Antibody against early driver of neurodegeneration cis P-tau blocks brain injury and tauopathy.

Traumatic brain injury (TBI), characterized by acute neurological dysfunction, is one of the best known environmental risk factors for chronic traumatic encephalopathy and Alzheimer's disease, the defining pathologic features of which include tauopathy made of phosphorylated tau protein (P-tau). However, tauopathy has not been detected in the early stages after TBI, and how TBI leads to tauopathy is unknown. Here we find robust cis P-tau pathology after TBI in humans and mice. After TBI in mice and stress in vitro, neurons acutely produce cis P-tau, which disrupts axonal microtubule networks a

Central nervous system delivery of helper-dependent canine adenovirus corrects neuropathology and behavior in mucopolysaccharidosis type VII mice.

Canine adenovirus type 2 vectors (CAV-2) are promising tools to treat global central nervous system (CNS) disorders because of their preferential transduction of neurons and efficient retrograde axonal transport. Here we tested the potential of a helper-dependent CAV-2 vector expressing β-glucuronidase (HD-RIGIE) in a mouse model of mucopolysaccharidosis type VII (MPS VII), a lysosomal storage disease caused by deficiency in β-glucuronidase activity. MPS VII leads to glycosaminoglycan accumulation into enlarged vesicles in peripheral tissues and the CNS, resulting in peripheral and neuronal dy…

Mitochondrial dysfunction and calcium deregulation by the RanBP9-cofilin pathway.

Mitochondrial dysfunction and synaptic damage are important features of Alzheimer's disease (AD) associated with amyloid β (Aβ) and tau. We … tion of cofilin into mitochondria and oxidative mechanisms. Further, RanBP9 retards the anterograde axonal transport of mitochondria in primary neurons and decreases synaptic mitochondrial activity in brain. These data indicate that RanBP9, cofilin, and Aβ mimic and potentiate each other to produce mitochondrial dysfunction, ROS overproduction, and calcium deregul …

Neurocircuitry of mood disorders.

This review begins with a brief historical overview of attempts in the first half of the 20th century to discern brain systems that underlie emotion and emotional behavior. These early studies identified the amygdala, hippocampus, and other parts of what was termed the 'limbic' system as central parts of the emotional brain. Detailed connectional data on this system began to be obtained in the 1970s and 1980s, as more effective neuroanatomical techniques based on axonal transport became available. In the last 15 years these methods have been applied extensively to the limbic system and prefron…

One hundred years of poliovirus pathogenesis.

Poliovirus was first isolated nearly 100 years ago in a landmark experiment that established the viral etiology of poliomyelitis. This disco … plication in the host, how the virus spreads to the central nervous system through the blood and by axonal transport, the determinants of viral tropism, and the basis for the attenuation phenotype of the Sabin vaccine strains. Is spinal muscular atrophy the result of defects in motor neuron processes?

The hereditary neurodegenerative disease spinal muscular atrophy (SMA) with childhood onset is one of the most common genetic causes of infa … vidence from animal and cell culture models of SMA points to roles for SMN in neurite outgrowth and axonal transport. Disruption of these functions might be particularly detrimental to motor neurons given their high metabolic demands and precise connectivity requirements, thus providing a possible explanation for the specificity of motor neuron susceptibility in S …

Localization of NADPH-diaphorase in pelvic afferent and efferent pathways of the rat.

The NADPH diaphorase histochemical reaction was used in combination with retrograde axonal transport of Fluorogold (FG) from the major pelvic ganglion (MPG) to determine if NADPH diaphorase is contained within afferent and preganglionic efferent pathways to pelvic visceral organs. In L6 and S1 dorsal root ganglia, 68.5% and 62.2%, respectively, of FG-labeled afferent neurons were NADPH-diaphorase positive. In the sacral parasympathetic nucleus (SPN) of the L6 and S1 spinal cord segments, 49.4% and 51.7%, respectively of FG labeled preganglionic efferent neurons were NADPH-diaphorase positive. …

[Afferent projections to the uvula in the cat. I. Climbing fiber projections].

The inferior olive afferents to the cerebellar uvula were studied by means of retrograde axonal transport of horseradish peroxidase (HRP) in the cat. Following large and small injections of HRP into the uvula, the distributions of labeled cells in the inferior olive were investigated. As to the climbing fiber projection, it is revealed that there are six longitudinal parasagittal zone extending throughout the dorsal and ventral uvula. That is, the caudal aspect of the nucleus beta projects to a most medially oriented zone (caudal beta zone), the rostral aspect of the nucleus beta projects to a…

Afferent projections from the brainstem to the three floccular zones in cats. I. Climbing fiber projections.

The cat's flocculus can be divided into 3 zones on the basis of differences in their efferent projection sites. In the present study, climbing fiber projections from the inferior olive to each zone of the flocculus were studied by means of retrograde axonal transport of horseradish peroxidase (HRP). Following large injections of HRP into the flocculus, labeled cells appear in the dorsal cap and the ventrolateral outgrowth of the principal olive. No HRP-labeled somata are present in other parts of the inferior olive. Following microinjections of HRP into the rostral of caudal zones of the flocc…

Incorporation of sialic acid into gangliosides and glycoproteins of the optic pathway following an intraocular injection of [N-3H]acetylmannosamine in the chicken.

Following an intraocular (i.o.) injection of [N-3H]acetylmannosamine, low molecular soluble radioactive label is distributed rapidly along the optic pathway. At the same time glycoproteins and gangliosides were found radioactively labelled in the distal parts of the optic pathway. Negligible amounts of radioactivity were incorporated into the same structures via the blood. The present data confirm, that glycoproteins are transferred in the retinal ganglion cells by a rapid axonal transport mechanism towards the nerve endings. Unlike glycoproteins labelled gangliosides accumulated in the optic …

Target muscles for retrograde gene delivery to specific spinal cord segments.

Targeted retrograde gene delivery into the injured spinal cord is less invasive for the damaged tissue. One of the advantages of this approach is the possible selection of target organs according to the level of spinal cord injury. We evaluated nine candidate target organs for retrograde delivery of an adenovirus vector carrying beta-galactosidase (AdV-LacZ) gene to cervical, thoracic and lumbar spinal cord segments. One week after vector injection into each muscle, we assessed the LacZ gene expression in the spinal cord by X-gal staining. The most appropriate target organs with high transduct

Hydrogen peroxide (H2O2) controls axon pathfinding during zebrafish development.

It is now becoming evident that hydrogen peroxide (H2O2), which is constantly produced by nearly all cells, contributes to bona fide physiological processes. However, little is known regarding the distribution and functions of H2O2 during embryonic development. To address this question, we used a dedicated genetic sensor and revealed a highly dynamic spatio-temporal pattern of H2O2 levels during zebrafish morphogenesis. The highest H2O2 levels are observed during somitogenesis and organogenesis, and these levels gradually decrease in the mature tissues. Biochemical and pharmacological approach

Sustained effect of bone marrow mononuclear cell therapy in axonal regeneration in a model of optic nerve crush.

In adult mammals, the regeneration of the optic nerve is very limited and at the moment there are several groups trying different approaches to increase retinal ganglion cell (RGC) survival and axonal outgrowth. One promising approach is cell therapy. In previous work, we performed intravitreal transplantation of bone-marrow mononuclear cells (BMMCs) after optic nerve crush in adult rats and we demonstrated an increase in RGC survival and axon outgrowth 14 days after injury. In the present work, we investigated if these results could be sustained for a longer period of time. Optic nerve crush

Peripheral nerve response to injury.

Oral and maxillofacial surgeons caring for patients who have sustained a nerve injury to a branch of the peripheral trigeminal nerve must possess a basic understanding of the response of the peripheral nerves to trauma. The series of events that subsequently take place are largely dependent on the injury type and severity. Regeneration of the peripheral nerve is possible in many instances and future manipulation of the regenerative microenvironment will lead to advances in the management of these difficult injuries.

Cadherin-7 and cadherin-6B differentially regulate the growth, branching and guidance of cranial motor axons.

Cadherin-7 (Cad7) and cadherin-6B (Cad6B) are expressed in early and late phases of cranial motoneuron development, respectively. Cad7 is expressed by cranial motoneurons soon after they are generated, as well as in the environment through which their axons extend. By contrast, Cad6B is expressed by mature cranial motoneurons. We demonstrate in chick that these cadherins play distinct roles in cranial motor axon morphology, branching and projection. Using in vitro approaches, we show that Cad7 enhances motor axon outgrowth, suppresses the formation of multiple axons and restricts interstitial
