate that ubiquitin in spermatozoa may have a role in semen quality control, as ubiquitinated defective spermatozoa in the epididymis are subsequently phagocytosed by epididymal epithelial cells. Moreover, a role both in the regulation of mitochondrial inheritance in mammals (paternal mitochondria are eliminated and their ubiquitination appears to be important for this process)

Phosphorylation, acetylation and ubiquitination: the molecular basis of RUNX regulation.

The RUNX family members play pivotal roles in normal development and neoplasia. RUNX1 and RUNX2 are essential for hematopoiesis and osteogenesis, respectively, while RUNX3 is involved in neurogenesis, thymopoiesis and functions as a tumor suppressor. Inappropriate levels of RUNX activity are associated with leukemia, autoimmune disease, cleidocranial dysplasia, craniosynostosis and various solid tumors. Therefore, RUNX activity must be tightly regulated to prevent tumorigenesis and maintain normal cell differentiation. Recent work indicates that RUNX activity is controlled by various extracell

Overexpression of ubiquilin decreases ubiquitination and degradation of presenilin proteins.

Mutations in presenilin proteins (PS1 and PS2) are associated with most cases of early-onset Alzheimer's disease. Several proteins appear to regulate accumulation of PS proteins in cells. One such protein is ubiquilin-1, which increases levels of coexpressed PS2 protein in a dose-dependent manner. We now report that overexpression of ubiquilin-2, which is 80% identical to ubiquilin-1, also increases the levels of coexpressed PS1 and PS2 proteins in cells. To investigate the mechanism by which ubiquilin proteins increase levels of PS proteins, we examined how overexpression of ubiquilin-1, whic

SMURFs regulate mesendoderm differentiation through SMAD2-dependent ubiquitination of CDK9.

Precise control of transcriptional timing is essential during embryonic development, where dynamic gene expression programs govern lineage specification. Promoter-proximal pausing of RNA polymerase II (Pol II) and its regulated release into productive elongation constitute a critical checkpoint in transcriptional regulation. Nodal, a transforming growth factor β (TGF-β) family morphogen, promotes mesendoderm differentiation through SMAD2-dependent transcription, in part by promoting cyclin-dependent kinase 9 (CDK9)-mediated release of paused Pol II. While mechanisms of transcriptional activati

Cell-cycle-driven noncanonical ubiquitination of METTL14 orchestrates RNA methylation.

The precise temporal control of global mRNA methylation during the cell cycle is critical, yet the underlying mechanism remains unknown. Here we elucidate a noncanonical ubiquitination-mediated regulatory mechanism whereby RLIM deposits predominantly K27-linked ubiquitin (K27-Ub) on the m6A methyltransferase METTL14 in a cell-cycle-programmed manner. This modification modulates enzymatic activity by controlling METTL3-METTL14 complex assembly and RNA substrate engagement, independent of protein stability. We discover that transcriptome-wide mRNA m6A level, K27-Ub intensity on METTL14 and RLIM

Proteasome Cap Targeting Chimeras for Ubiquitination-Independent Targeted Protein Degradation.

Selective degradation of a disease-associated protein of interest (POI) is a powerful therapeutic strategy. FDA-approved and investigational glue and degrader drugs function by recruiting a POI to an E3 ubiquitin ligase that mediates POI polyubiquitination and triggers proteasomal degradation. However, using E3 ligases as an intermediary and requiring POI polyubiquitination makes this mechanism of action complex and difficult to rationalize and optimize. These issues have led to interest in evaluating whether direct recruitment of non-ubiquitinated POIs to the proteasome might achieve the same

TRIM32-mediated ubiquitination of STING promotes glycolysis and tumor progression.

The Warburg effect, a hallmark of metabolic reprogramming, drives tumor progression, but its upstream regulation remains unclear. Using hepatocellular carcinoma (HCC) as a model, we identify TRIM32, an E3 ubiquitin ligase, as a potent activator of glycolysis that promotes malignancy. TRIM32 is upregulated in HCC tissues and cell lines, correlating with aggressive features and poor prognosis. Gain- and loss-of-function studies show that TRIM32 enhances proliferation, invasion, and migration in vitro and accelerates tumor growth and lung metastasis in xenografts. Mechanistically, TRIM32 mediates

TRIM33 loss reduces androgen receptor transcriptional output and H2BK120 ubiquitination.

The Androgen Receptor (AR) is a ligand-dependent transcription factor that drives prostate cancer development and progression. Although, a detailed effect on AR biology has been described for a number of interacting proteins, many AR coregulators remain to be characterized in relation to their distinct impact on AR function. Here, we describe TRIM33 as a conserved AR-interactor across multiple prostate cancer cell lines. We observed that TRIM33 and AR share overall chromatin interaction profiles, in which TRIM33 is involved in downstream responsive transcriptomic output. In contrast to prior r

Targeted ubiquitination of Na V 1.8 reduces sensory neuronal excitability.

Chronic pain and addiction are a significant global health challenge. Voltage-gated sodium channel Na V 1.8, a pivotal driver of pain signaling, is a clinically validated target for the development of novel, non-addictive pain therapeutics. Small molecule inhibitors against Na V 1.8 have shown promise in acute pain indications, but large clinical effect sizes have not yet been demonstrated and efficacy in chronic pain indications are lacking. An alternative strategy to target Na V 1.8 channels for analgesia is to reduce the number of channels that are present on nociceptor membranes. We genera

Phosphoribosyl ubiquitination of SNARE proteins regulates autophagy during Legionella infection.

Legionella pneumophila is an intracellular pathogen that causes Legionnaires' disease. The bacteria release effector proteins, some of which remodel host autophagic-lysosomal pathways. One such effector is RavZ, which delipidates ATG8 proteins, making compromising autophagy in Legionella-infected cells. Here we show that SidE effectors also affect these pathways, by mediating phosphoribosyl-ubiquitination (PR-Ub) of the autophagic SNARE proteins STX17 and SNAP29. STX17 modification induces recruitment of STX17-positive membranes from the endoplasmic reticulum to Legionella-containing phagosome

Dysregulated SYVN1 promotes CAV1 protein ubiquitination and accentuates ischemic stroke.

BACKGROUND: Stroke is a major cause of death and severe disability, and there remains a substantial need for the development of therapeutic agents for neuroprotection in acute ischemic stroke (IS) to protect the brain against damage before and during recanalization. Caveolin-1 (CAV1), an integrated protein that is located at the caveolar membrane, has been reported to exert neuroprotective effects during IS. Nevertheless, the mechanism remains largely unknown. Here, we explored the upstream modifiers of CAV1 in IS. METHODS: E3 ubiquitin ligases of CAV1 that are differentially expressed in IS

Diagnostic implications of ubiquitination-related gene signatures in Alzheimer's disease.

The purpose of this study was to explore the diagnostic implications of ubiquitination-related gene signatures in Alzheimer's disease. In this study, we first collected 161 samples from the GEO database (including 87 in the AD group and 74 in the normal group). Subsequently, through differential expression analysis and the iUUCD 2.0 database, we obtained 3450 Differentially Expressed Genes (DEGs) and 806 Ubiquitin-related genes (UbRGs). After taking the intersection, we obtained 128 UbR-DEGs. Secondly, by conducting GO and KEGG enrichment analysis on these 128 UbR-DEGs, we identified the main

Research Progress on the Role of Ubiquitination in Eye Diseases.

The occurrence and development of ophthalmic diseases are related to the dysfunction of eye tissues. Ubiquitin is an important form of protein post-translational modification, which plays an essential role in the occurrence and development of diseases through specific modification of target proteins. Ubiquitination governs a variety of intracellular signal transduction processes, including proteasome degradation, DNA damage repair, and cell cycle progression. Studies have found that ubiquitin can play a role in eye diseases such as cataracts, glaucoma, keratopathy, retinopathy, and eye tumors.

USP13 promotes breast cancer metastasis through FBXL14-induced Twist1 ubiquitination.

PURPOSE: Epithelial-to-mesenchymal transition (EMT) is an important cause of high mortality in breast cancer. Twist1 is one of the EMT transcription factors (EMT-TFs) with a noticeably short half-life, which is regulated by proteasome degradation pathways. Recent studies have found that USP13 stabilizes several specific oncogenic proteins. As yet, however, the relationship between Twist1 and USP13 has not been investigated. METHODS: Co-Immunoprecipitation, GST-pulldown, Western blot, qRT-PCR and immunofluorescence assays were used to investigate the role of USP13 in de-ubiquitination of Twist

Spartin: At the crossroad between ubiquitination and metabolism in cancer.

SPART is a gene coding for a multifunctional protein called spartin, localized in various organelles of human cells. Mutations in the coding region are responsible for a hereditary form of spastic paraplegia called Troyer syndrome while the epigenetic silencing has been demonstrated for some types of tumors. The main functions of this gene are associated to endosomic trafficking and receptor degradation, microtubule interaction, cytokinesis, fatty acids and oxidative metabolism. Spartin has been shown to be a target regulated by STAT3 and localizes also at the level of the mitochondrial outer
