Amelioration of 6-OHDA-Induced Parkinson's Symptoms in Zebrafish Larvae by an Almond Skin Acetonic Extract.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by mitochondrial dysfunction, oxidative stress, and apoptosis. Natural products rich in polyphenols have been investigated for their potential to modulate pathways associated with PD-related pathology. The present study evaluated the effects of an acetonic almond skin extract, an agri-food by-product, in a zebrafish (Danio rerio) larval model of PD induced by 6-hydroxydopamine (6-OHDA). Embryos were exposed to 250 µM 6-OHDA alone or in combination with the extract (5 and 25 µg/mL) from 48 to 120 h post-fertilization (hpf).

Neuroprotective Effects of Thymol-Loaded Selenium Nanoparticles against 6-OHDA-Induced Apoptosis and Oxidative Stress in an In Vitro Parkinson's Disease Model.

INTRODUCTION: Parkinson's disease (PD) is characterized by the degeneration of dopaminergic neurons within the substantia nigra, leading to progressive motor dysfunction. There are still limited diseasemodifying options that counteract the process of disease progression. This study aimed to evaluate the neuroprotective effects of thymol, both in its free form and when loaded onto selenium nanoparticles (SeNPs), in a 6-hydroxydopamine (6-OHDA)-induced PD model using SH-SY5Y cells. METHODS: SeNPs were synthesized using a chemical reduction method with ascorbic acid, achieving a 68% entrapment e

Olfactory bulb 6-OHDA neurotoxicity as a model of early cortical dysfunction.

The olfactory bulb (OB) is one of the earliest brain regions affected in neurodegenerative diseases such as Parkinson's disease (PD). Its high metabolic rate, dopaminergic modulation, and connectivity with the cortical and limbic regions make it particularly vulnerable to early neuroinflammatory and oxidative processes. This study aimed to investigate whether the administration of 6-hydroxydopamine (6-OHDA) into the OB induces behavioral, redox, and inflammatory alterations associated with early cortical disturbances. Male Wistar rats were randomly assigned to the sham or 6-OHDA groups and und

High-mobility Group Protein 1/ Receptor for Advanced Glycation End Products/ Nuclear Factor-κB Signalling Pathway Contributes to the Pathogenic Process of Striatal Neuron Impairment in the Rat Model of Parkinson's Disease.

BACKGROUND: High-mobility group protein 1 (HMGB1) is a ligand known to bind to the receptor for advanced glycation end products (RAGE), and it can activate nuclear factor-κB (NF-κB) to mediate cellular damage. RAGE and Parkinson's disease (PD) are closely associated, but it remains unclear whether the HMGB1/RAGE/NF-κB signaling pathway contributes to the pathophysiology of PD. METHODS: PD was induced by administration of 6-hydroxydopamine (6-OHDA), while RAGE was inhibited using an inhibitor, FPS-ZM1. The grip strength test and Morris water maze were used to evaluate sensorimotor and memory s

Targeting oxidative stress and mitochondrial dysfunction via URG7 overexpression in an in vitro Parkinson's disease neuronal model.

Neurodegenerative diseases represent a major healthcare challenge for aging populations, characterized by the progressive decline of neuronal function and subsequent brain atrophy. Despite differences in the brain regions affected and the underlying causes, these pathologies share common cellular and molecular mechanisms, including oxidative stress and mitochondrial dysfunction. Among neurodegenerative disorders, Parkinson’s disease (PD) is particularly associated with oxidative stress–driven dopaminergic neurodegeneration, in which reactive oxygen species (ROS) play a crucial role by contribu

TTP protects against 6-OHDA-induced dopaminergic neurodegeneration via astrocytic Nrf2/HO-1 activation in cellular and rat models of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neuronal loss driven by oxidative stress, mitochondrial dysfunction, and apoptosis. Adenosine A2A receptor (A2AR) antagonism is clinically validated for symptomatic relief; however, its potential disease-modifying mechanisms remain incompletely defined. We hypothesized that TTP, a novel and highly selective A2AR antagonist, would confer neuroprotection against 6-hydroxydopamine (6-OHDA)-induced toxicity by modulating oxidative stress and activating astrocytic Nrf2/HO-1 antioxidant signaling. In primary midbrain dopaminergic

Effects of 6-OHDA-Mediated Chemical Denervation of Sympathetic Axons in Inguinal Subcutaneous White Adipose Tissue.

AIMS: White adipose tissue (WAT) innervation by sensory and sympathetic axons contributes to tissue functions, but the consequences of selectively disrupting sympathetic input to WAT remain incompletely defined. MATERIALS AND METHODS: We utilised 6-hydroxydopamine (6-OHDA) as a validated approach to achieve bilateral sympathetic denervation of mouse inguinal subcutaneous WAT (ing-scWAT) and investigated impacts on adipose and whole-body metabolism. RESULTS: Sympathetic denervation did not impact body weight or adipose mass by 16 days post-denervation but significantly altered energy expendit

Efficacy and safety of needle-free jet injection of self-crosslinking hyaluronic acid for facial fine wrinkles and skin texture: 12-week clinical outcomes in Asian women.

INTRODUCTION: Facial aging involves progressive deterioration of the superficial dermal matrix, leading to fine wrinkles, surface irregularity, and reduced skin elasticity. Conventional hyaluronic acid (HA) formulations vary in their ability to achieve uniform superficial dermal dispersion and sustained tissue effects, and needle injections may lead to inconsistent depth and localized pooling. This study evaluated changes in facial skin texture and fine wrinkles following delivery of self-crosslinking HA formulation containing N-hyaluronoyl 5-hydroxydopamine through a needle-free jet injection

Matsupexole: A novel nonergot dopamine receptor agonist with sustained efficacy in a rat model of Parkinson's disease and limited off-target activity.

BACKGROUND AND PURPOSE: Dopamine receptor agonists, particularly targeting the dopamine D2L receptor (D2LR), have been used to treat Parkinson's disease (PD). However, valvular heart disease and somnolence, mainly caused by activating the serotonin 5-HT2B receptor (5-HT2BR) and dopamine D3 receptor (D3R), respectively, currently challenge their clinical use. Here, we aimed to develop a novel dopamine receptor agonist with superior therapeutic efficacy for PD, while minimising adverse reactions. EXPERIMENTAL APPROACH: Matsupexole, a novel nonergot dopamine receptor agonist, was evaluated by ag

Multiple stimulations of chemoluminescent-expressing mesenchymal-derived dopaminergic transplants safely reinforce limb-use control restoration during swimming in a unilateral Parkinson's disease model.

In Parkinson's disease (PD), dopaminergic (DA) neurons deteriorate within the substantia nigra pars compacta (SNc), causing movement difficulties. This deterioration can be modeled in rats given large, unilateral DA-depleting lesions of the nigrostriatal pathway using the neurotoxin 6-hydroxydopamine. The behavioral benefits promoted by classic L-DOPA pharmacotherapy typically do not include skilled limb use restoration. Among deteriorative diseases, the single neurotransmitter loss in PD allows for a more discrete or focused replacement goal. Stem cell therapy (SCT) would be more likely to he

Combined Vitamin D and Resveratrol Treatment Attenuates Motor Deficits and Neurodegeneration in a 6-OHDA Mouse Model of Parkinson's Disease in Male Mice: A Focus on Sirt1 and Lingo-1 Pathways in the Ventral Tegmental Area.

BACKGROUND: Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration, leading to severe motor impairments. Lingo-1 negatively regulates neuronal survival, while Sirtuin-1 (Sirt1) supports mitochondrial function and oxidative stress resistance. Given the neuroprotective properties of Vitamin D3 (VitD3) and resveratrol, this study examined whether their combined administration could modulate Sirt1 and Lingo-1 expression and attenuate neurodegeneration in a 6-hydroxydopamine (6-OHDA) model of PD. METHODS: Male Swiss mice were assigned to six groups: Control, Sham,

Therapeutic effect of formononetin in 6-OHDA induced Parkinson disease in rats.

Preclinical models of Parkinson's disease (PD) have been developed using intracerebroventricular (i.c.v.) injection of 6-hydroxydopamine (6-OHDA) to induce neurodegeneration and motor dysfunction. Formononetin, a phytoestrogen with known anti-aging and anti-apoptotic properties, was investigated for its potential neuroprotective effects against 6-OHDA-induced toxicity. In this study, the rats received a single i.c.v. injection of 6-OHDA and were then treated with formononetin at doses of 25, 50, and 100 mg/kg orally for 21 days. Motor coordination, grip strength, and gait were evaluated using

Effects and Mechanisms of Melatonin Receptor Agonist and Antagonist on Disease Progression in a 6-OHDA-Induced Parkinson's Disease Rat Model.

BACKGROUND: Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration. Melatonin (MLT) is implicated in neuroprotection, yet the effects of modulating its receptors remain unclear. This study investigated the impact of the MLT receptor agonist agomelatine (AG) and antagonist luzindole (LU) on motor behavior, serum MLT levels, and dopaminergic neuron survival in a 6-hydroxydopamine (6-OHDA) rat model of PD. METHODS: A PD model was induced by stereotaxic injection of 6-OHDA into the medial forebrain bundle. Rats received intraperitoneal AG or LU for 2 or 4 weeks. M

A model of L-DOPA-induced dyskinesia in parkinsonian mice produced by AAV vector-mediated overexpression of α-synuclein.

L-3,4-dihydroxyphenylalanin (L-DOPA) is the most effective drug for treating Parkinson's disease (PD); however, long-term L-DOPA therapy can lead to L-DOPA-induced dyskinesia (LID). While the 6-hydroxydopamine-lesioned rodent model for LID fails to reproduce the pathological hallmarks of PD, a newly introduced rodent model using adeno-associated virus (AAV)-mediated overexpression of α-synuclein results in α-synuclein aggregation and progressive loss of dopaminergic neurons. The present study aimed to provoke LID in parkinsonian mice generated by AAV vector-mediated overexpression of α-synucle

Cannabidiol improves L-DOPA-induced dyskinesia and modulates neuroinflammation and the endocannabinoid, endovanilloid and nitrergic systems.

Despite the widespread use of L-3,4-dihydroxyphenylalanine (L-DOPA) as the gold standard for dopamine (DA) replacement in Parkinson's Disease (PD), its prolonged administration frequently leads to L-DOPA-induced dyskinesia (LID), a significant therapeutic challenge. Modulating the endocannabinoid system has emerged as a promising approach for managing LID. This study explored whether cannabidiol (CBD), a non-psychoactive compound of Cannabis sativa, and PECS-101, a fluorinated derivative of CBD, could mitigate the onset and progression of LID. We used unilateral 6-hydroxydopamine-lesioned rats

Cryogel microcarriers loaded with glial cell line-derived neurotrophic factor enhance the engraftment of primary dopaminergic neurons in a rat model of Parkinson's disease.

Objective.Cryogel microcarriers made of poly(ethylene glycol) diacrylate and 3-sulfopropyl acrylate have the potential to act as delivery vehicles for long-term retention of neurotrophic factors (NTFs) in the brain. In addition, they can potentially enhance stem cell-derived dopaminergic (DAergic) cell replacement strategies for Parkinson's disease (PD), by addressing the limitations of variable survival and poor differentiation of the transplanted precursors due to neurotrophic deprivation post-transplantation in the brain. In this context, to develop a proof-of-concept, the aim of this study

Decrease of KATP channel expression through D3 receptor-mediated GSK3β signaling alleviates levodopa-induced dyskinesia (LID) in Parkinson's disease mouse model.

AIMS: The standard Parkinson's disease (PD) treatment is L-3,4-dihydroxyphenylalanine (L-DOPA); however, its long-term use may cause L-DOPA-induced dyskinesia (LID). Aberrant activation of medium spiny neurons (MSNs) contributes to LID, and MSN excitability is regulated by dopamine D3 receptor (D3R) and ATP-sensitive potassium (KATP) channel activity. Nevertheless, it remains unclear if D3R and KATP channels may be linked in the context of LID. METHODS: Wild-type and tyrosine hydroxylase (TH)-specific Kir6.2 knockout mice were injected with 6-hydroxydopamine (6-OHDA) to generate a PD mouse mo
