Understanding AL amyloidosis with a little help from in vivo models.

Monoclonal immunoglobulin (Ig) light chain amyloidosis (AL) is a rare but severe disease that may occur when a B or plasma cell clone secretes an excess of free Ig light chains (LCs). Some of these LCs tend to aggregate into organized fibrils with a β-sheet structure, the so-called amyloid fibrils, and deposit into the extracellular compartment of organs, such as the heart or kidneys, causing their dysfunction. Recent findings have confirmed that the core of the amyloid fibrils is constituted by the variable (V) domain of the LCs, but the mechanisms underlying the unfolding and aggregation of

Espin overexpression causes stereocilia defects and provides an anti-capping effect on actin polymerization.

Stereocilia are actin-based projections of hair cells that are arranged in a step like array, in rows of increasing height, and that constitute the mechanosensory organelle used for the senses of hearing and balance. In order to function properly, stereocilia must attain precise sizes in different hair cell types and must coordinately form distinct rows with varying lengths. Espins are actin-bundling proteins that have a well-characterized role in stereocilia formation; loss of function mutations in Espin result in shorter stereocilia and deafness in the jerker mouse. Here we describe the gene

Phosphomimetic Mutation at Ser165 of α-Tubulin Promotes the Persistence of GTP Caps in Microtubules.

Protein kinase C (PKC)-mediated phosphorylation of α-tubulin at Ser165 or expression of phosphomimetic (S165D)-α-tubulin stimulates microtubule (MT) polymerization (Cytoskeleton 2014, 71, 257-272). Ser165 lies near the interface between adjacent αβ-tubulin heterodimers and helix H8, which contains Glu254, the catalytic residue in α-tubulin that hydrolyzes the exchangeable GTP in β-tubulin (β:GTP) and triggers MT depolymerization. It was hypothesized that S165D, a phosphomimetic variant of α-tubulin, perturbs the alignment of α:Glu254 with respect to β:GTP, thereby impairing its hydrolysis. Mol…

Long-range mechanical signaling in biological systems.

Cells can respond to signals generated by other cells that are remarkably far away. Studies from at least the 1920's showed that cells move … ks of stiff or semiflexible filament such as those that form the extracellular matrix (ECM) and the cytoskeleton have unusual properties that suggest multiple mechanisms for long-range signaling in biological tissues. These include not only direct force transmission, but also highly non-uniform local deformations, and force-generated changes in fiber alignment …

Disease-associated keratin mutations reduce traction forces and compromise adhesion and collective migration.

Keratin intermediate filament (IF) proteins constitute the major cytoskeletal components in epithelial cells. Missense mutations in keratin 5 (K5; also known as KRT5) or keratin 14 (K14; also known as KRT14), highly expressed in the basal epidermis, cause the severe skin blistering disease epidermolysis bullosa simplex (EBS). EBS-associated mutations disrupt keratin networks and change keratinocyte mechanics; however, molecular mechanisms by which mutations shape EBS pathology remain incompletely understood. Here, we demonstrate that, in contrast to keratin-deficient keratinocytes, cells expre

Unusual dynamics of the divergent malaria parasite PfAct1 actin filament.

Gliding motility and host cell invasion by the apicomplexan parasite Plasmodium falciparum (Pf), the causative agent of malaria, is powered by a macromolecular complex called the glideosome that lies between the parasite plasma membrane and the inner membrane complex. The glideosome core consists of a single-headed class XIV myosin PfMyoA and a divergent actin PfAct1. Here we use total internal reflection fluorescence microscopy to visualize growth of individual unstabilized PfAct1 filaments as a function of time, an approach not previously used with this actin isoform. Although PfAct1 was tho

Cocaine addicted to cytoskeletal change and a fibrosis high.

Cocaine is one of the most widely abused illicit drugs due to its euphoric and addictive properties. Cocaine-mediated cognitive impairments are the result of dynamic cytoskeletal rearrangements involved in mediating structural and behavioural plasticity. Cytoskeletal changes initiated following cocaine abuse are regulated by the Rho family of GTPases with significant downstream activity in key actin binding proteins. Moreover, signalling via the endoplasmic reticulum chaperone protein, sigma-1 receptor has highlighted the possibility of cocaine regulated pathology in other organ systems. Howev

Beyond bouncy gaits: The role of multiscale compliance in skeletal muscle performance.

McNeill Alexander demonstrated that compliant tendons could improve locomotor performance by decoupling muscle length changes from joint movements and mechanical energy fluctuations. This was revolutionary for our understanding of animal locomotion, but also highlighted the limitations of our understanding of the contractile performance of muscle under the dynamic conditions relevant to movement. This review addresses the potential for biological compliance to not only alter the demands on muscle but also fundamentally change contractile performance. Compliance exists across all spatial scales

Consequences of Keratin Phosphorylation for Cytoskeletal Organization and Epithelial Functions.

Intermediate filaments are major phosphoproteins. The complex patterns of intermediate filament phosphorylation make up a poorly understood code reflecting cytoskeletal properties and cellular function through an intense crosstalk with multiple signaling pathways. This review focuses on the epithelial keratin intermediate filaments highlighting the tight-knit relationship of keratin phosphorylation and network organization during cell division and apoptosis, and the importance of keratin phosphorylation during epithelial stress responses. The occurrence of keratin phosphorylation in genetic sk

Proteomic identification of the molecular basis of mammalian CNS growth cones.

The growth cone, which is a unique structure with high motility that forms at the tips of extending axons and dendrites, is crucial to neuronal network formation. Axonal growth of the mammalian CNS is most likely achieved by the complicated coordination of cytoskeletal rearrangement and vesicular trafficking via many proteins. Before recent advances, no methods to identify numerous proteins existed; however, proteomics revolutionarily resolved such problems. In this review, I summarize the profiles of the mammalian growth cone proteins revealed by proteomics as the molecular basis of the growt

Suppression of anaphylactic shock in sensitized guinea pigs by cytochalasin D.

A new highly effective pathogenetic therapy for anaphylactic reaction in vivo has been suggested. Forty-three guinea pigs, presensitized with horse serum and divided into two groups, were injected intracardially with 1 ml of 1.6 x 10(-4) M cytochalasin D and 1 ml of 1.6 x 10(-5) M cytochalasin D in DMSO solution at various times before being challenged with the serum. Experimental data not only showed that anaphylactic reaction can be suppressed in vivo by blocking cytoskeletal activity but also demonstrated different pharmacodynamic characteristics of cytochalasin D on clinical course, gravit

Oncocytic bronchial adenoma. Histological, histochemical and ultrastructural study.

Benign adenomas derived from the bronchial mucous glands are uncommon. Probably the least common variety is the oncocytomatous adenoma, this case being the second known example. The tumour was situated in the right main bronchus in a man aged 75. An oncocytomatous adenoma should be clearly distinguished from the common oncocytomatous change which affects the normal bronchial mucous gland cells seen in adults. The eosinophilic, columnar tumour cells seen by light microscopy contained numerous prominent mitochondria by electron microscopy and granules of serous secretion. Other tumour cells cont

Possible role of myofibroblasts in the pathogenesis of Dupuytren's contracture.

In the contracting palmar aponeurosis of patients suffering from Dupuytren's disease myofibroblasts are revealed by electron microscopical examination. These cells display abnormal foldings of the nucleus, microfilaments resembling the myofilaments in smooth muscle cells, rough endoplasmic reticulum occupying circumscribed portions of the cytoplasm, free ribosomes, well-developed Golgi apparatus, numerous mitochondria, occasionally lipid droplets, cilia-like formations, hemidesmosomes and maculae adherens (desmosomes) between the cells and formation of basement membrane. High resolution scanning electron microscopy of elastic cartilage.

The elastic cartilage of the rat epiglottis was studied with a high-resolution SEM technique. The chondrocytes were found to be anchored in their lacunae by fibrils running in from the territorial matrix. This matrix exhibited a dense network of fibrils arranged tangentially around the lacunar cavity. The fibrils of the inter-territorial matrix however, formed a three dimensional network of sheets with interconnecting fibrils. The SEM has shown up for the first time a substructure in the fibrils in the form of circular protuberances arranged circumferentially around the fibrils and forming rid

Unusual gonadal stromal tumor of the testis. Case report with ultrastructural observations.

Ultrastructual observations on an unusual gonadal stromal tumor of the testis in a 4-year-old boy with gynecomastia revealed that the tumor cells lacked the abundant smooth endoplasmic reticulum usually found in neoplastic and nonneoplastic Sertoli and Leydig cells. The cells were rich in cytoplasmic microfilaments, with localized dense areas of the type seen in smooth muscle cells as well as intracytoplasmic lipid droplets. It is concluded that the tumor cells show important similarities to the immature fusiform interstitial cells and the peritubular contractile cells present in the normal te

Nebulette knockout mice have normal cardiac function, but show Z-line widening and up-regulation of cardiac stress markers.

AIMS: Nebulette is a 109 kDa modular protein localized in the sarcomeric Z-line of the heart. In vitro studies have suggested a role of nebulette in stabilizing the thin filament, and missense mutations in the nebulette gene were recently shown to be causative for dilated cardiomyopathy and endocardial fibroelastosis in human and mice. However, the role of nebulette in vivo has remained elusive. To provide insights into the function of nebulette in vivo, we generated and studied nebulette-deficient (nebl(-) (/-)) mice. METHODS AND RESULTS: Nebl(-) (/-) mice were generated by replacement of ex

The effects of cytoskeletal inhibitors on the collagen gel contraction by dog periodontal ligament fibroblasts in vitro.

Periodontal ligament fibroblasts (PLF) were incorporated into small collagen gel discs (100 microliters, dia 10.0 mm), cultured in media containing various concentrations of cytochalasin D or colchicine and the diameters of the discs then measured. Cytochalasin D (3.1-800 nM) inhibited contraction of the gel discs in a dose-dependent manner and 800 nM of cytochalasin D completely inhibited contraction. This complete inhibition was still observed when cell density in the gel was increased. Although colchicine at a concentration of 0.98-15.7 nM also inhibited the contraction in a dose-dependent

A plasma membrane integral sialoglycoprotein (Sgp 130) molecularly distinguishes nonjunctional dense plaque sites of microfilament attachment.

An integral sialoglycoprotein with Mr approximately 130,000 (Sgp 130) and highest expression in adult chicken gizzard smooth muscle has been recently identified as an excellent candidate for classification as a plasma membrane protein natively associated (directly or indirectly) with actin microfilaments (Rogalski, A.A., and S.J. Singer, 1985, J. Cell Biol., 101:785-801). In this study, the relative in situ distributions of the Sgp 130 integral species (a designation that also includes non-smooth muscle molecular forms) and the peripheral protein, vinculin, have been simultaneously revealed fo

Linggui Zhugan Decoction mitigates post-myocardial infarction heart failure through modulation of cardiomyocyte F-actin cytoskeletal organization.

BACKGROUND: Linggui Zhugan Decoction (LGZGD), a traditional Chinese formula for spleen-strengthening and yang-warming to resolve fluid retention, has consistently shown efficacy in preventing and treating heart failure. However, its underlying biological mechanisms remain incompletely understood. OBJECTIVE: This study aimed to investigate the therapeutic efficacy of LGZGD in a rat model of heart failure and in H9C2 cardiomyocytes, as well as to elucidate its effects on F-actin remodeling and the underlying mechanisms. METHODS: A rat model of heart failure post-myocardial infarction was estab
