The COVID-19 epidemic was not the first coronavirus epidemic of this century and represents one of the increasing number of zoonoses from wildlife to impact global health. SARS CoV-2, the virus causing the COVID-19 epidemic is distinct from, but closely resembles SARS CoV-1, which was responsible for the severe acute respiratory syndrome (SARS) outbreak in 2002. SARS CoV-1 and 2 share almost 80% of genetic sequences and use the same host cell receptor to initiate viral infection. However, SARS predominantly affected individuals in close contact with infected animals and health care workers. In

Changes in the epidemiology of infectious diseases are the direct result of ecological and evolutionary changes in hosts and parasites. Precisely what the causal processes are is rarely known in any particular case, and this hinders the design of appropriate control strategies. This is particularly so for emerging infections, as opportunity is rapidly lost to study the ecological parameters which might have affected initial emergence. However, molecular evolutionary studies of the pathogens can yield data which discriminate between possible causes. The current distribution of DNA sequence vari

The Zoonoses Anticipation and Preparedness Initiative (ZAPI) was set up to prepare for future outbreaks and to develop and implement new technologies to accelerate development and manufacturing of vaccines and monoclonal antibodies. To be able to achieve surge capacity, an easy deployment and production at multiple sites is needed. This requires a straightforward manufacturing system with a limited number of steps in upstream and downstream processes, a minimum number of in vitro Quality Control assays, and robust and consistent platforms. Three viruses were selected as prototypes: Middle East

The legal and illegal trade in wildlife for food, medicine and other products is a globally significant threat to biodiversity that is also responsible for the emergence of pathogens that threaten human and livestock health and our global economy. Trade in wildlife likely played a role in the origin of COVID-19, and viruses closely related to SARS-CoV-2 have been identified in bats and pangolins, both traded widely. To investigate the possible role of pangolins as a source of potential zoonoses, we collected throat and rectal swabs from 334 Sunda pangolins (Manis javanica) confiscated in Penin

Tickborne rickettsial diseases (TBRD) continue to cause severe illness and death in otherwise healthy adults and children, despite the availability of low cost, effective antimicrobial therapy. The greatest challenge to clinicians is the difficult diagnostic dilemma posed by these infections early in their clinical course, when antibiotic therapy is most effective. Early signs and symptoms of these illnesses are notoriously nonspecific or mimic benign viral illnesses, making diagnosis difficult. In October 2004, CDC's Viral and Rickettsial Zoonoses Branch, in consultation with 11 clinical and

About 70 % of zoonotic infectious diseases originate from wildlife reservoirs, particularly bats, primates, and rodents. Bats comprise about 20 % of all known mammal species worldwide and have been identified as reservoir and carrier hosts of various viral disease outbreaks. They also play a crucial role in viral adaptation and evolution. Given this inherent risk, this review focuses on how anthropogenic activities (habitat destruction, agricultural intensification, bushmeat hunting, and occupational exposure) and climate change are increasing the frequency and intensity of bat-human contact.

Research conducted over seven decades in the Brazilian Amazon has documented the emergence and re-emergence of arboviruses and zoonotic viruses associated with changes in natural ecosystems. Disturbances such as highway construction, deforestation for cattle ranching and soybean cultivation, dam construction for hydroelectric power, intensive mineral extraction, and other human activities can result in outbreaks of viruses maintained in zoonotic or enzootic cycles within forests. These disturbances facilitate the contact between humans and zoonotic hosts or vectors. The potential for a pandemi

Merbecovirus is a subgenus of betacoronaviruses and exhibits high genetic diversity with a capacity for cross-species transmission. However, beyond Middle East respiratory syndrome coronavirus (MERS-CoV), our knowledge of the ecology and pathogenic potential of these viruses remains limited. Merbecoviruses were once thought to rely exclusively on dipeptidyl peptidase 4 for cell entry, but recent discoveries have revealed that several members can also engage with angiotensin-converting enzyme 2 or aminopeptidase N, expanding their receptor repertoire and potential host range. Here we summarize

Henipaviruses, including the highly pathogenic Nipah virus and Hendra virus, represent a major zoonotic threat with high mortality rates and potential for human-to-human transmission. Recent discoveries of novel henipaviruses in China, Europe and other regions highlight the urgent need for enhanced surveillance in both wildlife reservoirs such as bats, shrews, rodents, and human populations, particularly in high-risk areas. Despite advancements in metagenomic sequencing, gaps in integrated surveillance, fragmented One Health implementation, and insufficient diagnostic infrastructure in large p

Zoonotic hepeviruses, particularly hepatitis E virus (HEV, species Paslahepevirus balayani) represent a major yet underestimated public health challenge in Europe. Despite being the leading cause of acute viral hepatitis, surveillance, diagnostic practices and prevention strategies remain heterogeneous across EU/EEA countries, limiting comparability and hindering accurate burden estimates. Underdiagnosis is further compounded by extrahepatic manifestations and the growing impact of chronic HEV infection in immunocompromised patients. At the human-animal-environment interface, zoonotic HEV circ

The increasing zoonotic potential of highly pathogenic avian influenza (HPAI) H5N1 poses a growing threat to global public health. This review examines the molecular and evolutionary mechanisms facilitating H5N1 adaptation in mammalian hosts, focusing on genetic reassortment events, key mutations, and transmission dynamics. Recent mammalian spillover cases, including infections in mink, sea lions, felines, and cattle, suggest a weakening species barrier, with mutations such as PB2-E627K and HA-Q226L enhancing viral replication and host receptor binding affinity. Despite these concerning develo

A systematic risk assessment approach is essential for evaluating the relative risk of influenza A viruses (IAVs) with pandemic potential. To achieve this, the Tool for Influenza Pandemic Risk Assessment (TIPRA) was developed under the Global Influenza Programme of WHO. Since its release in 2016 and update in 2020, TIPRA has been used to assess the pandemic risk of 11 zoonotic IAVs across ten evaluation rounds. Notably, A(H7N9), A(H9N2), and A(H5) clade 2.3.4.4 viruses were re-evaluated owing to changes in epidemiological characteristics or virus properties. A(H7N9) viruses had the highest rel

In the last 4 years, the world has experienced two pandemics of bat-borne viruses. Firstly, in 2019 the SARS-CoV-2 pandemic started and has been causing millions of deaths around the world. In 2022, a Monkeypox pandemic rose in various countries of the world. Those pandemics have witnessed movements and initiatives from healthcare and research institutions to establish a worldwide understanding to battle any future pandemics and biological threats. One Health concept is a modern, comprehensive, unifying ways to improve humans, animals, and ecosystems' health. This concept shows how much they a

The emergence of highly zoonotic viral infections has propelled bat research forward. The viral outbreaks including Hendra virus, Nipah virus, Marburg virus, Ebola virus, Rabies virus, Middle East respiratory syndrome coronavirus, SARS-CoV and the latest SARS-CoV-2 have been epidemiologically linked to various bat species. Bats possess unique immunological characteristics that allow them to serve as a potential viral reservoir. Bats are also known to protect themselves against viruses and maintain their immunity. Therefore, there is a need for in-depth understanding into bat-virus biology to u

Vampire bats became the main reservoir of rabies in Latin America, where the disease remains one of the most important viral zoonoses affecting humans and livestock. In Peru, the most affected livestock are cattle. The official data of 1,729 cases of bovine rabies were evaluated between 2003 and 2017 through a descriptive analysis, decomposition of the time-series and spatio-temporal analyses. Although the cases did not present a defined seasonality, the trend seemed to increase for several years. The bovine rabies cases are more frequent in the inter-Andean valleys than in other regions of th…

Reverse zoonosis or zooanthroponosis is the transfer of pathogens from humans to animals. Although less studied than zoonotic diseases, this phenomenon poses significant risks to both animal and public health. The increasing human-animal interactions driven by urbanization, globalization, and environmental changes have exacerbated the occurrence of reverse zoonosis. This review evaluated the global impact and transmission patterns of reverse zoonosis, highlighting the anthropogenic and intrinsic factors contributing to its emergence. The study performed a systematic review and included 91 scie

Highly pathogenic avian influenza (HPAI) H5N8 virus, first identified in late 2016 in Egypt, continues to circulate and has replaced the previously dominant HPAI H5N1 virus of clade 2.2.1. In this study, HPAI H5N8 was detected on 23 commercial poultry farms in Egypt. Complete genome sequences of three isolates collected in 2021 were obtained using next-generation sequencing (NGS) and subjected to genetic characterization. Phylogenetic analysis showed these isolates to belong to clade 2.3.4.4b, comprising two genotypes: EA-2021-Q and EA-2020-A. Molecular analysis of the haemagglutinin (HA) prot

Pigs are considered the main reservoir of the zoonotic Hepatitis E virus (HEV) world-wide. However, in countries where there is no pork industry or pork consumption, the adaptation of zoonotic HEV (genotype 3) to different species has remained a controversial issue. The objective of our study was to investigate wild rodents as a potential new reservoir or host animal for the transmission of HEV to humans. In this study, HEV RNA was investigated in liver, lung, and spleen samples from 391 rodents of the genus Microtus collected from areas close to rural settlements and in blood samples taken fr

Nanopore-based sequencing has emerged as a revolutionary tool for animal pathogen genomics, offering capabilities unattainable with Sanger and next-generation sequencing (NGS). Despite rapid technical progress, routine veterinary deployment still faces uncertainty in study design, sample preparation, and interpretation thresholds across diverse hosts and sample matrices. Accordingly, this review consolidates recent evidence and provides workflow-oriented guidance for veterinary diagnostics and One Health surveillance.
