Biological profiling of piperazinediones for the management of anxiety.

The anxiolytic effect of earlier reported piperazinediones was assessed by elevated plus maze (EPM), hole-board and open-field (OFT) tests. The rats were administered pretreatment of three different doses i.e. 2, 1 and 0.5 mg/kg of compounds 52, 53 and 55 for seven consecutive days. Compound 52 and diazepam showed increase in open arm entries, increase in time spent therein and total arm entries at 1 mg/kg dose. The compound also produced increase in the number of head dip, sniffing behavior and total number of squares crossed compared to diazepam. In OFT paradigm grooming behavior, number of

Early prenatal exposure to LPS results in anxiety- and depression-related behaviors in adulthood.

Maternal immune activation can result in different behavioral abnormalities and brain dysfunction, depending on the nature of the inflammogen and the timing of the challenge. Few studies report the possible link between prenatal exposure to inflammation and mood disorders. Here we aimed to evaluate the effects of a single low lipopolysaccharide (LPS) injection to the dam at gestational day 9 on the offspring behavior and hippocampal function. We found that mice exposed to LPS show anxiety- and depression-related behaviors. Specifically, we found that animals prenatally exposed to LPS avoided t

The antioxidant gallic acid induces anxiolytic-, but not antidepressant-like effect, in streptozotocin-induced diabetes.

The physiopathology of anxiety or depression related to diabetes is still poorly understood. The treatment with antidepressant drugs is a huge challenge due to theirs adherence low rate and many adverse effects. Thus, the seeking for a better treatment for these associated diseases is of utmost importance. Given that the oxidative stress in different tissues occurs in diabetes and anxiety or depression as well, the antioxidant gallic acid becomes an interesting compound to be investigated. Thus, the effects of long-term treatment with gallic acid (0, 10, 20 and 40 mg/kg; gavage) were evaluated

Quercetin and Tranylcypromine Improve Memory, Behavioral Performance, and Cholinergic Function in Male Rats Subjected to Chronic Restraint Stress.

Background/Objectives: Major depressive disorder (MDD) is a debilitating illness, and chronic stress is a contributing factor for depressive symptoms. However, despite intense research, the mechanisms of MDD remain substantially unidentified. Quercetin is a powerful flavonoid and could be used as a possible therapeutic strategy for depression. Acknowledging the potential benefits of quercetin, this study investigated its effect alone or in association with the standard drug tranylcypromine (TCP) in a rodent model of chronic restraint stress (CRS). Methods: Adult male rats were subjected to a C

Low emotional contagious behavior induces PTSD susceptibility in observers and is related to the regulation of oxytocin receptor in mice.

BACKGROUND: Post-traumatic stress disorder (PTSD) is a serious psychiatric disorder that occurs after an individual has witnessed or experienced a major traumatic event. Emotional contagion seems to play an important role in witnessing trauma, highlighting the importance of understanding the neurobiological consequences of psychological or emotional stress and its impact on the individual's mental health. Therefore, understanding the relationship between emotional contagion and PTSD susceptibility and the abnormal neurobiological and behavioral changes behind it could help find effective molec

A quantitative analysis of the roles of dosage, sequence, and duration of estradiol and progesterone exposure in the regulation of maternal behavior in the rat.

The regulation of the onset of maternal behavior in the rat is under hormonal control. This study reports a new endocrine model for the study of the hormonal regulation of maternal responsiveness. The model employs the administration of physiological amounts of the steroids estradiol (E2) and progesterone (P) via Silastic implants to inexperienced nulliparous rats and measurement of the effects of these implants on maternal behavior. In the first two experiments, the levels of E2 and P in the sera of pregnant and hormone-treated rats were measured by RIA. Using known physiological treatments o

Effects of chronic nicotine administration on behavioral parameters and Na⁺/K⁺-ATPase mRNA expression in the brains of male Swiss Albino mice exposed to chronic stress during adolescence.

BACKGROUND: Stress during adolescence leads to the development of major stress-related diseases. The use of psychoactive substances and most psychiatric disorders have been linked to cigarette smoking during adolescence. Na+/K + ATPase isoforms are related to memory formation, stress activity and psychological disorders. Moreover, nicotine downregulates Na+/K + ATPase α2 isoform in rats’ brains. However, its connection to social stress and nicotine addiction is still unclear. Here we aimed for elucidating the effects of nicotine on behavioral parameters and Na+/K + ATPase mRNA in brains of mal

Prenatal, but not neonatal, paracetamol exposure disrupts neuroendocrine regulation of stress and emotional behavior in adult male rats.

Paracetamol is a widely used analgesic and antipyretic, commonly administered during pregnancy and early postnatal life. Its mechanism of action involves inhibiting cyclooxygenase-2/3 (COX-2/3) enzymes, which play a critical role in normal brain development during the perinatal period. In this study, we investigated anxiety-like behavior, depression-like behavior, and corticosterone levels in adult male rats following either prenatal or neonatal paracetamol exposure. Pregnant dams received subcutaneous injections of paracetamol (60 mg/kg) or saline every 12 h from gestational day (GD) 16 to 20

Gut Microbiota from GAD patients with divergent early pharmacological responses transmits anxiety-like behaviors and distinct metabolic profiles in recipient mice.

The gut microbiota is implicated in the pathogenesis of generalized anxiety disorder (GAD), but whether microbial differences between GAD patients with remission (R-GAD) and those with poor early treatment response (P-GAD) causally contribute to variable therapeutic outcomes remains unclear. Here, we transplanted feces from R-GAD and P-GAD patients into antibiotic-pretreated male C57BL/6 mice. P-GAD recipients showed heightened anxiety-like behaviors (reduced central zone time in open field test and shorter open-arm duration in elevated plus-maze), lower gut microbial α-diversity, and distinct

Effects of Exercise Type and Sertraline on Anxiety and Social Behavior in a Female Rat Model of PTSD.

BACKGROUND: Post-traumatic stress disorder (PTSD) often co-occurs with anxiety and social interaction difficulties, particularly in females. This study examined the effects of voluntary and involuntary exercise, combined with sertraline, on PTSD-like symptoms, anxiety, and social behavior in female rats. METHODS: PTSD-like symptoms were induced using the Single Prolonged Stress (SPS) model, which involved sequential exposure to restraint stress (2 h), forced swimming (20 min), and ether anesthesia (2-3 min), followed by a 7-day incubation period. A 10-min restraint was then applied as an addi

Trifluoro-icaritin attenuates neuropathic pain-associated anxiety-like behaviors and hippocampal oxidative stress in rats.

Neuropathic pain is frequently accompanied by anxiety-like symptoms. Trifluoro-icaritin (ICTF), an icaritin derivative with reported central nervous system activity, has analgesic and anti-inflammatory effects, but its actions in neuropathic pain-associated anxiety remain unclear. We examined whether changes in sigma-1 receptor (Sigma-1R) expression and hippocampal oxidative stress accompany the behavioral effects of ICTF in spared nerve injury (SNI) rats. Mechanical hypersensitivity and anxiety-related behavior were assessed using the paw withdrawal threshold, open field test, and elevated pl

Chronic Chrysin, but Not Diazepam, Induces Anxiolytic-like Effects Without Behavioral Tolerance.

BACKGROUND: Chrysin (5-7-dihydroxyflavone) exerts anxiolytic-like effects via the GABAA/benzodiazepine receptor complex after acute administration, similar to diazepam. Chronic diazepam treatment produces pharmacological tolerance, likely due to structural receptor modification. Whether chronic chrysin treatment also produces pharmacological tolerance remains unknown. This study evaluated GABAA/benzodiazepine receptor-mediated behavioral responses after a pharmacological challenge with a sedative dose of diazepam (5 mg/kg) in rats chronically treated with chrysin or diazepam. METHODS: Thirty-

Stem cells from tooth apical papilla modulate fibromyalgia-like changes in mice.

BACKGROUND: The treatment of fibromyalgia remains a current challenge. While the analgesic effects of mesenchymal stem cells have been the subject of previous research, their potential in managing fibromyalgia symptoms needs further investigation. In this study, we investigated the effects of human stem cells from tooth apical papilla (SCAP), when administered systemically, in a mouse model of fibromyalgia induced by reserpine. METHODS: The effects of repeated intraperitoneal (i.p.) treatment with SCAP were evaluated in the mouse model of fibromyalgia induced by reserpine, by assessing behavi

Protective role of madecassoside from Centella asiatica against protein L-isoaspartyl methyltransferase deficiency-induced neurodegeneration.

Protein L-isoaspartyl methyltransferase (PIMT/PCMT1) could repair l-isoaspartate (L-isoAsp) residues formed by deamidation of asparaginyl (Asn) residues or isomerization of aspartyl (Asp) residues in peptides and proteins during aging. Aside from abnormal accumulation of L-isoAsp, PIMT knockout (KO) mice mirrors some neuropathological hallmarks such as anxiety-like behaviors, impaired spatial memory and aberrant synaptic plasticity in the hippocampus of neurodegenerative diseases (NDs), including Alzheimer's disease (AD) and related dementias, and Parkinson's disease (PD).
