Pan-cancer antagonistic inhibition pattern of ATM-driven G2/M checkpoint pathway vs other DNA repair pathways.

DNA repair mechanisms keep genome integrity and limit tumor-associated alterations and heterogeneity, but on the other hand they promote tumor survival after radiation and genotoxic chemotherapies. We screened pathway activation levels of 38 DNA repair pathways in nine human cancer types (gliomas, breast, colorectal, lung, thyroid, cervical, kidney, gastric, and pancreatic cancers). We took RNAseq profiles of the experimental 51 normal and 408 tumor samples, and from The Cancer Genome Atlas and Clinical Proteomic Tumor Analysis Consortium databases - of 500/407 normal and 5752/646 tumor sample

From atlas to sacrum: Morphological and morphometric characterization of the feline vertebral canal using computed tomography.

The vertebral canal plays a critical role in protecting the spinal cord and maintaining biomechanical stability. Although the vertebral canal has been extensively studied in dogs, equivalent analyses in cats are limited. This study provides a comprehensive morphological and morphometric assessment of the feline vertebral canal from atlas (C1) to sacrum (Sc) using computed tomography (CT). In 35 cats, predominantly European Shorthairs, cross-sectional area (CSA) and perimeter (P) were determined at cranial, central, and caudal levels of each vertebra. Vertebral canal circularity (CVC) was calcu

Single-cell transcriptome profiling reveals the heterogeneity of ossification of the posterior longitudinal ligament (OPLL) and its immune microenvironment.

BACKGROUND: Ossification of the posterior longitudinal ligament (OPLL) is a heterotopic ossification process of the posterior longitudinal ligament (PLL) that can compress the spinal cord and nerve roots, yet its cellular heterogeneity and pathogenesis remain unclear. METHODS: We performed single-cell RNA sequencing (scRNA-seq) with integrative computational analyses and histological validation on 4,683 cells from surgically resected cervical OPLL lesions obtained from human patients. RESULTS: We identified 15 major cell subsets, including chondrocyte-lineage populations-progenitor/prolifera

MMP1 Overexpression Promotes Cancer Progression and Associates with Poor Outcome in Head and Neck Carcinoma.

Matrix metalloproteinase-1 (MMP1) has been reported to play key roles in a variety of cancers by degrading the extracellular matrix. However, its carcinogenic roles have not been shown yet in head and neck squamous cell carcinoma (HNSCC). This study aimed to elucidate its expression pattern and functional roles as well as clinical significance in HNSCC. The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), and immunohistochemistry (IHC) were utilized to determine the MMP1 expression pattern and the associations between its expression and patients' outcome in HNSCC. Mice tongue squamou

Analysis of the immune landscape in virus-induced cancers using a novel integrative mechanism discovery approach.

BACKGROUND: The mechanisms of carcinogenesis from viral infections are extraordinarily complex and not well understood. Traditional methods of analyzing RNA-sequencing data may not be sufficient for unraveling complicated interactions between viruses and host cells. Using RNA and DNA-sequencing data from The Cancer Genome Atlas (TCGA), we aim to explore whether virus-induced tumors exhibit similar immune-associated (IA) dysregulations using a new algorithm we developed that focuses on the most important biological mechanisms involved in virus-induced cancers. Differential expression, survival

Posterior inferior cerebellar artery with extracranial origin harboring an extracranial aneurysm.

BACKGROUND: We report on a rare case of a saccular aneurysm on the TM segment of the right PICA. The aneurysm was located at the nonbifurcation region of the PICA. CASE DESCRIPTION: The patient underwent a brain CT scan that revealed a diffuse SAH and an IVH in the fourth ventricle with obstructive hydrocephalus. After external ventricular drainage to relieve the hydrocephalus, the aneurysm was demonstrated by cerebral digital subtraction angiography. CONCLUSIONS: The origin of the PICA aneurysm was extracranial and intradural, illustrating a rare location of such type of aneurysm.

Fractured cervical spine and aortic transection.

A 17-year-old victim of a road traffic accident presented. Following investigation diagnoses of fractured first cervical vertebra, aortic transection, diffuse cerebral oedema, fractured right ribs 2-4 and pubic rami were made. Management of this case presented a number of anaesthetic dilemmas: management of the airway, use of cross-clamp vs. shunting or heparinization and bypass, cardiovascular and neurological monitoring, maintenance of cardiovascular stability during and post cross-clamp, minimizing the risk of post-operative renal and neurological dysfunction.

SLC39A6 as a pan-cancer promising biomarker and actionable therapeutic target for CH5132799 sensitivity.

BACKGROUND: The zinc transporter SLC39A6, a member of the ZIP (Zrt-Irt-like protein) family, mediates zinc influx from the extracellular milieu into the cytosol and is indispensable for the function of numerous enzymes, transcription factors and signaling molecules. Previous studies have shown that SLC39A6 expression is associated with prognosis in esophageal squamous cell carcinoma and cervical cancer, indicating its potential impact on patient survival and tumor immunity. But a comprehensive pan-cancer analysis of SLC39A6 is still lacking. This study aimed to systematically delineate the onc

Unilateral posterior atlantoaxial transarticular screw fixation.

Bilateral posterior C 1-2 transfacet screw placement with associated posterior bone graft wiring is the accepted treatment for patients with atlantoaxial instability. This technique was modified to treat 19 patients with atlantoaxial instability and unilateral anomalies that prevented placement of a screw across the C1-2 facet. In these cases, a single contralateral transarticular screw was placed in conjunction with interspinous bone graft wiring to avoid neural or vertebral artery injury and to provide C1-2 stability. Postoperatively, all 19 patients were placed in Philadelphia collars (mean

Unilateral hypoglossal nerve palsy due to neurovascular conflict in a child.

A neurovascular conflict (NC) consists of a pathological contact between a vessel, generally an artery, and the root entry zone of a cranial nerve close to the brainstem. Even if NC of the V, VII and IX cranial nerve have been rarely described, to the best of our knowledge there is no report about the XII cranial nerve NC in the paediatric age. A three-year-old girl presented with right-sided tongue atrophy and fasciculation, of one-year-duration, consistent with a peripheral lesion of the right XII cranial nerve. Brain MRI and MRA documented a marked tortuosity of the vertebrobasilar arteries

Bulk and single cell RNA sequencing data reveal lactylation-related gene signatures for prognosis and immunity in cervical cancer.

Previous studies indicate that lactylation, a post-translational modification, may play an important role in the progression of cervical cancer (CC). However, the comprehensive roles of lactylation in influencing the tumor microenvironment, immune landscape, and prognosis of CC have yet to be fully elucidated. The bulk RNA-seq and single-cell RNA-seq datasets of CC patients were downloaded from the Cancer Genome Atlas and Gene Expression Omnibus databases, respectively. A total of 630 genes associated with lactylation activity were identified using AUCell algorithm, differential expression, an
