A prevascularized tissue engineering chamber supports growth and function of islets and progenitor cells in diabetic mice.

BACKGROUND: In this study, a natural compound quercetin (Qu) was investigated for its various antitumor effects. However, due to its poor water solubility and low bioavailability, its clinical application is limited. To overcome this constraint, a modification was to Qu, which resulted in the creation of novel flavonoid self-assembling nanoparticles (HCQ NPs). METHODS: HCQ NPs were synthesized by a self-assembly method and characterized using transmission electron microscopy, the Malvern Zetasizer instrument, X-ray photoelectron spectroscopy (XPS), the ultraviolet-visible spectrophotometric m

Progress in developing a living human tissue-engineered tri-leaflet heart valve assembled from tissue produced by the self-assembly approach.

The aortic heart valve is constantly subjected to pulsatile flow and pressure gradients which, associated with cardiovascular risk factors and abnormal hemodynamics (i.e. altered wall shear stress), can cause stenosis and calcification of the leaflets and result in valve malfunction and impaired circulation. Available options for valve replacement include homograft, allogenic or xenogenic graft as well as the implantation of a mechanical valve. A tissue-engineered heart valve containing living autologous cells would represent an alternative option, particularly for pediatric patients, but stil

PRP-driven biomimetic liver tissue engineering: A cost-effective platform for high-efficiency expansion of mouse primary hepatocytes.

Three-dimensional (3D) scaffold-based culture systems offer a promising approach for expanding primary hepatocytes in vitro, potentially overcoming donor shortages in liver failure treatment. In this study, we designed a multifunctional Alg1SBC scaffold that actively loads and activates platelet-rich plasma (PRP) as a bioinspired platform. This system effectively combines low-cost PRP with a dual-network matrix composed of alginate (Alg) and sulfonated bacterial cellulose (SBC). The negatively charged surface of the Alg1SBC scaffold mimics the electrostatic characteristics of hepatic sinusoids

Tenoxicam-loaded bioglass/chitosan composites for bone tissue engineering: in vitro characterization, sustained drug release, and antimicrobial activity.

The development of multifunctional biomaterials that combine structural support with therapeutic functions is a key challenge in bone tissue engineering. Bioglass and chitosan are widely studied for their bioactivity and biocompatibility, while non-steroidal anti-inflammatory drugs (NSAIDs) such as Tenoxicam provide pain relief and inflammation control. We synthesized novel Tenoxicam-loaded Bioglass/Chitosan (TXC/BG/Cs) composites via the sol–gel process, incorporating 1, 2, and 3 wt% drug concentrations. The composites were characterized by FTIR, XRD, SEM, and EDX before and after immersion i…

Fabrication of a calcium cement-incorporated composite hydrogel scaffold via extrusion-based 3D bioprinting for bone tissue engineering.

A composite hydrogel bio-ink designated CuraGel was developed for extrusion-based 3D bioprinting of bone tissue engineering scaffolds. The formulation comprised gelatin (3% w/v), guar gum (1% w/v), sodium alginate (4% w/v), and β-tricalcium phosphate (β-TCP; 1% w/v) in 10 mL distilled water. Guar gum was incorporated as a rheological modifier to enhance viscosity, shape fidelity, and water retention during the printing process. Post-printing ionic crosslinking in 2% calcium chloride (CaCl2) for 3 min yielded structurally stable scaffolds with well-defined porous architectures. Swelling kinetic…

Tissue engineering strategies for annulus fibrosus repair: a scoping review of repair methods, animal models, and evaluation techniques.

Background. intervertebral disc (IVD) degeneration is a common chronic disease and may lead to disc herniation. Microdiscectomy removes the herniated tissue, but cannot address altered IVD biomechanics or restore annulus fibrosus (AF) structural integrity. With the development of tissue engineering, AF repair has shown potential in preclinical studies. However, these studies have not been reviewed from a practical experimental perspective, particularly focusing on animal models and evaluation methods. Therefore, this review aims to summarise and compare tissue-engineered AF repair strategies f…

Fabrication and evaluation of pectin-gelatin skin tissue engineering scaffolds containing amoxicillin trihydrate-loaded UiO-66-NH2 nanoparticles.

In this study, a novel nanocomposite scaffold comprising pectin, gelatin, and amoxicillin trihydrate-loaded UiO-66-NH2 metal-organic frameworks (UiO-66-NH2@AMX) was fabricated for skin tissue engineering applications. UiO-66-NH2 nanoparticles were synthesized hydrothermally and subsequently loaded with amoxicillin trihydrate. Finally, UiO-66-NH2@AMX nanoparticles were incorporated into pectin-gelatin scaffolds using the freeze-drying method. FE-SEM imaging confirmed the octahedral morphology of the nanoparticles before and after drug loading. FTIR analysis demonstrated successful drug incorpor…

Bioprinting and bioinks are two of the game changers in bone tissue engineering. This review presents different bioprinting technologies including extrusion-based, inkjet-based, laser-assisted, light-based, and hybrid technologies with their own strengths and weaknesses. This review will aid researchers in the selection and assessment of the bioink; the discussion ranges from commercially available bioinks to custom lab-made formulations mainly based on natural polymers, such as agarose, alginate, gelatin, collagen, and chitosan, designed for bone tissue engineering. The review is centered on …

Hydrogel Based on Riclin Cross-Linked with Polyethylene Glycol Diglycidyl Ether as a Soft Filler for Tissue Engineering.

Hydrogels composed of natural polysaccharides have been widely used as filling materials, with a growing interest in medical cosmetology and skin care. However, conventional commercial dermal fillers still have limitations, particularly in terms of mechanical performance and durability in vivo. In this study, a novel injectable and implantable hydrogel with adjustable characteristics was prepared from succinoglycan riclin by introducing PEG diglycidyl ether as a cross-linker. FTIR spectra confirmed the cross-linking reaction. The riclin hydrogels exhibited shear-thinning behavior, excellent me

Lignin-cellulose complexes derived from agricultural wastes for combined antibacterial and tissue engineering scaffolds for cutaneous leishmaniasis wounds.

Bacterial infections in wounds significantly impair the healing process. The use of natural antibacterial products over synthetic antibiotics has emerged as a new trend to address antimicrobial resistance. An ideal tissue engineering scaffold to treat infected wounds should possess antibacterial properties, while simultaneously promoting tissue regrowth. Synthesis of hydrogel scaffolds with antibacterial properties using hemp shive (HT1/HT2) lignin, sugarcane bagasse (SCB) lignin and cellulose was carried out. All lignin samples had low molecular weights and were constituted of G-type β-5 dime…

Scaffold-based tissue engineering: Supercritical carbon dioxide as an alternative method for decellularization and sterilization of dense materials.

Development of ready-to-use biomaterials and scaffolds is vital for further advancement of scaffold-based tissue engineering in clinical practice. Scaffolds need to mimic 3D ultrastructure, have adequate mechanical strength, are biocompatible, non-immunogenic and need to promote tissue regeneration in vivo. Although decellularization of native tissues seems promising to deliver scaffolds that meet these criteria, adequate decellularization of hard, poorly penetrable and poorly diffusible tissues remains challenging whilst being a very time-consuming process. In this study, a method to decellul…
