Mitochondrial Toxicology of Heavy Metals and Pesticides: Transport Systems, Mitochondrial Dysfunction and Permeability Transition.

Mitochondrial transport systems are essential regulators of cellular bioenergetics, calcium homeostasis, and metabolic signaling, and have emerged as critical targets of environmental toxicants. Although heavy metals and pesticides act through distinct primary mechanisms, increasing evidence indicates that they converge on a common network of mitochondrial dysfunction characterized by oxidative stress, impaired metabolite transport, calcium dyshomeostasis, and sensitization to mitochondrial permeability transition. This review provides an updated overview of the major mitochondrial transport s

F-actin-dependent activation of NADPH oxidase 5 by the vacuolar-type ATPase.

NADPH oxidase 5 (NOX5) is a transmembrane oxidative signaling enzyme that produces superoxide in response to increases in intracellular calcium. NOX5 has been shown to play essential roles in processes such as specialized cell differentiation, proliferation, and cell contraction, while its overexpression and hyperactivation are associated with human diseases, including cardiovascular disorders, cancer, and diabetic kidney disease. Despite its importance in human health and disease, the regulatory factors and mechanistic signaling details of NOX5 remain incompletely resolved. We previously repo

Hypoxia-activated endothelial cells drive stemness and vasculogenic mimicry in NSCLC via a HIF-1α/ITPR3 axis.

Vasculogenic mimicry (VM) predicts poor prognosis in non-small cell lung cancer (NSCLC). Hypoxia is a critical tumor microenvironmental regulator, yet its detailed mechanism linking to VM formation remains poorly defined. This study aimed to elucidate the role of hypoxic endothelial cells in promoting NSCLC aggressiveness and VM formation. We integrated clinical data analysis, single-cell RNA sequencing (scRNA-seq), transcriptomic profiling, and multiple in vitro and in vivo functional validations to consolidate our findings. Clinical NSCLC data revealed a positive correlation between endothel

Are You a Friend or an Enemy? The Dual Action of Methylglyoxal on Brain Microvascular Endothelial Cells.

Methylglyoxal is a reactive dicarbonyl intermediate in the advanced glycation end-product (AGE) pathway, and alterations in its levels have been detected in the plasma, cerebrospinal fluid, and brain parenchyma in various pathologies, particularly in diabetes. In this study, we investigate the effects of methylglyoxal (MGO) on murine brain microvascular endothelial cells at both physiological and pathological concentrations. We evaluate molecular parameters, including reactive oxygen species (ROS) production, cytosolic calcium signaling, and ATP synthesis, as well as cellular responses such as…

Beef-Derived Peptides Mediated Desensitization of Bitter Taste Receptor T2R14 Through GPCR Kinase 2.

Background/Objectives: Humans have at least 26 bitter taste receptors (T2Rs), and among these, bitter taste receptor 14 (T2R14) is highly expressed in both oral and extraoral tissues. Over 100 bitter ligands can activate T2R14, including hormones, vitamins, plant compounds, and peptides. Previous studies suggest that bitter tastants such as quinine and caffeine can inhibit G protein-coupled receptor kinases (GRKs) and delay T2R signal termination. Our earlier research showed that peptides from alcalase and chymotrypsin hydrolysates of beef proteins inhibited quinine-dependent calcium release t

Calcium-Sensing Receptor Activation Disrupts Phosphatidylserine Asymmetry and Promotes Calcium Oxalate Crystal-Induced Epithelial Injury in Renal Tubular Epithelial Cells.

BACKGROUND: Calcium oxalate (CaOx) crystal retention on the renal tubular epithelium is a key step in urolithiasis. Phosphatidylserine (PS) exposure may facilitate crystal-cell adhesion, but the upstream signaling mechanisms and the relative contributions of impaired inward PS flipping versus outward PS redistribution remain unclear. MATERIALS AND METHODS: Global proteomic profiling using 2-dimensional electrophoresis and matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry (2-DE/MALDI-TOF/TOF) in an immortalized human proximal tubular epithelial cell li

Congenital Hypothyroidism Dysregulates TRPC6 to Mediate Abnormal Dendritic Spine Growth of Hippocampal Neurons.

BACKGROUND: Congenital hypothyroidism (CH) may lead to irreversible neurological dysfunction of offspring by affecting hippocampal morphogenesis. The dentate gyrus (DG) is primarily the affected tissue by the CH, in which the dendritic spine density of gyrus granule cells (DGCs) is significantly reduced, thereby resulting in the cognitive impairment. The CaMKIV/CREB signaling pathway has been shown to mediate the deficient growth of DGCs dendritic spines, but the mechanism of CH in modulating the Ca2+-dependent CaMKIV is still elusive. METHODS: CH model of rat pups was prepared by the supply

Activation of retinal glial (Müller) cells by extracellular ATP induces pronounced increases in extracellular H+ flux.

Small alterations in extracellular acidity are potentially important modulators of neuronal signaling within the vertebrate retina. Here we report a novel extracellular acidification mechanism mediated by glial cells in the retina. Using self-referencing H+-selective microelectrodes to measure extracellular H+ fluxes, we show that activation of retinal Müller (glial) cells of the tiger salamander by micromolar concentrations of extracellular ATP induces a pronounced extracellular H+ flux independent of bicarbonate transport. ADP, UTP and the non-hydrolyzable analog ATPγs at micromolar concentr

Listeriolysin O-mediated calcium influx potentiates entry of Listeria monocytogenes into the human Hep-2 epithelial cell line.

To investigate factors which modulate the entry of Listeria monocytogenes into mammalian cells, we have analyzed the role of Ca(2+). We show that L. monocytogenes induced Ca(2+) transients into the human Hep-2 epithelial cell line. The nonpathogenic species L. innocua or a L. monocytogenes mutant strain defective in listeriolysin O (LLO) production was unable to induce these calcium fluxes. Addition of plasma membrane calcium channel antagonists or chelation of extracellular calcium markedly reduced L. monocytogenes entry. In contrast, chelation of host cytosolic Ca(2+) or blockade of Ca(2+) r

The effects of 2-aminoethoxydiphenyl borate, a novel inositol 1,4, 5-trisphosphate receptor modulator on myometrial contractions.

These studies were performed to evaluate the effect of 2-aminoethoxydiphenyl borate (2-APB), a novel membrane-permeable inositol 1,4,5-trisphosphate-receptor inhibitor on agonist-induced, spontaneous, and KCl-stimulated in vitro myometrial contractions. 2-APB significantly inhibited spontaneous myometrial contractions as well as phasic contractions induced by various uterotonic agonists. Confiriming its effects on intracellular calcium release, 2-APB inhibited phasic contractions in the absence of extracellular calcium. 2-APB had little effect on the tonic response to KCl stimulation, implicat

Low endolymph calcium concentrations in deafwaddler2J mice suggest that PMCA2 contributes to endolymph calcium maintenance.

In vertebrates, transduction of sound into an electrochemical signal is carried out by hair cells that rely on calcium to perform specialized functions. The apical surfaces of hair cells are surrounded by endolymphatic fluid containing calcium at concentrations that must be maintained by active transport. The mechanism of this transport is unknown, but an ATP-dependent pump is believed to participate. Mutation of the Atp2b2 gene that encodes plasma membrane calcium ATPase type 2 (PMCA2) produces the deaf, ataxic mouse: deafwaddler2J (dfw2J). We hypothesized that PMCA2 might transport calcium i

Postprandial dynamics of sensory receptor and neural mechanisms underlying feeding regulation in mule ducks.

The gustatory system plays a central role in nutrient detection and feeding regulation in birds, yet the underlying molecular mechanisms in ducks remain poorly characterized. Here, we provide the first integrated analysis of nutrient detection pathways, from peripheral oral sensing to central brain integration, in mule ducks at 4 weeks of age. Nutritional status was confirmed via plasma free fatty acids and triglycerides. We quantified taste receptors mediating sweet (Taste Receptor type 1 member 1 and 3), bitter (Taste Receptor type 2 member 9 and 40), sour (Acid-Sensing Ion Channels), umami

Calcium-Triggered PuMYB44-PuERF118 Cascade Activates PuFAD8 to Promote Aroma Biosynthesis in Pear Fruit.

Fruit aroma is a key factor shaping the commercial value of pears. MYB and ERF transcription factors (TFs) are involved in regulating the biosynthesis of fruit aroma compounds, yet the mechanisms underlying their synergistic regulation of pear aroma formation remain elusive. Calcium signaling exerts an important role in fruit aroma biosynthesis; nevertheless, how calcium signaling mediates MYB-ERF transcription factors to participate in pear aroma biosynthesis remains unclear. Here, transcriptomic profiling of calcium-treated pear fruits screened two significantly upregulated TFs, PuMYB44 and …

Effects of Supplemental Calcium and Vitamin D on Expression of Toll-Like Receptors and Phospho-IKKα/β in the Normal Rectal Mucosa of Colorectal Adenoma Patients.

Chronic inflammation in the colorectum, a significant contributor to colorectal carcinogenesis, can be triggered by the activation of proinflammatory signaling pathways such as those initiated by Toll-like receptors (TLR) and nuclear factor κB (NF-κB). Although experimental evidence supports calcium and vitamin D potentially modifying these proinflammatory pathways in the colorectum, human data in these regards are scarce. We investigated supplemental calcium (1,200 mg daily) and/or vitamin D3 (1,000 IU daily) effects on inflammatory signaling pathway-related biomarkers in a subset of 105 part

Vitamin D-independent therapeutic effects of extracellular calcium in a mouse model of adult-onset secondary hyperparathyroidism.

Cell proliferation and PTH secretion in the parathyroid gland are known to be regulated by vitamin D and extracellular calcium. Here, we examined the vitamin D-independent effects of correction of extracellular calcium in an adult-onset secondary hyperparathyroidism (sHPT) model, using mice with a nonfunctioning vitamin D receptor (VDR). Wildtype and homozygous VDR mutant mice were kept on a rescue diet (RD) containing 2% calcium (Ca), 1.25% phosphorus (P), and 20% lactose until they were 4 mo or 1 yr of age. Subsequently, 4-mo-old mice were switched to a challenge diet (CD) containing the fol

Confocal microscopy-based radial profile analysis for submitochondrial protein localization.

Mitochondria are double-membrane organelles whose functions depend on membrane proteins. Identifying whether proteins localize to the outer mitochondrial membrane (OMM) or inner mitochondrial membrane (IMM) is important but often requires specialized techniques. Here, we developed a confocal laser scanning microscopy (CLSM)-based radial profile analysis for the localization of submitochondrial proteins. Based on this method, cytochrome c oxidase subunit 4 (COX4), adenine nucleotide translocase 2 (ANT2), mitochondrial calcium uniporter (MCU) and calcium-activated potassium channel subunit alpha

Distinct intracellular spatiotemporal expression of Calmodulin genes underlies functional diversity of Calmodulin-dependent signalling in cardiac myocytes.

AIMS: This study aims to resolve the mechanisms underlying Calmodulin (CaM)'s signalling diversity by investigating whether the three CaM genes-Calm1, Calm2, and Calm3-play distinct or redundant roles in cardiac myocytes, focusing on their spatial mRNA localization and interactions with key targets. METHODS AND RESULTS: We utilized single-molecule mRNA detection and three-dimensional imaging to map the spatial distribution of Calm1, Calm2, and Calm3 mRNAs within ventricular myocytes. These mRNAs were found to be consistently positioned within specific cellular zones, overlapping with their ta
