Energy landscapes and tunable electronic properties of polar and non-polar SiC nanofilms.

The energy landscapes and electronic properties of non-polar and polar nanofilms of silicon carbide are studied using periodic hybrid density functional calculations. Relative energies and electronic properties are reported as a function of film thickness for a set of structures derived from those of wurtzite, zinc blende and graphite. The energy landscape of the films is complex with pronounced nano-polymorphism. Across polar films different mechanisms in different cases remove or reduce the dipole moment, including charge transfer and phase transitions to the graphitic-like structures which

Distributions of energy imparted to a micro-volume by secondary charged fragments of nuclear interactions in space radiation field.

Visible Light-Induced C─C Bond Cleavage for the Construction of Ketone Using Molecular Oxygen.

The selective cleavage of carbon-carbon bonds by photocatalysis with the participation of oxygen for ketone synthesis and drug derivatization is reported. In this oxidation-based ketone formation reaction, molecular oxygen complexes with the substrate, which is subsequently photoexcited to generate a charge transfer (CT) state. This CT state facilitates energy transfer with molecular oxygen, producing singlet oxygen, which facilitates hydrogen atom process and promotes the subsequent cleavage of the carbon-carbon bond, ultimately leading to the formation of aryl ketone compounds. 48 aryl keton…

Interaction between tricyclic psychopharmacons and some antibiotics.

The tricyclic psychopharmacons, e.g. clozapine, promethazine and imipramine cure plasmids and inhibit plasmid transfer among bacteria due to the inhibition of supercoiling activity on DNA gyrase. In addition an interaction was found between clozapine, imipramine, promethazine and some antibiotics, e.g. penicillins and tetracycline in vitro. The nature of interaction is based on a charge transfer complex, which is formed between clozapine, imipramine, promethazine and penicillins. Differential spectrophotometry showed that ampicillin reduced the highest energy peaks of clozapine, promethazine a

Benchmarking DFT for Chain-Length and Water Effects in Thermal Conia-Ene Cyclization.

Reliable density functional theory (DFT) methods for thermally driven cyclization reactions remain insufficiently established, particularly for systems in which proton transfer, conformational preorganization, and weak environmental effects jointly shape the energy landscape. Here, we present a systematic benchmark of DFT exchange-correlation functional/basis-set combinations against CCSD-(T) reference energies for the thermal Conia-Ene cyclization, using chain length and explicit water assistance as chemically relevant probes of functional performance. Representative reaction profiles were co

Effect of osteoporosis-related reduction in the mechanical properties of bone on the acetabular fracture during a sideways fall: A parametric finite element approach.

PURPOSE: The incidence of acetabular fractures due to low-energy falls is increasing among the geriatric population. Studies have shown that several biomechanical factors such as body configuration, impact velocity, and trochanteric soft-tissue thickness contribute to the severity and type of acetabular fracture. The effect of reduction in apparent density and elastic modulus of bone as well as other bone mechanical properties due to osteoporosis on low-energy acetabular fractures has not been investigated. METHODS: The current comprehensive finite element study aimed to study the effect of r

Erythropoietic protoporphyria: photodynamic transfer of protoporphyrin from intact erythrocytes to other cells.

Erythrocytes in patients with erythropoietic protoporphyria (EPP) contain large amounts of protoporphyrin and are regarded as the main source of protoporphyrin in this disease. Cells in the skin of EPP patients accumulate protoporphyrin released from the erythrocytes and upon sun exposure endothelial cells are photodamaged. In the present study a light-induced transfer of protoporphyrin directly from EPP erythrocytes to cultured cells is demonstrated. Erythrocytes were layered upon cultured cells and irradiated. The nearness of erythrocyte and cultured cell membranes potentiated the transfer o

IMPORTANCE: Abnormalities in neural activity and cerebral bioenergetics have been observed in schizophrenia (SZ). Further defining energy metabolism anomalies would provide crucial information about molecular mechanisms underlying SZ and may be valuable for developing novel treatment strategies. OBJECTIVE: To investigate cerebral bioenergetics in SZ via measurement of creatine kinase activity using in vivo 31P magnetization transfer spectroscopy. DESIGN, SETTING, AND PARTICIPANTS: Cross-sectional case-control study in the setting of clinical services and a brain imaging center of an academic

Monitoring Calcium Dynamics in Pig Intestinal Organoids with FLIM and FRET.

Intracellular calcium signaling plays a crucial role in intestinal epithelial function. In this chapter, we describe a method to monitor calcium dynamics in live pig small intestinal organoids using stably expressed genetically encoded FRET-based biosensor in combination with intensity-based ratiometric and fluorescence lifetime imaging (FLIM) microscopy readouts. The calcium biosensor Twitch-2B was introduced into organoids by electroporation. Despite the modest lifetime contrast, the biosensor provides a highly reliable ratiometric signal and is compatible with both conventional fluorescence

Heterogeneous amyloid β dimerization revealed by single-molecule FRET and MD simulation.

The correlation between specific amyloid fibril polymorphs and diseases suggests that characterization of heterogeneity in the aggregation process is key to understanding disease mechanisms. In this work, we use single-molecule Förster resonance energy transfer (FRET) spectroscopy and molecular dynamics (MD) simulation to quantitatively characterize dimerization of amyloid β (Aβ), the first step of the aggregation process. We find that dimerization is extremely rare, with a high dissociation constant of >16 mM, yet there are multiple dimer states with distinct FRET efficiencies and stabilities…

DNA damage induces long range changes to duplex structure-a non-protein start to damage detection?

DNA-binding proteins must quickly locate specific sites on DNA to enable replication, repair, and transcription. While sequence-specific recognition is well understood, the physical basis of structure-specific recognition remains unclear, limiting our understanding of DNA damage repair. Proteins must distinguish damaged sites within largely undamaged DNA; however, studying this is challenging due to DNA's dynamic nature. We hypothesized that DNA damage causes changes in DNA structure, signalling protein recruitment. Using confocal single-molecule FRET, we analysed five DNA duplexes containing

Steric Modulation of Protein-Mediated Nanoparticle Assembly: Controlling Cluster Size, Polydispersity, and FRET Responses by Rebalancing Short- and Long-Range Interactions.

Protocol for isolating extracellular vesicles from human or mouse airway macrophages for functional assays and in vivo or in vitro experimentation.

Macrophages are key players in host defense and lung injury, and their crosstalk with other cells dictates the course of the inflammatory response and tissue remodeling. Here, we present a protocol for isolating extracellular vesicles (EVs) from human bronchoalveolar lavage fluid (BALF) and for generating EVs from primary human or murine alveolar macrophages following stimulation by cigarette smoke extract. Furthermore, we describe how to measure matrix metalloproteinase (MMP)-12 activity on macrophage-derived EVs using a Förster resonance energy transfer assay.
