Some observations on the intrinsic nervous mechanism in Hirschsprung's disease.

Both at rest and during transmural stimulation acetylcholine output from isolated longitudinal and circular muscle strips is significantly higher in the spastic segment than in the proximal dilated bowel. No difference has been found in the tissue concentration of acetylcholine between ganglionic and aganglionic specimens. The pattern of response to transmural stimulation is also similar in the spastic and dilated bowel. However, after cholinergic and adrenergic blockade transmural stimulation fails to induce relaxation in aganglionic specimens, as it does in normal colon. The hypotheses are a

Functional segregation within the pelvic nerve of male rats: a meso- and microscopic analysis.

The pelvic splanchnic nerves are essential for pelvic organ function and have been proposed as targets for neuromodulation. We have focused … e surface of the MPG to the accessory nerves and a third class entered the MPG, encircling specific cholinergic neurons projecting to the LUT. A subpopulation of preganglionic inputs to noradrenergic MPG neurons also showed CGRP immunoreactivity. Together, these studies reveal new molecular and structural features of the pelvic nerves and suggest functional targets o …

Trophic factor-induced excitatory synaptogenesis involves postsynaptic modulation of nicotinic acetylcholine receptors.

Neurotrophic factors have well established roles in neuronal development, although their precise involvement in synapse formation and plasticity is yet to be fully determined. Using soma-soma synapses between identified Lymnaea neurons, we have shown recently that trophic factors are required for excitatory but not inhibitory synapse formation. However, neither the precise site (presynaptic versus postsynaptic cell) nor the underlying mechanisms have yet been defined. In the present study, synapse formation between the presynaptic cell visceral dorsal 4 (VD4) and its postsynaptic partner right

Alterations in Phase-Related Prefrontal Activation During Cognitive Tasks and Nicotinic α4β2 Receptor Availability in Alzheimer's Disease.

BACKGROUND: Evidence shows that the cholinergic system plays an important role in regulating working memory and that working memory-related prefrontal activation decreases with age and neuronal degeneration, such as Alzheimer's disease (AD). However, the relation between attention-related α4β2 nicotinic cholinergic function and task-induced prefrontal activation especially time course-related activation remains to be explored. OBJECTIVE: We aimed to elucidate the relationship between changes in task-induced oxy-hemoglobin concentration (cerebral blood flow, CBF) in the prefrontal cortex and t

Beyond the cholinergic hypothesis: do current drugs work in Alzheimer's disease?

Alzheimer's disease (AD) is a neurodegenerative disease characterized by memory and cognitive loss, and represents the leading cause of dementia in elderly people. Besides the complex biochemical processes involved in the neuronal degeneration (formation of senile plaques containing Abeta peptides, and development of neurofibrillary tangles), other molecular and neurochemical alterations, like cholinergic deficit due to basal forebrain degeneration, also occur. Because acetylcholine has been demonstrated to be involved in cognitive processes, the idea to increase acetylcholine levels to restor

Curcumin attenuates diabetic encephalopathy in rats: behavioral and biochemical evidences.

Emerging epidemiological data indicates that diabetes is a potential predisposing factor for neuropsychiatric deficits as stroke, cerebrovascular diseases, diabetic encephalopathy, depression and anxiety. Diabetic encephalopathy, characterized by impaired cognitive functions and neurochemical and structural abnormalities, involves direct neuronal damage caused by intracellular glucose. Curcumin, a well-established phenolic antioxidant and anti-inflammatory molecule, is capable of playing an important role against amyloid and dendritic pathology and thus has neuroprotective properties. The aim

Donepezil treatment of vascular dementia.

Cholinergic deficits are clinicopathological hallmarks of Alzheimer's disease (DAT) and during the past decade have been the sole target for clinically effective treatments. By contrast, vascular dementia subtypes (VaD) are heterogeneous clinical syndromes, and therapeutic approaches have been directed toward control of vascular risk factors. Little attention has been paid to cholinergic deficits as a mechanism contributing to cognitive impairments in VaD as a potential target for treatment. The purpose of the study was to determine whether there are therapeutic benefits from long-term treatme

Cholinergic neurotransmission underlying visual hallucinations in Parkinson's disease: Integration of multimodal evidence and translational approaches.

Visual hallucinations (VH) represent a prevalent and clinically significant non-motor manifestation in Parkinson's disease (PD), often leading to accelerated cognitive decline, increased caregiver burden, and reduced quality of life. Despite their impact, the neurobiological basis of VH remains poorly defined, and effective treatments are limited. This review systematically synthesizes evidence from neuropathology, multimodal neuroimaging (PET, DTI, fMRI), neurophysiology, and computational modeling to establish a coherent mechanism of VH rooted in cholinergic system failure. We demonstrate th

Bilateral simultaneous acute angle closure glaucoma following anticholinergic therapy for vestibular neuritis.

Attacks of bilateral simultaneous acute angle closure glaucoma are rare. A 59-year-old man developed vertigo and vomiting, with nystagmus in primary gaze, left-beating nystagmus in left gaze and a positive head impulse test. He was diagnosed with acute vestibular neuritis and received standard therapy, using four drugs that have anticholinergic effects. He subsequently developed bilaterally red eyes, initially diagnosed as bacterial conjunctivitis, and then blurred vision with ocular pain. An ophthalmological assessment identified mid-dilated, non-reactive pupils with corneal oedema and marked

Morphological variability is greater at developing than mature mouse neuromuscular junctions.

The neuromuscular junction (NMJ) is the highly specialised peripheral synapse formed between lower motor neuron terminals and muscle fibres. Post-synaptic acetylcholine receptors (AChRs), which are found in high density in the muscle membrane, bind to acetylcholine released into the synaptic cleft of the NMJ, thereby enabling the conversion of motor action potentials to muscle contractions. NMJs have been studied for many years as a general model for synapse formation, development and function, and are known to be early sites of pathological changes in many neuromuscular diseases. However, inf

Neocortical 40 Hz oscillations during carbachol-induced rapid eye movement sleep and cataplexy.

Higher cognitive functions require the integration and coordination of large populations of neurons in cortical and subcortical regions. Oscillations in the gamma band (30-45 Hz) of the electroencephalogram (EEG) have been involved in these cognitive functions. In previous studies, we analysed the extent of functional connectivity between cortical areas employing the 'mean squared coherence' analysis of the EEG gamma band. We demonstrated that gamma coherence is maximal during alert wakefulness and is almost absent during rapid eye movement (REM) sleep. The nucleus pontis oralis (NPO) is criti

Structural changes to the basal forebrain cholinergic system in the continuum of Alzheimer disease.

In this chapter, we review evidence, derived predominantly from in vivo human MRI studies, that the basal forebrain (BF) and its projection system undergo structural changes across the continuum of Alzheimer disease (AD) progression. We examine how these changes are detectable from the earliest presymptomatic stages and continue into the prodromal and clinical phases of AD. The chapter begins with a brief overview of BF neuroanatomy before characterizing how changes to the BF and ascending cholinergic white matter projections parallel AD progression. In subsequent sections, we describe how the

Impaired signaling for neuromuscular synaptic maintenance is a feature of Motor Neuron Disease.

A central event in the pathogenesis of motor neuron disease (MND) is the loss of neuromuscular junctions (NMJs), yet the mechanisms that lead to this event in MND remain to be fully elucidated. Maintenance of the NMJ relies upon neural agrin (n-agrin) which, when released from the nerve terminal, activates the postsynaptic Muscle Specific Kinase (MuSK) signaling complex to stabilize clusters of acetylcholine receptors. Here, we report that muscle from MND patients has an increased proportion of slow fibers and muscle fibers with smaller diameter. Muscle cells cultured from MND biopsies failed

Neuronal control of extrajunctional acetylcholine receptor-channels induced by injury in frog skeletal muscle fibres.

(1) Frog Sartorius muscles were transected to form two muscles: a neural segment (tibial part) containing all the endplates, and an aneural segment (pelvic part). The acetylcholine receptor-channels which appear at both new ends of the muscle fibres were studied by spectral analysis of membrane current fluctuations elicited by acetylcholine (ACh). (2) During the first two weeks after muscle transection, the ACh-receptors at the new muscle-tendon junction of the neural segment opened channels which had a long open time, similar to that of extrajunctional ACh-receptor channels induced by denerva

The physiological properties of amine-containing neurones in the lobster nervous system.

1. Our previous studies have shown that octopamine and serotonin are found associated with a system of neurones in the connective tissue sheath of the second roots of lobster thoracic ganglia. To try to understand the mechanism of activation of these neurones, we undertook an examination of their general physiological properties. 2. All of the neurones receive excitatory synaptic input that has a cholinergic pharmacology, which suggests that it may be from sensory neurones. A very limited number of cells, possibly one, provdes the total synaptic input to all the cells in the roots of the secon

Rivastigmine for parkinsonian symptoms in dementia with Lewy bodies: a paradigm shift.

INTRODUCTION: Parkinsonian symptoms are one of the core features of Dementia with Lewy bodies (DLB), which has a prevalence of more than 80%. Management of DLB poses unique challenges as the treatment of one condition leads to the worsening of another. Rivastigmine's efficacy and tolerability for cognitive and neuropsychiatric symptoms are well-studied. However, its effect on parkinsonian symptoms of DLB is inconclusive. CLINICAL CASE SERIES: In this case series, we elucidated five clinically diagnosed subjects of DLB with varying severity of parkinsonian motor symptoms. Beyond the cardinal s

NGF content in the cerebral cortex of non-demented patients with amyloid-plaques and in symptomatic Alzheimer's disease.

There is increasing evidence that in Alzheimer's disease nerve growth factor (NGF) protein and NGF mRNA content in postmortem cortex is not decreased, but may even be elevated although the NGF-sensitive cholinergic basal forebrain neurons are preferentially affected. However, only little is known about the early pathophysiological events leading to Alzheimer's disease. We therefore measured the post-mortem NGF concentrations in temporal and frontal cortex of Alzheimer's disease patients, non-demented controls without Alzheimer's disease-related pathology, as well as non-demented patients with

Effects of cholinergic agents on anorectal physiology.

BACKGROUND: Despite their potential therapeutic benefit, the effects of cholinergic agents on anal function have been poorly investigated. AIM: To analyse the effects of neostigmine and atropine on anorectal responses to rectal isobaric distension. METHODS: This was a placebo-controlled, randomized, double-blind crossover study, performed in 12 healthy volunteers who received intravenously, on 3 separate days, neostigmine, atropine or the placebo. During each day of the experiment, seven pressure steps (ranging from 1 to 31 mmHg) in three different protocols of rectal isobaric distension (ph
