The variability of the large genomic segment of Ťahyňa orthobunyavirus and an all-atom exploration of its anti-viral drug resistance.

Ťahyňa virus (TAHV), a member of the Bunyaviridae family (California complex), is an important but neglected human mosquito-borne pathogen. The virus genome is composed of three segments, i.e., small (S), medium (M), and large (L). Previous studies on genetic variability of viruses within the California complex were focused on S and M segments, but the L segment remains relatively unstudied. To assess the genetic variation and the relation to virus phenotype we analyzed the L segment sequences of biologically diverse TAHV strains isolated in the Czech Republic and Slovakia. Phylogenetic analys

Autoimmune mechanisms in antibiotic treatment-resistant lyme arthritis.

In about 10% of patients with Lyme arthritis in the United States, joint inflammation persists for months or even several years after the apparent eradication of the spirochete, Borrelia burgdorferi, from the joint with antibiotic treatment. We propose a model of molecular mimicry affecting genetically susceptible individuals to explain this treatment-resistant course. The majority of patients with treatment-resistant Lyme arthritis have HLA-DRB1*0401 or related alleles, and the severity and duration of their arthritis correlate with cellular and humoral immune responses to outer-surface prote

Integrative analysis of mortality risk in SFTS using machine learning and genetic approaches.

This study presents a novel approach to predicting mortality risk in patients with severe fever with thrombocytopenia syndrome (SFTS) by integrating machine learning models and genetic analysis techniques. Utilizing clinical data from 107 confirmed cases, we developed several predictive models and identified the extreme gradient boosting (XGBoost) algorithm as the most effective, achieving an area under the curve (AUC) of 0.950. Further analysis was performed based on the 15 robust features selected and performance-optimized through the Boruta algorithm, integrating Shapley additive explanatio

Newborn Screening for Metachromatic Leukodystrophy in Tuscany: The Paradigm of a Successful Preventive Medicine Program.

Metachromatic leukodystrophy (MLD) is a rare inherited disorder of lysosomal storage, caused by a deficiency in the arylsulfatase A (ARSA) enzyme, leading to toxic accumulation of sulfatides, which progressively impair motor and cognitive function. MLD is a candidate for inclusion in newborn screening (NBS) programs, due to the narrow pre-symptomatic window for effective therapeutic intervention. We set up a prospective pilot NBS program for MLD in Tuscany, based on a two-step approach. The first-tier test quantified four sulfatides; if levels exceeded the cut-off, we performed the second-tier

Development of a rapid detection method for patch thickness based on machine vision and near infrared spectroscopy: a case study of curcumin patch.

Patches represent a significant dosage form within transdermal drug delivery systems. In the quality control of transdermal patches, thickness not only affects the drug loading per unit area and drug release behavior but also is associated with patient usability and product stability. Targeting curcumin patch, a method used for thickness distribution monitoring utilizing machine vision coupled with near infrared (NIR) spectroscopy was proposed. Twenty-nine batches of patches were prepared via the central composite design. The RGB color space data were extracted from the captured images of all

An Explainable Fuzzy Framework for Assessing Preeclampsia Classification.

Background: Preeclampsia remains a leading cause of maternal morbidity worldwide. There is a critical need for predictive systems that not only perform accurately but also provide interpretable insights for clinical decision-making. This work introduces SK-MOEFS, an explainable framework based on fuzzy logic and multi-objective evolutionary optimization, designed to classify preeclampsia risk while generating clinically interpretable rules. Methods: The model integrates fuzzy decision trees with a genetic algorithm to identify a compact and relevant set of rules, optimized for both accuracy an

Is It Time for a New Algorithm for the Pharmacotherapy of Steroid-Induced Diabetes?

Glucocorticoids (GS) are widely used in multiple medical indications due to their anti-inflammatory, immunosuppressive, and antiproliferative effects. Despite their effectiveness in treating respiratory, skin, joint, renal, and neoplastic diseases, they dysregulate glucose metabolism, leading to steroid-induced diabetes (SID) or a significant increase of glycemia in people with previously diagnosed diabetes. The risk of adverse event development depends on the prior therapy, the duration of the treatment, the form of the drug, and individual factors, i.e., BMI, genetics, and age. Unfortunately

Novel Multivariable Evolutionary Algorithm-Based Method for Modal Reconstruction of the Corneal Surface from Sparse and Incomplete Point Clouds.

Three-dimensional reconstruction of the corneal surface provides a powerful tool for managing corneal diseases. This study proposes a novel method for reconstructing the corneal surface from elevation point clouds, using modal schemes capable of reproducing corneal shapes using surface polynomial functions. The multivariable polynomial fitting was performed using a non-dominated sorting multivariable genetic algorithm (NS-MVGA). Standard reconstruction methods using least-squares discrete fitting (LSQ) and sequential quadratic programming (SQP) were compared with the evolutionary algorithm-bas

Estimating gene-level false discovery probability improves eQTL statistical fine-mapping precision.

Statistical fine-mapping prioritizes putative causal variants from a large number of candidate variants, and is widely used in expression quantitative loci (eQTLs) studies. In eQTL fine-mapping, the existence of causal variants for gene expression is not guaranteed, since the genetic heritability of gene expression explained by nearby (cis-) variants is limited. Here we introduce a refined fine-mapping algorithm, named Knockoff-Finemap combination (KFc). KFc estimates the probability that the causal variant(s) exist in the cis-window of a gene through construction of knockoff genotypes (i.e. a

Management of Blau syndrome: review and proposal of a treatment algorithm.

UNLABELLED: Blau syndrome is a rare genetic granulomatosis affecting children. It could be responsible for vision-threatening complications and articular deformation. Due to the rarity of this disease, there are no standardized guidelines for its management. This work aimed to provide an updated overview of the different therapeutic options for Blau syndrome. We conducted research in the PubMed database for the different treatments used in Blau syndrome patients, and we proposed a therapeutic algorithm for disease management. High doses of corticosteroids are considered as a bridging therapy i

Golgi_DF: Golgi proteins classification with deep forest.

INTRODUCTION: Golgi is one of the components of the inner membrane system in eukaryotic cells. Its main function is to send the proteins involved in the synthesis of endoplasmic reticulum to specific parts of cells or secrete them outside cells. It can be seen that Golgi is an important organelle for eukaryotic cells to synthesize proteins. Golgi disorders can cause various neurodegenerative and genetic diseases, and the accurate classification of Golgi proteins is helpful to develop corresponding therapeutic drugs. METHODS: This paper proposed a novel Golgi proteins classification method, wh

A new phenotypic classification system for dyslipidemias based on the standard lipid panel.

BACKGROUND: Dyslipoproteinemias can be classified by their distinct lipoprotein patterns, which helps determine atherosclerotic cardiovascular disease (ASCVD) risk and directs lipid management but this has required advanced laboratory testing. OBJECTIVE: To develop a new algorithm for classifying lipoprotein disorders that only relies on the standard lipid panel. METHODS: Lipid thresholds for defining the different lipoprotein phenotypes were derived for Non-High-Density Lipoprotein-Cholesterol (NonHDL-C) and Triglycerides (TG) to be concordant when possible with the current US Multi-Society

Machine learning-based identification of radiofrequency electromagnetic radiation (RF-EMR) effect on brain morphology: a preliminary study.

The brain of a human and other organisms is affected by the electromagnetic field (EMF) radiations, emanating from the cell phones and mobile towers. Prolonged exposure to EMF radiations may cause neurological changes in the brain, which in turn may bring chemical as well as morphological changes in the brain. Conventionally, the identification of EMF radiation effect on the brain is performed using cellular-level analysis. In the present work, an automatic image processing-based approach is used where geometric features extracted from the segmented brain region has been analyzed for identifyi

A Custom EOG-Based HMI Using Neural Network Modeling to Real-Time for the Trajectory Tracking of a Manipulator Robot.

Although different physiological signals, such as electrooculography (EOG) have been widely used in the control of assistance systems for people with disabilities, customizing the signal classification system remains a challenge. In most interfaces, the user must adapt to the classification parameters, although ideally the systems must adapt to the user parameters. Therefore, in this work the use of a multilayer neural network (MNN) to model the EOG signal as a mathematical function is presented, which is optimized using genetic algorithms, in order to obtain the maximum and minimum amplitude …

Survival models for familial aggregation of cancer.

It has recently been shown that the relative risks of the order of 2 to 4 that are frequently found for cancer among relatives of affected cases are unlikely to be explainable by shared environmental risk factors. Classical methods of epidemiological analysis are not well suited to such analysis because they assume that the outcomes of each individual are independent. Classical methods of genetic analysis, on the other hand, are limited in their handling of environmental factors and variable ages of onset. The recent development of random effects models for survival analysis, however, appears

A Gaussian process model for chemoinformatics with application to the hazard classification of organic solvents.

With the proliferation of screening tools for chemical testing, it is now possible to create vast databases of chemicals easily. However, rigorous statistical methodologies employed to analyze these databases are in their infancy, and further development to facilitate chemical discovery is imperative. In this paper, we address the challenge of predicting an organic solvent's water pollution class based on its chemical structure. We hypothesize that solvents with similar chemical structure are more likely to belong in the same hazard class. This chemical similarity is measured by the Tanimoto d
