How Three Self-Secreted Biofilm Exopolysaccharides of Pseudomonas aeruginosa, Psl, Pel, and Alginate, Can Each Be Exploited for Antibiotic Adjuvant Effects in Cystic Fibrosis Lung Infection.

In cystic fibrosis (CF), pulmonary infection with Pseudomonas aeruginosa is a cause of increased morbidity and mortality, especially in patients for whom infection becomes chronic and there is reliance on long-term suppressive therapies. Current antimicrobials, though varied mechanistically and by mode of delivery, are inadequate not only due to their failure to eradicate infection but also because they do not halt the progression of lung function decline over time. One of the reasons for this failure is thought to be the biofilm mode of growth of P. aeruginosa, wherein self-secreted exopolysa

In vitro and in vivo evaluation of poly-3-hydroxybutyric acid-sodium alginate as a core-shell nanofibrous matrix with arginine and bacitracin-nanoclay complex for dermal reconstruction of excision wound.

The protective layer of the body, the skin is often prone to damage due to several factors like trauma, accidents, stress and hazardous exposure. This requires the skin to regenerate itself which is a finely regulated process. To hasten the process and prevent further damage, the dressing material is of prime importance. Herein, we fabricated poly-3-hydroxybutyric acid (P)-sodium alginate (S)-(core-shell) nanofibrous matrix as protective scaffold for the skin tissue regeneration in excision wound model. The arginine (A) and layered double hydroxides-bacitracin (LB) were incorporated into the c

Novel synthesized zinc oxide nanoparticles loaded alginate-chitosan biofilm to enhanced wound site activity and anti-septic abilities for the management of complicated abdominal wound dehiscence.

Wound dehiscence is a surgical complication and its management is inevitable because 25% to 35% of patients suffered from post laparotomy wound dehiscence. The excellent biodegradability and biocompatibility of chitosan and alginate have provided ample space for future developments in biomedical applications. Hence, the present work is directed towards the synthesis of robust biofilm made up of chitosan (CS), zinc oxide (ZnO) nanoparticles and Alginate (Alg). Chitosan and alginate were used for their pore forming ability, and ZnO is for its antibacterial action. The proposed biofilm was charac

Human osteochondritis dissecans fragment-derived chondrocyte characteristics ex vivo, after monolayer expansion-induced de-differentiation, and after re-differentiation in alginate bead culture.

BACKGROUND: Autologous chondrocyte implantation (ACI) is a therapy for articular cartilage and osteochondral lesions that relies on notch- or trochlea-derived primary chondrocytes. An alternative cell source for ACI could be osteochondritis dissecans (OCD) fragment-derived chondrocytes. Assessing the potential of these cells, we investigated their characteristics ex vivo and after monolayer expansion, as monolayer expansion is an integral step of ACI. However, as monolayer expansion can induce de-differentiation, we asked whether monolayer-induced de-differentiation can be reverted through suc

A sodium alginate-dual-protein double-network gel based on whey protein isolate and Tiger nut protein: Preparation, characterization and its sustained release properties of curcumin.

This study developed a sodium alginate-tiger nut protein/whey protein isolate double-network gel, and its sustained-release properties for curcumin were investigated. A series of sodium alginate (SA) concentrations (0-0.3%) were screened, and 0.25% SA was determined as the optimal level, which yielded superior gel hardness (401.67 ± 16.04 g) and water-holding capacity (81.49 ± 1.96%). Structural characterizations verified a double-network structure formation: one relied on protein aggregation, while the secondary network was formed via calcium ions-mediated cross-linking of SA. Owing to the du

Kinetical approach of release and antioxidant performances of ι-carrageenan-based films modified by sodium alginate and Ca2+ and incorporating sea fennel essential oil and its major components.

Understanding whether antioxidant efficiency in active films is governed by release kinetics or intrinsic radical-scavenging reactivity remains insufficiently explored in hydrophilic biopolymer networks. This study investigates the coupling between release mechanisms and antioxidant reaction kinetics in ι-carrageenan-based films loaded with bioactive (chlorogenic acidor d-limoneneor sea fennel essential oil) to evaluate their release behavior and antioxidant activity. Release experiments were conducted in food simulant D1 (50% ethanol) over 48 h, while antioxidant activity (AA%) was monitored

Enzyme- and ion-induced high internal phase emulsion gels based on soy protein isolate, chitosan, and alginate as fat analogues: Tunable texture and controlled flavor release.

Fat analogues have attracted increasing attention in food science because of their potential benefits for environmental sustainability, animal welfare, and human health. However, a deeper understanding of the textural design and flavor regulation of such systems is still needed. In this study, a high-internal-phase emulsion gel (HIPEG) with desirable textural properties was developed as a plant-based fat analogue with efficient flavor encapsulation and controlled release behavior. Briefly, hydrophobically modified chitosan gel network (h-CSG) was incorporated into a soy protein isolate (SPI) s

Multifunctional injectable hydrogels with self-healing and electroconductive properties as a potential platform for neural tissue engineering: Integration of polypyrrole and IKVAV peptide into modified alginate-hyaluronic acid networks.

Injectable biomaterials have emerged as a promising option for minimally invasive tissue repair, especially in cases with small and irregular lesions. In combination with electroconductivity and self-healing abilities, these materials provide unique benefits for neural tissue engineering. They promote adaptive defect filling, assist in post-injection structural repair, and improve bioelectrical signaling. The current study focuses on the synthesis and characterization of injectable, self-healing, electroconducting hydrogels, which are intended to serve as a potential platform for addressing sm

Innovative pH-responsive alginate-coated rosuvastatin-loaded chitosan nanoparticles: a targeted approach to inhibiting HMGB1-activated RAGE/TLR4-NFκB signaling in colonic inflammation in rats.

This study developed and optimized an innovative oral pH-dependent drug delivery system utilizing rosuvastatin-loaded chitosan nanoparticles (RSV-CSNPs) coated with sodium alginate (ALG). The goal was to protect RSV-CSNPs from degradation in the acidic gastric environment and facilitate targeted sustained release in the colon to address inflammatory bowel disease. Nanoparticles were initially prepared by ionic gelation. A subsequent ALG coating process was optimized using a 23 factorial design. The optimal ALG-coated formulation demonstrated minimal drug loss (0.88% ± 0.09%), desirable particl

Polylactic-Co-Glycolic Acid/Alginate/Neem Oil-Reduced Graphene Oxide as a pH-Sensitive Nanocarrier for Hesperidin Drug Delivery: Antimicrobial and Acute Otitis Media Assessments.

Background/Objectives: Hesperidin (HSP) is a potent phytochemical antioxidant and anti-inflammatory agent that protects against otitis media. However, due to its low solubility and bioavailability, a suitable delivery method is needed to overcome these problems. A hydrogel is a promising nanocarrier for controlled drug delivery in response to external stimuli, such as pH variations. Methods: Graphene oxide (GO)-based nanocarriers that encapsulate hesperidin (HSP) were further coated with a polylactic-co-glycolic acid/alginate (PLGA-Alg) hydrogel before being integrated into a green neem oil (N

3D printed sodium Alginate-Gelatin hydrogel loaded with Santalum album oil as an antibacterial Full-Thickness wound healing and scar reduction Scaffold: In vitro and in vivo study.

Managing wounds and accompanying consequences like exudation and microbiological infections is challenging in clinical practice. Bioactive compounds from traditional medicinal plants help heal wounds, although their bioavailability is low. This study uses sodium alginate (SA), gelatin (G), and Santalum album oil (SAL) to 3D print a polymeric hydrogel scaffold to circumvent these difficulties. The 3D printed scaffolds showed hydrophilicity, an average pore size of 221.30 ± 19.83 µm, adequate swelling, higher mechanical strength with tensile strength (σ) of 13.5 ± 1.08 MPa, a Young's modulus of

Study on fabrication of calcium sulfate hemihydrate/tricalcium phosphate based injectable bone cement modified by sodium alginate-carboxymethyl chitosan gel network and its resistance to collapse.

The collapsibility of bone cement may cause blood vessel embolism, blocking blood flow and causing serious complications such as pulmonary embolism or spinal cord injury, especially when implantation by injection. Therefore, it is of great significance to develop an artificial bone graft with excellent collapse resistance performance. Calcium sulfate and calcium phosphate complex bone cements can be formulated as injectable materials, making them particularly suitable for treating irregular bone defects. However, its clinical application is limited by poor collapsibility resistance and mechani

Intraperitoneal Administration of a Cisplatin-Loaded Nanogel through a Hybrid System Containing an Alginic Acid-Based Nanogel and an In Situ Cross-Linkable Hydrogel for Peritoneal Dissemination of Ovarian Cancer.

Intraperitoneal chemotherapy demonstrates potential applicability in the treatment of peritoneally disseminated ovarian cancer because the disseminated tumors can directly receive exposure to high concentrations of anticancer drugs. However, a considerable proportion of drugs, particularly micromolecular and hydrophilic drugs, such as cisplatin (CDDP), are often excreted through glomerular filtration for a short period. To effectively deliver CDDP into peritoneally disseminated ovarian cancer tissues, we developed an alginate (AL)-based hybrid system in which a CDDP-loaded AL nanogel (AL/CDDP-

Encapsulation of Escherichia coli strain Nissle 1917 in a chitosan-alginate matrix by combining layer-by-layer assembly with CaCl2 cross-linking for an effective treatment of inflammatory bowel diseases.

Escherichia coli strain Nissle 1917 (EcN) has been widely shown to effectively treat inflammatory bowel diseases (IBDs). Unfortunately, after oral administration, EcN viability dramatically decreases due to severe environmental factors, including low gastric pH, temperature and osmotic pressure. To address these challenges and improve oral bio-availability, this study utilized layer-by-layer assembly (LbL) and ionic cross-linking with CaCl2 as a method of EcN encapsulation (GEcN). Upon examination, GEcN cells were shown to maintain their ability to grow and proliferate, but had a slightly long

[Preventive effect of polaprezinc suspension dispersed in sodium alginate solution (P-AG) for stomatitis induced by Docetaxel/Cisplatin/Fluorouracil (DCF) chemotherapy in patients with head and neck cancer].

We measured the effectiveness of the prophylactic administration of a polaprezinc suspension dispersed in sodium alginate solution (P-AG) by dividing it into two courses in the same patients, and measured the stomatitis induced by Docetaxel/Cisplatin/Fluorouracil (DCF) chemotherapy. We then evaluated the results. We defined the therapeutic course as the course where P-AG was given therapeutically for stomatitis induced after DCF chemotherapy. We defined the prophylactic course as when P-AG was prophylactically given before any incidences of stomatitis after the therapeutic course. We compared

Succinylated nori protein-alginate complex particles prepared using temperature-controlled ultrasonication for the formation of high internal phase Pickering emulsion gels as plant-based fat analogues.

In this study, succinylated nori protein (SNP), sodium alginate (SA), and flaxseed oil were used to develop novel high-internal-phase Pickering emulsion (HIPPE) gels with potential application as plant-based fat analogues. Stable composite particles were successfully fabricated through ultrasonication combined with temperature control. The ultrasonic treatment condition was identified as 450 W for 30 min, under which the Pickering particles exhibited the smallest particle size and superior emulsifying ability. The optimal formulation parameters were identified as an SNP-to-SA ratio of 1:5, an
