Hormone supplemented media for cloning human breast cancer: increased colony formation without alteration of chemosensitivity.

We tested the ability of hormones and growth factors to enhance the colony formation in soft agarose of breast carcinoma using two human breast carcinoma cell lines, MCF-7 and MDA-MB231, MCF-7 could clone in a basal medium supplemented only by insulin, transferrin, prostaglandin F2 alpha, and fibronectin. Combining oestradiol, dexamethasone, insulin, transferrin, and triiodothyronine with a basal medium supplemented with 5% (v/v) foetal bovine serum (FBS) increased colony forming efficiency (CFE) two-to three-fold over the best obtained in serum supplemented medium without hormones. While opti

A study on prevalence of scabies in a resettlement colony (slum area) and its association with some sociocultural and environmental factors.

A house to house survey as a part of mass treatment of scabies was done in one of the resettlement colonies of Delhi with a population of 9341 in 2035 families, to study some of the sociocultural and environmental factors in families having scabies patients. It was observed that overcrowding, sleeping habits due to limited sleeping space, sharing of linen, clothes and towels were some of the factors associated with scabies. There was no association between the size of the family and persons affected by scabies. The majority of the population were ignorant about the etiology and mode of spread

The post-UV colony-forming ability of normal fibroblast strains and of the xeroderma pigmentosum group G strain.

In xeroderma pigmentosum, an inherited disorder of defective DNA repair, post-UV colony-forming ability of fibroblasts from patients in complementation groups A through F correlates with the patients' neurological status. The first xeroderma pigmentosum patient assigned to the recently discovered group G had the neurological abnormalities of XP. We have determined the post-UV colony-forming ability of cultured fibroblasts from this patient and from 5 more control donors. Log-phase fibroblasts were irradiated with 254 nm UV light from a germicidal lamp, trypsinized, and replated at known densit

Cytologie and cytochemistry of colony cells in soft agar gel culture from normal and leukemic bone marrow.

In order to judge differentiation of cells in soft agar colonies, cytological and cytochemical classification of single cells within these colonies is necessary. In this study, 1,026 colonies from 15 normal and 95 leukemic bone marrows have been evaluated using cytological, cytochemical, and immunocytochemical techniques. In 180 colonies from 15 normal controls no segmented neutrophils have been observed. The colonies mostly consisted of monocytes and macrophages, rarely pure eosinophil colonies were observed. The number of monocyte/macrophage colonies in untreated AML and the percentage of pu

A predictive ant colony optimization routing protocol for correlated link stability in wireless body area networks.

BACKGROUND: Wireless Body Area Networks (WBANs) are essential for continuous health monitoring but face critical challenges due to dynamic link instability caused by body movements and strict bio-safety requirements. The fluctuating wireless channel leads to high packet loss, while radio frequency (RF) energy absorption, measured as Specific Absorption Rate (SAR), must remain below 1.6 W/kg to prevent tissue damage. Existing routing protocols often react to failures rather than predict them and rarely co-optimize reliability with thermal safety. METHODS: This paper proposes PACO-CLS (Predicti

Therapeutic role of granulocyte-colony stimulating factor and aspirin in unexplained recurrent pregnancy loss: a retrospective study.

BACKGROUND: Recurrent pregnancy loss (RPL), the loss of two or more clinically confirmed pregnancies, is observed in approximately 1%-5% of couples trying to conceive. Immune dysfunction is increasingly recognized as a contributing factor. This study evaluated pregnancy outcomes among women with unexplained RPL treated with combined recombinant human granulocyte-colony stimulating factor (rhG-CSF) and low-dose aspirin, while acknowledging that the evidence base for rhG-CSF in unexplained RPL remains uncertain. METHODS: In this retrospective study, 300 women aged 18-45 years with at least two

Avian cholera causes mass mortality at penguin mega-colonies amid H5N1 HPAI outbreaks in Antarctica.

The characterization of pathogens affecting Antarctic wildlife has been historically limited and sporadic. With the introduction of H5N1 high pathogenicity avian influenza virus (HPAIV) in Antarctica in the austral summer 2023/2024, concerns about the health of Antarctic seabirds and mammals prompted immediate surveillance efforts. In March 2024, we visited the Danger Islands archipelago, that hosts two mega-colonies of Adélie penguins, and observed an unusual mortality estimated in thousands of Adélie penguins and other species. Swabs and tissues were collected for molecular detection of infe

A wspA mutation in a clinical isolate of Pseudomonas aeruginosa results in rugose small colonies and enhances biofilm formation.

Pseudomonas aeruginosa is a major multidrug-resistant pathogen whose biofilm formation complicates treatment. Rugose small colony variants (RSCVs) exhibit enhanced biofilm production and resistance, posing significant clinical challenges; however, their formation mechanisms in clinical strains remain incompletely understood. Paired clinical isolates AR8023-1 (wild type) and AR8023-2 (RSCV) were chronologically collected from urine samples of a neurosurgery inpatient. Genome comparison via Breseq and plasmid complementation constructed the complemented strain AR8023-2RifR::wspAAR8023-1. All str

A Phase II Trial of Pembrolizumab plus Granulocyte-Macrophage Colony-Stimulating Factor in Advanced Biliary Cancer.

PURPOSE: Immune checkpoint inhibitors have limited activity as monotherapy in biliary cancers. Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a pleiotropic immune cell growth factor which achieved prolonged survival when combined with ipilimumab in melanoma. We conducted a single-center, phase II trial to evaluate the efficacy and safety of combining GM-CSF with pembrolizumab in patients with advanced biliary cancers after prior chemotherapy but no prior immune checkpoint inhibitor. EXPERIMENTAL DESIGN: Pembrolizumab 200 mg was administered intravenously in 21-day cycles, along

Seasonal dynamics of seven viruses and Varroa destructor in Apis mellifera colonies: a 4-year survey in Tokyo.

Long-term monitoring of viruses and Varroa destructor within the same honey bee colonies is rare in Japan. Thus, we conducted a 4-year fixed-point survey of three Apis mellifera colonies in Tokyo with monthly screening for seven viruses (using polymerase chain reaction) and Varroa infestation. Deformed wing virus was detected year-round, whereas sacbrood virus and black queen cell virus peaked in spring and summer. Israeli acute paralysis virus appeared sporadically, without seasonal trends. Varroa infestation rates (monitored during the latter 2.5 years) did not correlate with the number of d

Granulocyte colony-stimulating factor acts through calcium-permeable AMPA receptors to potentiate cocaine reward.

Neuroimmune interactions have emerged as critical modulators of substance use disorders and may represent promising translational therapeutic targets. In prior work, we demonstrated that the cytokine granulocyte colony stimulating factor (G-CSF) is elevated in mice following cocaine exposure, with circulating levels correlating with cocaine intake and locomotor sensitization. Additionally, exogenous G-CSF enhances cocaine reward and increases low-dose cocaine self-administration. We have further shown that repeated G-CSF administration alters expression of glutamatergic synapse-associated prot

Characterizing the Reactive Metabolites of Colony-Stimulating Factor 1 Receptor Inhibitor PLX5622 in Liver Microsomes and Mice.

Colony-stimulating factor 1 receptor (CSF1R) is a receptor tyrosine kinase involved in cell growth and differentiation, particularly in macrophages and microglia. CSF1R inhibitors are under investigation for various diseases, including cancer, autoimmune/inflammatory diseases, and neurodegenerative disorders. PLX5622 is a highly specific, brain-penetrant, and orally bioavailable CSF1R inhibitor that is being evaluated in a clinical trial for rheumatoid arthritis and considered as an attractive candidate for the treatment of Alzheimer's disease (AD). Drug metabolism significantly influences bot

Colony-Forming Unit Assay Adapted For Patient-Derived Bone Marrow and Peripheral Blood Mononuclear Cells.

The colony-forming unit (CFU) assay is a foundational technique for studying hematologic diseases, including myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML). This assay employs semisolid media supplemented with defined cytokine combinations to support three-dimensional growth and lineage-specific differentiation of erythroid and myeloid progenitor cells. However, patient-derived peripheral blood and bone marrow samples present technical challenges due to reduced viability, increased sensitivity to handling, and variability in colony growth. Furthermore, existing guidelines for

Varroa control strategies of different beekeeper groups and corresponding honey bee colony mortality in Central Europe.

INTRODUCTION: The parasitic mite Varroa destructor remains a major threat to Apis mellifera globally, contributing significantly to high colony losses despite various control interventions. Understanding the range of Varroa control strategies employed by beekeepers is crucial for mitigating these losses and improving honey bee health. OBJECTIVES: This study aimed to comprehensively analyze the diverse Varroa destructor control strategies implemented by beekeepers across Austria, Czechia, Poland, and Slovakia, and to evaluate their association with winter honey bee colony mortality. Our primar

Spatiotemporal clustering and ecological niche modeling of Varroa destructor infestations in honey bee colonies across Türkiye.

Varroa destructor is the most important ectoparasite affecting honey bee (Apis mellifera) colonies worldwide and represents a major threat to apiculture through its direct pathogenic effects and its role in the transmission of viral diseases. Despite its widespread occurrence in Türkiye, information regarding its large-scale spatial distribution and environmental determinants remains limited. This study aimed to investigate the spatiotemporal distribution patterns of V. destructor infestations across Türkiye using retrospective Space-Time Permutation Scan Statistics (SaTScan) and ecological ni

Controlling Varroa destructor infestations in managed honeybee colonies limits virus spillover into wild pollinator communities.

Given recent pathogen-mediated pollinator declines, there is concern regarding virus spillover from managed honeybees to wild pollinators. While the parasitic mite Varroa destructor specializes on honeybees, the viruses it transmits are widespread throughout pollinator communities. Here, we investigated whether beekeeper control of varroa mites influences the magnitude of deformed wing virus (DWV) spillover into wild pollinators. We sampled 27 apiaries along a gradient of mite infestation, screening nearly 1000 pollinators and 350 flowers for DWV via quantitative PCR. We found that controlling

Phase variable colony morphotypes of Clostridioides difficile elicit distinct host responses during acute infection.

Many Clostridioides difficile strains can form two colony morphotypes: rough and smooth. The rough and smooth morphotypes differ in multiple phenotypes, including cell length and chaining, motility, biofilm production, and virulence in the hamster model of C. difficile infection (CDI). Colony morphology undergoes phase variation and is determined by the ON/OFF expression of cmrRST, which encodes a signal transduction system. Here, we test the hypothesis that differences in colony morphology and the associated phenotypes influence pathogenesis and the host response to infection. We first compar
