First serological investigation of Toxoplasma gondii, Neospora caninum, Leishmania infantum and Leptospira spp. in dogs from a Fulni-ô Indigenous community in Pernambuco, Brazil: a One Health perspective.

Zoonotic diseases pose a major public health threat in indigenous communities due to geographic isolation, inadequate sanitation, and close contact between humans, domestic animals, and wildlife. This study aimed to investigate the presence of antibodies against Toxoplasma gondii, Neospora caninum, Leishmania infantum, and Leptospira spp. in dogs from the Fulni-ô indigenous community, located in Águas Belas, Pernambuco, Brazil. Blood samples were collected from 66 free-roaming or semi-domiciled dogs for serological analysis. Indirect immunofluorescence tests (IFAT) were used to detect antibodi

Maternal immune activation affects socio-communicative behavior in adult rats.

A wide body of evidence suggests a relationship between maternal immune activation (MIA) and neurodevelopmental disorders such as autism spectrum disorder (ASD). Since social and communicative deficits are included in the first diagnostic criterion of ASD, we aimed to characterize socio-communicative behaviors in the MIA model based on prenatal exposure to poly(I:C). Our previous studies demonstrated impaired socio-communicative functioning in poly(I:C)-exposed adolescent rats. Therefore, the current study sought to clarify whether these changes would persist beyond adolescence. For this purpo

Establishing causality in Salmonella-microbiota-host interaction: The use of gnotobiotic mouse models and synthetic microbial communities.

Colonization resistance (CR), the ability to block infections by potentially harmful microbes, is a fundamental function of host-associated microbial communities and highly conserved between animals and humans. Environmental factors such as antibiotics and diet can disturb microbial community composition and thereby predispose to opportunistic infections. The most prominent is Clostridioides difficile, the causative agent of diarrhea and pseudomembranous colitis. In addition, the risk to succumb to infections with genuine human enteric pathogens like nontyphoidal Salmonella (NTS) is also incre

Dog Ecology, Bite Incidence, and Disease Awareness: A Cross-Sectional Survey among a Rabies-Affected Community in the Democratic Republic of the Congo.

Despite the existence of safe and efficacious human and animal rabies vaccines, millions of people remain at risk of exposure to this deadly zoonotic disease through bites of infected dogs. Sub-Saharan African countries, such as the Democratic Republic of the Congo (DRC), bear the highest per capita death rates from rabies where dog vaccination and availability of lifesaving post-exposure prophylaxis (PEP) is scarce. Mass dog vaccination is the most cost-effective and sustainable approach to prevent human rabies deaths. We conducted a cross-sectional household survey in a rabies-affected commu

Comparative analysis of the fecal bacterial community of five harbor seals (Phoca vitulina).

The gut microbiota has many beneficial effects on host metabolism and health, and its composition is determined by numerous factors. It is also assumed that there was a co-evolution of mammals and the bacteria inhabiting their gut. Current knowledge of the mammalian gut microbiota mainly derives from studies on humans and terrestrial animals, whereas those on marine mammals are sparse. However, they could provide additional information on influencing factors, such as the role of diet and co-evolution with the host. In this study, we investigated and compared the bacterial diversity in the fece

Detection of Brucella abortus in Chiredzi district in Zimbabwe.

Brucellosis is an endemic disease in Zimbabwe caused by the genus Brucella. Brucella seroprevalence was recently reported to be high in the wildlife-livestock interface in the Chiredzi district and the neighbouring Gonarezhou National Park (GNP) in Zimbabwe, and higher amongst communal cattle with an abortion history and access to grazing in GNP than amongst communal cattle with no abortion history or access to grazing in GNP. The aim of this study was to investigate Brucella species in brucellosis seropositive cattle in the Chiredzi district with access to GNP using isolation and identificati

Predominance of Blastocystis sp. subtype 4 in rural communities, Nepal.

Blastocystis sp. is a common intestinal parasite. To date, there have been sporadic and scanty studies on Blastocystis sp. carried out in rural communities in Nepal. We surveyed the prevalence of Blastocystis sp. and its possible associated risk factors, and reported the predominant Blastocystis sp. subtype in two rural communities, Bolde Phediche and Bahunipati, in Nepal. Human faecal samples were collected from 241 participants, cultured using in vitro cultivation and examined for Blastocystis sp. The presence of Blastocystis sp. in faecal samples was further confirmed by polymerase chain re

Gut microbiome shifts during frog development parallel host-association patterns across vertebrate lineages.

The developmental habitat transitions between aquatic and terrestrial environments in frogs offer a compelling model to investigate how environmental shifts shape gut microbiota. However, our understanding of how these transitions influence gut microbial communities across frog development remains limited. Here, we investigated how habitat changes during frog development affect gut microbiota composition and compared these patterns with host-microbiome interactions across vertebrates. Frog gut microbiota composition changed across developmental stages, and the proportion attributable to the su

Unique microbial communities and phylosymbiosis signals in herpetofauna.

Microbial symbionts are closely related to the internal and external factors of their host. However, the prevalence of phylosymbiosis (the presence of host phylogenetic signal in microbial community composition) remains controversial, especially in animals collectively referred to as herpetofauna. To expand our understanding of host-microbiota interactions, we analysed 11 697 symbiotic microbiota samples from of 337 herpetofaunal species, covering skin, oral cavity, gut, cloaca, feces, and other body sites. The composition of the microbial communities gradually changes along the digestive trac

Association of Anthropogenic Disturbances and Intestinal Parasitism in Ecuadorian Mantled Howler Monkeys, Alouatta palliata aequatorialis.

Forest disturbance and human encroachment have the potential to influence intestinal parasite communities in animal hosts by modifying nutritional health, physiological stress, host densities, contact rates, and ranging patterns. Anthropogenic disturbances also have the ability to affect the ecological landscape of parasitic disease, potentially impacting the health of both wildlife and people. Our research investigated the association of forest disturbance and human encroachment on intestinal parasite communities in mantled howler monkeys, Alouatta palliata aequatorialis. We found that indivi

Developments in the synthesis of the antiplatelet and antithrombotic drug (S)-clopidogrel.

S-(+)-Methyl 2-(2-chlorophenyl)-2-(6,7-dihydrothieno[3,2-c]pyridin-5(4H)-yl)acetate, also known as (S)-clopidogrel, is marketed under the trade names Plavix and Iscover. It is a potent thienopyridine-class of antithrombotic and antiplatelet drug (antiaggregant). Among the two available stereoisomers of clopidogrel, for pharmaceutical activities only the S-enantiomer is applicable, as no antithrombotic activity is observed in the R-enantiomer and causes political upheavals and social turmoil in animal experiments. Worldwide sales of Plavix amounted to $6.4 billion yearly, which ranks second. At

Discovery of Linvencorvir (RG7907), a Hepatitis B Virus Core Protein Allosteric Modulator, for the Treatment of Chronic HBV Infection.

Described herein is the first-time disclosure of Linvencorvir (RG7907), a clinical compound and a hepatitis B virus (HBV) core protein allosteric modulator, for the treatment of chronic HBV infection. Built upon the core structure of hetero aryl dihydropyrimidine, RG7907 was rationally designed by combining all the drug-like features of low CYP3A4 induction, potent anti-HBV activity, high metabolic stability, low hERG liability, and favorable animal pharmacokinetic (PK) profiles. In particular, the chemistry strategy to mitigate CYP3A4 induction through introducing a large, rigid, and polar su

The Disease Ecology, Epidemiology, Clinical Manifestations, Management, Prevention, and Control of Increasing Human Infections with Animal Orthopoxviruses.

Zoonotic orthopoxvirus outbreaks have occurred repeatedly worldwide, including monkeypox in Africa and the United States, cowpox in Europe, camelpox in the Middle East and India, buffalopox in India, vaccinia in South America, and novel emerging orthopoxvirus infections in the United States, Europe, Asia, and South America. Waning smallpox immunity may increase the potential for animal-to-human transmission followed by further community transmission person-to-person (as demonstrated by monkeypox and buffalopox outbreaks) and by contact with fomites (as demonstrated by camelpox, cowpox, and, po

Animal and translational models of SARS-CoV-2 infection and COVID-19.

COVID-19 is causing a major once-in-a-century global pandemic. The scientific and clinical community is in a race to define and develop effective preventions and treatments. The major features of disease are described but clinical trials have been hampered by competing interests, small scale, lack of defined patient cohorts and defined readouts. What is needed now is head-to-head comparison of existing drugs, testing of safety including in the background of predisposing chronic diseases, and the development of new and targeted preventions and treatments. This is most efficiently achieved using

Acute toxic kidney injury.

Substances toxic to the kidney are legion in the modern world. The sheer number and variety, their mutual interactions and, metabolism within the body are a challenge to research. Moreover, the kidney is especially prone to injury owing to its physiology. Acute kidney injury (AKI) induced by poisonous or primarily nephrotoxic substances, may be community acquired with ingestion or inhalation or nosocomial. Many nephrotoxic plants, animal poisons, medications, chemicals and illicit drugs can induce AKI by varying pathophysiological pathways. Moreover, the epidemiology of toxic AKI varies depend
