A p50 surface antigen restricted to human urinary bladder carcinomas and B lymphocytes.

We have previously described the derivation of a monoclonal antibody, S2C6, to a novel 50 Kdalton antigen associated with human urinary bladder carcinoma. No reactions were obtained with carcinomas of unrelated origin or with normal urothelial cells. However, the antibody also reacted with a similar antigen on some cell lines of B lymphocyte origin. Using large panels of target cells we have now shown that this reactivity was entirely restricted to cells of the B lineage within the haematopoietic system. As opposed to its apparent restriction to malignant cells of the urothelium, the S2C6 anti…

Osmotic permeabilities across corneal endothelium and antidiuretic hormone-stimulated toad urinary bladder structures.

Osmotic permeabilities of several epithelial structures have been determined with novel optical procedures based on specular microscopy. The osmotic permeabilities of several tissue layers were determined by continuously monitoring the position of the apical tissue borders while an osmotic flow was imposed across those layers. The values found were (in micrometer/s; mean +/- SE): corneal epithelium, 137 +/- 30 (n = 5): antidiuretic hormone stimulated toad bladder, 429 +/- 64 (n = 6); and corneal endothelium, 711 +/- 34 (n = 7). In addition, the osmotically-induced transient change in thickness

Effect of vasopressin on cyclic AMP-dependent protein kinase in toad urinary bladder.

The effect of vasopressin on the toad urinary bladder has been shown to be mediated by cyclic AMP. It has been assumed that, as demonstrated for other systems, this involves activation of cyclic AMP-dependent protein kinase. In order to test this hypothesis we investigated the effect of vasopressin on cyclic AMP-dependent protein kinases in epithelial cells of toad bladders. About 80% of protein kinase activity and cyclic AMP-binding capacity was found to be in the cytosol. DEAE-cellulose chromatography showed a pattern of 15--20% type I and 80--85% type II cyclic AMP-dependent protein kinase.…

Vasopressin-stimulated movement of drugs and uric acid across the toad urinary bladder.

Vasopressin is known to increase the permeability of the toad bladder, an analogue of the mammalian collecting duct, to water and hydrophilic solutes such as urea. In the present study, the effect of vasopressin on the permeability of a series of lipophilic compounds, including many commonly used drugs, has been determined. In all cases, permeability increased from 50 to 100%. The response to vasopressin was mediated by cyclic adenosine monophosphate (cAMP), and was generally not altered by phloretin, an agent that inhibits amide movement through the amide transport pathway. Evidence that thes

Urinary Biomarkers Objectively Measure Minimal Residual Disease in Non-Muscle-Invasive Bladder Cancer.

PURPOSE: To determine whether carcinoma in situ (CIS) is a sufficient measure of minimal residual disease (MRD) in non-muscle-invasive bladder cancer (NMIBC) and evaluate alternative measures of MRD. EXPERIMENTAL DESIGN: We evaluated the concordance of CIS on transurethral resection of bladder tumor (TURBT) and radical cystectomy (RC) to determine eradication rates of clinical CIS (cCIS) by TURBT alone and rates of occult pathologic CIS (pCIS) seen only in RC specimens. We studied a prospective cohort of patients with BCG-naïve high-grade NMIBC to evaluate pretreatment urinary cytology and ur

Benign Glandular Lesions of the Urinary Bladder: Diagnostic Criteria and Differential Diagnosis Considerations.

CONTEXT.—: The urinary bladder is a unique organ that may be involved by both benign and malignant glandular lesions. Urothelial carcinoma may possess glandular differentiation, and other glandular malignancies may arise from or secondarily involve the urinary bladder. However, many benign glandular lesions may also involve the urinary bladder, and these can mimic malignancy. Relatively common benign lesions of the urinary bladder that may demonstrate glandular or tubular features include cystitis glandularis, glandular metaplasia, nephrogenic adenoma, Müllerian lesions, prostatic-type polyp, …

Glycovariant-based diagnosis of bladder cancer using urinary mucin 1 and carcinoembryonic antigen.

Glycan alterations, or glycovariants (GV), are well-recognized features of malignant transformation and are commonly observed in several cancers, including bladder cancer (BlCa). However, their integration into practical diagnostic assays remains limited. Here, we investigated whether glycan modifications on the urinary glycoproteins mucin 1 (MUC1) and carcinoembryonic antigen (CEA) could serve as diagnostic indicators for BlCa diagnosis. MUC1 and CEA were captured directly from urine samples of patients with benign prostate disease (n = 20) and BlCa (n = 19) by immobilizing monoclonal antibod

PD-L1 expression in schistosomal granuloma: an immunohistochemical study in urinary bladder schistosomiasis.

AIMS: Schistosomiasis remains endemic in various parts of the world, and insights into pathogen immunobiology are mainly based on experimental models, while studies on human tissues are limited. METHODS: We explored the role of immune checkpoint pathway by evaluating the immunohistochemical expression of programmed death-ligand 1 (PD-L1) in a retrospective cohort of patients with bilharzial cystitis. Inflammation severity by conventional histology and staining intensity by immunohistochemistry were assigned three-tier scores (0/1+/2+), and a cut-off for staining percentage was set at 5%. RES

Urinary protein semi-quantification in immunochemotherapy for bladder cancer: markers of clinical outcomes.

OBJECTIVE: PD-1 based immunochemotherapy has improved bladder cancer (BC) outcomes, yet individual responses remain heterogeneous. Low-cost, non-invasive predictive biomarkers are urgently needed. Urinary protein semi-quantification (UPSQ) correlates with BC tumor load, but its predictive value during PD-1 based immunochemotherapy remains unclear. We conducted a post-hoc analysis of the TRUCE-01 (NCT04730219) and TRUCE-02 (NCT04730232) trials to explore the association between UPSQ and clinical outcomes. METHODS: 104 patients received 3 cycles of tislelizumab plus nab-paclitaxel. UPSQ at base

Non-coding RNAs in urinary bladder cancer microenvironment: Diagnostic, therapeutic, and prognostic perspective.

Urinary bladder cancer (UBC) is the ninth most common cancer worldwide. Despite the reliance of UBC therapy on definite pathological grading and classifications, the clinical response among patients varies widely. The molecular basis of this type of cancer appeals to considerable research; hence, new diagnostic and therapeutic options are introduced. Convenient keywords were searched in Google Scholar, PubMed, the Egyptian Knowledge Bank (EKB), and Web of Science. The recent era of UBC research is concerned with non-coding RNAs (ncRNAs), predominantly, microRNAs (miRNAs) and long non-coding RN…

Trained Immunity in Bladder ILC3s Enhances Mucosal Defense Against Recurrent Urinary Tract Infections.

Background: Urinary tract infections (UTIs) rank among the most prevalent infectious diseases globally, with recurrent UTIs (rUTIs) posing substantial therapeutic challenges due to the lack of durable protective immunity. While trained immunity augments innate immune responses, its induction and functional significance in bladder-resident group 3 innate lymphoid cells (ILC3s) remain unknown. This study investigates whether ILC3s develop trained immunity following uropathogenic Escherichia coli (UPEC) exposure and how they contribute to mucosal defense against rUTIs. Methods: The ILC3 counts we

Bladder mucosal smoothness predicts early recovery of urinary continence after laparoscopic radical prostatectomy.

BACKGROUND: To propose the bladder mucosal smoothness (BMS) grade and validate a predictive model including MRI parameters preoperatively that can evaluate the early recovery of urinary continence (UC) after laparoscopic radical prostatectomy (LRP). METHODS: A retrospective analysis was conducted on 203 patients (83 patients experienced UI at the three-month follow-up) who underwent LRP in our medical center and were diagnosed with prostate cancer (PCa) from June 2016 to March 2020. Patients' clinicopathological data were collected. Prostate volume (PV), membranous urethra length (MUL), intra

Macroplastique, a urethral bulking agent for stress urinary incontinence mimicking a bladder tumour.

Urinary incontinence is a stigmatised condition with high prevalence and significant patient morbidity. Stress urinary incontinence (SUI) is a subtype of incontinence particularly prevalent in middle-aged to older women-often seen as a sequelae of childbirth or pelvic surgery. Treatment has proved challenging with a chequered history. Urethral bulking agents have been developed as a minimally invasive treatment option for mild to moderate SUI. Following injection, the agent causes mild deformation of the urethral anatomy leading to reduced incontinence as higher pressures are subsequently requ
