Age at menarche and lung function: a Mendelian randomization study.

A trend towards earlier menarche in women has been associated with childhood factors (e.g. obesity) and hypothesised environmental exposures (e.g. endocrine disruptors present in household products). Observational evidence has shown detrimental effects of early menarche on various health outcomes including adult lung function, but these might represent spurious associations due to confounding. To address this we used Mendelian randomization where genetic variants are used as proxies for age at menarche, since genetic associations are not affected by classical confounding. We estimated the effe

Global analysis of the human pathophenotypic similarity gene network merges disease module components.

The molecular complexity of genetic diseases requires novel approaches to break it down into coherent biological modules. For this purpose, many disease network models have been created and analyzed. We highlight two of them, "the human diseases networks" (HDN) and "the orphan disease networks" (ODN). However, in these models, each single node represents one disease or an ambiguous group of diseases. In these cases, the notion of diseases as unique entities reduces the usefulness of network-based methods. We hypothesize that using the clinical features (pathophenotypes) to define pathophenotyp

Genetic Evidence Linking Inflammatory Cytokines and Blood Metabolites to Heart Failure Risk.

BACKGROUND: Heart failure (HF) is a critical condition characterized by the heart's inability to pump blood effectively, leading to significant morbidity and mortality. Inflammation and metabolic disturbances play key roles in its progression. This study is aimed at elucidating the causal relationships between inflammatory cytokines, metabolites, and HF using Mendelian randomization (MR). METHODS: A three-step, two-sample MR analysis was performed using genetic data from genome-wide association studies. The study included data on 91 inflammatory cytokines, over 1400 metabolites, and HF cases

Neuropsychiatric Disorders and Constipation: Unraveling Causal Links Through Genetic Analysis.

BACKGROUND: Numerous observational studies have suggested a relationship between neuropsychiatric disorders and constipation. However, the specific causal relationships remain unclear. Mendelian randomization (MR) serves as a proven strategy for examining the causal relationships between genetic exposures and outcomes. In the present study, we used a two-sample Mendelian randomization (TSMR) analysis to thoroughly explore the potential bidirectional genetic causal effects between neuropsychiatric disorders and constipation. METHODS: We utilized the R11 data from Finnish genome-wide associatio

Causal relationship between interleukin-6 levels and the risk of synovitis and tenosynovitis: A two-sample Mendelian randomization study.

Synovitis and tenosynovitis are common musculoskeletal disorders with multifactorial etiologies involving mechanical loading and inflammatory responses. Previous studies have suggested that interleukin-6 (IL-6) may play a critical role in tendon injury and repair; however, definitive evidence regarding a causal relationship between IL-6 levels and the risk of tenosynovitis remains lacking. This study employed a two-sample Mendelian randomization (MR) approach to evaluate the potential causal association between circulating IL-6 levels and the risk of tenosynovitis. Genetic instruments for IL-6

Severe infantile epileptic encephalopathy due to mutations in PLCB1: expansion of the genotypic and phenotypic disease spectrum.

Homozygous deletions of chromosome 20p12.3, disrupting the promoter region and first three coding exons of the phospholipase C β1 gene (PLCB1), have previously been described in two reports of early infantile epileptic encephalopathy (EIEE). Both children were born to consanguineous parents, one presented with infantile spasms, the other with migrating partial seizures of infancy. We describe an infant presenting with severe intractable epilepsy (without a specific EIEE electroclinical syndrome diagnosis) and neurodevelopmental delay associated with compound heterozygous mutations in PLCB1. Pathogenic genes associated with immune-related genes in graves' disease: a multi-omics Mendelian randomization analysis.

Immune-related genes (IRGs) have been implicated in autoimmune diseases, but their causal role in GD remains unclear. We conducted a multi-omics Mendelian randomization (MR) analysis integrating genome-wide association study (GWAS) data from GD patients with blood-based methylation quantitative trait loci(mQTLs), expression QTLs(eQTLs), and protein QTLs(pQTLs). SMR analysis was performed to assess the causal associations between IRGs and GD, followed by HEIDI tests to screen for pleiotropy. Colocalization analysis identified shared genetic determinants. We further validated the findings in Fin
