The present study was designed to determine the selected monoamine changes that occur during infantile hydrocephalus. Obstructive hydrocepha … and serotonin were increased 70% and 50%, respectively. These increases may indicate impairment of axonal transport or damage to projections from the locus ceruleus and raphe region. These preliminary results suggest that infantile hydrocephalus causes perturbations in the levels of different monoamines in several brain regions. Such changes may critically influe …

The cerebellar afferent projection from the trigeminal sensory nuclei (TSN) was studied by means of retrograde axonal transport of horseradish peroxidase (HRP). The projection is almost exclusively ipsilateral. Three cortical regions, viz., the intermediate-lateral part of lobulus simplex with the adjacent area of lobule V, the rostralmost folia of the paramedian lobule with the surrounding parts of crus I and II, and lobule IX, especially its rostral two folia, are the main targets for the cerebellar afferent fibres. A few fibres reach also the other cerebellar regions, as shown in Fig. 3. Mo…

The spread of neurodegeneration through the human brain network is reported as underlying the progression of neurodegenerative disorders. Ho … d prior to the appearance of amyloid precursor protein accumulation, an indicator of disrupted fast axonal transport. This study demonstrated strong topographical spread of neurodegeneration along recognized neural projections and showed that myelin and glial pathology precedes axonal loss in the process, suggesting that the mechanism of trans-synaptic damage may …

Riboflavin transporter deficiency (RTD) is a neurodegenerative disorder that presents from infancy to adulthood with a progressive axonal neuropathy characterized by a variety of neurologic symptoms including hearing loss, weakness, bulbar palsy, and respiratory insufficiency. Pathogenic variants in SLC52A2 and SLC52A3 are implicated in the pathogenesis of RTD type 2 and 3, respectively. Early identification of this disorder is critical, as it is treatable with riboflavin supplementation. We describe a 16-year-old female with a phenotype consistent with RTD3 found to have a novel heterozygous

Hereditary spastic paraplegias (HSPs) are neurodegenerative motor neuron diseases characterized by progressive age-dependent loss of corticospinal motor tract function. Although the genetic basis is partly understood, only a fraction of cases can receive a genetic diagnosis, and a global view of HSP is lacking. By using whole-exome sequencing in combination with network analysis, we identified 18 previously unknown putative HSP genes and validated nearly all of these genes functionally or genetically. The pathways highlighted by these mutations link HSP to cellular transport, nucleotide metabo

Herpes simplex virus type 1 (HSV) of a highly or slightly neuropathogenetic strain was inoculated into the tongues of mice. After appropriate survival times, the animals were killed and portions of the facial and trigeminal nerves as well as the brain stem were examined with virus isolation and immunocytochemistry for the presence of HSV. Both virus strains were demonstrated in the geniculate and trigeminal ganglia and in the CNS portion of the facial and trigeminal nerves, as well as in various brain stem areas including the trigeminal tract and nucleus, the facial and hypoglossal nuclei, and

Effects of metabolic inhibitors and ouabain on the rate of afferent discharges from isolated muscle spindle of the bullfrog were examined in vitro to elucidate the mechanism for generating afferent action potentials. Dinitrophenol, NaCN, iodoacetic acid, and ouabain gradually reduced the rate of discharges. These drugs were without direct effect on the nerve axon. The rate of discharges was also reduced by Li+, which is known to be minimally extruded from the interior of cells by an active transport mechanism. The rate of discharges remained approximately 50% in the presence of NaCN or in the

Immunofluorescence for gonadotropin releasing hormone (GnRH) in combination with retrograde labeling from the interpeduncular nucleus, as well as from the vasculature confirms that, in the rat, certain GnRH neurons project from the septum-diagonal band to the interpeduncular nucleus. However, about one half of these GnRH neurons also project to fenestrated capillaries as evidenced by uptake and retrograde transport of both peripherally injected Fluoro-Gold and centrally injected rhodamine labeled microspheres. The results indicate that the endocrine effects of GnRH are exerted in part by neuro

This study examines the thalamo-cortical projections to the most superficial neocortical layers in the hedgehog (Erinaceus europaeus) after small injections of horseradish peroxidase and horseradish peroxidase conjugated to wheat germ agglutinin in the somato-sensory cortex. The injections were limited to layers I, II and upper parts of layer III/IV. Retrogradely labeled cells were plotted in serial sections through the thalamus. Injections in the somato-sensory cortex gave a pattern of elongated columns of labeled cells, extending rostro-caudally in the nucleus ventralis thalami. In the neoco

A patient with subdural hematoma and right amaurosis resulting from traumatic injury to the optic nerve developed papilledema in both eyes a few days after the trauma. This was interpreted as supplementary proof of the existence of axonal flow in the first weeks after occurrence of the optic axon lesion. In the amaurotic eye, the papilledema was less pronounced and disappeared more rapidly (after 3 weeks, as opposed to 6 weeks for the other eye). The nerve fiber layer disappeared between 4 and 6 weeks after the trauma.

The thalamo-cortical and cortico-cortical afferents of the cat's insular cortex were investigated with the retrograde horseradish peroxidase technique. The most prominent loci of thalamic labeling were the suprageniculate nucleus and parts of the posterolateral nucleus. Injections into the anterior part of the insular cortex also resulted in labeled cells in the ventromedial posterior nucleus and in the intralaminar nuclei, while injections into posterior parts revealed projections from the medial and dorsal parts of the medial geniculate nucleus. Only the anterior and most ventral parts of th

Cells of origin of the pontine afferents from the inferior colliculus (IC) of cats have been identified by means of retrograde axonal transport of horseradish peroxidase (HRP). Following injections of HRP in the pontine nuclei, many labeled neurons were found ipsilaterally in the caudal parts of the external and pericentral nuclei, while a few cells were found in the central nucleus. Additional neurons occurred in the nuclei of the middle and rostral parts. This organization contrasts with that of other collicular descending systems. Thus, neurons projecting to the superior olivary complex and…

Horseradish peroxidase (HRP) was injected into the vitreous of the eye, the orbital cavity, or the optic tectum of Caiman crocodilus. Following intravitreal injections, retrograde transport of the enzyme was observed bilaterally, but predominantly contralaterally, in a large oblong field of cells at the isthmic level of the midbrain, bounded medially by the trochlear nucleus and laterally by the nucleus isthmi. Control injections of HRP into the orbital cavity and eye muscles labelled motoneurons of the extraocular muscles but not cells of this isthmic field. The field is therefore the source

Introduction: Myosin proteins interact with filamentous actin and translate the chemical energy generated by ATP hydrolysis into a wide variety of mechanical functions in all cell types. The classic function of conventional myosins is mediation of muscle contraction, but myosins also participate in processes as diverse as exocytosis/endocytosis, membrane remodeling, and cytokinesis. Myosin 5a (Myo5a) is an unconventional motor protein well-suited to the processive transport of diverse molecular cargo within cells and interactions with multiprotein membrane complexes that facilitate exocytosis.

Neurons extend long axons that traverse distinct microenvironments, yet how these subcellular compartments acquire and maintain specialized identities remains unclear. Here, we use spatial translatomics to define the local translatomes of somatosensory dorsal root ganglion (DRG) neurons. Translating Ribosome Affinity Purification and RNA sequencing (TRAP-seq) reveal thousands of mRNAs preferentially translated within central axons, peripheral axons, or DRG somata, establishing compartment-specific translational programs. Many of these transcripts encode ion channels and neurotransmitter recept

Several arguments exist in various animal species and man for the presence of a sympathetic component in the pelvic nerve, classically regarded as parasympathetic. We tested this hypothesis in the male rat. Nerve bundles issued from the sacral region of the paravertebral sympathetic chain and reaching the S1 spinal nerve were identified. Neurons in the sacral parasympathetic nucleus of the L6-S1 spinal cord and in the L2-S1 paravertebral sympathetic chain were retrogradely labeled from the pelvic nerve. Radioautography evidenced labeling of unmyelinated fibers in the pelvic nerve following in

Central nervous system neurons which have access to the general circulation were identified by injecting the retrograde tracer Fluoro-Gold peripherally. Fluoro-Gold does not penetrate the blood-brain barrier but is taken up by nerve terminals which project to areas supplied by fenestrated capillaries or to the periphery. Fluoro-Gold-accumulating neurons were present in the following regions or cell groups of the central nervous system: diagonal band of Broca; medial preoptic area; organum vasculosum of the lamina terminalis; subfornical organ; anterior periventricular area; paraventricular nuc
