Predictive biomarkers and tumor microenvironment in female genital melanomas: a multi-institutional study of 55 cases.

Female genital melanomas are rare. At diagnosis, most affected patients have advanced disease. Surgery remains the primary treatment, and adjuvant therapy is largely ineffective. Recently, immune checkpoints and the mitogen-activated protein kinase pathway have been explored as treatment targets. However, evaluation of these biomarkers in genital melanomas is limited. We evaluated the clinicopathological features of 20 vulvar, 32 vaginal, and three cervical melanomas and assessed programmed cell death ligand 1 (PD-L1) expression, CD8 tumor-infiltrating lymphocyte density, mismatch repair prote

Intraperitoneal oxaliplatin administration inhibits the tumor immunosuppressive microenvironment in an abdominal implantation model of colon cancer.

Recent studies have demonstrated that some chemotherapeutic drugs can enhance antitumor immunity by eliminating and inactivating immunosuppressive cells. Oxaliplatin (OXP) induces immunogenic cell death by increasing the immunogenicity of cancer cells. However, the effects of OXP on the tumor immunosuppressive microenvironment remain unclear. The aim of the present study was to evaluate the antitumor activity of OXP by intraperitoneal (i.p.) administration in an abdominal implantation model of colon cancer and tested the tumor immune microenvironment to observe whether OXP affects the local im

Molecular and Histopathological Characterization of the Tumor Immune Microenvironment in Advanced Stage of Malignant Pleural Mesothelioma.

INTRODUCTION: Malignant pleural mesothelioma (MPM) is a rare, highly aggressive, and relatively chemoresistant and radioresistant malignancy with limited therapeutic options. Our objective was to investigate the prevalence of programmed death ligand 1 (PD-L1) and the characteristics of the immune environment in this disease. METHODS: A total of 99 archival tumors from advanced-stage MPM were immunohistochemically tested in parallel for PD-L1 in two different laboratories, and 87 of them were profiled for immune gene expression by NanoString analysis for 800 genes. A prior study on the same sa

Tumor Microenvironment Modulation by Cyclopamine Improved Photothermal Therapy of Biomimetic Gold Nanorods for Pancreatic Ductal Adenocarcinomas.

Due to the rich stroma content and poor blood perfusion, pancreatic ductal adenocarcinoma (PDA) is a tough cancer that can hardly be effectively treated by chemotherapeutic drugs. Tumor microenvironment modulation or advanced design of nanomedicine to achieve better therapeutic benefits for PDA treatment was widely advocated by many reviews. In the present study, a new photothermal therapy strategy of PDA was developed by combination of tumor microenvironment modulation and advanced design of biomimetic gold nanorods. On one hand, biomimetic gold nanorods were developed by coating gold nanorod…

Microenvironment Tumor Metabolic Interactions Highlighted by qMSI: Application to the Tryptophan-Kynurenine Pathway in Immuno-Oncology.

Inhibition of NK and effector T-cell functions and activation of regulatory cell populations are the main immunosuppressive effects of indoleamine-2,3-dioxygenase1 (IDO1). By converting tryptophan (Trp) into kynurenine (Kyn), IDO1 is involved in the immune response homeostasis, and its dysregulated expression is described in immune-related pathologies, as tumors that hijack it to evade immune destruction. Thereby, IDO1 inhibitors are being developed to stimulate antitumor immune responses. Existing and standard quantitation methods of IDO1 substrate and metabolite(s) are based on the total lev

Development of a prosaposin-derived therapeutic cyclic peptide that targets ovarian cancer via the tumor microenvironment.

The vast majority of ovarian cancer-related deaths are caused by metastatic dissemination of tumor cells, resulting in subsequent organ failure. However, despite our increased understanding of the physiological processes involved in tumor metastasis, there are no clinically approved drugs that have made a major impact in increasing the overall survival of patients with advanced, metastatic ovarian cancer. We identified prosaposin (psap) as a potent inhibitor of tumor metastasis, which acts via stimulation of p53 and the antitumorigenic protein thrombospondin-1 (TSP-1) in bone marrow-derived ce

SH3-domain binding protein 1 in the tumor microenvironment promotes hepatocellular carcinoma metastasis through WAVE2 pathway.

SH3-domain binding protein-1 (SH3BP1) specifically inactivating Rac1 and its target WAVE2 is required for cell motility. The present study shows SH3BP1 expression patterns in human HCC tissues and cell lines were examined. The regulation of SH3BP1 on HCC cell migration and invasion related to Rac1-WAVE2 signaling was characterized using in vitro and in vivo models. SH3BP1 overexpressed in HCC tissues and highly metastatic HCC cells was significantly associated vascular invasion (VI). SH3BP1 promoted VEGF secretion via Rac1-WAVE2 signaling, so as to exert an augmentation on cell invasion and mi

Microenvironment of the murine mammary carcinoma 4T1: endogenous IFN-gamma affects tumor phenotype, growth, and metastasis.

IFN-gamma has a profound influence on growth and metastasis of solid tumors. This is true for the murine mammary carcinoma 4T1 which grows faster and metastasizes much more readily when transplanted into the mammary fatpads of IFN-gamma(-/-) mice. We were interested in determining which infiltrating hematopoietic cells produce IFN-gamma within the 4T1 tumor microenvironment. 4T1 tumors were infiltrated with progressively increasing numbers of F4/80(+)/CD11c(+) myeloid cells, many of which were also Gr-1(+), and Gr-1(+)/CD11b(+) granulocytes. Only small numbers of CD4 T cells, CD8 T cells, NK c…

FATP5 loss in hepatocellular carcinoma cells orchestrates tumor-promoting microenvironment by inducing passive long-chain fatty acid accumulation within tumor-associated endothelial cells.

Hepatocellular carcinoma (HCC) is characterized by severe immunosuppression and metabolic reprogramming, yet the spatial dynamics of lipid flux between malignant and stromal cells remain poorly understood. Here, we identify a profound, type-specific metabolic rewiring in HCC driven by the widespread downregulation of fatty acid transport protein 5 (FATP5). FATP5 deficiency in malignant cells selectively attenuates the uptake of long-chain fatty acids (LCFAs), creating an extracellular lipid surplus that is passively sequestered by neighboring FATP3-expressing tumor-associated endothelial cells

Multi-omics study on tumor-associated macrophages remodeling the tumor microenvironment via the CXCL5-CXCR2 axis to drive immune escape in bladder cancer.

BACKGROUND: PD-1/L1 inhibitors improve the prognosis of patients with advanced bladder cancer, but the clinical remission rate remains below 25%. Tumor-associated macrophages (TAMs) and chemokines are critical in the tumor microenvironment (TME), affecting tumor progression, immunotherapeutic efficacy, and patient prognosis; however, their underlying mechanisms remain unclear. This study exhibits innovation by adopting a tumor microenvironment perspective to investigate the interaction mechanism between bladder cancer cells and tumor-associated macrophages, as well as factors affecting the eff…

FOXP3 and CXCR4-positive regulatory T cells in the tumor stroma as indicators of tumor immunity in the conjunctival squamous cell carcinoma microenvironment.

Conjunctival squamous cell carcinoma (SCC) is the most common ocular surface neoplasia. The purpose of this retrospective study was to examine the role of regulatory T cell (Treg) activity in tumor immunity and investigate the tumor microenvironment as a new treatment focus in conjunctival SCC. Cancer progression gene array and immunohistochemical analyses of FOXP3 as a Treg marker, CD8 as a tumor-infiltrating lymphocyte marker, and CXCR4 expression on activated Tregs were conducted in a series of 31 conjunctival SCC cases. The objective was to investigate the immunoreactive response in tumor …

Intratumoral Canine Distemper Virus Infection Inhibits Tumor Growth by Modulation of the Tumor Microenvironment in a Murine Xenograft Model of Canine Histiocytic Sarcoma.

Histiocytic sarcomas refer to highly aggressive tumors with a poor prognosis that respond poorly to conventional treatment approaches. Oncolytic viruses, which have gained significant traction as a cancer therapy in recent decades, represent a promising option for treating histiocytic sarcomas through their replication and/or by modulating the tumor microenvironment. The live attenuated canine distemper virus (CDV) vaccine strain Onderstepoort represents an attractive candidate for oncolytic viral therapy. In the present study, oncolytic virotherapy with CDV was used to investigate the impact …

Carbamoylphosphonates control tumor cell proliferation and dissemination by simultaneously inhibiting carbonic anhydrase IX and matrix metalloproteinase-2. Toward nontoxic chemotherapy targeting tumor microenvironment.

Carbamoylphosphonates (CPOs) have been identified as inhibitors of matrix metalloproteinases (MMPs) and as orally active, bioavailable, and safe antimetastatic agents. In this article, we focus on the direct antitumor activity of the CPOs. We discovered that CPOs also inhibit carbonic anhydrases (CAs), especially the IX and XII isoforms identified as cancer promoting factors. Thus, CPOs can be regarded as novel nontoxic drug candidates for tumor microenvironment targeted chemotherapy acting by two synergistic mechanisms, namely, inhibiting CAs and MMPs simultaneously. We have also demonstrated…

PFKP in malignant tumors: bridging metabolic reprogramming, post-translational modifications, and the immunosuppressive microenvironment.

Metabolic reprogramming is a hallmark of malignant tumors. Platelet-type phosphofructokinase (PFKP), a rate-limiting enzyme in glycolysis, is aberrantly overexpressed across diverse cancers and promotes tumor proliferation, invasion, and metastasis through both metabolic and non-metabolic pathways. The enzymatic activity, stability, and subcellular localization of PFKP are dynamically regulated by post-translational modifications-including phosphorylation, ubiquitination, acetylation, and lactylation-as well as by protein-protein interaction networks. Upon nuclear translocation, PFKP dissociat

Synthetic inducible bacteria normalize the microenvironment of refractory tumors, potentiating checkpoint inhibitor responses.

Immunotherapy has transformed cancer treatment but remains ineffective in many solid tumors, largely due to the immunosuppressive tumor microenvironment (TME). A dense/stiff extracellular matrix (ECM) can hinder immune cell infiltration, limiting therapeutic success. To overcome this challenge, we developed a live biotherapeutic platform using engineered programmable bacteria that offer advantages for delivering enzymes that normalize the TME. These non-pathogenic, commensal-derived strains are equipped with tumor-inducible promoters to deliver hyaluronidase locally and safely within the TME. …

Profiling of the TNBC Tumor Microenvironment's Functional Heterogeneity by CITE-seq to Guide Personalized Medicine.

UNLABELLED: Introduction / Objective: Triple-negative breast cancer (TNBC) is an aggressive subtype with few effective treatments and frequent resistance to immune checkpoint inhibitors. The heterogeneous and plastic tumor microenvironment (TME) is a dynamic, complex ecosystem. This study aims to deconstruct the TNBC immune ecosystem and generate a cellular atlas of its tumor microenvironment. METHODS: In this study, we apply three CITE-seq datasets from murine TNBC tumors, totaling 2,356 cells. After correcting for technical batch effects, we perform clustering to identify major cell types, …

Dual roles of DDR1 in stromal remodeling and immune escape: a regulator of the tumor immune microenvironment.

The discoidin domain receptor 1 (DDR1) is a collagen-activated receptor tyrosine kinase that orchestrates multiple cellular processes and remodels the tumor immune microenvironment (TIME). Notably, DDR1 promotes immune exclusion predominantly through its extracellular domain, which regulates collagen fiber alignment to form a physical barrier that impedes cytotoxic T cell infiltration, a function independent of its kinase activity. Moreover, DDR1 mediates cancer cell adhesion, migration, and metabolic reprogramming, while modulating cancer-associated fibroblasts, macrophages, and neutrophils t
