Selective calcium imaging of olfactory interneurons in a land mollusk.

The procerebrum (PC) of the land slug Limax is the olfactory center involved in olfactory discrimination and learning. The PC contains two types of neurons, bursting (B) and nonbursting (NB) neurons. B neurons are local interneurons and show spontaneous bursting while NB neurons directly receive input from olfactory nerve fibers, and are possibly involved in odor coding. In order to observe activity of NB neurons, selective calcium imaging was made by retrograde staining of NB neurons with the calcium sensitive dye, rhod-2 AM, from the internal mass (IM) of the PC. The stained cells, presumabl

Reading cinnamon activates olfactory brain regions.

Some words immediately and automatically remind us of odours, smells and scents, whereas other language items do not evoke such associations. This study investigated, for the first time, the abstract linking of linguistic and odour information using modern neuroimaging techniques (functional MRI). Subjects passively read odour-related words ('garlic', 'cinnamon', 'jasmine') and neutral language items. The odour-related terms elicited activation in the primary olfactory cortex, which include the piriform cortex and the amygdala. Our results suggest the activation of widely distributed cortical

A battery of olfactory-based screens for phenotyping the social and sexual behaviors of mice.

We provide a battery of screens that reflect the social and sexual behavior displayed by both captive and feral mice. Some screens focus on measuring aspects of chemical communication, providing information about whether or not mice will interact and if they do so, predict the nature of the interaction. Other screens measure direct interactions between target mice and same- and opposite-sex conspecifics, providing an indication of the social status and sexual responsiveness, respectively, of target mice. The battery of screens yields a high-throughput bioassay of a mouse's relative social stat

Chemosensory and hormone information are relayed directly between the medial amygdala, posterior bed nucleus of the stria terminalis, and medial preoptic area in male Syrian hamsters.

In many rodent species, including Syrian hamsters, the expression of appropriate social behavior depends critically on the perception and identification of conspecific odors. The behavioral response to these odors is mediated by a network of steroid-sensitive ventral forebrain nuclei including the medial amygdala (Me), posterior bed nucleus of the stria terminalis (BNST), and medial preoptic area (MPOA). Although it is well-known that Me, BNST, and MPOA are densely interconnected and each uniquely modulates odor-guided social behaviors, the degree to which conspecific odor information and ster

Distinct electrophysiological properties in subtypes of nonspiking olfactory local interneurons correlate with their cell type-specific Ca2+ current profiles.

A diverse population of local interneurons (LNs) helps to process, structure, and spatially represent olfactory information in the insect antennal lobe. In Periplaneta americana, we identified two subtypes of nonspiking local interneurons (type II LNs) by their distinct morphological and intrinsic electrophysiological properties. As an important step toward a better understanding of the cellular mechanisms that mediate odor information processing, we present a detailed analysis of their distinct voltage-activated Ca(2+) currents, which clearly correlated with their distinct intrinsic electroph

Pathogenesis of aerosolized Eastern Equine Encephalitis virus infection in guinea pigs.

Mice and guinea pigs were experimentally exposed to aerosols containing regionally-distinct strains (NJ1959 or ArgM) of eastern equine encephalitis virus (EEEV) at two exclusive particle size distributions. Mice were more susceptible to either strain of aerosolized EEEV than were guinea pigs; however, clinical signs indicating encephalitis were more readily observed in the guinea pigs. Lower lethality was observed in both species when EEEV was presented at the larger aerosol distribution (> 6 mum), although the differences in the median lethal dose (LD50) were not significant. Virus isolation

Neural substrates of olfactory processing in schizophrenia patients and their healthy relatives.

Odorants represent powerful stimuli capable of eliciting various emotional responses. In schizophrenia patients and their non-affected relatives, olfactory and emotional functions are impaired, revealing a familial influence on these deficits. We aimed at determining the neural basis of emotional olfactory dysfunctions using odors of different emotional valence for mood induction and functional magnetic resonance imaging (fMRI) by comparing 13 schizophrenia patients, their non-affected brothers and 26 matched healthy controls. Blood-oxygen-level-dependent (BOLD) effects and subjective mood cha

Calretinin immunoreactivity in the prenatally developing olfactory systems of the tree shrew Tupaia belangeri.

The prenatal patterns of calretinin immunoreactivity were studied in the olfactory systems of Tupaia belangeri. We investigated the peripheral and primary central parts of the vomeronasal system and of the main olfactory system from the 19th to the 43rd (last) day of gestation and compared the findings with the known calretinin immunoreactivity patterns in adult T. belangeri and the published data on other mammals. The onset of calretinin immunoreactivity was noted in the main olfactory system on the 23rd day of gestation and, in the vomeronasal system, on the 25th day of gestation: single pre

Functional mapping of human brain in olfactory processing: a PET study.

This study describes the functional anatomy of olfactory and visual naming and matching in humans, using positron emission tomography (PET). One baseline control task without olfactory or visual stimulation, one control task with simple olfactory and visual stimulation without cognition, one set of olfactory and visual naming tasks, and one set of olfactory and visual matching tasks were administered to eight normal volunteers. In the olfactory naming task (ON), odors from familiar items, associated with some verbal label, were to be named. Hence, it required long-term olfactory memory retriev

Contrasting neurochemical interactions of tiletamine, a potent phencyclidine (PCP) receptor ligand, with the N-methyl-D-aspartate-coupled and -uncoupled PCP recognition sites.

Neurochemical interactions of tiletamine, a potent phencyclidine (PCP) receptor ligand, with the N-methyl-D-aspartate (NMDA)-coupled and -uncoupled PCP recognition sites were examined. Tiletamine potently displaced the binding of [3H]1-(2-thienyl)cyclohexylpiperidine with an IC50 of 79 nM without affecting sigma-, glycine, glutamate, kainate, quisqualate, or dopamine (DA) receptors. Like other PCP ligands acting via the NMDA-coupled PCP recognition sites, tiletamine decreased basal, harmaline-, and D-serine-mediated increases in cyclic cGMP levels and induced stereotypy and ataxia. Tiletamine

Cortical interneurons become activated by deafferentation and instruct the apoptosis of pyramidal neurons.

Unlike peripheral nervous system neurons and certain groups of nerve cells in the CNS, cortical projection neurons are tolerant of axonal lesions. This resistance is incongruent with the massive death of pyramidal neurons in age-associated neurodegenerative diseases that proceed along corticocortical connections. Some insights have emerged from our previous work showing that pyramidal cells in piriform cortex undergo classical apoptosis within 24 h after bulbectomy via transsynaptic, but not retrograde, signaling. These findings allow the investigation of cellular and molecular changes that ta

Balenine alleviates neurodegeneration and inflammation in a mouse model of Parkinson's disease.

Carnosine, an imidazole dipeptide, has potential for treating neurodegenerative diseases, including Parkinson's disease. However, carnosine-degrading enzymes limit its bioavailability. In this study, we established a mouse model of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine hydrochloride (MPTP)-induced Parkinson's disease and intranasally administered balenine, a carnosine analog featuring a methylated imidazole that is resistant to degradation enzymes, to target the brain via the olfactory epithelium. MPTP + balenine-treated mice demonstrated improved recognition scores in the object locati

Nose to brain delivery of nanosuspensions with first line antiviral agents is alternative treatment option to Neuro-AIDS treatment.

Intranasal drug delivery is one of the uprising areas of the research in targeting drug to the brain. Nose to brain drug delivery follows the olfactory pathway and purportedly known to be more efficient to deliver neuro-therapeutics to the brain by circumventing the BBB and thereby increasing bioavailability of drugs in the brain. The advantage of this method is non-invasiveness, rapid onset of action and helps to achieve site specific delivery. In this research work nanosuspension were prepared using combination of antiretroviral agents for Neuro-AIDS treatment. Nanosuspensions were prepared

Proliferation activity in the adult rat brain following exposure to ionizing radiation.

BACKGROUND: The aim of our study was to investigate radiationinduced shortterm effects on the rat forebrain. MATERIAL AND METHODS: Adult male Wistar rats received whole body exposure with fractionated doses of gamma rays (a total dose of 3 Gy) and were investigated seven and 14 days later. Immunohistochemistry and confocal microscopy were used to determine proliferating cells derived from anterior subventricular zone (SVZa) and distributed along the subventricular zone olfactory bulb axis (SVZ OB axis). Cell counting was performed in four anatomical parts along the welldefined pathway, known

Effect of non-selective dopaminergic receptor agonist on disrupted maternal behavior in olfactory bulbectomized mice.

Olfactory bulbectomy (OBX) animals are considered a putative model of depression that produces behavioral, physiological, and neurochemical alterations resembling clinical depression. Depression is a critical cause of child abuse and neglect, and it has been reported that maternal behavior involves dopaminergic neurons of the mesolimbic pathway. In this study, we investigated the effect of apomorphine, a non-selective dopaminergic receptor agonist, on maternal behavior to examine the influence of activated brain dopaminergic function in OBX mice. In addition, we conducted the sucrose preferenc

X chromosome-linked Kallmann syndrome: stop mutations validate the candidate gene.

Kallmann syndrome represents the association of hypogonadotropic hypogonadism with anosmia. This syndrome is from a defect in the embryonic migratory pathway of gonadotropin-releasing hormone synthesizing neurons and olfactory axons. A candidate gene for the X chromosome-linked form of the syndrome was recently isolated by using a positional cloning strategy based on deletion mapping in the Xp22.3 region. With the PCR, two exons of this candidate gene were amplified on the genomic DNAs from 18 unrelated patients affected with the X chromosome-linked Kallmann syndrome. Three different base tran

On the Larval Sensory Organs and Central Nervous System of the Zoea 1 of the Asian Shore Crab Hemigrapsus sanguineus (Decapoda, Brachyura).

Many decapod crustaceans occupy different habitats during their ontogeny and undergo a niche shift during the transition from the larval to the juvenile phase. Their life cycle comprises pelagic larvae and, in most species, benthic juvenile-adult stages. Already at hatching, decapod larvae possess a wealth of organ systems, necessary to autonomously survive and develop in the plankton. They also exhibit a rich behavioral repertoire to appropriately respond to environmental stimuli such as light, gravity, hydrostatic pressure, tidal currents, temperature, salinity, chemical cues, and food conce

Assessing the feasibility of intranasal radiotracer administration for in brain PET imaging.

INTRODUCTION: The development of clinically useful tracers for PET imaging is enormously challenging and expensive. The intranasal (IN) route of administration is purported to be a viable route for delivering drugs to the brain but has, as yet, not been investigated for the delivery of PET tracers. If the intranasal (IN) pathway presents a viable option, it extends the PET imaging field by increasing the number of tracers available for human use. Here we report the results of a rodent study testing the feasibility of the IN route to administer radiotracers for brain PET imaging. METHODS: We u
