GLP-1 receptor agonists at the crossroads of obesity and addiction: A review of shared neurobiology and translational evidence.

Obesity and substance use disorders (SUDs) are chronic relapsing conditions that may co-occur through convergent pathophysiological mechanisms involving dysregulated reward circuitry. Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) - originally developed for type 2 diabetes and obesity - act on both peripheral metabolic and central reward pathways, positioning them as potential candidates for repurposing in addiction medicine. In this narrative review, we sought to synthesize the shared neurobiology of obesity and SUDs and critically evaluate emerging translational evidence supporting GL

Gut dysbiosis associated with neonatal respiratory distress syndrome and biological plausibility of disease-specific probiotic intervention: a translational study.

BACKGROUND: Neonatal respiratory distress syndrome (RDS) is among the most prevalent morbidities in late preterm and term infants. Although the gut-lung axis has been implicated in neonatal respiratory disease, the relationship between RDS and early gut microbiome composition remains poorly characterized. This study aimed to characterize gut microbiome alterations associated with RDS and surfactant replacement therapy (SRT), and to evaluate the biological plausibility of a disease-specific probiotic intervention. METHODS: Two complementary cohorts were prospectively enrolled. In the clinical

Heart-Type Fatty Acid-Binding Protein (H-FABP) as a Candidate Adjunctive Biomarker for Immune Checkpoint Inhibitor-Related Cardiotoxicity: Linking Early Immune-Metabolic Myocardial Injury with Translational Cardio-Oncology.

Immune checkpoint inhibitors (ICIs) have transformed the therapeutic landscape of oncology but are increasingly associated with cardiovascular immune-related adverse events (irAEs), including myocarditis, heart failure, arrhythmias, and vascular complications. Among these, ICI-associated myocarditis represents the most severe manifestation, often characterized by high mortality and challenging early diagnosis. Detecting subclinical myocardial injury before irreversible cardiomyocyte necrosis occurs remains a major unmet need in contemporary cardio-oncology. This narrative expert review critica

Translational Nanomedicine for Osteoporosis: Bridging Bone Signaling Networks and Targeted Delivery Systems.

Osteoporosis is a complex skeletal disorder characterized by reduced bone mineral density and microarchitectural deterioration, leading to an increased risk of fractures. Conventional pharmacotherapies, such as bisphosphonates and selective estrogen receptor modulators, are constrained by poor bioavailability, lack of targeting specificity, and systemic side effects. Unlike previous reviews that examine signaling pathways and nanocarrier design in isolation, this review presents an integrated mechanistic framework wherein specific therapeutic nodes within key bone remodeling pathways-including

Smart alginate-based biomaterials for neurodegenerative disease therapy: Innovations in delivery, regeneration, and clinical translation.

Neurodegenerative diseases and central nervous system (CNS) injuries remain among the most challenging disorders to treat due to their complex pathophysiology, limited regenerative capacity, and the presence of the blood-brain barrier (BBB), which severely restricts therapeutic delivery. Despite extensive research efforts, most current interventions are palliative and fail to modify disease progression. Biomaterial-based strategies have emerged as promising adjuncts to conventional therapies, with alginate-based systems attracting increasing attention due to their biocompatibility, mild aqueou

Mitochondrial-targeted therapy for osteoarthritis: Challenges and opportunities from basic research to clinical translation.

Osteoarthritis is a high-burden degenerative joint disease. Existing therapies only alleviate symptoms but fail to halt disease progression. Studies have identified mitochondrial dysfunction as a core driver of cartilage degeneration in OA. Key mechanisms include mitochondrial reactive oxygen species bursts that activate inflammatory and cell death pathways; imbalances in mitochondrial dynamics leading to fragmentation; autophagy defects causing damage accumulation; and reduced biogenesis coupled with hyperglycolysis, which exacerbates the energy crisis. Collectively, these processes accelerat

Translational tumor control probability modeling for NSCLC: A two-dimensional maximum-likelihood framework.

BACKGROUND: Tumor control probability (TCP) modeling for early-stage non-small cell lung cancer (NSCLC) is usually performed by one-dimensional (1D) fitting that assumes error-free dose and assigns all uncertainty to the local control (LC) axis. Neglecting dose-axis uncertainties leads to a classical errors-in-variables (EIV) problem that can bias both the dose required for 50% TCP ( TC D 50 ) and curve steepness. Furthermore, the mechanistic origins of TCP steepness remain incompletely characterized, limiting the translational link between cellular radiobiology and clinical dose-response.

Mirvetuximab Soravtansine: Mechanism of Action, Clinical and Translational Science.

Mirvetuximab soravtansine (MIRV) is an antibody-drug conjugate (ADC) composed of the DM4 payload conjugated to a folate receptor α (FRα)-targeting antibody via the cleavable sulfo-SPDB linker. MIRV targets and binds to FRα with high affinity and specificity, releasing the DM4 payload intracellularly following MIRV-FRα complex internalization and degradation. DM4 and its metabolite S-methyl-DM4 suppress microtubule dynamic instability, which triggers cell cycle arrest and apoptosis. Selective FRα-overexpression in ≥ 90% of epithelial ovarian tumor cells and its ability to internalize large mole

Stem cells as emerging regenerative approaches for post-traumatic stress disorder: Mechanisms and translational challenges.

Post-traumatic stress disorder (PTSD) is a debilitating neuropsychiatric condition triggered by severe trauma, characterised by dysregulated fear circuitry, hippocampal atrophy with impaired neurogenesis, chronic neuroinflammation, neuroendocrine dysregulation, and disrupted prefrontal-limbic connectivity. Existing treatments are largely symptomatic, failing to address underlying neurobiological deficits. Emerging regenerative approaches using human stem cells, particularly induced pluripotent stem cell-derived neural progenitor cells (iPSC-NPCs), human embryonic stem cells (hESCs), mesenchyma

GLP-1 Receptor Agonists in Obstructive Sleep Apnea: A Translational Perspective on Mechanisms and Clinical Implications.

Obstructive sleep apnea (OSA) affects nearly one billion adults worldwide and involves mechanical upper-airway obstruction alongside metabolic dysfunction, systemic inflammation, and gut-brain axis disruption. Despite the efficacy of continuous positive airway pressure (CPAP), suboptimal long-term adherence highlights the need for complementary pharmacological strategies. This narrative review synthesizes preclinical and clinical evidence on GLP-1-based therapies, encompassing selective GLP-1 receptor agonists and tirzepatide, a dual GIP/GLP-1 receptor agonist, in obesity-associated OSA. Genome Editing for Familial Hemophagocytic Lymphohistiocytosis: Design Principles, Challenges, and Translational Perspectives.

Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome caused by genetic defects in cytotoxic lymphocyte function. Current therapies can control disease activity, but transplantation of allogeneic hematopoietic stem and progenitor cells (HSPCs) remains the only curative option and is associated with substantial risks. These limitations have accelerated development of genome editing approaches enabling precise correction of disease-causing mutations in autologous cells. Familial HLH (FHL) represents a compelling target for genome editing, but successful and sa

GLP-1 receptor agonists and cancer: current clinical evidence and translational opportunities for preclinical research.

Cancer diagnoses are prevalent in people with obesity and type 2 diabetes, and abundant clinical evidence supports the protective effects of weight loss for cancer prevention. Glucagon-like peptide-1 (GLP-1) receptor agonists have revolutionized obesity and type 2 diabetes medicine and alleviate many comorbidities of these metabolic diseases. In this Review, we summarize the current clinical evidence for GLP-1 receptor agonists and cancer risk, including thyroid, pancreatic, gastrointestinal, and hormone-dependent malignancies. With few exceptions, recent meta-analyses report that GLP-1 recept

Translating a Preclinically Tested 15 Hz rTMS Protocol to Humans With Chronic Spinal Cord Injury: A Safety and Feasibility Study.

OBJECTIVES: Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive brain stimulation strategy with a demonstrated potential to reinforce the residual pathways after a spinal cord injury (SCI). A preclinically tested high-frequency (15 Hz) rTMS (15 Hz rTMS) protocol was shown to induce corticospinal tract axon regeneration growth and sprouting, resulting in improved voluntary motor control and performance. In a translational perspective, we aimed to investigate the safety and feasibility of the 15 Hz rTMS paradigm as an adjunct therapeutic intervention in individuals with chronic

[Biological characteristics and translational research of dental stem cells].

Dental stem cells (DSCs), a distinct subset of mesenchymal stem cells (MSCs), are isolated from dental tissues, such as dental pulp, exfoliated deciduous teeth, periodontal ligament, and apical papilla. They have emerged as a promising source of stem cell therapy for tissue regeneration and autoimmune disorders. The main types of DSCs include dental pulp stem cells (DPSCs), stem cells from human exfoliated deciduous teeth (SHED), periodontal ligament stem cells (PDLSCs), and stem cells from apical papilla (SCAP). Each type exhibits distinct advantages: easy access via minimally invasive proced

Modeling Postoperative Pathologic Ileus in Mice: A Simplified and Translational Approach.

BACKGROUND: Postoperative Ileus (POI) is an iatrogenic complication characterized by a temporary paralysis of gastrointestinal transit, leading to food intolerance, nausea, vomiting, and thus prolonged hospitalization. The severity of POI is influenced by surgical trauma, particularly in intestinal surgeries, which have a high complication rate. To date, no animal model has precisely replicated POI in the context of digestive sutures or anastomoses, procedures common in human digestive resections. METHODS: To induce POI, mice underwent different surgeries. The surgical procedure involved a mi

Cell Counting and Cell Cycle Analysis of Simple Non-Cultured Endothelial Cell Injection (SNEC-I) Therapy: Characterization for Clinical Translation.

Human corneal endothelial cell therapy has recently emerged as a novel solution to treat corneal endothelial diseases. We previously demonstrated the potential of utilizing non-cultured primary corneal endothelial cells (CEnCs) isolated from donor corneas with low endothelial cell density for simple non-cultured endothelial cell injection (SNEC-I) therapy. This study aimed to develop a robust and semi-automated approach for cell counting, characterize the extent of cellular manipulation, and evaluate the translational workflow. To address this, we evaluated manual and automated cell counting a
