Yersinia species display a tropism for lymphoid tissues during infection, and the bacteria select innate immune cells for delivery of cytotoxic effectors by the type III secretion system. Yet, the mechanism for target cell selection remains a mystery. Here we investigate the interaction of Yersinia pestis with murine splenocytes to identify factors that participate in the targeting process. We find that interactions with primary immune cells rely on multiple factors. First, the bacterial adhesin Ail is required for efficient targeting of neutrophils in vivo. However, Ail does not appear to dir

The complex hide-and-seek game between HIV-1 and the host immune system has impaired the development of an efficient vaccine. In addition, the high variability of the virus impedes the long-term control of viral replication by small antiviral drugs. For more than 20 years, phage display technology has been intensively used in the field of HIV-1 to explore the epitope landscape recognized by monoclonal and polyclonal HIV-1-specific antibodies, thereby providing precious data about immunodominant and neutralizing epitopes. In parallel, biopanning experiments with various combinatorial or antibod

In order to increase the plasma half-life and tissue specificity of IL-1 receptor antagonist, a recombinant fusion protein IL-1Ra-HSA, linked by a rigid peptide linker PAPAP, was engineered and expressed by the Pichia pastoris host cells. The fusion protein was secreted to the host cells culture, identified by Western blot, and purified by affinity chromatography. This was followed by a further examination of its bioactivity and pharmacokinetics. Our results demonstrated that the fusion protein retained the antagonist activity of IL-1Ra, capable of binding specifically to the IL-1 receptor on

Paragonimus westermani is a tissue-invading trematode parasite that causes inflammatory lung disease as well as systemic infections including cerebral invasion in carnivorous mammals. While aminopeptidases play important roles in trematodes in the catabolism of host hemoglobin, an essential source of nutrient for the parasite, little is known about aminopeptidase in Paragonimus. Presently, we isolated a cDNA encoding a 58 kDa P. westermani leucine aminopeptidase (PwLAP). Deduced amino acid sequence of PwLAP exhibited significant sequence homology with LAP from Schistosoma spp. and Fasciola hep

The host range of retroviral vectors including lentiviral vectors can be expanded or altered by a process known as pseudotyping. Pseudotyped lentiviral vectors consist of vector particles bearing glycoproteins (GPs) derived from other enveloped viruses. Such particles possess the tropism of the virus from which the GP was derived. For example, to exploit the natural neural tropism of rabies virus, vectors designed to target the central nervous system have been pseudotyped using rabies virus-derived GPs. Among the first and still most widely used GPs for pseudotyping lentiviral vectors is the v

Naegleria fowleri is the etiologic agent of primary amebic meningoencephalitis, a rare but rapidly fatal disease of humans. It invades the central nervous system via nasal mucosa and cribriform plate. Once in brain tissue, the organism induces an acute hemorrhagic, necrotizing meningoencephalitis. We hypothesize that a protease released by the parasite contributes to tissue destruction and facilitates host invasion. Analysis of conditioned media of N. fowleri cultures revealed a major 30-kDa protease with substrate and inhibitor specificity consistent with cysteine proteases. Amino-terminal am

Cospeciation has been observed multiple times between parasites and their hosts. Here we compare the phylogeny of 2 different groups of chewing lice (Phthiraptera), one known for being host specific (Amblycera: Myrsidea) and one known for including many generalist species (Ischnocera: Brueelia-complex, specifically Guimaraesiella and Priceiella) with that of their songbird hosts (Passeriformes), which are participants in mixed-species feeding flocks in South China. Using event- (Jane) and distance-based (ParaFit) analyses we found that both groups of lice have phylogenies that are more similar

Like all pathogenic infectious agents, Plasmodium falciparum, the deadly malaria parasite, modulates its host environment as a part of its evolutionary adaptation to thrive, proliferate, and ultimately sustain its transmission. During their asexual intraerythrocytic developmental cycle, the parasites remodel the cytoplasm and surface of the host red blood cell (RBC) and alter its deformability. There is, however, also growing evidence that Plasmodium engages, modifies, and imports specific RBC proteins to facilitate specific biological functions essential for its growth and development within

Legionellae are opportunistic facultative intracellular pathogens with several antigens and toxic components which may function synergistically in contribution to pathogenesis and immunity. The serogroup specific antigens are associated with endotoxic activities of the bacteria and are potent adjuvants in inducing hypersensitivity and potentiating antibody responses to various antigens. Two protein toxins interfere with oxidative metabolism of host cells and are potential inhibitors of intracellular bactericidal activities. Tissue damage in the host appears to be caused by lysis of infected ce

The complete development of Cryptosporidium in host cell-free medium first described in 2004, represented a significant advance that can facilitate many aspects of Cryptosporidium research. A current limitation of host cell-free cultivation is the difficulty involved in visualising the life-cycle stages as they are very small in size, morphologically difficult to identify and dispersed throughout the media. This is in contrast to conventional cell culture methods for Cryptosporidium, where it is possible to focus on the host cells and view the foci of infection on the host cells. In the presen

Diabetic foot infections (DFIs) represent a major and increasing complication of diabetes mellitus, often leading to hospitalization, osteomyelitis, and lower limb amputation. The rising prevalence of multidrug-resistant pathogens in DFIs has limited the effectiveness of conventional antibiotic therapy, emphasizing the need for alternative or adjunctive approaches. Bacteriophage therapy has emerged as a promising strategy due to its specificity to target bacteria, ability to penetrate and disrupt biofilms, and activity against multidrug-resistant organisms, while generally demonstrating a favo

Helminth parasites suppress immune responses to prolong their survival within the mammalian host. Thereby not only helminth-specific but also nonhelminth-specific bystander immune responses are suppressed. Here, we use the murine model of Litomosoides sigmodontis infection to elucidate the underlying mechanisms leading to this bystander T-cell suppression. When OT-II T cells specific for the third-party antigen ovalbumin are transferred into helminth-infected mice, these cells respond to antigen-specific stimulation with reduced proliferation compared to activation within non-infected mice. Polyomaviruses are a growing family of small DNA viruses with a narrow tropism for both the host species and the cell type in which they productively replicate. Species host range may be constrained by requirements for precise molecular interactions between the viral T antigen, host replication proteins, including DNA polymerase, and the viral origin of replication, which are required for viral DNA replication. Cell type specificity involves, at least in part, transcription factors that are necessary for viral gene expression and restricted in their tissue distribution. In the case of the huma

This study focuses on the parasitic associations of mites and ticks infesting reptiles and amphibians through a multifocal approach. Herein, reptiles (n= 3,596) and amphibians (n= 919) were examined to ensure representativeness of the Brazilian herpetofauna megadiversity. The overall prevalence was calculated to better understand which were the preferred hosts for each order of Acari (Trombidiformes, Mesostigmata and Ixodida), as well as to determine which orders frequently parasitize reptiles and amphibians in Brazil, and their host specificity. Infestation rates were calculated [prevalence, …

Helicobacter pylori is a Gram-negative bacterium which causes ulcer, atrophic gastritis, adenocarcinoma, or mucosa-associated lymphoid tissue lymphoma. A comparative proteomic and immunoproteomic analysis was chosen to identify the antigenic patterns of three different H. pylori strains. These strains were probed against single sera from H. pylori-positive patients affected by gastric adenocarcinoma or duodenal ulcer. We found a quite heterogeneous antigenic pattern, both from strain and sera points of view, thus underlying both a strain- and a host-specificity. The different antigenic reperto
