Effects of xenoestrogen bisphenol A on uterine and pituitary weight, serum prolactin levels and immunoreactive prolactin cells in ovariectomized Wistar rats.

Considerable attention has currently been focused on bisphenol A (BPA), an environmental endocrine disrupting chemical that has oestrogenic activity. In vitro and in vivo short-term assays have shown that BPA is weakly estrogenic. In addition, the issue of species- and strain-differences in susceptibility to BPA was raised. The treatment of ovariectomized (OVX) Wistar rats with BPA at doses of 11-250 mg/kg per day, s.c., for 7 days, resulted in significant dose-dependent regrowth of uterus in uterotrophic assay. Additionally, the stimulation of anterior pituitary gland growth and induction of

Exposure to non-persistent chemicals in consumer products and fecundability: a systematic review.

BACKGROUND: Exposure to non-persistent chemicals in consumer products is ubiquitous and associated with endocrine-disrupting effects. These effects have been linked to infertility and adverse pregnancy outcomes in some studies and could affect couple fecundability, i.e. the capacity to conceive a pregnancy, quantified as time to pregnancy (TTP). OBJECTIVE AND RATIONALE: Few epidemiologic studies have examined the impact of non-persistent chemicals specifically on TTP, and the results of these studies have not been synthesized. We undertook a systematic review to summarize the strength of evid

Phosphonium Chloride-Enhanced Ionization for High-Throughput Screening and Imaging of Carbohydrate Metabolites: Unveiling Disruptors of Sugar Metabolism.

Carbohydrates with high structural complexity are involved in key biological processes and serve as susceptible targets of environmental metabolic disruptors. However, the lack of a high-throughput and sensitive method for profiling carbohydrates limits our mechanistic understanding of the disruption of sugar metabolic pathways. In this study, we found that postcolumn addition of 10 μM (chloromethyl)triphenylphosphonium chloride (Ph3(CH2Cl)PCl) enabled the efficient ionization of saccharides and sugar alcohols as [M+Cl]- adducts in high-resolution mass spectrometry analysis, lowering instrumen

Simultaneous Enhancement of Flame Resistance and Antimicrobial Activity in Epoxy Nanocomposites Containing Phosphorus and Silver-Based Additives.

The design and manufacture of innovative multifunctional materials possessing superior characteristics, quality and standards, rigorously required for future development of existing or emerging advanced technologies, is of great importance. These materials should have a very low degree of influence (or none) on the environmental and human health. Adjusting the properties of epoxy resins with organophosphorus compounds and silver-containing additives is key to the simultaneous improvement of the flame-resistant and antimicrobial properties of advanced epoxy-based materials. These environmentall

An optimized processing method for polar organic chemical integrative samplers deployed in seawater: Toward a maximization of the analysis accuracy for trace emerging contaminants.

Passive sampling of emerging contaminants (ECs) in seawater represents a challenge in environmental monitoring. A specific protocol for Polar Organic Chemical Integrative Sampler (POCIS) processing may be necessary when dealing with marine applications, due to the peculiarity of the considered matrix. Herein, both the instrumental LC-MS/MS analysis and the sampler processing for the determination of 22 ECs in seawater were carefully optimized. The study entailed a test simulating POCIS sorbent exposure to seawater as well as the processing of replicated field POCIS with different elution solve

Protective effects of biogenic selenium nanoparticles synthesized by Chlorella Vulgaris against bisphenol a-induced metabolic disturbances in rats.

BACKGROUND: Bisphenol A (BPA), a xenoestrogen and endocrine disruptor, is widely used in plastic and polycarbonate products, contributing to metabolic disorders such as dyslipidemia and obesity. Algae, like Chlorella vulgaris (CV), contain highly active compounds that act as natural reducing agents, converting inorganic selenium into nanoparticle form. METHODS: In this study, we developed novel biologically synthesized selenium nanoparticles using CV (SeNPs-CV) and investigated their protective properties in comparison to physically synthesized selenium nanoparticles (SeNPs) against BPA-induc

Expression of heat shock protein and hemoglobin genes in Chironomus tentans (Diptera, chironomidae) larvae exposed to various environmental pollutants: a potential biomarker of freshwater monitoring.

To identify a sensitive biomarker of freshwater monitoring, we evaluated pollutant-induced expression of heat shock proteins (HSPs) and hemoglobins (Hbs) genes in the larvae of the aquatic midge Chironomus tentans (Diptera, Chironomidae). As pollutants, we examined nonylphenol, bisphenol-A, 17alpha-ethynyl estradiol, bis(2-ethylhexyl) phthalate, endosulfan, paraquat dichloride, chloropyriphos, fenitrothion, cadmium chloride, lead nitrate, potassium dichromate, benzo[a]pyrene and carbon tetrachloride. We also investigated larval growth as a physiological descriptor by measuring changes in the b

Chemicals in plastics differentially affect the transcriptome of MCF-7 breast cancer cells.

Humans are exposed to chemicals that leach from plastics, yet many remain data-poor and lack toxicological evaluation. High-throughput transcriptomics (HTTr) provides a scalable way to screen chemicals and generate mechanistic insights relevant to human health risk assessment. We applied HTTr in MCF-7 breast cancer cells to assess chemicals across several concentrations (0.001-50 µM) used in plastics and dyes. Transcriptomic points of departure (tPODs) were derived from general gene perturbations, including pathway-level and estrogen receptor α (ERα)-specific changes. We also used transcriptom

Sex-specific associations of prenatal and postnatal exposure to 15 endocrine-disrupting chemical exposures with visual impairment at age three: the Korean Children's Environmental Health Study (Ko-CHENS).

BACKGROUND: Endocrine-disrupting chemicals (EDCs) have been linked to pediatric outcomes, but evidence on visual development and sex differences is scarce. We examined whether prenatal and postnatal EDC exposures were associated with visual impairment at age 3. METHODS: Participants were drawn from the Korean Children's Environmental Health Study, with urinary biomarkers measured at early pregnancy (n = 340), late pregnancy (n = 454), 24 months (n = 344), and 36 months (n = 713). A subset with complete data at all windows (n = 115) was analyzed using generalized estimating equations. Fifteen

Bisphenol F Exposure Induces Ovarian Ferroptosis via the ERβ-ATM-Ferritinophagy Axis.

Bisphenol F (BPF) has rapidly emerged as a primary substitute for bisphenol A (BPA) and a ubiquitous environmental contaminant, yet its reproductive toxicity remains poorly understood, particularly at low doses. Here, we demonstrate that oral exposure to BPF (as low as 250 μg/kg) induces ovarian hypertrophy, disrupted follicular architecture and cumulus-oocyte communication, and reduced serum estradiol and progesterone in mice by triggering ferroptosis. Integrating multiomics approaches, we identified NCOA4-mediated ferritinophagy as the central cellular driver of this process. In granulosa ce

In Vivo Investigation of the Effects of Nonylphenol on the Pituitary-Adrenal Axis and Pineal Gland in Male Rats.

The pineal gland secretes melatonin, which regulates various physiological processes; damage to this gland disrupts these functions. This study aimed to investigate the effect of nonylphenol on the pineal gland and the pituitary-adrenal axis, which is associated with this system. The study was initiated using Wistar albino male rats on their postnatal 21st day, a critical developmental stage for endocrine regulation. Nonylphenol was administered via oral gavage at doses of 5, 25 and 125 mg/kg/day, while bisphenol-A was given at 50 mg/kg/day as a positive control. At the end of the treatment pe

Long-term exposure to bisphenol-A causes oxidative stress-related alterations at the genetic and cellular levels in the mature ovary of adult zebrafish.

Bisphenol-A (BPA) is categorized as a major endocrine-disrupting chemical (EDC) used to manufacture many plastic products. BPA affects reproductive performance and promotes infertility by causing hormonal imbalance, mitochondrial dysfunction, and altered gene expression. The present investigation aimed to evaluate the effects of BPA exposure for 28 days on the activity or level of antioxidant response elements (AREs), mRNA expressions of antioxidant genes, and histomorphological changes in the ovary of adult zebrafish. The adult female zebrafish were randomly divided into four experimental gro

Associations of maternal and paternal preconception and maternal pregnancy urinary phthalate biomarker and bisphenol A concentrations with offspring autistic behaviors: The PEACE study.

BACKGROUND: Environmental chemical exposures in utero may play a role in autism development. While preconception risk factors for autism are increasingly being investigated, little is known about the influence of chemical exposures during the preconception period, particularly for paternal exposures. METHODS: In 195 children from the Preconception Environmental exposures And Childhood health Effects (PEACE) cohort born to parents recruited from a fertility clinic in Boston, Massachusetts between 2004 and 2017, we quantified concentrations of 11 phthalate metabolites and bisphenol A (BPA) in u

Perinatal bisphenol A exposure has an age- and dose-dependent association with thyroid allostasis adaptive response, as well as anxiogenic-depressive-like and asocial behaviors in juvenile and adult male rats.

Thyroid hormones are essential for brain development, and a shortage throughout the fetal and postnatal periods can result in mood disorders. Perinatal exposure to bisphenol A (BPA) affects thyroid activity and dependent processes indirectly during pregnancy or early postnatal life. This is particularly important because it may cause changes in tissue ontogeny, increasing the risk of developing disorders later in life. The study aimed to investigate the consequences of thyroid hormone deficiency on anxiety, social, and depressive behaviors, as well as disruption in thyroid peroxidase (TPO) gen

Prenatal Bisphenol a Exposure and Postnatal Trans Fat Diet Alter Small Intestinal Morphology and Its Global DNA Methylation in Male Sprague-Dawley Rats, Leading to Obesity Development.

In this study, we aimed to determine whether a postnatal trans fat diet (TFD) could aggravate prenatal bisphenol A (BPA) exposure effects on offspring’s small intestine and adulthood obesity, due to the relatively sparse findings on how the interaction between these two variables interrupt the small intestinal cells. Twelve pregnant rats were administered with either unspiked drinking water (control; CTL) or BPA-spiked drinking water throughout pregnancy. Twelve weaned pups from each pregnancy group were then given either a normal diet (ND) or TFD from postnatal week (PNW) 3 until PNW14, divid

Bisphenol A attenuates the therapeutic effect of the selective G protein-coupled estrogen receptor agonist G-1 on allergic rhinitis inflammation in mice.

BACKGROUND: Bisphenol A (BPA) is found in many plastics widely used in everyday life and affects the immune system. Previous studies found that the selective G protein coupled estrogen receptor (GPER) agonist G-1 can reduce the inflammation associated with asthma and allergic rhinitis (AR). BPA also interferes with the protective effect of estradiol against myocardial ischemia-reperfusion injury. OBJECTIVE: We explored whether BPA attenuates the effect of G-1 on inflammation in a mouse AR model. METHODS: The AR model was established by sensitizing and stimulating female BALB/c mice with oval

Tetramethyl bisphenol a inhibits leydig cell function in late puberty by inducing ferroptosis.

Tetramethyl bisphenol A (TMBPA) is a commonly used bisphenol analog, used as a fire retardant. However, whether it inhibits the function of Leydig cells in late puberty remains unclear. In this study, 35-day-old male Sprague-Dawley rats were gavaged with 0, 10, 100, and 200 mg/kg body weight TMBPA for 21 days. TMBPA significantly reduced serum testosterone levels at 10 mg/kg and higher doses without altering serum luteinizing hormone and follicle-stimulating hormone levels. TMBPA significantly increased serum iron concentraion while reducing the ratio of serum glutathione (GSH) and GSH/GSSG (o

Bisphenol A and rotenone induce S-nitrosylation of protein disulfide isomerase (PDI) and inhibit neurite outgrowth of primary cultured cells of the rat hippocampus and PC12 cells.

Bisphenol A (BPA) interferes the function and development of the central nervous system (CNS), resulting in behavioral abnormalities and memory loss. S-nitrosylation of protein disulfide isomerase (PDI) is increased in brains with sporadic Alzheimer's disease and Parkinson's disease. The aim of the present study was to clarify the role of nitric oxide (NO) in BPA-induced neurotoxicity. Since rotenone induces NO-mediated neurodegeneration through S-nitrosylation of PDI, it was used as a positive control. First, rats were treated with BPA and rotenone, and S-nitrosylation of PDI was detected in

Transgenerational effect of parental obesity and chronic parental bisphenol A exposure on hormonal profile and reproductive organs of preadolescent Wistar rats of F1 generation: A one-generation study.

There is a global concern about adverse health effects of endocrine-disrupting chemicals (EDCs). Bisphenol A (BPA), an estrogenic and obesogenic compound, used in the plastic and medical industry has a dominant position among EDCs as far as human health and regulatory scenario are concerned. Due to its omnipresence across the biosphere, population of all age groups and health status is unavoidably exposed to BPA. Transgenerational exposure to BPA and its effects have also been recognized. However, there is no report on the transgenerational effect of BPA on metabolically disordered parents, su
