Evaluating Dried Blood Spot and Dried Blood Matrix as Alternatives to Venipuncture for Diagnosing Haemoglobinopathies in Adults: A Mass Spectrometric Approach.

Microsampling techniques such as dried blood spot (DBS) and dried blood matrix (DBM) offer several advantages over venipuncture sampling tec … and δ/β (%) for β-thalassemia-were analyzed. All the sampling techniques successfully distinguished sickle cell trait from sickle cell disease, β-thalassemia trait from β-thalassemia major and detected HbE and HbD-Punjab traits. There were no statistically significant differences between sampling techniques in the qualitative detection of HbS and HbD-Punjab variant …

Candida splenic abscess in the absence of obvious immunodeficiency.

Focal hepatosplenic candidiasis has been increasingly recognized as a variant of disseminated candidiasis in immunocompromised patients. It rarely presents as Candida splenic abscess. Its occurrence in non-leukemic patients is quite rare. A patient with diabetes mellitus and previously undiagnosed sickle cell trait presented with upper gastrointestinal bleeding. Candida splenic abscess was diagnosed. She was successfully treated with percutaneous drainage and fluconazole. The clinical presentation, diagnosis, and treatment of Candida splenic abscess is reviewed, and its relationship to diabete…

[Clinical hematological and biochemical pattern of three SN Baltimore hemoglobin carriers (author's transl)].

The clinical, hematologic and biochemical study of three children, carriers of the double heterozygotous SN Baltimore hemoglobin from Guadeloupe, have not allowed to detect any typical anomaly and specially no sign of anemia or hemolysis. An iron deficiency of parasitologic origin has only been detected. This absence of pathological symptoms is related to the fact that SN red cells tend to sickle less rapidly than those from persons with sickle cell trait AS.

Sickle cell rheology is determined by polymer fraction--not cell morphology.

Sickle cell disease pathophysiology is mediated by acute and chronic impairment of cell flexibility due to the formation of intracellular sickle hemoglobin (Hb S) polymer as cells are partially deoxygenated in the microcirculation. We have recently developed a method to measure the relationship between the formation of intracellular polymerized Hb S and cell filtration. In this study, we have used this method to examine whether sickle cell morphology, independent of Hb S polymer fraction, had an effect on cell rheology. We primarily use sickle trait (AS) and Hb S-beta(+)-thalassemia (S-beta(+)

Alpha-thalassemia in blacks: genetic and clinical aspects and interactions with the sickle hemoglobin gene.

Recent advances in molecular genetics have permitted detailed study of the human alpha-globin genes and the causes of alpha-thalassemia. In this review, we examine the causes of alpha-thalassemia in the black population and the consequences of the interactions between alpha-thalassemia and structural hemoglobin variants.

Textural differences between AA and SS blood specimens as detected by image analysis.

High-resolution image analysis was employed in the analysis of round (discoid) erythrocytes from hematologically normal (AA) individuals, AA individuals with nonspecific anemia, individuals with sickle cell trait (AS), individuals with SC disease (SC), and individuals with sickle cell anemia (SS). The shape feature Form Factor (4 pi Area/Perimeter2) was used to select round cells and to exclude sickle and other abnormal cells. Textural features extracted from round cells of SS and SC patients were found to differ from those derived from cells of normal andanemic AA individuals. Two textural fe…

Compound heterozygosity for hemoglobin S and hemoglobin E in a family of Proto-Australoid origin: a case report.

BACKGROUND: Hemoglobin S and E are commonly occurring hemoglobin variants among distinctly separate tribal populations of Central and Northeast India, respectively. Combined heterozygosity for hemoglobin S and E or hemoglobin SE disease is a benign clinical condition with rare incidence. Reports of approximately 46 hemoglobin SE cases are available worldwide. We conducted a screening program to study the prevalence of hemoglobin variants among the tribal population working in the tea estates of Northeast India. A total of 551 subjects were screened, and complete blood count was performed. Base

Discovering the genetics underlying foetal haemoglobin production in adults.

Sickle cell disease (SCD) and beta thalassaemia, caused by lesions that affect the HBB (beta globin gene), form the most common human genetic disorders world-wide, and represent a major public health problem. Inter-individual variation in foetal haemoglobin (HbF) expression is a known and heritable disease modifier; high HbF levels are correlated with reduced morbidity and mortality in both diseases. This review traces our progress in the understanding of the persistence of HbF in adults as a quantitative trait and the genetic approaches used in teasing out the loci contributing to its variabi

Erythrocyte membrane enzymes in sickle cell anemia. 2. Acetylcholinesterase and ATPase activities.

The activity level of acetylcholinesterase in the erythrocytes of 32 patients homozygous for sickle cell anemia was determined and compared with that of normal AA controls as well as with that of AS individuals. Acetylcholinesterase activity was markedly higher in erythrocyte membrane from SS individuals than in those from AS individuals or AA controls. Additionally, ATPase activities were also significantly higher in sickle cell erythrocytes as compared to normal cells. These higher values of acetylcholinesterase and ATPase activities in SS erythrocytes may be explained as a consequence of th

Massive sequestration of human sickle cells after transfusion to a baboon.

A baboon was exchange-transfused with sickle cell anemia patients' blood. The animal died suddenly, and postmortem examination showed widespread red cell sequestration, particularly in the spleen and liver. The clinical and pathological findings were similar to those in children with sickle cell anemia who die of acute splenic sequestration syndrome. A control animal, exchange-transfused with normal human blood, tolerated the procedure without difficulties for a period of 4 days, when a delayed transfusion reaction occurred. Thus the baboon can be used as a model for the abnormal circulatory b

[José Martins da Cruz Jobim: his priority in the detection of an hereditary hematologic disorder in African Negro slaves (author's transl)].

A brazilian physician, José Martins da Cruz Jobim, may be considered the author of one of the earliest medical reports with a story of the prevalence of the sikle cell trait among african negroes who came to Brazil as slaves, by observing their rather remarkable resistance against malarial infections than other sets of our population. Naturally, his report, published in 1835, must be considered by the actual scientific concepts.
