MFN2-associated astrocyte dysfunction contributes to neuronal hyperexcitability and depressive-like behaviors.

Astrocyte pathology is increasingly recognized as a key factor in the pathogenesis of major depressive disorder (MDD), where disrupted gluta … tofusin-2 (MFN2)-associated astrocyte dysfunction in depression. SMART-seq profiling of hippocampal astrocytes from chronic social defeat stress (CSDS) mice revealed Mfn2 up-regulation, accompanied by excessive mitochondrial fusion, impaired mitophagy, and astrocyte loss. Elevated MFN2 expression was detected in plasma, postmortem brain tissue, and iPSC-deri …

Tissue Dissociation Workflows Influence Reactive Astrocyte Recovery in Single-Cell Transcriptomics.

Single-cell RNA sequencing has advanced our understanding of glial diversity, yet tissue dissociation remains an important technical variable. We revealed that a widely used commercial dissociation protocol can underrepresent reactive astrocytes in an acute ischemic stroke model, with preliminary observations indicating a comparable trend in a chronic tauopathy model. Benchmarking against an optimized, low-shear workflow demonstrates that different tissue dissociation workflows can substantially alter the recovery and apparent transcriptomic profile of disease-associated astrocytes, selectivel…

Rescuing anxiety by targeting the neuronal-astrocytic iron signaling axis in male mice.

Anxiety disorders affect more than one-quarter of the global population, but current first-line therapies suffer from slow onset and limited efficacy, highlighting the urgent need for novel pathogenic targets. Psychological stress disrupts iron homeostasis in the brain and alters neural circuit excitability; however, the mechanism underlying these two processes remains elusive. Here, we identify a cell type-specific iron signaling pathway between neurons and astrocytes that mediates stress-induced anxiety by regulating synaptic iron transport in the ventral hippocampal-medial prefrontal cortex…

Microglia-derived extracellular vesicles attenuate acute a-synuclein induced astrocyte inflammation.

Aggregates of misfolded α-synuclein (Syn) and neuroinflammation are pathological features of Parkinson's disease (PD). These, misfolded conformations of Syn promote cytokine and chemokine signaling in the surrounding microenvironment by triggering activation of glial cells through pattern recognition receptors. Microglia and astrocytes act as innate mediators of the neuroimmune response in the brain by regulating inflammatory signaling via paracrine and autocrine forms of cell communication. Extracellular vesicles (EVs) represent a form of glial cell to cell communication that can regulate the…

ROS-autocatalytic nanoplatform interfering astrocyte-cancer crosstalk and enhancing glioblastoma therapy.

Glioblastoma (GBM) remains incurable and exhibits exceptionally high metabolic demands, driving the evolution of hyperactive mitochondrial systems within tumors. In parallel, mitochondrial transfer from astrocytes to cancer cells fuels metabolic reprogramming and enhances GBM tumorigenicity. These malignant traits markedly constrain therapeutic efficacy and patient survival. Herein, we report a dehydroascorbic acid (DHA)-functionalized, ROS-responsive, ferrocene-integrated polymer nanoplatform (GS@DFP) co-loaded with S-Gboxin and Galunisertib, simultaneously targeting GBM mitochondria and mito…

Neuronal and astrocytic adaptations in the lateral habenula during withdrawal from chronic ethanol.

The lateral habenula (LHb) encodes aversive states and negative affect, positioning it as a candidate region for the negative reinforcement that drives alcohol withdrawal. However, little is known about how chronic ethanol exposure affects LHb neuronal function and glial biology during withdrawal. Here, we used chronic intermittent ethanol (CIE) vapor exposure, a well-established model of alcohol dependence that reliably produces somatic and affective signs of withdrawal, to examine LHb physiology and astrocytic markers during acute withdrawal in male and female rats. Whole-cell and cell-attac

Astrocytic PI3Kα controls synaptic plasticity and cognitive function via serine metabolism.

Astrocytes modulate neuronal activity by gliotransmission and through metabolic regulation, yet the relationship between these functions remains poorly understood. Here we show that phosphatidylinositol 3-kinase (PI3K) signaling in astrocytes controls synaptic plasticity and memory by regulating cellular metabolism. Deletion of the p110α isoform of PI3K in hippocampal astrocytes impairs NMDA receptor activation during the induction of hippocampal long-term potentiation (LTP). This defect is rescued by either D-serine (co-agonist of synaptic NMDA receptors) or its precursor L-serine, indicating…

Soluble TREM2 Mediates Blood Brain Barrier Permeability through Astrocyte Reactivity.

The Triggering receptor expressed on myeloid cells 2 (TREM2) is a transmembrane protein predominantly expressed by cells of the myeloid lineage. In the CNS, it is mainly expressed by microglia and macrophages, where it regulates their reactive state. This protein can be shed to release soluble TREM2 (sTREM2), which is increased in the cerebrospinal fluid of people with neurocognitive disorders. However, it remains unknown how sTREM2 contributes to neuropathogenesis. We examined the effects of sTREM2 on the inflammatory state of astrocytes, and how these impact blood-brain barrier (BBB) integri…

Deficiency of Mesencephalic Astrocyte-Derived Neurotrophic Factor Aggravates Acute Pancreatitis in Mice.

Acute pancreatitis (AP) is a common, potentially severe inflammatory disease of the pancreas. Although environmental triggers such as alcohol and gallstones are well known, only a subset of exposed individuals develop AP, suggesting that genetic or intrinsic factors contribute to disease onset and severity. Endoplasmic reticulum (ER) stress has emerged as a key pathogenic mechanism in AP due to the essential role of the ER in protein synthesis, folding, and degradation (proteostasis). Mesencephalic astrocyte-derived neurotrophic factor (MANF) is an ER stress-inducible protein highly expressed

Astrocytic lysosome deficits reduce alpha-synuclein degradation and induce the spread of pathology.

Parkinson's Disease (PD) is a neurodegenerative disorder that results from a loss of dopaminergic neurons in the substantia nigra. A pathological hallmark of PD is proteinaceous inclusions called Lewy body aggregates, which consist primarily of misfolded neuronal alpha-synuclein (αSyn). PD pathology progression is thought to be driven by a prion-like spread of αSyn aggregates between adjacent neurons; however, the role of other cell types, such as pathology bearing astrocytes, in this process is still elusive. αSyn pathology has been observed in PD patient astrocytes, suggesting that astrocyte…

Astrocyte Ca2+ Dysregulation in Alzheimer's Disease Mouse Models: Revisiting the Dogma of Hyperactivity.

Astrocytes are essential gatekeepers of brain homeostasis, and the disruption of their functions can contribute to the development of several neurological diseases. Among astrocyte signaling pathways, the intracellular second messenger Ca2+ plays a pivotal role in regulating the release of gliotransmitters, which actively modulate fundamental processes in the brain such as synaptic plasticity and memory function. Several studies over the years support the idea that dysregulated astrocytic Ca2+ homeostasis represents a relevant mechanism in Alzheimer's disease pathogenesis. Early works in trans…

Astrocyte-to-microglia purinergic signaling mediates synaptic shielding and promotes neuronal activity.

How glial cells sense and regulate neuronal hypoactivity remains poorly understood. Using in vivo imaging in anesthetized, sleeping, and behaving mice, we identified a glial purinergic signaling mechanism that converts neuronal hypoactivity into local circuit regulation. Cortical hypoactivity increased spatially confined ATP release through astrocytic pannexin-1 hemichannels, which were structurally enriched near perisomatic parvalbumin-positive boutons. Local ATP release directed microglial process movement and stabilized bulbous endings through microglial P2Y12 signaling, accompanied by loca

Dorsal striatal circuit mechanisms contributing to astrocyte modulation of alcohol-related behaviors.

BACKGROUND: The dorsolateral striatum (DLS) is a key site for coordinating the alcohol-induced stimulant response, a behavioral marker predi … cyte calcium activity under control (saline) or EtOH conditions. Together, this work identifies DLS astrocytes as key contributors to EtOH-induced stimulation and highlights the importance of considering astrocyte-neuron interactions in evaluating alcohol effects.

Striatal cholinergic and dopaminergic driven astrocyte Ca2+ activity is disrupted in Parkinsonian mice.

Brain neuromodulatory systems exert powerful effects on local neuronal circuit function and behavior. In addition to classical actions directly on neurons, growing evidence indicates that neuromodulators also recruit Ca2+-dependent astrocyte mechanisms to regulate synaptic plasticity and network function. The dorsal striatum integrates cortical and thalamic inputs under strong dopamine (DA) and acetylcholine (ACh) neuromodulatory control. To what extent the circuit and behavioral effects of striatal ACh and DA depend on astrocyte Ca2+ activity remains unclear. We show that locomotion elicits r

Sex-specific associations between astrocytic reactivity and cognitive decline in unimpaired elderly.

INTRODUCTION: Astrocyte reactivity may contribute to increased Alzheimer's disease (AD) risk in women. Although higher plasma glial fibrillary acidic protein (GFAP) levels have been reported in women, few studies have investigated sex-specific relationships with other plasma AD biomarkers and cognition. METHODS: Participants were enrolled in the Memory Education and Research Initiative, a longitudinal community-based cohort, and underwent evaluation including blood biomarker sampling. The Alzheimer's Disease Neuroimaging Initiative (ADNI) was included as a replication cohort. RESULTS: Unimpa

The astrocyte marker ALDH1L1 also identifies a stromal cell population in the lymph node.

ALDH1L1 is widely used as a marker of astrocytes in the central nervous system (CNS), but its expression and potential roles in the periphery, particularly in lymphoid organs, remain poorly understood. Here, we found that ALDH1L1+ cells comprise approximately 5–9% of the lymph node (LN). To better understand their identity, we investigated whether these cells share characteristics with glial, immune, endothelial or stromal cell populations under homeostatic conditions. Using ALDH1L1/TdTomato reporter mice, immunofluorescence, and flow cytometry, we found that TdTomato+ cells in LNs do not coex…

Carboplatin alleviates astrocytic TDP-43 neurotoxicity by inhibiting NF-κB activation.

Amyotrophic lateral sclerosis (ALS) is a rare and progressive motor neuron disease; however, its exact pathogenic mechanisms remain unclear. Currently, no effective treatments are available for this disease. Therefore, in this study, we investigated the anti-inflammatory effects of the anti-cancer agent, carboplatin, on neuronal cells and its potential therapeutic effects against ALS. Carboplatin inhibited NF-κB phosphorylation in the transactive response DNA-binding protein (TDP)-43-transfected astrocytes, reducing pro-inflammatory cytokine levels, without affecting the TDP-43 protein levels.…

Zeb2 Controls Retinal Physiological and Pathological Angiogenesis by Regulating Astrocyte Proliferation and Differentiation.

Retinal angiogenesis relies on a precisely timed interaction between astrocytes and endothelial cells (ECs), yet the transcriptional regulatory program underlying this complex neurovascular crosstalk remains poorly characterized. Here, we define Zeb2 as a pivotal transcriptional modulator of retinal astrocyte function that coordinates developmental and pathological vascular growth. It is transiently expressed in retinal astrocyte progenitor cells during development and re-induced under pathological hypoxia. Conditional ablation of Zeb2 in retinal astrocytes enhanced their proliferation, migrat…
