Microfilariae of Wuchereria bancrofti in a lymph node aspirate. A case report.

Microfilariae of Wuchereria bancrofti were detected by fine needle aspiration biopsy (FNAB) in an axillary lymph node from a 27-year-old female. The patient also had intestinal tuberculosis. A routine hemogram did not show any eosinophilia, and three consecutive night blood samples also were negative for microfilariae. This was an early case of filariasis without microfilaremia and was detected by FNAB of the lymph node. The association of filariasis and tuberculosis was purely coincidental.

Natural infections of Dirofilaria immitis in Aedes (Stegomyia) polynesiensis and Aedes (Finlaya) samoanus and their implication in human health in Samoa.

Dirofilaria immitis infections were observed in Aedes polynesiensis and Ae. samoanus in Samoa, together with Wuchereria bancrofti infections, in a study on sub-periodic bancroftian filariasis during 1978-1980. In the 4 indicator villages, the infection rate in Ae. polynesiensis was 0.46% and the infective rate 0.09% (15,223 mosquitoes were dissected). The infection rate in Ae. samoanus was 0.20% and the infective rate 0.08% (10,089 dissected). In 45 selected villages throughout the country, Ae. polynesiensis infection and infective rates were 0.92% and 0.29% (7575 dissected) and the rates for

Seasonality of adult Culex quinquefasciatus and transmission of bancroftian filariasis in Pondicherry, south India.

Seasonal variations in biting Culex quinquefasciatus and transmission of bancroftian filariasis were investigated in Pondicherry, South India. The biting density of C. quinquefasciatus, the principal vector species, was lowest in the summer months and higher during the monsoon and winter months. The survival of the vectors as indicated by the proportion of parous mosquitoes was found to be less in the summer season. Biting mosquitoes with infective stage larvae were not encountered during the hottest months of May, June and July and the early monsoon month of August indicating seasonality of t

The resting and house frequenting behavior of Mansonia annulifera, Ma. uniformis and Ma. indiana, the vectors of Malayan filariasis in Kerala State, India.

Mansonia annulifera, was recorded to be an endophilic species, preferring to rest indoors, while Ma. uniformis was exophilic, having a predilection for outdoor resting habitats, eg bushes and shrubs. Ma. indiana did not show a clear preference to either of these biotopes. In indoor resting collections, the unfed proportion of Ma. uniformis was significantly higher during post-dusk compared to day hours (p < 0.05), indicating that this exophilic species enters houses during dusk hours for feeding. The full fed proportion was higher during day hours compared to dusk/night hours. The semigravid p

Experimental Wuchereria bancrofti infection of Culex quinquefasciatus and Aedes aegypti.

Laboratory-derived females of Culex quinquefasciatus and Aedes aegypti were experimentally infected with Wuchereria bancrofti by allowing the insects to feed on infected volunteers of known microfilariae density between 24.00 and 2.00 hours. Of the 240 mosquitoes used for the experiments only 67 (55.8%) of C. quinquefasciatus and 83 (69.2%) of the Ae.aegypti actually fed on the volunteers' blood. After 16 days post-infection period, 50 (74.62%) of the engorged C. quinquefasciatus harboured infective L3 larval stages of W. bancrofti, while 16 (23.9%) and 1 (1.49%) harboured L2 and L1 pre-infect

Tropical pulmonary eosinophilia.

Tropical pulmonary eosinophilia is one of the many PIE syndromes [pulmonary infiltrates with eosinophilia (of the peripheral blood)]. It is caused by immunologic hyperresponsiveness to the filarial parasites Wuchereria bancrofti or Brugia malayi. Its clinical presentation includes nocturnal cough, dyspnea, wheezing, fever, weight loss, fatigue, interstitial mottling on chest radiograph, predominantly restrictive but also obstructive lung function abnormalities, and peripheral blood eosinophilia of more than 3000 per microliter. It can be distinguished from other PIE syndromes by the patient's

Monitoring of filarial antigens in jirds and patients after treatment.

We developed a sandwich ELISA with monoclonal antibodies to monitor filarial antigens in animals and patients after infection and treatment. Levels of antimicrofilarial antibodies and parasite antigens were measured periodically in 40 B. malayi infected jirds. In all animals L3 HC11 antigen was detected earlier than Mf ES34 antigen, while antimicrofilarial antibodies appeared much more slowly. These serologic changes precede the onset of patent infections. After 3 courses of treatment with DEC and M170, the levels of parasite antigen in sera and of Mf in peritoneal cavities were monitored in 2

Observations on experimental infection of periodic Brugia malayi in man.

Three volunteers were inoculated with different numbers of infective larvae of periodic Brugia malayi from an artificially infected Meriones unguiculatus. At different times after inoculation, the volunteers developed clinical manifestations such as chills, fever, cough, asthma, skin itching, edema, adenolymphangitis and eosinophilia. Microfilaremia was first detected at 41 and 46 weeks after inoculation in two subjects. At 11 weeks, the percentage of E-rosette forming lymphocytes in these subjects was below the normal level. Specific antibody was first detected in three volunteers at 2, 3 and

Circulating parasite antigen in Brugia pahangi-infected jirds.

The Mongolian jird is used widely in filariasis research for studies of protective immunity, pathogenesis, and therapy. The purpose of this study was to evaluate parasite antigen detection as a means of noninvasively monitoring Brugia pahangi infection in jirds. A parasite antigen with Mr of 105-110 kDa was identified in sera from i.p.- and s.c.-infected jirds by immunoblot with a monoclonal antibody to phosphorylcholine. The same antibody was used in a direct sandwich enzyme immunoassay to measure antigen in jird sera. Parasite antigen was detectable as early as 2 wk after i.p. or s.c. inject

A mathematical analysis of various factors involved in transmission of bancroftian filariasis in Pondicherry.

The variation of clumping factor in microfilaraemia among different age groups was observed in Pondicherry for 1981 and 1986. The clumping factor was minimum in the age group 0-5 yr and it was maximum in 16-20 yr group in 1981 and 21-25 yr in 1986. The variation in clumping factor resembled the changes in microfilaria (mf) rate. Multiple linear regression analysis showed that the multiple correlation between mf rate and the other parameters i.e., median microfilarial density (MFD50), clumping factor in human population, and, infection rate and infectivity rate in vector population was highly s

Sex differentials in susceptibility to lymphatic filariasis and implications for maternal child immunity.

This paper reviews epidemiological data to see if there are sex differentials in prevalence, density and clinical pathology due to lymphatic filariasis. Of 53 studies from Africa, South East Asia, the Indian Subcontinent and The Americas, 43 showed a lower mean prevalence of infection in females than in males. Prevalence is consistently lower in women of reproductive age and this is statistically significant in 16 of 32 studies classified by age and sex. Density of infection is also lower in the reproductive age but may be higher in children and in older women. Clinical disease is also lower i

Non-specific immune suppression by CD8+ T cells in Brugia pahangi-infected rats.

Non-specific suppression of the immune response was investigated in Brugia pahangi-infected Lewis rats. The proliferative response of peripheral blood lymphocytes or splenic non-adherent cells to mitogens was significantly reduced by B. pahangi infection. The degree of hyporesponsiveness of splenic non-adherent cells to mitogens was comparable between microfilaremic and non-microfilaremic animals. The suppressed proliferative response of splenic non-adherent cells was restored by blocking with anti-CD8 monoclonal antibody. After separation of T cells into CD4+ and CD8+ subpopulations, only CD8

Giant gourami (Osphronemus goramy: Anabantoidei) as a potential agent for control of weeds, the breeding source for the vectors of Brugia malayi.

The biocontrol efficacy of giant gourami against various aquatic weeds, which form the breeding source of Mansonioides, the vectors of B. malayi, was explored. This fish showed a feeding predilection towards Pistia plant which is the most favoured host of immatures of Mansonioides. A single fish consumed this weed on an average of 206.25 +/- 19.09 g and 316.85 +/- 26.55 g a day under laboratory and field conditions respectively. Techniques developed for the culturing of this fish are also described.

Impact of 50 years of vector control on the prevalence of Brugia malayi in Shertallai area of Kerala state.

This article examines the long term effects of vector control on the prevalence of B. malayi infection and disease, by comparing the results of 3 earlier studies (1934, 1955, 1976) in one area of south India with a recent (1986) survey. The data indicate that disease and infection prevalence have declined continuously over the last 50 years. Infection has declined (from 21 to 2%) more markedly than disease (from 24 to 10%). Age-specific data indicate that this difference is due to the irreversibility of the clinical signs and the long term survival of diseased cohorts. The results indicate tha

Bancroftian filariasis in coastal north Trinidad: the effects of mass chemotherapy using spaced doses of diethylcarbamazine citrate on human microfilaraemias and vector infection rates.

The effects of a mass chemotherapy programme using spaced doses of diethylcarbamazine citrate on Wuchereria bancrofti microfilaraemias and vector infection rates were studied in an isolated rural community of 650 people in north Trinidad where a microfilaria rate of 15% had previously been recorded. A single oral dose of 6 mg/kg body weight was given at monthly intervals for one year. After 6 months, 79% (52/66) of persons previously microfilaraemic had become negative. After 12 months the negative proportion had increased to 90% (63/70). The geometric mean microfilaria density of those still
