Animal Models for Swine Influenza Virus Research: Pathology, Viral Dynamics, and Immune Responses.

Swine influenza virus (SIV) continues to evolve and possesses notable zoonotic potential, making it an important respiratory pathogen of concern for both the global swine industry and public health. Owing to antigenic drift, genetic reassortment, and regional lineage diversity, vaccine efficacy against SIV shows marked variability across different epidemiological contexts. Therefore, establishing appropriate animal models to dissect its pathogenic mechanisms, transmission characteristics, and immune response patterns is of critical importance. This review systematically summarises the animal m

Using ecological niche models to describe the geographical distribution of the myiasis-causing Cochliomyia hominivorax (Diptera: Calliphoridae) in southern South America.

In southern South America, namely Argentina and Chile, Cochliomyia hominivorax (Coquerel) is the main myiasic agent on humans and domestic animals. The distribution pattern of the species is poorly known and the southern limit of its geographic distribution is unclear. The aims of this study are to elucidate the basic environmental factors associated with occurrence of this myiasic species, evaluation of models constructed on the basis of occurrence data based on adult specimen records to predict geographic occurrence of myiasis, evaluation of unsurveyed sites of high potential of occurrence o

Malaria shaped human spatial organization for the past 74 thousand years.

While climate is often seen as the main driver of early humans' spatial organization in Africa, genetic and archaeological studies also suggest diseases as key selective forces in the Pleistocene. We explored whether Plasmodium falciparum-induced malaria drove habitat choice in human societies 74,000 to 5000 years ago. Combining species distribution models of mosquito complexes, palaeoclimatic and epidemiological data, we estimated an index of malaria transmission risk in sub-Saharan Africa through time. We then correlated it with an independent reconstruction of the human niche, demonstrating

Comorbid management of chronic obstructive pulmonary disease and heart failure.

Chronic obstructive pulmonary disease (COPD) and heart failure (HF) frequently coexist, driven by shared risk factors (smoking, inflammation) and bidirectional pathophysiological interactions, worsening prognosis. This review synthesizes evidence from PubMed, Web of Science, EMBASE, and Scopus (2005-2025), using keywords: COPD, heart failure, mechanism, and biomarkers. Included studies encompassed epidemiological, mechanistic, diagnostic, and therapeutic aspects. COPD-HF comorbidity complicates diagnosis due to overlapping symptoms and limitations of spirometry/imaging. Selective β1-blockers a

Pathogen infection as a possible cause for autoimmune hepatitis.

Autoimmune disorders afflicting the liver comprise the bona fide autoimmune diseases, primary biliary cirrhosis, primary sclerosing cholangitis, and autoimmune hepatitis as well as drug-induced autoimmune-like diseases, such as halothane hepatitis. Whereas drug-induced forms of acute or chronic hepatitis often have a clear triggering factor, the etiology of classical autoimmune liver diseases is only poorly understood. Besides a genetic component present in disease susceptible individuals, environmental triggering factors are likely to play a role in the initiation and/or propagation of the di

Immunomodulation by helminth parasites: defining mechanisms and mediators.

Epidemiological and interventional human studies, as well as experiments in animal models, strongly indicate that helminth parasitic infections can confer protection from immune dysregulatory diseases such as allergy, autoimmunity and colitis. Here, we review the immunological pathways that helminths exploit to downregulate immune responses, both against bystander specificities such as allergens and against antigens from the parasites themselves. In particular, we focus on a highly informative laboratory system, the mouse intestinal nematode, Heligmosomoides polygyrus, as a tractable model of

Preventive effects of an enriched environment on rodent psychiatric disorder models.

Interplay between genetic and environmental factors plays a key role in psychiatric disorders, as well as other brain diseases, cancer, and metabolic syndrome. In accordance with epidemiological findings, animal studies have pointed out the importance of a variety of environmental factors, such as viral infection during pregnancy or infancy, early parental loss or separation, and physical or sexual abuse in early life, in the etiology of psychiatric disorders. Conversely, positive effects of environmental factors against the pathogenesis of psychiatric disorders are also demonstrated, in which

Pain and emotional processing in psychological trauma.

Extreme psychological and physical traumas cause dramatic symptom patterns which are insufficiently described by the psychiatric diagnostic criteria of post traumatic stress disorders (PTSD). Additionally, due to the neurobiological proximity and similarity of processing mechanisms of physical and psychological pain stimulation and extremely negative emotions, the patients often suffer from persistent pains even after the somatic healing process is completed. Epidemiological studies confirm the joint occurrence of pain and PTSD. The close relationship and the etiological and behavioral similar

Strategies to identify genes for complex diseases.

Genes underlie numerous human diseases and traits. Although we have witnessed a great deal of success in identifying disease-susceptible genes, the task remains challenging for most of the complex diseases. This paper reviews evidence for the role of genetic factors in complex diseases including breast cancer, diabetes and multiple sclerosis. We then describe strategies that can potentially optimize our chance of success in identifying genes involved in complex diseases. Advances in molecular biology, particularly mapping of the human genome, statistical methods that provide more accurate mode

Cancer families: human models of susceptibility to neoplasia--the Richard and Hinda Rosenthal Foundation Award lecture.

Members of cancer families are at exceptionally high risk and can be studied as human models of cancer susceptibility. These patients represent rare "experiments of nature" that can reveal new insights into carcinogenic processes. We use clinical observations to identify the high risk patient, epidemiological studies to quantitate the excess risk, and laboratory investigations to examine the biological basis of susceptibility. We have uncovered a series of new family cancer syndromes that are under study for molecular mechanisms involved in the pathogenesis of cancers in general. This presenta

Testis Development and Fertility Potential in Boys with Klinefelter Syndrome.

Klinefelter syndrome (KS) is the leading genetic cause of primary hypogonadism and infertility in men. The clinical phenotype has expanded beyond the original description of infertility, small testes, and gynecomastia. Animal models, epidemiologic studies, and clinical research of male subjects with KS throughout the lifespan have allowed the better characterization of the variable phenotype of this condition. This review provides an overview on what is known of the epidemiology, clinical features, and pathophysiology of KS, followed by a more focused discussion of testicular development and t

Cardiometabolic Disease in Rural Southeastern Communities.

BACKGROUND: Cardiometabolic disease disproportionately affects rural communities, but early behavioral and physiological risk indicators are understudied in these populations. OBJECTIVES: The aim of this study was to characterize cardiometabolic health in a rural southeastern U.S. community. Primary objectives included assessing the prevalence of blood pressure and glycemic alterations and examining associations between modifiable factors such as sleep, nutrition, and educational attainment. A secondary objective was to assess community interest in future hypertension research. METHODS: A cr

The global burden of migraine and tension-type headache, 1990-2021: a multi-model Bayesian meta-regression with decomposition of age-period-cohort effects, health inequalities, and frontier analysis.

BACKGROUND: The Global Burden of Disease Study 2021 (GBD 2021) indicates that headache disorders constitute a major global public health burden. Based on its data, this study constructed models of migraine and tension-type headache across 204 countries and 21 regions to systematically analyze their epidemiological characteristics. METHODS: The analysis stratified prevalence by age, sex, and Socio-demographic Index (SDI), and applied decomposition methods to quantify the attributable burden. Age-Period-Cohort models and spatiotemporal models of health inequality were constructed to assess burd

A Microdosimetric Dose Response Model for Monoenergetic Ions and Doses Relevant for Space Radiation Carcinogenesis.

The radiation environment in space consists of a complex mixture of particles and energies that are characteristically different from any natural Earth radiation source. Projections of space radiation cancer risk are obtained by scaling or adjusting epidemiological models derived from terrestrially exposed cohorts to account for differences in radiation quality, dose rate, and other factors. Radiation quality and dose-rate effects introduce significant uncertainty, thereby obfuscating risk communication and hindering the ability to evaluate the efficacy of mitigation strategies such as medical…

Clinical Utility of Protein Language Models in Resolution of Variants of Uncertain Significance in KCNQ1, KCNH2, and SCN5A Compared With Patch-Clamp Functional Characterization.

BACKGROUND: Genetic testing for cardiac channelopathies is the standard of care. However, many rare genetic variants remain classified as variants of uncertain significance (VUS) due to lack of epidemiological and functional data. Whether deep protein language models may aid in VUS resolution remains unknown. Here, we set out to compare how 2 deep protein language models perform at VUS resolution in the 3 most common long-QT syndrome-causative genes compared with the gold-standard patch clamp. METHODS: A total of 72 rare nonsynonymous VUS (9 KCNQ1, 19 KCNH2, and 50 SCN5A) were engineered by s

Scalable gradients enable Hamiltonian Monte Carlo sampling for phylodynamic inference under episodic birth-death-sampling models.

Birth-death models play a key role in phylodynamic analysis for their interpretation in terms of key epidemiological parameters. In particular, models with piecewise-constant rates varying at different epochs in time, to which we refer as episodic birth-death-sampling (EBDS) models, are valuable for their reflection of changing transmission dynamics over time. A challenge, however, that persists with current time-varying model inference procedures is their lack of computational efficiency. This limitation hinders the full utilization of these models in large-scale phylodynamic analyses, especi

The Gut-Brain Axis in Neurodegenerative Diseases and Relevance of the Canine Model: A Review.

Identifying appropriate animal models is critical in developing translatable in vitro and in vivo systems for therapeutic drug development and investigating disease pathophysiology. These animal models should have direct biological and translational relevance to the underlying disease they are supposed to mimic. Aging dogs not only naturally develop a cognitive decline in many aspects including learning and memory deficits, but they also exhibit human-like individual variability in the aging process. Neurodegenerative processes that can be observed in both human and canine brains include the p

Neuropsychiatric consequences of childhood group A streptococcal infection: A systematic review of preclinical models.

In recent years, clinical studies have shown strong epidemiological evidence of an increased risk of developing neuropsychiatric disorders after childhood exposure to streptococcal infection, including the Pediatric Autoimmune Neuropsychiatric Disorders Associated with Streptococcal infection (PANDAS). New preclinical studies on group A streptococcus (GAS) exposure investigate how to disentangle the influences of immune activation to induce long-term neurobehavioral effects associated with neuropsychiatric disorders such as obsessive-compulsive disorder, schizophrenia or autism. The present sy
