Sex Differences in the Association Between Cyberbullying Victimization and Mental Health, Substance Use, and Suicidal Ideation in Adolescents.

OBJECTIVE: To examine sex differences in the association between cyberbullying victimization and mental health (psychological distress and delinquency), substance use-related outcomes (drug and tobacco use, binge drinking), and suicide ideation among adolescents. METHOD: Data were obtained from the Ontario Student Drug Use and Health Survey (OSDUHS; 2013, N=10,272, grade 7 to 12). The sample for analysis included 4,940 students with a mean age of 15.1 years (43.3% male). A series of multi-level, binary, logistic regression models were conducted separately for female and male adolescents to qu…

Chronic binge alcohol exposure during pregnancy impairs rat maternal uterine vascular function.

BACKGROUND: Alcohol exposure during pregnancy results in an array of structural and functional abnormalities called fetal alcohol spectrum disorders (FASD). Alcohol dysregulates the exquisite coordination and regulation of gestational adaptations at the level of the uterine vasculature. We herein hypothesized that chronic binge-like alcohol results in uterine vascular dysfunction and impairs maternal uterine artery reactivity to vasoconstrictors and dilators. METHODS: We utilized a once-daily binge alcohol (4.5 g/kg body weight) exposure paradigm (gestational day 7 to 17) in a pregnant rat mo

11β-hydroxysteroid dehydrogenase inhibition as a new potential therapeutic target for alcohol abuse.

The identification of new and more effective treatments for alcohol abuse remains a priority. Alcohol intake activates glucocorticoids, which have a key role in alcohol's reinforcing properties. Glucocorticoid effects are modulated in part by the activity of 11β-hydroxysteroid dehydrogenases (11β-HSD) acting as pre-receptors. Here, we tested the effects on alcohol intake of the 11β-HSD inhibitor carbenoxolone (CBX, 18β-glycyrrhetinic acid 3β-O-hemisuccinate), which has been extensively used in the clinic for the treatment of gastritis and peptic ulcer and is active on both 11β-HSD1 and 11β-HSD

Chronic binge alcohol consumption alters myogenic gene expression and reduces in vitro myogenic differentiation potential of myoblasts from rhesus macaques.

Chronic alcohol abuse is associated with skeletal muscle myopathy. Previously, we demonstrated that chronic binge alcohol (CBA) consumption by rhesus macaques accentuates skeletal muscle wasting at end-stage of simian immunodeficiency virus (SIV) infection. A proinflammatory, prooxidative milieu and enhanced ubiquitin proteasome activity were identified as possible mechanisms leading to loss of skeletal muscle. The possibility that impaired regenerative capacity, as reflected by the ability of myoblasts derived from satellite cell (SCs) to differentiate into myotubes has not been examined. We

Validity Concerns About the Heartbeat Counting Task Extend to Alcohol Use disorder: Evidence From Subclinical and Clinical Samples.

Theoretical models propose that interoception plays a role in addictive disorder. However, this assumption has been mostly tested using the heartbeat counting task (HCT), which is known to be contaminated by estimation strategies. An adapted version of the HCT (in which respondents report only felt heartbeats) has been developed to reduce estimation biases. Here, we examined the validity of the classical and adapted HCT versions in samples presenting alcohol use disorders. We recruited a clinical sample of 48 patients with severe alcohol use disorder (SAUD), matched with 41 healthy controls (H

In Vivo Administration of Phosphatidic Acid, a Direct Alcohol Target Rescues Fetal Growth Restriction and Maternal Uterine Artery Dysfunction in Rat FASD Model.

Fetal growth restriction is a hallmark of Fetal Alcohol Syndrome (FAS) and is accompanied by maternal uterine circulatory maladaptation. FAS is the most severe form of Fetal Alcohol Spectrum Disorder (FASD), a term for the range of conditions that can develop in a fetus when their pregnant mother consumes alcohol. Alcohol exerts specific direct effects on lipids that control fundamental developmental processes. We previously demonstrated that direct in vitro application of phosphatidic acid (PA, the simplest phospholipid and a direct target of alcohol exposure) to excised uterine arteries from

Alcohol Impairs Bioenergetics and Differentiation Capacity of Myoblasts from Simian Immunodeficiency Virus-Infected Female Macaques.

Alcohol misuse and HIV independently induce myopathy. We previously showed that chronic binge alcohol (CBA) administration, with or without simian immunodeficiency virus (SIV), decreases differentiation capacity of male rhesus macaque myoblasts. We hypothesized that short-term alcohol and CBA/SIV would synergistically decrease differentiation capacity and impair bioenergetic parameters in female macaque myoblasts. Myoblasts from naïve (CBA-/SIV-), vehicle [VEH]/SIV, and CBA/SIV (N = 4-6/group) groups were proliferated (3 days) and differentiated (5 days) with 0 or 50 mM ethanol (short-term). C

Machine-Learning Models of Cognitive Test Performance Incorporating Exposomic Pesticide Biomarkers in Older U.S. Adults.

Background/Objectives: Environmental exposures may contribute to heterogeneity in cognitive aging, yet population-scale datasets integrating exposure biomarkers with cognitive testing remain underused. Methods: Using NHANES 2011-2014 data, we analyzed adults aged ≥60 years with complete cognitive assessments, demographic/lifestyle covariates, Patient Health Questionnaire-9 scores, binge drinking status, and urinary concentrations of eight…

Disentangling the effects of corticotrophin releasing factor and GABA release from the bed nucleus of the stria terminalis on ethanol self-administration in mice.

Excessive alcohol use causes a great deal of harm and negative health outcomes. Corticotrophin releasing factor (CRF), a stress-related neuropeptide, has been implicated in binge ethanol intake and ethanol dependence in rodents. CRF containing neurons in the bed nucleus of the stria terminalis (BNSTCRF) can influence ethanol consumption. These BNSTCRF neurons also release GABA, raising the question, is it CRF release, GABA release, or both that is regulating alcohol consumption. Here, we used viral vectors to separate the effects of CRF and GABA release from BNSTCRF neurons on the escalation o

Exacerbated cardiac dysfunction from combined alcohol binge and synthetic cannabinoid use.

Alcohol remains the most frequently used intoxicant, posing a significant global health concern. Binge drinking has been linked to acute cardiovascular complications, including reduced cardiac performance, arrhythmias, and blood pressure instability. Additionally, there is a growing number of clinical reports describing severe adverse cardiac events associated with the recreational use of synthetic cannabinoids. Recent surveys reveal a troubling rise in polydrug misuse, particularly among young adults, with an increasing number of cases linked to fatal outcomes. This study aimed to characteriz…
