Associations of prenatal dietary exposure to multiple trace elements and child neurodevelopment in the ELFE mother-child cohort.

BACKGROUND: Prenatal exposure to neurotoxic trace elements may disrupt early brain development, with diet as a major exposure pathway during pregnancy. This study examined the associations between maternal dietary exposure to trace elements during late pregnancy and child neurodevelopment at 3.5 years. METHODS: We analyzed 10,080 mother-child pairs from the Étude Longitudinale Française depuis l'Enfance (ELFE) study. Maternal dietary exposure to 14 trace elements was assessed by combining a food frequency questionnaire with trace element content data in foodstuffs. Child neurodevelopment was

Exposure risks to phthalates by cosmetics and personal care products in pregnant women and early life: Urinary levels, mechanisms, biomarkers, and mitigation strategies: a review.

The increasing use of personal care products (PCPs) and cosmetics have increased the risk of exposure to phthalates, especially in the early stages of life for maternal-fetal-infant. This review aimed to determine the phthalates exposure risk through the consumption of PCPs and cosmetics in Pregnant women and early life, determine the mechanisms, biomarkers, and mitigation strategies for phthalates. The highest phthalates exposure levels of pregnant women (as well as fetal exposure through placenta and infants through breast milk) who used PCPs and cosmetics continuously or in larger quantitie

Exposure to methylergonovine maleate as a cause of sirenomelia.

BACKGROUND: Sirenomelia is a rare, but deadly condition characterized by fusion of the lower limbs, lower spinal column defects, severe malformations of the urogenital and lower gastrointestinal tract, and an aberrant abdominal umbilical artery. METHODS: The two main hypotheses, not mutually exclusive, that have been advanced to explain the pathogenesis of sirenomelia are the blastogenetic theory and the vascular disruption theory. RESULTS: We describe a case of sirenomelia, probably associated with the use of methylergonovine maleate, an ergot alkaloid, during the first weeks of pregnancy.

Small extracellular vesicles: connecting early life exposure outcomes to air pollution during pregnancy to early childhood health.

Exposure to air pollution poses a serious threat to maternal and child health, particularly during critical developmental periods. Extracellular vesicles (EVs), as key mediators of intercellular communication, have emerged as a novel mechanism through which environmental exposures, including air pollutants, exert systemic effects. This review synthesizes current evidence on the role of small EVs (sEVs) in mediating the biological impacts of prenatal air pollution exposure, with a focus on their molecular profile, including the presence of signaling molecules like miRNAs and proteins, and their

Neuronal substrates and functional consequences of prenatal cannabis exposure.

Cannabis remains one of the world's most widely used substance of abuse amongst pregnant women. Trends of the last 50 years show an increase in popularity in child-bearing women together with a constant increase in cannabis potency. In addition, potent herbal "legal" highs containing synthetic cannabinoids that mimic the effects of cannabis with unknown pharmacological and toxicological effects have gained rapid popularity amongst young adults. Despite the surge in cannabis use during pregnancy, little is known about the neurobiological and psychological consequences in the exposed offspring.

Developmental programming: Sex-specific effects of prenatal exposure to a real-life mixture of environmental chemicals on liver function and transcriptome in sheep.

Humans are chronically exposed to a mixture of environmental chemicals (ECs), many with metabolic and endocrine disrupting potential, contributing to non-communicable disease burden. Understanding the effects of chronic exposure to low-level mixtures of ECs requires an animal model that reflects real-world conditions, lags behind studies on single ECs. Biosolids, from wastewater treatment, offers a real-life model to investigate the developmental health risks from EC mixtures. Prenatal biosolids exposure studies have documented metabolic perturbations including heavier thyroid glands in male f

Prenatal pesticide exposure and adverse reproductive outcomes: Epidemiological evidence and mechanistic insights.

The widespread use of pesticides has led to ubiquitous environmental residues, posing risks to vulnerable populations, including pregnant women and fetal/neonatal developmental stages. Numerous epidemiologic studies have demonstrated that maternal exposure to pesticides is associated with adverse reproductive outcomes, including pregnancy complications and neonatal birth adverse effects. This article reviewed the effects of maternal exposure to pesticides (including insecticides, herbicides and fungicides) on pregnancy complications including spontaneous abortion, recurrent miscarriage, gestat…

Global air pollution exposure and congenital anomalies: an updated systematic review and meta-analysis of epidemiological studies.

A systematic review and meta-analysis was conducted to evaluate recent epidemiological evidence on the association of air pollution with congenital anomalies (CAs). Of 11,014 records, 49 were finally included in this meta-analysis. Per 10 μg/m3 increase in air pollutant, PM10 exposure during the 1st month of pregnancy and at the first trimester (T1) was associated with increased overall CAs. Further, exposure to PM10 was associated with congenital heart disease (OR = 1.055, 95% CI: 1.035, 1.074) and patent ductus arteriosus (OR = 1.094, 95% CI: 1.020, 1.168) at T1, with chromosomal anomalies d

Exploring associations between prenatal exposure to multiple endocrine disruptors and birth weight with exposure continuum mapping.

BACKGROUND: Improved understanding of how prenatal exposure to environmental mixtures influences birth weight or other adverse outcomes is essential in protecting child health. OBJECTIVE: We illustrate a novel exposure continuum mapping (ECM) framework that combines the self-organizing map (SOM) algorithm with generalized additive modeling (GAM) in order to integrate spatially-correlated learning into the study mixtures of environmental chemicals. We demonstrate our method using biomarker data on chemical mixtures collected from a diverse mother-child cohort. METHODS: We obtained biomarker c

Developmental toxicity of lead in rats after gestational exposure and the protective role of taurine.

The present study was conducted to investigate the potential adverse effect of Pb on pregnant Sprague-Dawley rats and their fetuses after maternal exposure, on gestational days (GD) 7-16. The possible protective role of taurine (TA), administered throughout the gestation period (GD 1-20) against Pb toxicity, was also evaluated. Pregnant rats were divided into four groups: Group 1 (control) was given distilled water; Group 2 was exposed to Pb (250 ppm) in drinking water (GD 7-16), whereas Group 3 received TA (50 mg/kg/day) by oral gavage (GD 1-20); Group 4 was exposed to Pb (GD 7-16), whereas p…

Fetal androgen exposure is a determinant of adult male metabolic health.

Androgen signalling is a critical driver of male development. Fetal steroid signalling can be dysregulated by a range of environmental insults and clinical conditions. We hypothesised that poor adult male health was partially attributable to aberrant androgen exposure during development. Testosterone was directly administered to developing male ovine fetuses to model excess prenatal androgenic overexposure associated with conditions such as polycystic ovary syndrome (PCOS). Such in utero androgen excess recreated the dyslipidaemia and hormonal profile observed in sons of PCOS patients. 1,084 o

Growth hormone reverses dyslipidemia in adult offspring after maternal undernutrition.

The abnormal intrauterine milieu of fetal growth retardation could lead to dyslipidemia in adulthood. Studies have shown that growth hormone (GH) therapy in small for gestational age (SGA) children would be beneficial for metabolic parameters. Here we investigated whether GH treatment introduced at adolescent period in SGA could reverse dyslipidemia during later life. SGA rat model was established by using semi-starvation treatment during the whole pregnancy. SGA or appropriate for gestational age (AGA) offspring were assigned to receive GH or normal saline (NS). Once-daily subcutaneous inject

Life-long programming implications of exposure to tobacco smoking and nicotine before and soon after birth: evidence for altered lung development.

Tobacco smoking during pregnancy remains common, especially in indigenous communities, and likely contributes to respiratory illness in exposed offspring. It is now well established that components of tobacco smoke, notably nicotine, can affect multiple organs in the fetus and newborn, potentially with life-long consequences. Recent studies have shown that nicotine can permanently affect the developing lung such that its final structure and function are adversely affected; these changes can increase the risk of respiratory illness and accelerate the decline in lung function with age. In this r
