AQP7 suppresses cell stemness and metastasis by targeting Wnt signaling in basal-like breast cancer.

BACKGROUND: Breast cancer is a leading cause of cancer-related death among women worldwide. Aquaporin 7 (AQP7), a transmembrane protein facilitating the transport of glycerol, and/or water across plasma membranes, has been found the aberrant expression and dysfunction in various types of human cancer. RESEARCH DESIGN AND METHODS: In this study, we revealed an aberrant downregulation of AQP7 in basal-like breast cancer. We performed functional assays in vitro and in vivo using basal-like human breast cancer cells with or without expression of AQP7, including cancer cell stemness, cell migratio

LINC00908 Inactivates Wnt/β-Catenin Signaling Pathway to Inhibit Prostate Cancer Cell Stemness via Upregulating GSK3B and FBXW2.

BACKGROUND: Chemotherapy and androgen-deprivation treatment are the curative approaches utilized to suppress prostate cancer (PCa) progression. However, drug resistance and metastasis are extensive and hard to overcome even though remarkable progress has been made in recent decades. The cancer stem cell-related theoretical model explains the distinct molecular characteristics of cancer, its relapse, metastasis, and drug resistance. Meanwhile, noncoding RNA functions in the formation of drug resistance and metastasis in most cancers. The long intergenic nonprotein coding RNA 908 (LINC00908) has

Ovulation and extra-ovarian origin of ovarian cancer.

The mortality rate of ovarian cancer remains high due to late diagnosis and recurrence. A fundamental step toward improving detection and treatment of this lethal disease is to understand its origin. A growing number of studies have revealed that ovarian cancer can develop from multiple extra-ovarian origins, including fallopian tube, gastrointestinal tract, cervix and endometriosis. However, the mechanism leading to their ovarian localization is not understood. We utilized in vitro, ex vivo, and in vivo models to recapitulate the process of extra-ovarian malignant cells migrating to the ovari

Cancerous ovarian stem cells: obscure targets for therapy but relevant to chemoresistance.

Chemotherapy with platinum and taxanes is the first line of treatment for all epithelial ovarian cancer (EOC) patients after debulking surgery. Even though the treatment is initially effective in 80% of patients, recurrent cancer is inevitable in the vast majority of cases. Emerging evidence suggests that some tumor cells can survive chemotherapy by activating the self-renewal pathways resulting in tumor progression and clinical recurrence. These defined population of cells commonly termed as "cancer stem cells" (CSC) may generate the bulk of the tumor by using differentiating pathways. These

A locoregional program of photothermal debulking, stemness suppression, and dendritic cell activation for durable glioblastoma immunotherapy.

Glioblastoma (GBM), especially in large or unresectable tumors, remains difficult to treat because of the lack of effective debulking, the persistence of therapy-resistant glioma stem cells (GSCs), and a profoundly immunosuppressive tumor microenvironment. Here, a locoregional hydrogel platform is developed to integrate photothermal debulking, post-ablation GSC suppression, and dendritic-cell-mediated immune activation for GBM therapy. The brain-compliant injectable hydrogel (MIN-PPIC@iGel) co-encapsulates indocyanine green/napabucasin-loaded micelles and poly(I:C)-loaded polymersomes, enablin

Targeting Lung Cancer Stem Cells with Trimebutine: Synergistic Antitumor Activity in Combination with Cisplatin.

Lung cancer recurrence and therapeutic resistance are driven by cancer stem cells (CSCs), yet clinically available CSC-targeted therapies remain lacking. To identify actionable anti-CSC agents, we screened 1018 FDA-approved drugs using a 3D sphere culture system enriching lung CSC-like cells. Trimebutine, a gastrointestinal motility regulator targeting G-protein-coupled receptors (GPCRs), was identified as a potent agent with over 49.5-fold higher selectivity toward lung CSC-like cells (SI > 49.55) compared with standard therapies such as cisplatin (SI = 1.39) and gefitinib (SI = 1.00). In H46

Exploiting metabolic vulnerability in glioblastoma using a brain-penetrant drug with a safe profile.

Glioblastoma is one of the most treatment-resistant and lethal cancers, with a subset of self-renewing brain tumour stem cells (BTSCs), driving therapy resistance and relapse. Here, we report that mubritinib effectively impairs BTSC stemness and growth. Mechanistically, bioenergetic assays and rescue experiments showed that mubritinib targets complex I of the electron transport chain, thereby impairing BTSC self-renewal and proliferation. Gene expression profiling and Western blot analysis revealed that mubritinib disrupts the AMPK/p27Kip1 pathway, leading to cell-cycle impairment. By employin

M2 Receptor Activation Counteracts the Glioblastoma Cancer Stem Cell Response to Hypoxia Condition.

Glioblastoma multiforme (GBM) is the most malignant brain tumor. Hypoxic condition is a predominant feature of the GBM contributing to tumor growth and resistance to conventional therapies. Hence, the identification of drugs able to impair GBM malignancy and aggressiveness is considered of great clinical relevance. Previously, we demonstrated that the activation of M2 muscarinic receptors, through the agonist arecaidine propargyl ester (Ape), arrests cell proliferation in GBM cancer stem cells (GSCs). In the present work, we have characterized the response of GSCs to hypoxic condition showing

p75 neurotrophin receptor: A potential surface marker of tongue squamous cell carcinoma stem cells.

The present study detected p75 neurotrophin receptor (p75NTR) expression in tongue squamous cell carcinoma (TSCC) cell lines, in order to define the biological properties of p75NTR+ cells and to confirm the use of p75NTR+ as a surface marker for TSCC stem cells. p75NTR+ cells were separated from Tca‑8113 and CAL‑27 TSCC cells by fluorescence‑activated cell sorting. Colony formation, MTT and scratch assays, and a tumorigenicity analysis were performed to measure self-renewal and proliferation, multidirectional differentiation, and tumorigenicity of p75NTR+ cells. p75NTR+ cells comprised 3.1 and

Changes in the Relative Number of SP Cells of Melanoma Line B16 after Radiation Exposure in vivo.

The quantitative regularities of changes of cancer stem cell (CSC) population were explored after local γ-irradiation of experimental tumors (murine melanoma line B16). CSCs were detected by the ability of these cells to exclude Hoechst 33342 fluorescent dye and to form a so-called side population (SP) under flow cytometry study. In the control group of unexposed mice a positive correlation was found between the proportion of CSCs (SP) and tumorweight at the initial stage of growth (R = 0.77, p = 0.009). In the advanced stages of tumor growth similar relationship was not revealed. Statisticall

9-O-acetylated sialic acids differentiating normal haematopoietic precursors from leukemic stem cells with high aldehyde dehydrogenase activity in children with acute lymphoblastic leukaemia.

Источник пометил публикацию как отозванную

Childhood acute lymphoblastic leukaemia (ALL) originates from mutations in haematopoietic progenitor cells (HPCs). For high-risk patients, treated with intensified post-remission chemotherapy, haematopoietic stem cell (HSC) transplantation is considered. Autologous HSC transplantation needs improvisation till date. Previous studies established enhanced disease-associated expression of 9-O-acetylated sialoglycoproteins (Neu5,9Ac2-GPs) on lymphoblasts of these patients at diagnosis, followed by its decrease with clinical remission and reappearance with relapse. Based on this differential express

PHA for aplastic anemias: the alpha but not the omega of mitogen therapies.

This manuscript updates an analysis of the foremost clinical investigation of PHA for aplastic anemias and other disorders associated with impaired hematopoiesis during the years 1963-1967. Humble displayed remarkable insight in motivating the trials long before the recent era during which these disorders have become much better understood. That investigation actually consisted of a series of small pilot studies totaling only 44 patients involving all categories in the aplastic group and 15 miscellaneous disorders mostly neoplastic or premalignant. The most serious of several faults with the s

Sarcomatoid neoplasms of the lung and pleura.

Sarcomatoid neoplasms of the lung and pleura are rare tumors that present a complex differential diagnosis, making them challenging for surgical pathologists. In the lung, the main tumors are the sarcomatoid carcinomas, including pleomorphic carcinoma, spindle cell carcinoma, giant cell carcinoma, carcinosarcoma, and pulmonary blastoma. They are characterized by histologic heterogeneity; molecular data support their origin from a pluripotent stem cell that undergoes neoplastic transformation with divergent epithelial and sarcomatous differentiation. Diagnosis is difficult in small biopsy speci

Three-dimensional platform-dependent modulation of stemness and EMT-associated gene expression in primary and metastatic uveal melanoma models.

Uveal melanoma is the most common primary intraocular malignancy in adults and is associated with high rates of metastatic progression. Although local tumour control is often achieved, effective therapies for metastatic disease remain limited. Three-dimensional culture systems more closely replicate aspects of the tumour microenvironment than conventional two-dimensional monolayers; however, differences between platforms and their impact on stemness- and epithelial-to-mesenchymal transition-associated transcription in uveal melanoma remain incompletely defined. Primary-derived and metastatic-d

METTL3-mediated m6A modification of LINC00857 enhances stemness and metastasis of ovarian cancer cells by activating the YAP-TEAD pathway.

This study was designed to illustrate the mechanism underlying the up-regulation of LINC00857 expression and to identify novel molecular targets for the treatment of ovarian cancer (OC). The LINC00857 and methyltransferase-like 3 (METTL3) expression levels were observed in clinical OC tissues and cells, and their correlation within tissues was analyzed. To further explore the relationship between LINC00857 and METTL3, a combined transfection of pcDNA3.1-LINC00857 and METTL3 siRNA was performed. Subsequently, cell invasion, viability, migration, and sphere-forming capabilities were assessed usi
