This study aimed to identify kiwi kefir-like beverages with high levels of viable probiotic cells and low levels of calories, acids, and alcohol. To achieve this, microbiological and chemical characterizations were conducted on beverages inoculated with varying amounts of kefir grains (GW) and incubated at different agitation speeds (A), following a second-order orthogonal factorial design. For each experimental condition, three 24-h batch cultures were performed using three successive passages of kefir grains. Higher GW levels promoted greater nutrient consumption and metabolite production. H

Hard kombucha is an alcoholic beverage produced by the fermentation of a 40% (v/v) kombucha solution with selected yeast strains and sugar. Among 35 yeast strains examined, Lachancea thermotolerans, known for its high ethanol production, was selected to prepare hard kombucha using tap and deep ocean water. After 21 days of fermentation, the pH of the filtered tap water hard kombucha (TWHK) and deep ocean water hard kombucha (DOWHK) decreased to 2.6-2.69. In contrast, their titratable acidity (TA) increased to 93.68 g acetic acid/L and 107.73 g acetic acid/L, respectively. The ethanol content r

The global demand for premium-quality coffee is growing, as more consumers appreciate its distinct sensory attributes. Fermentation plays a key role in producing natural (dry-processed) coffees with distinct and appealing flavors and aromas. However, improper fermentation conditions can lead to defects that negatively impact the quality of the coffee. This research aimed to compare various fermentation methods for producing natural coffees and assess their impact on both sensory attributes and chemical composition. Five fermentation processes were evaluated: exposure to air for 24 and 48 h, fe

Salvia miltiorrhiza Bge. (S. miltiorrhiza) is an important herbal medicine, and the researches mainly focused on the roots of S. miltiorrhiza rather than the above-ground parts. Therefore, the detailed research on the above-ground parts of S. miltiorrhiza is still unclear. This study aimed to elucidate the constituents of the above-ground parts of S. miltiorrhiza extract (SME) and evaluate its effect on endothelial cell injury. For this purpose, the constituents of SME were identified by ultra performance liquid chromatography tandem mass spectrometry, and 41 compounds were determined, includi

Rice (Oryza sativa L.) endosperm accumulates huge amounts of starch. Rice starch is highly digestible, potentially enhancing the occurrence of blood sugar- and intestine-related diseases such as type 2 diabetes. Resistant starch (RS) is hardly digestible in small intestine but can be converted into beneficial short-chain fatty acids in large intestine, potentially reducing the incidence of these diseases. However, it is still difficult to produce a high RS rice variety. Here, we report that simultaneous deficiency of soluble starch synthases IIa and IIIa confers high RS content in rice endospe

Bacillus megaterium is particularly known for its abundance in soils and its plant growth promotion. To characterize the metabolites excreted by this specie, we performed successive liquid/liquid extractions from bacteria culture medium with different polarity solvents (cyclohexane, dichloromethane, ethyl acetate and butanol) to separate the metabolites in different polarity groups. The extracts were characterized regarding their total phenolic content, the amount of reducing sugar, the concentration of primary amines and proteins, their chromatographic profile by HPLC-DAD-ELSD and their chemi

In this study, Fe conversion during hydrothermal carbonization (HTC) of coking sludge were investigated, and the effect mechanism of Fe component on the adsorption performance of coking sludge hydrochar (CHC) was explored. The results showed that after HTC treatment, more than 95 % of Fe remained in the CHC. Fe3+ was reduced to Fe2+ by sugar and amino acids. Fe was stabilized during the HTC process and was still predominantly in the Fe manganese oxidation state. The CHC prepared at 270 °C exhibited excellent adsorption capacities for Congo red (CR), tetracycline (TC), and Cr (VI). Their maximu

Antisense oligonucleotides (ASOs) are single-stranded nucleic acid strings that can be used to selectively modify protein synthesis by binding complementary (pre-)mRNA sequences. By specific arrangements of DNA and RNA into a chain of nucleic acids and additional modifications of the backbone, sugar, and base, the specificity and functionality of the designed ASOs can be adjusted. Thereby cellular uptake, toxicity, and nuclease resistance, as well as binding affinity and specificity to its target (pre-)mRNA, can be modified. Several neurodegenerative diseases are caused by autosomal dominant t

Numerous lectins act as pattern recognition receptors (PRRs) in the innate immune system of invertebrates. Here, a galectin (FmGal) was isolated from hemocytes of Fenneropenaeus merguiensis. FmGal contained one open reading frame encoding a peptide of 338 amino acids. The primary sequence of FmGal comprised a carbohydrate recognition domain with a specific galactose binding site. The FmGal transcripts were found mostly in hemocytes of healthy shrimp. The expression of FmGal was up-regulated upon challenge with Vibrio parahaemolyticus and white spot syndrome virus (WSSV). Gene-silencing with Fm

Nucleos(t)ide analogues (NA) belong to a family of compounds widely used in anticancer/antiviral treatments. They generally exhibit a cell toxicity limited by cellular uptake levels and the resulting nucleos(t)ides metabolism modifications, interfering with the cell machinery for nucleic acids synthesis. We previously synthesized purine nucleos(t)ide analogues N7-coordinated to a platinum centre with unaltered sugar moieties of the type: [Pt(dien)(N7-dGuo)]2+ (1; dien = diethylenetriamine; dGuo = 2'-deoxy-guanosine), [Pt(dien)(N7-dGMP)] (2; dGMP = 5'-(2'-deoxy)-guanosine monophosphate), and [P

The retained placenta is a common pathology of dairy cows. It is associated with a significant drop in the dry matter intake, milk yield, and increased susceptibility of dairy cows to metritis, mastitis, and displaced abomasum. The objective of this study was to identify metabolic alterations that precede and are associated with the disease occurrence. Blood samples were collected from 100 dairy cows at -8 and -4 weeks prior to parturition and on the day of retained placenta, and only 16 healthy cows and 6 cows affected by retained placenta were selected to measure serum polar metabolites by a

Reduced nutrient intake is a widely conserved manifestation of sickness behavior with poorly characterized effects on adaptive immune responses. During infectious challenges, naive T cells encountering their cognate antigen become activated and differentiate into highly proliferative effector T cells. Despite their evident metabolic shift upon activation, it remains unclear how effector T cells respond to changes in nutrient availability in vivo. Here, we show that spontaneous or imposed feeding reduction during infection decreases the numbers of splenic lymphocytes. Effector T cells showed ce

The naturally occurring isotope of hydrogen ((1)H), deuterium ((2)H), could have an important biological role. Deuterium depleted water delays tumor progression in mice, dogs, cats and humans. Hydratase enzymes of the tricarboxylic acid (TCA) cycle control cell growth and deplete deuterium from redox cofactors, fatty acids and DNA, which undergo hydride ion and hydrogen atom transfer reactions. A model is proposed that emphasizes the terminal complex of mitochondrial electron transport chain reducing molecular oxygen to deuterium depleted water (DDW); this affects gluconeogenesis as well as fa

Cocoa (Theobroma cacao L., Sterculiaceae) is a widely consumed food ingredient. Although typically found in high-fat, high-sugar foods such as chocolate, cocoa is rich in polyphenols, methylxanthines, and monounsaturated fatty acids. There is increasing evidence that moderate consumption of cocoa and cocoa-containing foods may have beneficial effects on the health including vasodilatory, antioxidant, and anti-inflammatory effects. Polyphenols in cocoa, including monomeric flavanols, as well as polymeric proanthocyanidins, may play a role in these observed beneficial effects. Chronic inflammati

Campylobacter jejuni is a major foodborne pathogen of animal origin and a leading cause of bacterial gastroenteritis in humans. During the past decade, especially since the publication of the first C. jejuni genome sequence, major advances have been made in understanding the pathobiology and physiology of this organism. It is apparent that C. jejuni utilizes sophisticated mechanisms for effective colonization of the intestinal tracts in various animal species. Although Campylobacter is fragile in the environment and requires fastidious growth conditions, it exhibits great flexibility in the ad

This study addresses the histomorphology, the histochemistry and the distribution of glycoconjugates (GCs) in the mucosa of the digestive tract of the weakfish Cynoscion guatucupa. The histological characterization of the buccopharyngeal cavity and the esophagus revealed that they are lined by a stratified epithelium where the mucosa is thrown into longitudinal folds. The stomach is lined with a simple columnar secretory epithelium with tubular glands. GCs were analyzed using a range of histochemical methods. They were identified as oxidizable vicinal diols; sialic acids and some of their chai

Listeria monocytogenes is a Gram-positive intracellular parasite and the causative organism of human listeriosis. In this article we demonstrate that L. monocytogenes encodes a functional member of the CodY family of global regulatory proteins that is responsive to both GTP and branched chain amino acids. By transcript analyses we identified the CodY regulon in L. monocytogenes and demonstrated that it comprises genes involved in amino acid metabolism, nitrogen assimilation as well as genes involved in sugar uptake and incorporation, indicating a role for CodY in L. monocytogenes in both carbo
