The ameliorative effect of depolymerized sulfated polysaccharides from Eucheuma serra (DESP) on ovalbumin (OVA)-caused induced food allergy was investigated in this work. Results showed that OVA stimulated the secretion of allergy-related cytokines (OVA-specific IgE, mMCP-1, IgA, TNF-α) and led to diarrhea, intestinal epithelial damage, and intestinal microflora dysbiosis in sensitized mice. After the administration of DESP, however, the anaphylactic symptoms (shortness of breath, hypothermia, diarrhea), along with the allergy-related cytokines, were effectively suppressed. Moreover, the reduc

Intestinal L cells regulate a wide range of metabolic processes, and L-cell dysfunction has been implicated in the pathogenesis of obesity and diabetes. However, it is incompletely understood how luminal signals are integrated to control the development of L cells. Here we show that food availability and gut microbiota-produced short-chain fatty acids control the posttranslational modification on intracellular proteins by O-linked β-N-acetylglucosamine (O-GlcNAc) in intestinal epithelial cells. Via FOXO1 O-GlcNAcylation, O-GlcNAc transferase (OGT) suppresses expression of the lineage-specifyin

Experimental IgE-mediated food allergy depends on intestinal anaphylaxis driven by interleukin-9 (IL-9). However, the primary cellular source of IL-9 and the mechanisms underlying the susceptibility to food-induced intestinal anaphylaxis remain unclear. Herein, we have reported the identification of multifunctional IL-9-producing mucosal mast cells (MMC9s) that can secrete prodigious amounts of IL-9 and IL-13 in response to IL-33, and mast cell protease-1 (MCPt-1) in response to antigen and IgE complex crosslinking, respectively. Repeated intragastric antigen challenge induced MMC9 development

In intestinal helminth infections, Th2 immune respones are generally associated with mucin secretion for worm expulsion from the host intestine. In particular, IL-4 and IL-13 are the important cytokines related with intestinal mucus production via STAT6 signalling in nematode infections. However, this perspective has never been studied in Gymnophalloides seoi infection. The present study aimed to observe the STAT6 signalling and cytokine responses in C57BL/6 mice, a mouse strain resistant to infection with this trematode. The results showed that worm expulsion occurred actively during days 1-2

Dietary fiber (DF) consists of non-starch polysaccharides (NSPs) and lignin, and is categorized into soluble and insoluble DF. DF is often regarded as an antinutritional element; however, the intake of both soluble and insoluble fiber can induce physiological alterations in the gastrointestinal tract (GIT). These adaptations include increased synthesis and secretion of digestive enzymes, altered gastrointestinal motility and development, improved gizzard functionality, and the proliferation of intestinal and caecal microbiota. The induced adaptations interfere with the digestibility of energy

Diabetes is now a global chronic disease, with the number of people with diabetes expected to reach 643 million by the end of 2030. Semaglutide, a human glucagon-like peptide-1 (GLP-1) analogue with 94% similarity to human GLP-1, can promote insulin secretion and repress glucagon secretion in a glucose concentration-dependent manner, resulting in substantial improvement of blood glucose levels and reducing the risk of hypoglycemia in patients with type 2 diabetes. To improve the absorption efficiency of semaglutide in oral delivery, we developed chitosan hydrochloride-coated and nonionic surfa

Lutzomyia longipalpis Lutz & Neiva, 1912 (Diptera, Psychodidae), is the primary vector of Leishmania infantum Nicole, 1908, the etiological agent of American visceral leishmaniasis. During their development, sandfly larvae pass through four instars, consuming soil particles enriched with microorganisms and decomposing organic material. In numerous insect species, the intestinal epithelium not only secretes digestive enzymes and absorbs digested nutrients but also carries out additional functions, such as regulating luminal pH and facilitating the absorption or secretion of ions and water. Vancomycin-resistant Enterococcus (VRE) poses a serious threat in hospitals where they densely colonize the intestinal tracts of patients. In vulnerable hosts, these pathogens may translocate to the bloodstream and become lethal. The ability to selectively reduce VRE in the intestinal tracts of patients could potentially prevent many of these translocation events and reduce the spread of the pathogen. Herein, we have engineered Escherichia. coli Nissle 1917 to produce and secrete three antimicrobial peptides, Enterocin A, Enterocin B, and Hiracin JM79, to specifically target and kill Enterococ

Salmonella enterica serovar Enteritidis is a common cause of foodborne illness in the United States. The bacterium can be transmitted to humans via contaminated chicken meat and eggs, and virulence in humans requires type III secretion system 1 (TTSS-1), encoded on Salmonella pathogenicity island 1 (SPI-1). Chickens often carry S Enteritidis subclinically, obscuring the role of SPI-1 in facilitating bacterial colonization. To evaluate the role of SPI-1 in the infection of chicks by Salmonella, we created and utilized strains harboring a stable fluorescent reporter fusion designed to quantify S

INTRODUCTION: SGLT1 is the primary transporter responsible for the absorption of glucose and galactose in the intestine, while SGLT2 and SGLT1 are both involved in the renal reabsorption of glucose. SGLT2 inhibitors are a new class of oral antidiabetic drugs, acting by increasing urinary glucose excretion (UGE). They offer the advantages of a reduced risk of hypoglycaemia, a decrease in body weight and blood pressure and an efficacy at all stages of type 2 diabetes (T2DM).
AREAS COVERED: Herein, the authors focus specifically on sotagliflozin (LX4211), the first-in-class dual SGLT1/SGLT2 inhi

Despite the introduction 20-30 years ago of potent inhibitors of gastric acid secretion and anti-inflammatory drugs that preferentially inhibit cyclo-oxygenase (COX)-2, the GI adverse effects of nonsteroidal anti-inflammatory drugs (NSAIDs) remain a significant clinical concern and a considerable economic burden. Inhibitors of acid secretion and selective COX-2 inhibitors reduce damage only in the proximal GI tract (stomach and proximal duodenum), but NSAIDs produce injury and bleeding throughout the GI tract. The small intestinal damage caused by NSAIDs is common, difficult to diagnose, and t

The influence of vitamin A on the functions of the small intestine was examined in rats made vitamin A deficient for 40 days by feeding a special diet after weaning and in pair fed vitamin A deficient rats that were given supplementary vitamin A (240 IU/day) in their drinking water. The basal and stimulated electrogenic secretory and absorptive functions of the jejunum and proximal and distal ileum removed from these rats were examined in vitro using the short circuit current as the index of transport activity. The basal short circuit current in the jejunum and proximal ileum was not significa

We studied two aspects of the human gastric mucosa: the surface morphology of mucous cells, as viewed by scanning electron microscopy (SEM); the glycosidic components of intracellular mucins, characterized by means of lectins. The latter were conjugated with fluorescein isothiocyanate and with colloidal gold-silver for the visualization of the reaction products in light microscopy (LM) and in SEM (backscattered mode) respectively. The surface morphology of mucous cells appears to be correlated to the secretory state. In gastric ulcers we found a prevalence of non-secreting cells. A decrease in

The submandibular salivary gland of the male mouse and its secretions contain growth-promoting factors that accelerate cell proliferation in vitro and also have effects on the gastrointestinal tract in vivo. We therefore stimulated salivary secretion with isoproterenol for studies of the effects of submandibular saliva on intestinal cell proliferation; only submandibular glands respond to isoproterenol by releasing epidermal growth factors and other growth factors into the saliva. Submandibular sialadenectomized and sham sialadenectomized male mice on pair-feeding schedules were given isoprote

Enteroendocrine cells (EECs) are specialized cells located throughout the gastrointestinal (GI) tract, where they have an essential role in regulating various physiological processes related to digestion, metabolism, and gut physiology. EECs secrete different hormones in response to food intake and the presence of nutrients in the gut, which regulate digestion, appetite, insulin secretion, and energy balance. One of the most well-studied hormones is glucagon-like peptide-1 (GLP-1), which is produced by L cells in both the small intestine and the colon. Colonic GLP-1-secreting L cells are not i

A number of species of euryhaline teleosts have the remarkable ability to adapt and survive in environments of extreme salinity, up to two or even three times the osmolality of seawater. This review looks at some of the literature describing the adaptive changes that occur, primarily with intestinal water absorption and with the properties of the gill epithelium. While there is much that is still not completely understood, recent work has begun to look at these adaptations at the cellular and molecular level. As with seawater osmoregulation, fish adapting to hypersaline conditions generally in

OBJECTIVES: Intestinal mucositis is the major side effect during abdominal or pelvic radiotherapy, but the underlying immunogen remains to be further characterised and few radioprotective agents are available. This study investigated the role of dsDNA-triggered inflammasomes in intestinal mucositis during radiotherapy.
METHODS: Pro-inflammatory cytokines were detected by ELISA. Radiation-induced intestinal injury in mice was analyzed by means of survival curves, body weight, HE staining of intestines, and intestinal barrier integrity. Western blot, immunofluorescence staining, co-immunoprecip

Macronutrients in the gastrointestinal (GI) lumen are able to activate "intestinal brakes", feedback mechanisms on proximal GI motility and secretion including appetite and energy intake. In this review, we provide a detailed overview of the current evidence with respect to four questions: (1) are regional differences (duodenum, jejunum, ileum) present in the intestinal luminal nutrient modulation of appetite and energy intake? (2) is this "intestinal brake" effect macronutrient specific? (3) is this "intestinal brake" effect maintained during repetitive activation? (4) can the "intestinal bra

Deoxynivalenol (DON), a prevalent Fusarium mycotoxin, poses a significant threat to intestinal health in piglets. This study established a deoxynivalenol (DON) intoxication model in piglets, revealing compromised intestinal barrier function accompanied by inflammatory responses and oxidative stress. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS) on intestinal samples from piglets (n = 6 per group), we identified 8,084 proteins, of which 124 showed significantly altered expression following DON exposure. Bioinformatic analysis indicated that the differentially expressed protein

Pineal melatonin is a potent synchronizer within the circadian system. Certain extra-pineal tissues also synthesize melatonin, including the gastrointestinal organs, whose melatonin contents can exceed that of pineal severalfold. Considering the role of pineal melatonin as a general synchronizer, we investigated whether the intestinal melatonin could serve a similar role in the local organization of the metabolic events, as a core element of the Food-Entrainable Oscillator (FEO). To be considered so, the intestinal secretory activity should meet some requirements: it should have self-sustained
