Integrative genetic analysis identifies shared regulation of DNA methylation and gene expression in migraine risk.

BACKGROUND: Migraine affects approximately 14% of adults, but the molecular mechanisms underlying susceptibility remain incompletely understood. Although genome-wide association studies (GWAS) have identified many migraine risk loci, translating these associations into biological insight remains challenging. Environmental and physiological migraine triggers, including hormones, diet and stress, may act partly through epigenetic mechanisms such as DNA methylation. Because many migraine risk variants are non-coding and may influence disease through regulatory effects, integrating genetic associa

Gene regulation in response to host sex and infection route in Brugia pahangi with new genome annotation.

Male rodents are almost exclusively used for experimental maintenance of filarial nematodes because they have higher susceptibility to parasitism than female rodents. This study investigates the effects that rodent host sex has on the worm transcriptome for subcutaneous (SQ) infections and intraperitoneal (IP) infections. Significantly more adult worms were recovered from IP-infected gerbils (median = 78 worms/infected animal) relative to SQ-infected gerbils (median = 2 worms/infected animal). Microfilaria production in male gerbils infected via the SQ route (mean = 6.62 microfilariae (mf)/20

Microglia up‑regulate thromboxane A2 synthesis genes in response to C6 glioma‑conditioned medium.

Microglia accumulate in malignant gliomas and play a pivotal role in tumor progression. Using single‑cell RNA sequencing studies researchers have probed gene expression in the myeloid cells in experimental gliomas at relatively late stages of the tumor development. Therefore, the early changes in gene expression in microglia in response to glioma are not fully characterized. We have previously reported distinct profiles of gene expression in the rat primary microglia cultures treated for 6 hours with either rat C6 glioma‑conditioned medium (GCM) or lipopolysaccharide. In the current study, usi

Identification of miR's regulating oncogenes and tumor suppressor genes in acute myeloid leukemia: A bioinformatic approach.

BACKGROUND: Acute myeloid leukemia (AML) is one of the most aggressive hematological malignancies, characterized by mutations in progenitor cells that disrupt normal apoptosis and cellular differentiation, leading to the uncontrolled proliferation of immature myeloid blasts. Mutations in oncogenes and tumor suppressor genes play a crucial role in this process by promoting unchecked cell division and compromising regulatory control over the cell cycle. This study aimed to identify microRNAs (miRNAs) that regulate these oncogenes and tumor suppressor genes in AML. METHOD: We analyzed RNA-seq da

Duplication of a conserved mitochondrial enzyme gene arms parasitoid wasps with venom cytotoxicity and oogenesis regulation.

Gene duplication, followed by neofunctionalization, is a key mechanism driving the emergence of evolutionary novelties. Despite its significance, the molecular and functional processes underlying this phenomenon remain incompletely understood. By tracing the evolutionary history of cysteine-S-conjugate beta-lyase genes within the kynurenine aminotransferase family, we identified a gene duplication event in parasitoid wasps of the Chalcidoidea superfamily. Notably, a single-copy, highly conserved mitochondria-localized physiological gene underwent a significant duplication, resulting in one cop

Microbiome affects mice metabolic homeostasis via differential regulation of gene expression in the brain and gut.

The gut microbiome (GMB) regulates digestion, metabolism, immunity, and energy homeostasis. This study investigates how gut microbiota integrate the regulation in the neuroendocrine and enteroendocrine systems, with a focus on G protein-coupled receptors (GPCRs) in the brain-gut axis and sex differences. Germ-free (GF) mice exhibited increased hypothalamic expression of the anorexigenic neuropeptide and decreased expression of the negative regulator of leptin signaling. GF males had significantly lower serum leptin levels compared to conventional (CON) males, highlighting a potential link betw

[Heterogeneity in the regulation of cellular stress responses by FUS gene mutations associated with amyotrophic lateral sclerosis].

OBJECTIVES: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease characterized by the selective death of motor neurons, exhibiting marked clinical heterogeneity and lacking effective treatment. The etiology and pathogenic mechanisms remain incompletely understood. The FUS (fused in sarcoma) gene is one of the key causative genes in ALS. Pathogenic mutations in the encoded protein are predominantly clustered in the C-terminal nuclear localization signal (NLS) region, and distinct NLS mutation sites show considerable differences in pathogenic potency, clinical phenotype

Pseudomonas aeruginosa extracellular vesicles affect gene regulation and lung inflammation and immunity in cystic fibrosis.

Bacterial extracellular vesicles (EVs) are important mediators of host infection. People with cystic fibrosis (CF) often suffer from chronic infection with Pseudomonas aeruginosa, an opportunistic pathogen. However, the relative abundance of P. aeruginosa is not associated with the onset of increased pulmonary symptoms, known as a pulmonary exacerbation. We hypothesized that the cargo of P. aeruginosa EVs is different at times of baseline wellness and pulmonary exacerbation onset in persons with CF. This is the first study to characterize and compare P. aeruginosa EVs at these two time points,

The Alzheimer's disease risk genes MS4A4A and MS4A6A cooperate to negatively regulate TREM2 and microglia states.

Genetic variations in MS4A4A and MS4A6A Triggering receptor expressed on myeloid cells 2 (TREM2) are linked to the regulation of cerebrospinal-fluid-soluble TREM2 levels and are associated with Alzheimer's disease (AD) risk and progression. By modulating MS4A4A using knockout, overexpression, and degrading antibodies in macrophages, microglia, non-human primates (NHPs), and a mouse model of amyloid pathology, we provide evidence that MS4A4A and MS4A6A are negative regulators of both the transmembrane and soluble TREM2 proteins. Additionally, MS4A4A limits microglia viability, phagocytosis, and

Multi-omics reveals that genes linked to succinylation regulate the onset of epilepsy through metabolic reprogramming.

The relationship between succinylation modification and epilepsy is not yet well defined, and the potential mediation of metabolic imbalance in its regulatory pathways requires deeper investigation. This study combines Mendelian randomization (MR) and single-cell transcriptomic techniques to investigate the causal interplay between succinylation-related genes, plasma metabolites, and epilepsy. Specifically, the eQTLGen and plasma metabolite databases are utilized for two-sample MR analysis, which evaluates genetic instrumental variables and quantifies causal effects. The two-step MR approach i

Cellular and molecular targets in β-Thalassemia: advances in iron Regulation, Erythropoiesis, and Gene-Based therapies.

β-Thalassemia is a prevalent autosomal recessive genetic disorder caused by mutations in the β-globin gene, leading to impaired hemoglobin production and chronic anemia. While traditional treatments such as regular blood transfusions, iron chelation, and supportive care have significantly improved patient survival, these methods remain palliative. The availability of curative options is limited, with allogeneic hematopoietic stem cell transplantation (HSCT) presenting risks due to donor constraints and procedure-related complications. Recent advances in the molecular understanding of β-thalass

NO and melatonin interplay augment Cd-tolerance mechanism in eggplants: ROS detoxification and regulation of gene expression.

Global crop productivity is consistently threatened by cadmium (Cd) soil contamination. Crosstalk between nitric oxide (NO) and melatonin (MT) in enhancing crop resilience to heavy metal stress has gained significant attention. Here, we evaluated the joint effect of sodium nitroprusside (SNP; 200 μM as NO donor) and MT (100 μM) on Cd-stressed eggplant (Solanum melongena L.). Cd stress significantly reduced plant growth, biomass yield, leaf pigments, and biochemical properties. Conversely, SNP, MT, and particularly their combined (SNP + MT) application reduced Cd toxicity and enhanced growth an

Identification of the MAP4K gene family reveals GhMAP4K13 regulates drought and salt stress tolerance in cotton.

Mitogen-activated protein kinase kinase kinase kinases (MAP4Ks) are a class of highly conserved serine/threonine-protein kinases in eukaryotes. They participate in the typical MAPK cascade system and various signal transduction pathways regulating biological processes in plants, during stressful conditions. To date, genome-wide identification of MAP4Ks in cotton has not been reported. In this study, 77 MAP4K genes were identified in four Gossypium species. Protein characteristics, gene structures, conserved motifs and gene expression analysis were carried out. Genome-wide or fragment duplicati
