The processing of adult mosquitoes collected in the field for studies of natural infection with arboviruses of public health interest (genera: Flavivirus, Alphavirus, Orthobunyavirus) follows the assembly of homogenized mosquito pools and extraction of genetic material using commercial or manual techniques, and the detection of viral RNA by polymerase chain reaction (nested RT-PCR) [11]. This paper describes the procedures for capturing, taxonomic determination, obtaining genetic material, and molecular techniques for detecting alphaviruses.

Muscovy duck parvovirus (MDPV) and Goose parvovirus (GPV) are highly contagious waterfowl viruses causing significant harm to the poultry industry. This study designed primers and probes targeting the non-structural genomic region of MDPV and GPV, optimizing amplification for both quantitative polymerase chain reaction (qPCR) and duplex crystal digital polymerase chain reaction (dPCR). The resulting duplex crystal dPCR method showed strong specificity, detecting MDPV and GPV without cross-reactivity with other waterfowl viruses. Sensitivity tests revealed that the lower detection limit for qPC

Brazil has the largest buffalo herd outside the Asian continent, with more than 1.5 million head. As buffalo farming has become an economically important activity, studies on the occurrence of pathogens shared between cattle and buffalo are necessary. The aim of this study was to investigate the molecular occurrence of piroplasmids and bacterial agents (Anaplasmataceae and Coxiellaceae) potentially transmitted by vectors in buffaloes in southeastern Brazil using molecular methods. DNA was extracted from blood samples of 81 buffaloes, 165 Rhipicephalus microplus ticks, and 92 Haematopinus tuber

Staphylococcus aureus (S.aureus) is a common Gram-positive human pathogen that causes community-acquired and nosocomial infections, with diverse clinical manifestations ranging from local superficial lesions and food poisoning to fatal systemic infections. The discovery of antibiotics significantly reduced the mortality rate, but the problem of drug resistance has since become increasingly prominent. Since the first identification of MRSA in 1960, this strain has emerged as a global public health threat. MRSA is a major pathogen of nosocomial infections, capable of causing various severe disea

To investigate the origin and pathogenesis of acquired immunodeficiency syndrome (AIDS)-related lymphoma (ARL), we studied 14 cases in which Epstein-Barr virus (EBV) infection was not an etiologic factor. By histology, 8 of the specimens were of the small noncleaved cell type and 6 consisted of the large diffuse cell type. Southern analysis using a J(H) probe was consistent with a monoclonal B-cell tumor in 13 cases. To characterize the expressed Ig genes, we performed reverse transcriptase-polymerase chain reaction (RT-PCR) and direct sequencing of PCR products. Eight cases expressed IgM and

Enteroviruses cause infections that present in diverse ways and affect people of all ages. Infections peak during summer and fall epidemics and cause 10 to 15 million symptomatic infections annually in the United States. The 70 enteroviral serotypes cause illness that ranges from nonspecific fevers and rashes to life-threatening myocarditis or central nervous system disease. These common infections create a significant burden on our society and healthcare system. New developments in rapid diagnosis of enterovirus infections using polymerase chain reaction (PCR) positively affect patient manage

Because the Bacillus Calmette-Guérin (BCG) prevents infants from contracting miliary tuberculosis and tuberculosis meningitis, BCG vaccination is recommended for those under 6 months old in Japan. Complications such as favorable local inflammatory reactions including redness, induration, and abscess formation may occur, but severe adverse effects such as osteomyelitis, periostitis, and disseminated BCG infection are generally rare. We report an 11-month-old boy with severe combined immunodeficiency dying of serious disseminated BCG infection despite anti-tuberculosis therapy and blood stem cel

Among all other viruses, human cytomegalovirus (HCMV) is the most frequent cause of congenital infection worldwide. Strain variation in HCMV may predict severity or outcome of congenital HCMV disease. Previous studies have associated a particular genotype with specific sequelae or more severe illness, but the results were contradictory. There are no previous studies addressing the genotype of HCMV in Iraq. Therefore, the present study is aimed at molecular detection and genotyping of HCMV isolated from symptomatic congenitally/perinatally infected neonates. This prospective study comprised 24

Hepatitis C virus (HCV) chronic infection can be associated with extrahepatic manifestations such as mixed cryoglobulinaemia and lymphoproliferative disorders that are endowed with increased rates of morbidity and all-cause mortality. In this study, we used flow cytometry to evaluate the effect of interferon-free antiviral treatment on peripheral blood lymphocytes in HCV-infected patients with or without associated lymphoproliferative disorders. Flow cytometry analysis of peripheral blood lymphocytes was performed at baseline and at the end of treatment. In HCV-infected patients with lymphopro

Human cytomegalovirus (HCMV) replication requires host metabolism. Infection alters the activity in multiple metabolic pathways, including increasing fatty acid elongation and lipid synthesis. The virus-host interactions regulating the metabolic changes associated with replication are essential for infection. While multiple host factors, including kinases and transcription factors, important for metabolic changes that occur following HCMV infection have been identified, little is known about the viral factors required to alter metabolism. In this study, we tested the hypothesis that pUL37x1 is

Poly(hexamethylene biguanide) (PHMB) is a polycationic antimicrobial polymer exhibiting broad-spectrum activity against bacteria, fungi, and viruses, and is widely used in medical settings for infection prevention and control. However, the relationship between chemical structure and antimicrobial activity remains unclear. In this study, we synthesised and characterised a series of polymeric biguanides with systematically varied alkyl chain lengths to examine the effects of structural variation on physicochemical properties and antimicrobial activity. A 28-year-old man presented with selective immunoglobulin A deficiency and severe diarrhea responding to a gliadin-free diet. Biopsy samples of the small intestine showed dense T-cell infiltrations in the lamina propria and a slight increase of intraepithelial T-lymphocytes. No clonal rearrangement of the T-cell receptor c-beta chain genes was detectable by Southern blotting. Four years later, at the age of 32, the patient was hospitalized again with liver failure, abdominal lymphadenopathy, pancytopenia, and recurrent bacterial infections. The influence of salicylic (SA) and jasmonic (JA) acids as signaling systems mediators on the generation of H2O2 and expression of genes encoding protective proteins was studied in the leaves of wheat Triticum aestivum L. upon infection with the pathogen of septoriosis Septoria nodorum Berk. It was found that presowing treatment of seeds with SA and JA decreased the development of the fungus on the leaves of wheat and had a stimulating effect on the production of H2O2 in the area of infection. An increased expression of genes encoding oxalate oxidase AJ556991.1 and anionic peroxidase TC 151917

Molecular biology techniques play a significant role in diagnostics of many infectious diseases. Polymerase chain reaction (PCR) is used to confirm tick-borne diseases e.g. Borrelia burgdorferi infection. Extension of PCR method in laboratory diagnostics of Lyme disease (LD) gives a possibility of confirmation of spirochete infection before patient body managed to produce antibodies. Diversity of material, which may be tested (blood, cerebrospinal fluid, skin biopsies, synovial fluid) and possibility of using several conservative genes for Borrelia genospecies additionally widen a diagnostic u

Toxoplasma gondii is an obligate intracellular parasite recognized as a causal agent of toxoplasmosis; zoonotic disease endemic in many countries worldwide, including Mexico. Different species of animals participate in the wild cycle infection, including opossums of the species Didelphis virginiana. Thirteen D. virginiana were captured in Yucatan, Mexico. Detection of T. gondii was achieved by Polymerase Chain Reaction, which determined an infection of 76.9% (10/13) in brains. Positive amplicons were sequenced for analysis, this produced results similar to T. gondii with identity and coverage

To study the relation of neonatal hyaline membrane disease (HMD) and congenital human cytomegalovirus (HCMV) infection, two pairs of primers were synthesized according to the sequence of HCMV conserved region of the immediate--early(IE) gene. The nested polymerase chain reaction (nested-PCR) was used for the detection of HCMV-DNA of the paraffin-embedded lung tissues from neonatal HMD and non-neonatal HMD. 11 of 30 neonatal HMD patients were positive, the positive rate was 36.7%. Specimens of non-neonatal HMD patients were all negative. The difference between the two groups was significant (ch

Peripheral neuropathy is a frequent complication in patients suffering from type II mixed cryoglobulinaemia (mCGII), a sort of vasculitis that is strongly associated with hepatitis C virus (HCV) infection and characterised by high concentrations of anti-HCV antibodies and HCV RNA in the cryoprecipitates. We report the finding of HCV RNA in homogenates of nerve biopsies from five such patients, by reverse transcription-polymerase chain reaction (RT-PCR) amplification of different regions of the viral genome. HCV RNA was localized in epineurial cells by in situ RT-PCR. Our data suggest that HCV
