Acute stress persistently enhances estrogen levels in the female rat.

Here we tested whether exposure to either tailshock or swim stress alters ovarian hormone levels, estrogen and progesterone, in females and whether the effects are persistent. Adrenal hormone levels were also measured in males and females. Estradiol levels were elevated in unstressed females during proestrus relative to females in other stages of estrous, and exposure to the stressors enhanced estradiol beyond basal levels. For females stressed during diestrus 2, estradiol levels were elevated immediately after stressor cessation and up to 24 hrs. Exposure to tailshock, but not swim-stress, tr

Conditioned taste aversion and c-fos expression in the rat brainstem after administration of various USs.

Conditioned taste aversion (CTA) is a form of association learning established when an animal associates a conditioned stimulus (taste) with a subsequent unconditioned stimulus (US, illness). We have studied the relationship between the efficacy of inducing CTA to 0.1% saccharin and c-fos expression in the lower brain stem following administration of 13 different USs in rats. The effective USs were grouped into abdominal irritants, rewarding drugs and emetic agents. Regardless of the properties of USs, good correlation was detected between the strength of CTA and c-fos expression within the ar

Restraint stress-induced changes in saccharin preference: the effect of antidepressive treatment and diazepam.

The effect of antidepressive treatment and of diazepam on saccharin preference has been studied in a model of acute restraint stress-induced saccharin preference deficit. It has been shown that 1-hr stressor produces short-term, and significant decrease of saccharin preference in a two-bottle test, measured at 24-hr periods of time. Single doses of desipramine and citalopram (10 mg/kg, IP) given prior to stress session significantly attenuated the deficit in saccharin preference. Less strong, but similar effects appeared after postshock antidepressant administration. On the other hand, electro

Behavioral studies with anxiolytic drugs. I. Interactions of the benzodiazepine antagonist Ro 15-1788 with chlordiazepoxide, pentobarbital and ethanol.

Lever pressing by squirrel monkeys was maintained under two behavioral procedures known to be sensitive to anxiolytic drugs. Under one procedure, responding maintained by food was suppressed by electric shock (punishment). Under a second procedure, responding was maintained under a multiple schedule in which the first response after 5 min produced either food or shock depending on the stimulus that was present throughout the interval (fixed-interval schedule). Under the punishment schedule, chlordiazepoxide (1.0-100 mg/kg), pentobarbital (1.0-17.0 mg/kg) and ethanol (0.5-2.5 g/kg) increased re

[Memory confinement versus generalization decrement on retrograde amnesia produced by cortical spreading depression].

An examination of memory confinement vs. generalization decrement under unilateral cortical spreading depression was undertaken using a licking suppression to a tone previously paired with a shock. In Experiment I, rats that were trained and tested under normal cortical conditions showed an impairment of memory when KCl-induced depression was given to an unilateral hemisphere within 15 min after the training. In Experiment II, where rats were trained under normal cortical conditions followed by unilateral depression and tested under unilateral cortical depression, the degree of impairment was

Aging-related changes in retrograde amnesia for mice.

Aging mice showed a retrograde amnesia for passive avoidance behavior when a single electroconvulsive shock (ECS) was given at post-training intervals up to 1 h, whereas adult mice showed no significant amnesia when ECS was given 10 min or more after training. Catecholamines (norepinephrine and dopamine), but not serotonin (5-HT) changed with aging in the several brain regions, but only 5-HT was significantly changed (elevated) by ECS. Protein synthesis was significantly inhibited by ECS, notably in limbic structures. The increased susceptibility of aging mice to the amnesic effect of ECS coul

Reflex conditioning in a spinal man.

A man with a completely transected spinal cord and a spastic neurogenic bladder was conditioned to void upon the presentation of an external stimulus. A classical conditioning paradigm was employed in which strong abdominal shock (unconditioned stimulus) was paired with an initially neutral mild electrical stimulation of the thigh (conditioned stimulus; CS). After the pairing trials, a reliable conditioned response of urination was elicited by the CS alone. The conditioned response did not extinguish over time, and the procedure left the bladder with clinically safe residual amounts of urine.

Anticonvulsant and reproductive toxicological studies of the imidazole-based histamine H3R antagonist 2-18 in mice.

The imidazole-based H3R antagonist 2-18 with high in vitro H3R antagonist affinity, excellent in vitro selectivity profile, and high in vivo H3R antagonist potency was tested for its anticonvulsant effect in maximal electroshock (MES)-induced convulsions in mice having valproic acid (VPA) as a reference antiepileptic drug (AED). Additionally, H3R antagonist 2-18 was evaluated for its reproductive toxicity in the same animal species. The results show that acute systemic administration (intraperitoneal; i.p.) of H3R antagonist 2-18 (7.5, 15, 30, and 60 mg/kg, i.p.) significantly and dose depende…

Chronic Electroconvulsive Shock Disrupts Hippocampal Physiology and Impairs Associative Memory in Mice.

Electroconvulsive therapy (ECT) is a highly effective treatment for depression but it has undesirable side effects on various aspects of cognition, including memory. Patients may display retrograde amnesia for autobiographical events and anterograde amnesia in tests of recall and recognition memory. These amnestic effects are likely to depend, at least in part, on changes in the hippocampus. ECT-induced volume increases in the hippocampus and dentate gyrus subregion in particular correlate with the cognitive deficits. However, the precise cellular and behavioral effects of ECT remain poorly un

Maximal electroconvulsive shock-induced seizures elicit long-lasting analgesia and short-lasting hyperoxia in a sex dependent manner.

BACKGROUND: Following a seizure, both humans and non-human animals exhibit postictal analgesia; decreased perception of nociceptive stimuli. Despite several brain regions having been implicated in postictal analgesia, a convergent mechanism has not been determined. Previous studies have found that, following a seizure, there is a period of altered oxygen dynamics that is the mechanism for several seizure comorbidities. We examined oxygen dynamics in the locus coeruleus (LC) to determine its potential role in postictal analgesia. METHODS: We employed maximal electroconvulsive shock (MES)-induc

Marked sex differences are observed in heroin acquisition and affective states in rats but converge to similar levels of footshock stress-induced reinstatement.

RATIONALE: Negative affective states, particularly those driven by stress, are known to trigger the development of opioid addiction and relapse. Previous preclinical research has identified sex differences in vulnerability to stress and resulting compulsion to consume drugs. OBJECTIVES: To investigate sex differences in heroin self-administration, stress-induced reinstatement, and related affective state in rats. METHODS: 22- and 50-kHz ultrasonic vocalizations (USVs), reflecting negative and positive affective state, respectively, were recorded during a heroin self-administration and footsh

Posterior parietal cortex mediates fear renewal in a novel context.

The return of fear following extinction therapy is an important issue associated with the treatment of many fear-related disorders. Fear renewal is a suitable model, with which context-dependent modulation of the fear response can be examined. In this model, any context outside of an extinction context (e.g., novel or familiar contexts) could evoke relapse of the fear response. However, brain regions associated with context-dependent modulation are not fully understood. The posterior parietal cortex (PPC) is considered a center for integrating multisensory information and making decisions. To

Acute foot-shock stress decreased seizure susceptibility against pentylenetetrazole-induced seizures in mice: Interaction between endogenous opioids and cannabinoids.

BACKGROUND: Stressful conditions affect the brain's neurotransmission and neural pathways that are involved in seizure susceptibility. Stress alters the intensity and/or frequency of seizures. Although evidence indicates that chronic stress exerts proconvulsant effects and acute stress has anticonvulsant properties, the underlying mechanisms which mediate these effects are not well understood. In the present study, we assessed the role of endogenous opioids, endocannabinoids, as well as functional interaction between opioid and cannabinoid systems in the anticonvulsant effects of acute foot-sh

Active avoidance learning differentially activates ERK phosphorylation in the primary auditory and visual cortices of Roman high- and low-avoidance rats.

Understanding the mechanisms underlying conditioned avoidance is a critical step toward the development of novel treatments of anxiety. In this context, the two-way active avoidance (2WAA) task is a validated paradigm to investigate uncontrolled avoidance, a hallmark of anxiety disorders. The outbred Roman high- (RHA) and low-avoidance (RLA) rat lines are selected for respectively rapid vs. poor acquisition of active avoidant behavior, and emotional reactivity appears to be the most prominent behavioral difference between the two lines, with RLA rats being more fearful/anxious than their RHA c

A comparison of cannabidiolic acid with other treatments for anticipatory nausea using a rat model of contextually elicited conditioned gaping.

RATIONALE: The effectiveness of cannabidiolic acid (CBDA) was compared with other potential treatments for anticipatory nausea (AN), using a rat model of contextually elicited conditioned gaping reactions. OBJECTIVE: The potential of ondansetron (OND), Δ(9)-tetrahydrocannabinol (THC), chlordiazepoxide (CDP), CBDA, and co-administration of CBDA and tetrahydrocannabinolic acid (THCA) to reduce AN and modify locomotor activity was evaluated. MATERIALS AND METHODS: Following four pairings of a novel context with lithium chloride (LiCl), the rats were given a test for AN. On the test trial, they

Bisphosphonates in phenytoin-induced bone disorder.

Chronic administration of phenytoin (PHT) has been associated with bone loss. Bisphosphonates [alendronate (ALD), ibandronate (IBD) and risedronate (RSD)] are potential candidates to prevent PHT-induced bone disorders, and the present study evaluated their effect on the antiepileptic efficacy of PHT. The PHT-induced depletion in folic acid (FA), vitamin B6 and vitamin B12 results in hyperhomocysteinemia. The elevated circulating homocysteine (hcy) could be a risk indicator for micronutrient-deficiency-related osteoporosis via generation of free radicals. Thus, an attempt was also made to unrav
