Intratympanic injection of emulsion-like dispersions to co-deliver cinnarizine and morin hydrate-lipoid E80 complex in a rabbit inner ear model.

BACKGROUND: Drug delivery to perilymph after crossing the round window membrane is paramount important for inner ear disease management. Intratympanic (IT) injection of emulsion-like dispersions augments cinnarizine (CNZ) and morin hydrate (MH)-Lipoid E80 complex permeation into perilymph in a healthy rabbit inner ear model. METHODS: A Box-Behnken design (BBD) followed by artificial neural network (ANN)-linked Levenberg - Marquardt (LM) algorithm was used for optimizing the injection formula. Immediately after 30-120 minutes post-IT injections, the concentration levels of CNZ and MH in both p

A mouse model of bilateral acute inner-ear (labyrinthine) injury with vestibular involvement reveals functional, behavioral, and histological correlates of vestibular compensation.

Bilateral vestibular dysfunction disrupts balance and spatial orientation, yet mechanisms of injury and compensation remain incompletely defined. We established a mouse model of bilateral acute inner-ear (labyrinthine) injury with vestibular involvement by sequentially incising the round and oval windows, and evaluate auditory brainstem responses (ABR), vestibular sensory-evoked potentials (VsEP), behavior, and histology across postoperative days 1-28. Threshold elevations and behavioral impairment peaked on day 3, with hair-cell degeneration most prominent early and partial morphological reco

S-100 protein in human inner ear: use of a novel immunohistochemical technique on routinely processed, celloidin-embedded human temporal bone sections.

The authors recently developed a new antigen retrieval technique which allows immunohistochemical detection of proteins in routinely processed, celloidin-embedded human temporal bone sections. This technique was used in the present study, which reports the occurrence of S-100 protein in the human inner ear. Fifteen celloidin-embedded human temporal bone sections, obtained from the Eastern Temporal Bone Bank at the Massachusetts Eye and Ear Infirmary, were tested with the monoclonal antibody to S-100. This protein was found in the spiral ligament, in Reissner's membrane, in the spiral limbus, a

Longitudinal outcomes of auditory and spoken language skills following unilateral and bilateral cochlear implantation in children and young adults with inner ear malformations.

INTRODUCTION: Severe-to-profound sensorineural hearing loss significantly impairs spoken language acquisition and cognitive development. Cochlear implants (CIs) bypass damaged inner ear structures to directly stimulate the cochlear nerve and represent an effective rehabilitation strategy. Inner ear malformations (IEMs) are frequently associated with congenital hearing loss; however, the influence of anatomical variability on postoperative functional outcomes remains incompletely understood. OBJECTIVE: To investigate the longitudinal trajectories of auditory skills, spoken language development

The segregation of Calb1, Calb2, and Prph neurons reveals distinct and mixed neuronal populations and projections to hair cells in the inner ear and central nuclei.

BACKGROUND: Knockin mouse models expressing calbindin (Calb1), calretinin (Calb2), and peripherin (Prph) exhibit changes in hair cells (HCs), spiral ganglion neurons (SGN), vestibular ganglion neurons (VGNs), and their central projections. RESULTS: Developing cristae HCs show strong Calb1-positive expression, but adult HCs are mainly Calb2-positive. Utricle and saccule initially have Calb2-positive HCs and later develop Calb1-positive HCs in the striola region. Inner hair cells (IHCs) and outer hair cells (OHCs) in the cochlea express Calb2 early on. Calb1 expression in OHCs overlaps with Cal

Local inner ear application of dexamethasone in cochlear implant models is safe for auditory neurons and increases the neuroprotective effect of chronic electrical stimulation.

Dexamethasone (DEX) can reduce fibrous tissue growth as well as loss of residual hearing which may occur after cochlear implantation. Little is known about the effect of local inner ear DEX treatment on the spiral ganglion neurons (SGN), which are the target of the electrical stimulation with a cochlear implant (CI). Three different clinically relevant strategies of DEX-delivery into the inner ear were used. DEX was either eluted from the electrode carriers' silicone, released from a reservoir by passive diffusion, or actively applied using a pump based system. The effect of the locally applie

HR 3 Tesla MRI for the diagnosis of endolymphatic hydrops and differential diagnosis of inner ear tumors--demonstrated by two cases with similar symptoms.

The synchronous appearance of different inner ear pathologies with a nearly equivalent clinical manifestation such as Menière's disease and vestibular schwannoma is very rare but leads to a relevant dilemma concerning therapy options. MRI is the method of choice to detect intralabyrinthine tumors. Since endolymphatic hydrops is considered the morphological equivalent of Menière's disease, magnetic resonance imaging including hT2w-FLAIR sequences 4 h after i. v. administration of gadolinium-based contrast agents (GBCA) allows the diagnosis and grading of endolymphatic hydrops in vivo synchronou

A pilot study of the safety and performance of the Otologics fully implantable hearing device: transducing sounds via the round window membrane to the inner ear.

OBJECTIVES: The safety and performance of the Otologics fully implantable hearing device were assessed in adult patients with mixed conductive and sensorineural hearing loss. METHODS: The subcutaneous microphone of this fully implantable device picks up ambient sounds, converts them into an electrical signal, amplifies the signal according to the user's needs, and sends it to an electromechanical transducer. The transducer tip is customized with a prosthesis in order to be in contact with the round window membrane and is protected by fascia; this translates the electrical signal into a mechan

Homozygous mutations in fibroblast growth factor 3 are associated with a new form of syndromic deafness characterized by inner ear agenesis, microtia, and microdontia.

We identified nine individuals from three unrelated Turkish families with a unique autosomal recessive syndrome characterized by type I microtia, microdontia, and profound congenital deafness associated with a complete absence of inner ear structures (Michel aplasia). We later demonstrated three different homozygous mutations (p.S156P, p.R104X, and p.V206SfsX117) in the fibroblast growth factor 3 (FGF3) gene in affected members of these families, cosegregating with the autosomal recessive transmission as a completely penetrant phenotype. These findings demonstrate the involvement of FGF3 mutat

A long-term high-fat diet increases oxidative stress, mitochondrial damage and apoptosis in the inner ear of D-galactose-induced aging rats.

In humans, chronic dyslipidemia associated with elevated triglycerides may reduce auditory function. However, there is little evidence available in the literature concerning the effects of a long-term high-fat diet (HFD) on the inner ears of animals. The purpose of this study was to investigate the effect of 12 month-HFD on the inner ear of Sprague-Dawley rats and on the D-galactose (D-gal)-induced aging process in the inner ear. We found that 12 month-HFD markedly elevated the auditory brainstem response (ABR) threshold in the high-frequency region. The HFD significantly increased the generat

An injectable bioadhesive hyaluronic acid hydrogel formulated with drug-loaded microparticle cross-linkers for co-delivery of dexamethasone and lidocaine to the inner ear.

Combination therapies with two or more drugs administrated by intratympanic (IT) injection represent a promising strategy for the treatment of sensorineural hearing loss (SNHL), an inner ear disease imposing a substantial burden on patients. However, the therapeutic outcomes of conventional liquid formulations have been hindered by poor drug retention in the middle ear cavity. In the present study, a versatile hydrogel (CT Gel) was engineered for co-delivery of dexamethasone and lidocaine with drug-loaded microparticles as both cross-linkers and modular drug reservoirs. Specifically, dexametha

Inner-ear delivery of AAV2-retro robustly labels lateral olivocochlear efferents but results in off-target transfection in the contralateral cochlea and the brain.

Gene delivery via adeno-associated viruses (AAVs) is a valuable tool for understanding the organization and function of auditory neuronal circuitry. In this study, we explored the use of AAVs injected into the inner ear to transfect the lateral olivocochlear (LOC) pathway, a poorly understood ipsilateral component of the auditory efferent feedback system. AAV serotypes exhibit a wide range of properties, including cell type selectivity and the capacity for retrograde transport, but none of the AAV serotypes used for inner ear delivery have been reported to successfully transfect the LOC pathwa
