Tuning the optical and magnetic properties of Tb3+-doped ZnO quantum dots: structural investigation and Judd-Ofelt theoretical analysis.

Tb3+-doped ZnO quantum dots (ZnO:Tb3+ QDs) with Tb3+ concentrations of 0-7 mol% were synthesized by a wet-chemical precipitation method to tune their optical and magnetic properties. XRD and Rietveld refinement confirmed the formation of hexagonal wurtzite ZnO without detectable secondary phases, while lattice expansion and increased microstrain suggested the incorporation of Tb3+ ions into the ZnO matrix. XPS and EDX mapping verified the presence and homogeneous distribution of Tb species. UV-vis absorption spectra showed a red shift of the absorption edge and bandgap narrowing with increasin

Mitochondrial-targeted DNA nanostructures deliver NAD+ to attenuate mitochondrial oxidative injury in renal ischemia-reperfusion.

Renal ischemia-reperfusion injury (IRI) is a leading cause of acute kidney injury (AKI) and is associated with substantial morbidity, delayed recovery, and impaired quality of life. Its progression is closely linked to mitochondrial oxidative stress, nicotinamide adenine dinucleotide (NAD+) depletion, and insufficient tubular repair. NAD+ is a key cofactor that sustains mitochondrial bioenergetics and redox homeostasis and supports sirtuin-dependent repair signaling. Therefore, restoring NAD+ metabolism is an attractive therapeutic strategy for IRI. Nevertheless, direct NAD+ administration is

Biotransformation-derived metabolites from Astragalus membranaceus and Cordyceps militaris alleviate hyperuricemia via multi-target regulation.

Hyperuricemia is a common metabolic disorder, and current pharmacological treatments are often associated with adverse effects. This study evaluated the anti-hyperuricemic activity of an ethanol extract obtained from bidirectional solid-state fermentation of Astragalus membranaceus and Cordyceps militaris (AMC-BFE) in a mouse model induced by potassium oxonate and adenine. AMC-BFE treatment significantly lowered serum uric acid levels, suppressed xanthine oxidase (XOD) activity, and ameliorated markers of liver and kidney function, including alanine aminotransferase, aspartate aminotransferase

Ubiquitin-Specific Protease 2 (USP2) as a Modulator of Energy Metabolism: A Review of Studies Using Animal and Cellular Models.

Ubiquitin-specific protease 2 (USP2) is a deubiquitinase that controls various cellular events, including cell cycle progression and tumorigenesis. Along with cell culture models, mouse models induced using chemical blockers and gene engineering have substantially contributed to our knowledge of the crucial roles of USP2 in energy metabolism and metabolic disorders. This review summarizes the evidence of the role of USP2 in regulating energy metabolism in mice and cells under physiological and pathological conditions. In hepatocytes, a short isoform of USP2, USP2b, aggravates type 2 diabetes a

Safety and toxicity assessment of antisense oligonucleotides in brain-relevant human cellular models.

Antisense oligonucleotides (ASOs) have become an established therapeutic modality for central nervous system (CNS) disorders, enabling selective modulation of gene expression through splice-switching or steric-hindrance mechanisms. Backbone and sugar chemistry modifications improved nuclease resistance, tissue retention, and pharmacodynamic durability, facilitating clinical translation of intrathecally delivered ASOs. This review summarizes current in vitro approaches for assessing safety and toxicity of CNS-targeted ASOs, alongside the evolving regulatory landscape shaping their preclinical e

Bisphenol AF Induces Hepatic Steatosis via Succinate-SUCNR1-Mediated Macrophage-Hepatocyte Interactions: An Adverse Outcome Pathway Study in Male C57BL/6 Mice.

Bisphenol AF (BPAF) exposure is increasingly linked to metabolic disorders, yet the molecular initiating events (MIE) and key events (KE) leading to hepatic steatosis remain unclear. We constructed an adverse outcome pathway (AOP) to mechanistically connect BPAF-triggered macrophage-hepatocyte crosstalk to liver fat accumulation. Male C57BL/6 mice received daily oral gavage of 0, 0.5, 4, or 32 mg kg-1 BPAF for 90 d, and Transwell co-cultures of RAW264.7 macrophages and AML12 hepatocytes were used for in vitro validation. Targeted metabolomics, western blotting, and lipid staining quantified su

Palmitoylation of PKCδ by ZDHHC5 in hypothalamic microglia presents as a therapeutic target for fatty liver disease.

The hypothalamus plays a fundamental role in controlling lipid metabolism through neuroendocrine signals. However, there are currently no available drug targets in the hypothalamus that can effectively improve human lipid metabolism. In this study, we found that the antimalarial drug artemether (ART) significantly improved lipid metabolism by specifically inhibiting microglial activation in the hypothalamus of high-fat diet-induced mice. Mechanically, ART protects the thyrotropin-releasing hormone (TRH) neurons surrounding microglial cells from inflammatory damage and promotes the release of T

Biochemical and cellular characterization of the CISD3 protein: Molecular bases of cluster release and destabilizing effects of nitric oxide.

The NEET proteins, an important family of iron-sulfur (Fe-S) proteins, have generated a strong interest due to their involvement in diverse diseases such as cancer, diabetes, and neurodegenerative disorders. Among the human NEET proteins, CISD3 has been the least studied, and its functional role is still largely unknown. We have investigated the biochemical features of CISD3 at the atomic and in cellulo levels upon challenge with different stress conditions i.e., iron deficiency, exposure to hydrogen peroxide, and nitric oxide. The redox and cellular stability properties of the protein agree o

Growing interest but limited evidence on the usefulness of cannabidiol in treating ophthalmic disease in dogs: a review.

The 2 most known cannabinoids are Δ9-tetrahydrocannabidiol (THC) and cannabidiol (CBD). Both chemicals are extracted from the cannabis plant but can also be synthetically produced. Δ9-Tetrahydrocannabidiol is extracted from the subspecies of the cannabis plant known as the marijuana plant, which contains a high concentration of THC (0.3% to 30%). Δ9-Tetrahydrocannabidiol is a major psychoactive and intoxicating component of the cannabis plant and is not recommended for use in dogs due to its toxic effect. Cannabidiol is extracted from the subspecies of the cannabis plant known as the hemp plan

Synthesis and Biological Activity of Homohypotaurine Obtained by the Enzyme-Based Conversion of Homocysteine Sulfinic Acid Using Recombinant Escherichia Coli Glutamate Decarboxylase.

l-Homocysteine, formed from S-adenosyl methionine following demethylation and adenosine release, accumulates when the methionine recycling pathway and other pathways become impaired, thus leading to hyperhomocysteinemia, a biomarker in cardiovascular diseases, neurological/psychiatric disorders, and cancer. The partial oxidation of the l-homocysteine thiol group and its decarboxylation on C-alpha lead to the formation of l-homocysteinesulfinic acid (l-HCSA) and homohypotaurine (HHT), respectively. Both compounds are not readily available from commercial suppliers, which hinders the investigati

Genipin rescues developmental and degenerative defects in familial dysautonomia models and accelerates axon regeneration.

The peripheral nervous system (PNS) is essential for proper body function. A high percentage of the world's population suffers from nerve degeneration or peripheral nerve damage. Despite this, there are major gaps in the knowledge of human PNS development and degeneration; therefore, there are no available treatments. Familial dysautonomia (FD) is a devastating disorder caused by a homozygous point mutation in the gene ELP1. FD specifically affects the development and causes degeneration of the PNS. We previously used patient-derived induced pluripotent stem cells (iPSCs) to show that peripher

Gastroprotective role of a flavonoid-rich subfraction from Fridericia chica (Bonpl.) L. G. Lohmann: a medicinal plant used in the Amazon region.

Fridericia chica is an Amazonian plant used to treat stomach disorders. However, the pharmacological activity of flavonoids in the extract has yet to be investigated. Therefore, we considered that a flavonoid-rich F. chica subfraction (FRS) has gastroprotective functions. For this, before the induction of gastric ulcers with ethanol or piroxicam, the rats received vehicle (water), omeprazole (30 mg/kg), or FRS (30 mg/kg), and the ulcer area was measured macro and microscopically, and the antisecretory action was investigated in pylorus-ligated rats. In addition, the roles of nitric oxide (NO)

Adipose-Derived Mesenchymal Stem Cells Attenuate Immune Reactions Against Pig Decellularized Bronchi Engrafted into Rat Tracheal Defects.

Decellularized scaffolds are promising biomaterials for tissue and organ reconstruction; however, strategies to effectively suppress the host immune responses toward these implants, particularly those without chemical crosslinking, remain warranted. Administration of mesenchymal stem cells is effective against immune-mediated inflammatory disorders. Herein, we investigated the effect of isogeneic abdominal adipose-derived mesenchymal stem/stromal cells (ADMSCs) on xenogeneic biomaterial-induced immunoreactions. Peripheral bronchi from pigs, decellularized using a detergent enzymatic method, we

The role of sympathetic innervation in neonatal muscle growth and neuromuscular contractures.

Neonatal brachial plexus injury (NBPI), a leading cause of pediatric upper limb paralysis, results in disabling and incurable muscle contractures that are driven by impaired longitudinal growth of denervated muscles. A rare form of NBPI, which maintains both afferent and sympathetic muscle innervation despite motor denervation, protects against contractures. We have previously ruled out a role for NRG/ErbB signaling, the predominant pathway governing antegrade afferent neuromuscular transmission, in modulating the formation of contractures. Our current study therefore investigated the contribu

The Congenital and Acquired Mechanisms Implicated in the Etiology of Central Precocious Puberty.

The etiology of central precocious puberty (CPP) is multiple and heterogeneous, including congenital and acquired causes that can be associated with structural or functional brain alterations. All causes of CPP culminate in the premature pulsatile secretion of hypothalamic GnRH and, consequently, in the premature reactivation of hypothalamic-pituitary-gonadal axis. The activation of excitatory factors or suppression of inhibitory factors during childhood represent the 2 major mechanisms of CPP, revealing a delicate balance of these opposing neuronal pathways. Hypothalamic hamartoma (HH) is the

Bioinspired pH-Responsive Microalgal Hydrogels for Oral Insulin Delivery with Both Hypoglycemic and Insulin Sensitizing Effects.

The oral form of insulin is more convenient and has better patient compliance than subcutaneous or intravenous insulin. Current oral insulin preparations, however, cannot overcome the enzyme barrier, chemical barrier, and epithelial barrier of the gastrointestinal tract completely. In this study, a microalgae-based oral insulin delivery strategy (CV@INS@ALG) was developed using Chlorella vulgaris (CV)-based insulin delivery system cross-linking with sodium alginate (ALG). CV@INS@ALG could overcome the gastrointestinal barrier, protect insulin from harsh gastric conditions, and achieve a pH-res

Comparative Hepatotoxicity of a Novel Perfluoroalkyl Ether Sulfonic Acid, Nafion Byproduct 2 (H-PFMO2OSA), and Legacy Perfluorooctane Sulfonate (PFOS) in Adult Male Mice.

Nafion byproduct 2 (H-PFMO2OSA) has been detected in the environment, but little is known about its toxicities. To compare the hepatotoxicity of H-PFMO2OSA with legacy perfluorooctane sulfonate (PFOS), male adult mice were exposed to 0.2, 1, or 5 mg/kg/d of each chemical for 28 days. Results showed that, although H-PFMO2OSA liver and serum concentrations were lower than those of PFOS, the relative liver weight in the H-PFMO2OSA groups was significantly higher than that in the corresponding PFOS groups. In addition, the increase in alanine transaminase and aspartate aminotransferase activity wa
