Alterations in the intestinal glycocalyx and bacterial flora in response to oral indomethacin.

Nonsteroidal anti-inflammatory drugs (NSAIDs), used extensively in clinical medicine, tend to cause adverse effects in the gastrointestinal tract. Earlier work has shown that oral administration of indomethacin produced oxidative damage in the small intestine and attenuation of the glycocalyx layer of the mucosa. The present study assessed, in greater detail, the alterations produced in the glycocalyx of rat small intestinal mucosa in response to indomethacin, with specific reference to surfactant-like particles (SLP) and brush border membranes (BBM). Changes in gut flora in response to the dr

Glomerular hyperfiltration and the salt paradox in early [corrected] type 1 diabetes mellitus: a tubulo-centric view.

Diabetes mellitus contributes greatly to morbidity, mortality, and overall health care costs. In major part, these outcomes derive from the high incidence of progressive kidney dysfunction in patients with diabetes making diabetic nephropathy a leading cause of end-stage renal disease. A better understanding of the early dysfunctions observed in the diabetic kidney may permit the development of new strategies to prevent diabetic nephropathy. This review proposes a "tubulo-centric" view of glomerular function in early type I diabetes mellitus. The following are particularly discussed (1) the pr

Astroglia-microvessel relationship in the developing human telencephalon.

The telencephalon of 12 and 18 week-old human foetuses was examined for evidence of astroglia-microvessel relationship. Immature astroglia cells (radial glia and astroblasts) and astrocytes were immunostained using antibodies to the cytoskeletal proteins vimentin (VIM) and glial fibrillary acidic protein (GFAP). The microvessels were detected using an antibody to the blood-brain barrier (BBB)-specific glucose transporter GLUT1. Two extracellular matrix (ECM) glycoproteins, laminin (LM), an endothelial-derived molecule, and tenascin-C (TN-C), a glia-derived molecule, were also analyzed. In the

Genetic determinants of type 2 diabetes mellitus: lessons learned from family studies.

Non insulin dependent diabetes mellitus (Type 2) is a multifactorial disease, with a polygenic inheritance and environmental factors contributing to its clinical expression. The search for the genetic determinants of Type 2 diabetes began when several genes involved in the mechanisms of insulin secretion or action were cloned, localized in the human genome, and when informative polymorphisms were described within or in the vicinity of these genes. It then became possible to compare, in groups of patients and normoglycaemic controls from various populations, the frequency of the different allel

Polyamine regulation of Na+/glucose symporter expression in LLC-PK1 cells.

Addition of polyamines or their analogs to newly confluent LLC-PK1 cells resulted in down-regulation of Na(+)-dependent glucose transport (symport) activity. Polyamines prevented the induction of this symporter by the differentiation inducer hexamethylene bisacetamide (HMBA) but did not influence induction by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX). Partial depletion of endogenous polyamines after addition of alpha-difluoromethylornithine (DFMO) resulted in a 4 to 5-fold increase in symporter expression. Symporter induction by either HMBA or DFMO was inhibited by the

Reduced glucose-6-phosphorylase and NADPH generating enzyme activities associated with glibenclamide induced hypoglycemia and hypoinsulinemia in genetically obese mice.

Addition of phenobarbital, an inducer of the liver mixed function oxidase system, to sulphonylurea regimen improves insulin sensitivity and intracellular glucose handling in patients with non-insulin dependent diabetes mellitus. The inducer also activates liver NADPH synthesis and its availability for mono-oxygenase reactions. In this study we further evaluated the mutual relationship between glucose and drug metabolism and the effect of sulphonylurea therapy by using genetically obese female mice. The mice were treated with glibenclamide, phenobarbital or both. Glibenclamide reduced blood glu

Ultrasonic modification on the physicochemical properties, structure characterizations, and intestinal barrier-protective activities of raspberry polysaccharides.

Natural polysaccharides possess diverse biological functions, yet their macromolecular architecture frequently restricts molecular accessibility and limits their application as health-promoting ingredients. Here, we employed an ultrasound-driven strategy to remodel the molecular structure of raspberry polysaccharide (RAP) and investigated how controlled depolymerization governs its intestinal barrier-protective properties. Native RAP was subjected to ultrasonic irradiation for 2 and 5 h, generating two structurally tailored derivatives, RAP-2 and RAP-5. Ultrasonic processing progressively decr

Polysaccharide-gut microbiota interactions in metabolic diseases: Structural selectivity, mediating mechanisms, and evidence deficiencies: A review.

Polysaccharides from food and medicinal sources are promising candidates for nutritional interventions in chronic metabolic diseases. Because intact polysaccharides are generally poorly absorbed after oral administration, their systemic effects cannot be fully explained by conventional models of absorption and direct action on target organs. Increasing attention has therefore focused on their gastrointestinal fate and on how microbial utilization and gut-derived metabolites may influence host metabolism. This review examines how molecular weight, monosaccharide composition, glycosidic linkage

Oleum cinnamomi disrupts Cutibacterium acnes adhesion and metabolism by covalently targeting enolase and coenzyme A.

BACKGROUND: Preventing Cutibacterium acnes (C. acnes) adhesion to human skin is essential for acne control, yet the molecular determinants of adhesion-associated drug resistance remain undefined and no pharmacological agents specifically blocking C. acnes attachment are available. OBJECTIVE: To evaluate oleum cinnamomi (OC), a natural cinnamon oil, as a potential anti-adhesive modality against C. acnes. METHODS: In vitro antibacterial assays, quantitative chemoproteomics, and keratinocyte infection models were integrated to quantify OC-mediated inhibition of C. acnes adhesion and biofilm for

Distinct structural features of neutral and acidic flaxseed gum polysaccharides confer complementary anti-hyperuricemic effects via multi-dimensional regulation of urate metabolism.

This study characterized flaxseed gum (FG) and its fractions, neutral (NFG) and acidic (AFG), elucidating their structure-activity relationships in hyperuricemia (HUA) mitigation. Monosaccharide, methylation, and NMR analyses identified NFG as a highly branched (1 → 4)-β-D-xylan, with its O-2/O-3 positions substituted with α-L-arabinofuranosyl, α-d-glucopyranosyl, and α-D-galactopyranosyl residues, while AFG exhibited a rhamnogalacturonan-I architecture comprising a repeating →2)-α-L-Rhap-(1 → 4)-α-D-GalpA-(1→ core. In HUA mice, oral administration of FG significantly decreased serum urate lev

From extreme environments to skin interfaces: thermophilic exopolysaccharides as smart and sustainable polymers for advanced cosmeceutical and transdermal applications.

Microbial exopolysaccharides (EPS) possess strong potential for next-generation biomaterials due to their sustainability, biodegradability, and biocompatibility as the promising replacements for fossil-derived polymers. Conventional synthetic and semi-synthetic polymers have been used for pharmaceutical and cosmetic applications but due to their limitations-particularly in advanced skin-interface technologies-the need for safe and environmentally friendly alternatives has emerged. Thermophilic microorganisms, which have high growth activity in extreme conditions and synthesize extracellular po

Environmental antibiotic and fungicide exposure differentially affects survival and carbohydrate-fueled flight performance in Apis mellifera.

Environmental contamination by antibiotics and fungicides is an emerging concern due to their widespread occurrence in agricultural ecosystems. The western honey bee, Apis mellifera, a key managed pollinator, is exposed to these contaminants through multiple pathways, including treated crops, contaminated water, and residues from apiculture, livestock waste, agricultural runoff, and wastewater discharges. The agrochemicals tetracycline and boscalid disrupt cellular energy metabolism and mitochondrial function in non-target organisms and may consequently compromise bee survival and the sustaine

Novel acidic β-glucans from Ganoderma lucidum: Structural characterization and molecular-weight-related antioxidant activity.

Ganoderma lucidum is a widely consumed edible-medicinal fungus and an important source of bioactive polysaccharides. However, how molecular weight and peripheral structural features relate to the antioxidant activity of its acidic β-glucans remains unclear. Here, three homologous acidic β-glucans, GLP-2a-2c, with molecular weights of 4.69-15.57 kDa, were isolated from G. lucidum fruiting bodies. HPGPC, monosaccharide profiling, UV/FT-IR spectroscopy, methylation-GC-MS, and 800 MHz 1D/2D NMR analyses identified multiple β-linked Glcp residue types, GlcA, and minor Fuc-, Ara-, and Man-related re

Medicinal Plant Polysaccharides as Microbiota-Directed Modulators of Immunosenescence: Structural Determinants, Metabolite Reprogramming, and Host Immune Regulation.

Immunosenescence is a major contributor to age-associated morbidity, yet microbiota-directed strategies capable of restoring immune homeostasis remain insufficiently validated. Medicinal plant polysaccharides (MPPs) are structurally diverse macromolecules that often resist host digestion and undergo microbial transformation in the colon, but their effects cannot be interpreted as those of a homogeneous intervention class. In this structured narrative review, we critically synthesize evidence across structural carbohydrate biology, microbial ecology, metabolite signaling, and immune aging and p

Irritable Bowel Syndrome: Contemporary Management Approaches, Limitations, and Future Directions.

Irritable bowel syndrome (IBS) is a common disorder of gut-brain interaction characterized by recurrent abdominal pain associated with altered bowel habits in the absence of structural disease. Despite its high prevalence and substantial impact on quality of life, healthcare utilization, and long-term symptom burden, IBS management remains challenging because of its heterogeneous pathophysiology and variable treatment response. Current evidence supports a multifactorial model involving altered motility, visceral hypersensitivity, dysregulated brain-gut signaling, mucosal immune activation, mic

Compositional and Functional Characterization of Glycolipid Biosurfactants from an Olive-Derived Lactiplantibacillus plantarum.

Biosurfactants from lactic acid bacteria are attractive as safe, multifunctional ingredients for food and cosmetic applications, yet many remain only partially characterized. An olive-derived strain was identified as Lactiplantibacillus plantarum J2K-1229 by 16S rRNA gene and genome-based phylogeny. Grown in rapeseed oil-containing medium, it secreted extracellular material with surface/interfacial activity, and its cell-free supernatant emulsified diverse plant oils (emulsification index after 24 h, 26.67-41.33%). A purified biosurfactant fraction gave carbohydrate- and lipid-positive, ninhyd
