Identification of potential novel proteomic markers of Leishmania spp.-derived exosomes.

INTRODUCTION: Extracellular vesicles (EVs) are heterogenous cell-derived membrane-bound structures which can be subdivided into three distinct classes according to distinct morphological characteristics, cellular origins, and functions. Small EVs, or exosomes, can be produced by the protozoan parasite Leishmania through the evolutionarily conserved ESCRT pathway, and act as effectors of virulence and drivers of pathogenesis within mammalian hosts. Techniques for the identification of EVs of non-mammalian origin, however, remain inaccurate in comparison to their well-characterized mammalian cou

Fluorescent nano- and microparticles for sensing cellular microenvironment: past, present and future applications.

The tumor microenvironment (TME) demonstrates distinct hallmarks, including acidosis, hypoxia, reactive oxygen species (ROS) generation, and altered ion fluxes, which are crucial targets for early cancer biomarker detection, tumor diagnosis, and therapeutic strategies. Various imaging and sensing techniques have been developed and employed in both research and clinical settings to visualize and monitor cellular and TME dynamics. Among these, ratiometric fluorescence-based sensors have emerged as powerful analytical tools, providing precise and sensitive insights into TME and enabling real-time

Parasocial interactions with media characters: the role of perceived and actual sociodemographic and psychological similarity.

INTRODUCTION: Similarity between media character and viewer is an important predictor of parasocial interactions. Thereby, similarities are often limited to single characteristics or to the similarities viewers perceive between themselves and characters. This article expands the existing literature in two ways. First, the effects of actual and perceived similarity on parasocial interactions are compared. Second, similarity is understood in a broad way. With age, gender, job, relationship, and living situation are assessed for sociodemographic similarities. Psychological similarities are consid

Design and Analysis of XY Large Travel Micro Stage Based on Secondary Symmetric Lever Amplification.

This study presents a newly developed piezoelectric drive mechanism for the purpose of designing, analyzing, and testing a micro-positioning platform driven by piezoelectric actuators. The platform incorporates a piezoelectric ceramic actuator and a flexible hinge drive and features a symmetrical two-stage lever (STSL) amplification mechanism and a parallelogram output structure. The implementation of this design has led to notable enhancements in the dynamic properties of the platform, thereby eliminating the undesired parasitic displacement of the mechanism. An analytical model describing th

Retina Oculomics in Neurodegenerative Disease.

Ophthalmic biomarkers have long played a critical role in diagnosing and managing ocular diseases. Oculomics has emerged as a field that utilizes ocular imaging biomarkers to provide insights into systemic diseases. Advances in diagnostic and imaging technologies including electroretinography, optical coherence tomography (OCT), confocal scanning laser ophthalmoscopy, fluorescence lifetime imaging ophthalmoscopy, and OCT angiography have revolutionized the ability to understand systemic diseases and even detect them earlier than clinical manifestations for earlier intervention. With the advent

Olfactory Drug Aerosol Delivery with Acoustic Radiation.

Nose-to-brain (N2B) drug delivery is a new approach to neurological disorder therapy as medications can bypass the blood-brain barrier and directly enter the brain. However, the delivery efficiency to the olfactory region using the conventional delivery method is impractically low because of the region's secluded position in a convoluted nasal cavity. In this study, the acoustic radiation force was explored as an N2B delivery alternative in a wide frequency range of 10-100,000 Hz at an increment of 50 Hz. Numerical simulations of the particle deposition in the olfactory region of four nasal co

Space and Time Coherent Mapping for Subcellular Resolution of Imaging Mass Spectrometry.

Space and time coherent mapping (STCM) is a technology developed in our laboratory for improved matrix-assisted laser desorption ionization (MALDI) time of flight (TOF) imaging mass spectrometry (IMS). STCM excels in high spatial resolutions, which probe-based scanning methods cannot attain in conventional MALDI IMS. By replacing a scanning probe with a large field laser beam, focusing ion optics, and position-sensitive detectors, STCM tracks the entire flight trajectories of individual ions throughout the ionization process and visualizes the ionization site on the sample surface with a subce

Historic and Prehistoric Epidemics: An Overview of Sources Available for the Study of Ancient Pathogens.

Since life on earth developed, parasitic microbes have thrived. Increases in host numbers, or the conquest of a new species, provide an opportunity for such a pathogen to enjoy, before host defense systems kick in, a similar upsurge in reproduction. Outbreaks, caused by "endemic" pathogens, and epidemics, caused by "novel" pathogens, have thus been creating chaos and destruction since prehistorical times. To study such (pre)historic epidemics, recent advances in the ancient DNA field, applied to both archeological and historical remains, have helped tremendously to elucidate the evolutionary t

Next-Generation Cancer-Specific Hybrid Theranostic Nanomaterials: MAGE-A3 NIR Persistent Luminescence Nanoparticles Conjugated to Afatinib for In Situ Suppression of Lung Adenocarcinoma Growth and Metastasis.

The rate of lung cancer has gradually increased in recent years, with an average annual increase of 15%. Afatinib (AFT) plays a key role in preventing non-small cell lung carcinoma (NSCLC) growth and spread. To increase the efficiency of drug loading and NSCLC cell tracking, near infrared-persistent luminescence nanomaterials (NIR PLNs), a silica shell-assisted synthetic route for mono-dispersal, are developed and used in the nanovehicle. After optimizing their physical and chemical properties, the NIR PLNs are able to absorb light energy and emit NIR luminescence for several hours. In this re

Setting the stage for a Global Programme to Eliminate Lymphatic Filariasis: the first 125 years (1875-2000).

The development of the World Health Organization's Global Programme to Eliminate Lymphatic Filariasis (GPELF) can be interpreted through many different lenses-e.g. one focusing on the health or economic plight of affected individuals and populations, another tracking the individuals and organizations responsible for building the programme or, as in this review, one identifying each of the critical requirements and specific hurdles that need to be addressed in order to successfully construct the programme. For almost 75 y after the life cycle of LF was first described, the principal tool for co

Dynamic off-resonance correction for spiral real-time MRI of speech.

PURPOSE: To improve the depiction and tracking of vocal tract articulators in spiral real-time MRI (RT-MRI) of speech production by estimating and correcting for dynamic changes in off-resonance. METHODS: The proposed method computes a dynamic field map from the phase of single-TE dynamic images after a coil phase compensation where complex coil sensitivity maps are estimated from the single-TE dynamic scan itself. This method is tested using simulations and in vivo data. The depiction of air-tissue boundaries is evaluated quantitatively using a sharpness metric and visual inspection. RESULT

Antibody repertoire analysis in polygenic autoimmune diseases.

High-throughput sequencing of the DNA/RNA encoding antibody heavy- and light-chains is rapidly transforming the field of adaptive immunity. It can address key questions, including: (i) how the B-cell repertoire differs in health and disease; and (ii) if it does differ, the point(s) in B-cell development at which this occurs. The advent of technologies, such as whole-genome sequencing, offers the chance to link abnormalities in the B-cell antibody repertoire to specific genomic variants and polymorphisms. Here, we discuss the current research using B-cell antibody repertoire sequencing in three

Closure of a Traumatic Esophagomediastinal Fistula in a Child by Endoscopic Fulguration and Fibrin Injection.

Posttraumatic esophagomediastinal fistula is an uncommon clinical entity that warrants surgical awareness due to its life-threatening potential. Its management, especially in previously operated field, is controversial and several endoscopic methods are being proposed as alternatives. Ours is the first report of endoscopic fulguration and fibrin injection in successful closure of such fistula. A 9-year-old female sustained complete tracheoesophageal transection from a gunshot wound to the neck and underwent immediate primary repair. She presented nine months later with fevers and swelling over

Estimation of Cardiopulmonary Parameters From Ultra Wideband Radar Measurements Using the State Space Method.

Ultra wideband (UWB) Doppler radar has many biomedical applications, including remote diagnosis of cardiovascular disease, triage and real-time personnel tracking in rescue missions. It uses narrow pulses to probe the human body and detect tiny cardiopulmonary movements by spectral analysis of the backscattered electromagnetic (EM) field. With the help of super-resolution spectral algorithms, UWB radar is capable of increased accuracy for estimating vital signs such as heart and respiration rates in adverse signal-to-noise conditions. A major challenge for biomedical radar systems is detecting

Low-power wearable respiratory sound sensing.

Building upon the findings from the field of automated recognition of respiratory sound patterns, we propose a wearable wireless sensor implementing on-board respiratory sound acquisition and classification, to enable continuous monitoring of symptoms, such as asthmatic wheezing. Low-power consumption of such a sensor is required in order to achieve long autonomy. Considering that the power consumption of its radio is kept minimal if transmitting only upon (rare) occurrences of wheezing, we focus on optimizing the power consumption of the digital signal processor (DSP). Based on a comprehensiv

Modelling sea lice dispersion under varying environmental forcing in a Scottish sea loch.

The spread of infectious larval sea lice, Lepeophtheirus salmonis (Krøyer, 1838), between wild salmonids and farmed Atlantic salmon, Salmo salar, remains a contentious area of uncertainty. However, as laboratory and field experiments increase our knowledge of sea lice behaviour under environmental forcing, numerical modelling tools can be used to predict the spread of infectious sea louse larvae from a point source. A three-dimensional numerical model has been developed and recently validated within Loch Torridon, a fjordic sea loch on the west coast of Scotland. Output from the numerical mode

EyeSim--an ophthalmic response simulator.

This paper pertains to the design, construction and development of an ophthalmic response simulator, intended to augment current neurological examination teaching practices in medical schools. Neurological examination of the eyes informs a practitioner of the functional state of certain cranial nerves (II, III, IV and VI) through the assessment of visual acuity, visual fields, ocular movements, external and internal eye structures and ophthalmoscopic examination. For the medical student, the interpretation of these indicators may be difficult, compounded by the infrequence with which such pati

Safety effects of relaxing California's clean-record requirement for driver license renewal by mail.

A pilot program was established in California in 1982 authorizing mail renewal of driver licenses for persons under age 70 lacking clean (accident- and conviction-free) 4-year driving records, but showing clean 2-year records at selection. Such drivers constitute about 14% of California's renewal population, or about 500,000 renewees per year. Eligible drivers whose licenses expired in 1983 were subjects of the present study. Half of them were randomly assigned to a group whose members were sent offers of renewal by mail and the other half were sent standard renewal notices requiring appearanc

How fast can PPRV spread? Analyzing the results of an experimental PPRV infection of goats equipped with Ultra-Wideband sensors.

Peste des Petits Ruminants (PPR) is a highly contagious disease affecting goats and sheep. The speed and extent of its spread depend on contact patterns and the virus's intrinsic characteristics. To estimate its propagation speed and the role of inter-individual contacts, we conducted PPR virus transmission experiments in goats that were equipped with Ultra-Wideband (UWB) sensors to continuously track inter-individual distances alongside clinical and virological monitoring. Using Bayesian modeling, we integrated contact data and health status to estimate the basic reproduction number (R0), the
