Paraisariamides (Psds) are a family of fungal N-methylated cyclic heptapeptides with structural similarity to the translation eukaryotic elongation factor 1A inhibitor ternatin. Initial characterization of the eight known Psds A-H revealed a complex biological activity profile; Psd A and Psds E-H showed differential cytotoxicity to HCT116 colon cancer cells, whereas Psds B-D were inactive. Here, we report that Psd A is a weak inhibitor of total protein synthesis and that three synthetic diastereomers of Psd A retain comparable pharmacological activity. Psd A and all structural variants decreas

Invasive fungal infections pose a significant health threat. Yarrowia lipolytica (formerly known as Candida lipolytica) is an opportunistic yeast with pathogenic potential, but its virulence and impact on the host remain poorly understood. This study aimed to investigate the infection characteristics associated with this relatively unknown pathogen by establishing reliable animal models. We developed intravenous challenge models in immunocompetent and immunosuppressed BALB/c mice by injecting fungal cells directly into the bloodstream. In immunocompetent mice, Y. lipolytica was effectively cle

Building renovations increase the concentration of Aspergillus conidia in the air. In 2010, one wing of the hospital building was imploded due to structural problems. To evaluate the impact of building implosion on the concentration of fungi in the air, the demolition was performed in two phases: mechanical demolition of 30 m of the building, followed by implosion of the wing. Patients at high risk for aspergillosis were placed in protected wards. Air sampling was performed during mechanical demolition, on the day of implosion and after implosion. Total and specific fungal concentrations were

The threat posed by microorganisms to food safety in development necessitates the discovery of new antimicrobial agents. Here, we unravel the antifungal activity of 6-methoxydihydrosanguinarine (6-ME) derived from Hylomecon japonica. 6-ME disrupted the biofilm formation of Candida albicans (C. albicans). 6-ME could cause the blockage of filamentation and classical cyclic adenosine monophosphate (cAMP) signaling pathway is involved in this process. 6-ME downregulated the expression of EFG1, CDC35, RAS1, and TPK2, all of which are critical components of the cAMP pathway. Furthermore, 6-ME change

The emergence of drug-resistant Candida species has created an urgent need for non-toxic molecules that inhibit fungal growth, biofilm development, and hyphal formation. In this study, fifty multi-halogenated indole derivatives were screened against ten Candida species, including azole-resistant C. albicans, C. auris, C. glabrata, and C. parapsilosis. Among them, 4,6-dibromoindole and 5-bromo-4-chloroindole exhibited the strongest antifungal and antibiofilm effects, with minimum inhibitory concentration (MIC) values of 10-50 µg/mL, outperforming ketoconazole and comparable to miconazole. Fungal infections are a major contributor to infectious disease-related deaths across the globe. Candida species are among the most common causes of invasive mycotic disease, with Candida albicans reigning as the leading cause of invasive candidiasis. Given that fungi are eukaryotes like their human host, the number of unique molecular targets that can be exploited for antifungal development remains limited. Currently, there are only three major classes of drugs approved for the treatment of invasive mycoses, and the efficacy of these agents is compromised by the development of drug resistance

Sporotrichosis is a neglected fungal infection caused by Sporothrix spp., which have a worldwide distribution. The standard antifungal itraconazole has been recommended as a first-line therapy. However, failure cases in human and feline treatment have been reported in recent years. This study aimed to synthesize several α- and β-2,3-dihydrofuranaphthoquinones and evaluate them against Sporothrix schenckii and Sporothrix brasiliensis-the main etiological agents of sporotrichosis in Brazil. The stability of these compounds was also investigated under different storage conditions for 3 months. Th

The Q cycle mechanism of the bc1 complex requires two quinone reaction centers, the hydroquinone oxidation (QP) and the quinone reduction (QN) center. These sites can be distinguished by the specific binding of inhibitors to either of them. A substantial body of information about the hydroquinone oxidation site has been provided by the analysis of the binding of QP site inhibitors to the bc1 complex in different redox states and to preparations depleted of lipid or protein components as well as by functional studies with mutant bc1 complexes selected for resistance toward the inhibitors. The r

Interkingdom competition between bacteria and fungi in nutrient-limited environments drives microbial community dynamics, yet the molecular mechanisms enabling bacterial dominance during bacterial-fungal interaction remain poorly characterized. This study reveals a paradigm-shifting role for bacterial type IV secretion system (T4SS) by demonstrating that a specialized bacterial-killing T4SS (T4SSBK) in Lysobacter enzymogenes serves as a universal cross-kingdom antifungal weapon. Combining genetic, biochemical, and bioinformatical approaches, we show that T4SSBK mediates direct contact-dependen

The red blood cell (RBC) membrane is unique and crucial for maintaining structural-functional relationships. Maternal smoking induces significant changes in the morphological, rheological, and functional parameters of both maternal and foetal RBCs, mainly due to the continuous generation of the free radicals. The major aim of this study was to follow the consequences of a secondary stressor, like fungal infection, on the already compromised RBC populations. The impact of Candida infection, a growing health concern, was investigated on four blood sample groups: mothers and their neonates origin

Plastics are used widely in almost every field of life, but their synthetic and persistent nature makes them harmful for the environment. The aim of this research was to evaluate the degradation abilities of Aspergillus niger, Candida albicans, and Acremonium sclerotigenum on microplastics (MPs). MP pieces of 4 ± 1 mm, including polyethylene, polyethylene terephthalate, and polystyrene, were incubated with fungal inoculums for 30 days. The degradation of treated MPs was determined by biofilm formation, weight loss, scanning electron microscopy (SEM), and Fourier transform analyses. The results

The rise of multidrug resistant fungal infections highlights the need to identify and develop novel antifungal agents. Occidiofungin is a nonribosomally synthesized glycolipopeptide that has a unique mechanism of action, disrupting actin-mediated functions and inducing cellular apoptosis. Antifungal activity has been observed in vitro against various fungal species, including multidrug resistant Candida auris, and in vivo efficacy has been demonstrated in a murine vulvovaginal candidiasis model. Occidiofungin, a cyclic glycolipopeptide, is composed of eight amino acids and in previous studies,

Currently, the spread of fungal infections is becoming an urgent problem. Fungi of the Candida genus are opportunistic microorganisms that cause superficial and life-threatening systemic candidiasis in immunocompromised patients. The list of antifungal drugs for the treatment of candidiasis is very limited, while the prevalence of resistant strains is growing rapidly. Therefore, the search for new antimycotics, including those exhibiting immunomodulatory properties, is of great importance. Plenty of natural compounds with antifungal activities may be extremely useful in solving this problem. T

Candida parapsilosis is an opportunistic fungal pathogen with increasing incidence in hospital settings worldwide; however, we lack a comprehensive understanding of the mechanisms promoting its virulence and drug resistance. Bergin et al. systematically quantify the frequency and effect of copy number variation (CNV) across 170 diverse clinical and environmental isolates of C. parapsilosis (Bergin SA, Zhao F, Ryan AP, Müller CA, Nieduszynski CA, Zhai B, Rolling T, Hohl TM, Morio F, Scully J, Wolfe KH, Butler G, 2022, mBio, https://doi.org/10.1128/mbio.01777-22). Cryptococcus neoformans is an important opportunistic human fungal pathogen that causes cryptococcosis in immunocompromised patients. However, the number of drugs for the treatment of cryptococcosis is restricted, and the development of novel antifungal drugs and innovative strategies for the treatment of cryptococcosis is urgently needed. In this study, we validated that DvAMP is a novel antimicrobial peptide with antimicrobial activity and that it was obtained by pre-screening from the UniProt database of more than three million unknown functional sequences based on the quantitative structur

Onychomycosis (OM) is a fungal infection, responsible for about 50% of nail diseases. OM has been attributed to the ability of fungi to naturally organize themselves into biofilms on nail surfaces. However, little is known about the exact role of the biofilm in the etiopathogenesis of OM, as well as its influence in the permeation of a topical treatment. The objectives of this study were to review the literature for topical OM treatments in clinical trials, assess the efficiency of these treatments, and discuss factors that could affect the success of these treatments. First, a systematic sear

Symbionts in the abdomen fat body of brown planthopper (BPH) play an important role in the growth and reproduction of their host, Nilaparvata lugens Stål (Hemiptera: Delphacidae). Thus, controlling BPH infection on rice by inhibiting symbionts with antimicrobials is feasible. However, the effect of antimicrobials on the microbiome in the fat body and the relationship between microbial community and mortality have not been fully elucidated. A decrease in the total number of yeast-like symbiotes in the fat body and elevated mortality were observed after exposure to toyocamycin, tebuconazole, and

Chiral primary amines are important intermediates in the synthesis of pharmaceutical compounds. Fungal reductive aminases (RedAms) are NADPH-dependent dehydrogenases that catalyse reductive amination of a range of ketones with short-chain primary amines supplied in an equimolar ratio to give corresponding secondary amines. Herein we describe structural and biochemical characterisation as well as synthetic applications of two RedAms from Neosartorya spp. (NfRedAm and NfisRedAm) that display a distinctive activity amongst fungal RedAms, namely a superior ability to use ammonia as the amine partn
