FGFR3 mutation causes abnormal membranous ossification in achondroplasia.

FGFR3 gain-of-function mutations lead to both chondrodysplasias and craniosynostoses. Achondroplasia (ACH), the most frequent dwarfism, is due to an FGFR3-activating mutation which results in impaired endochondral ossification. The effects of the mutation on membranous ossification are unknown. Fgfr3(Y367C/+) mice mimicking ACH and craniofacial analysis of patients with ACH and FGFR3-related craniosynostoses provide an opportunity to address this issue. Studying the calvaria and skull base, we observed abnormal cartilage and premature fusion of the synchondroses leading to modifications of for…

Anesthesia for cesarean delivery in an achondroplastic dwarf: a case report.

There are more than 100 different types of dwarfism. Achondroplasia is the most common form of this rare condition. The incidence of achondroplasia in the United States is about 15 per 1 million births. Although inherited as an autosomal dominant condition, 80% of cases result from spontaneous mutation. Underdevelopment and premature ossification of bones result in characteristic craniofacial and spinal abnormalities. Limited neck extension, foramen magnum stenosis, a large tongue, large mandible, and atlanto-axial instability can lead to increased difficulty of airway management. Severe kypho…

The ocular manifestations of Weissenbacher-Zweymuller syndrome.

PURPOSE: Weissenbacher-Zweymuller syndrome (WZS) is an autosomal recessive disorder of delayed skeletal maturation. Its characteristic features include rhizomelic dwarfism with metaphyseal and vertebral changes. It has been challenged whether WZS is a part of the spectrum of Stickler syndrome. We report ocular findings in the largest ever-presented series of patients with WZS. METHODS: Patients underwent a paediatric examination, including assessment of growth and development, genetic work-up and X-ray of vertebra and long bones. All had a complete ophthalmic examination, cycloplegic refracti…

Pulmonary hypoplasia in Jarcho-Levin syndrome.

Jarcho-Levin syndrome, also known as spondylothoracic dysplasia and characterized by short trunk dwarfism, "crab-like" rib cage, with ribs and vertebral defects; it is not uncommon in Puerto Ricans. Many patients die in early infancy due to respiratory compromise associated to lung restriction and the reported cases emphasize mostly the skeletal malformations associated to the syndrome. We report the autopsy findings in a newborn with isolated Jarcho-Levin syndrome emphasizing pulmonary pathology. He was a pre-term male who died of respiratory failure at three hours old and, autopsy findings c…

Clinical features of symptomatic Rathke's cleft cyst.

To investigate the clinical features of Rathke's cleft cysts (RCCs), we retrospectively analyzed 15 cases with histologically confirmed RCCs … enough to compress the infundibular system. Our series showed relative high incidence of pituitary dwarfism and diabetes insipidus (DI). These facts indicated that RCCs could evoke hyposecretion of growth hormone in young patients and DI in aged patients by direct compression of the pituitary gland in the early stage of progression. All cases who had head …

Growth hormone therapy in achondroplasia.

Achondroplasia is one of the most common causes of severe rhizomelic dwarfism. We have previously reported the growth-promoting effect of growth hormone (GH) in this disorder. In this expanded clinical study, dose dependency and the long-term effect of GH were also investigated. Prepubertal children with achondroplasia (82 males and 63 females) were randomly divided into 2 groups. Patients were treated with 0.5 IU/kg per week or 1.0 IU/kg per week subcutaneous recombinant human GH. Of 75 patients, the mutational analysis of fibroblast growth factor receptor-3 revealed that G1138A was detected …

[Cockayne's syndrome with unusual retinal involvement (report of one family)].

Cockayne's syndrome is a very rare autosomal recessive affection. Ocular involvement is an essential element for positive diagnosis; the retina shows a typical salt and pepper retinitis with optical atrophy. We report a family with four brothers who had Cockayne's syndrome with unusual retinal involvement. The patients' parents were first cousins. Ophthalmologic examination of the mother showed unilateral left pigmentary retinopathy localized in the peripapillary region. The father's ophthalmological examination was normal. The four brothers presented disharmonious dwarfism, cutaneous hyperpig…

[Idiopathic hypoparathyroidism (isolated)].

X-linked recessive, autosomal dominant and autosomal recessive types of familial isolated hypoparathyroidism have been reported, and there are a variety of syndromes of isolated hypoparathyroidism in association with other developmental anomalies involving dwarfism, cortical thickening of tubular bones, nephropathy, sensoryneural deafness and lymphedema. The molecular genetic basis of these forms of idiopathic hypoparathyroidism has been investigated recently. Several genetic abnormalities were elucidated; mutations of the parathyroid hormone gene in some families of autosomal dominant and aut…

[Reinhardt-Pfeiffer mesomelic dysplasia or dyschondrosteosis? Is the distinction well-founded? Apropos of a familial case with variable expression].

A familial observation of Reinhardt-Pfeiffer type mesomelic chondrodysplasia spanning three generations is reported. This case clearly shows that expression of the disease can vary widely within a given family. One member affected as a fetus had a severe form suggesting Langer mesomelic dwarfism syndrome, whereas his mother was free of clinical symptoms and his maternal aunt had a typical form of Reinhardt-Pfeiffer syndrome. Clinical manifestations in the other affected family members were perfectly consistent with dyschondrosteosis syndrome. The very broad spectrum of clinical patterns in thi…

Variation of quantitative and qualitative changes of enchondral ossification in heterozygous achondroplasia.

Whereas the radiologic features of achondroplasia--the most common type of skeletal dysplasia in adults--are clearly defined there is still some debate about the severity and the type of histologic changes. Earlier descriptions reported severe disturbance of enchondral ossification to be typical of achondroplasia: They are, however, misleading by dealing mostly with cases of lethal neonatal dwarfism (e.g. thanatophoric dysplasia). Newer findings confirmed that only minor, quantitative lesions are typical of heterozygous achondroplasia. But even in recent years some observers noted more severe …

PtCP1 Is an Extraplastidial Cysteine Protease Involved in Leaf Protein Degradation of Populus tomentosa Carr.

Protein turnover is essential for cellular metabolism, organelle biogenesis, stress adaptation, and ultimately the viability of cells and tissues. Papain-like cysteine proteases (PLCPs) are one of the vital components in protein degradation. PLCPs have been reported to act in senescence-associated proteolysis, but their roles in vegetative growth remain unclear. We identified PtCP1, an AALP-like PLCP in Populus tomentosa, localized to the vacuole and acid-triggered activated. CRISPR/Cas9-generated loss-of-function mutant (d7) showed dwarfism and non-stomatal photosynthetic limitations. On the …

SlTWD1 Affects Leaf Growth Primarily via Cell Expansion and Contributes to Leaf and Fruit Size Control in Tomato.

Leaf growth is a major determinant of plant architecture and productivity. TWISTED DWARF1 (TWD1/FKBP42) is a conserved immunophilin involved in plant growth and development, but its function in tomato remains unclear. Here, we identified and characterized SlTWD1, the tomato homolog of TWD1. SlTWD1 is highly conserved among land plants and functionally complemented the developmental defects of an Arabidopsis thaliana cr-twd1 mutant. SlTWD1 was broadly expressed in vegetative and reproductive tissues. CRISPR/Cas9-mediated disruption of SlTWD1 caused severe dwarfism and reduced leaf and fruit siz…

CGF1 and CGF2 jointly participate in iron homeostasis in chloroplasts.

Iron (Fe) plays a crucial role in chloroplast function, and its deficiency results in Fe chlorosis, a syndrome characterized by reduced biomass production and plant mortality. In this study, Fe supplementation rescued the dwarfism and chlorotic phenotype of cgf1 cgf2 mutant, suggesting that CGF1 and CGF2 are essential regulators of Fe homeostasis. Subcellular localization analysis revealed that both CGF1 and CGF2 localize to the chloroplast envelope. Their absence led to a decreased photosynthetic protein abundance and reduced photosynthetic supercomplex levels, demonstrating that CGF1 and CGF…

Allelic variations and interactive feedback in major regulators of plant architecture confer high-altitude adaptation in Arabidopsis thaliana.

Ideal plant architectures are crucial for adapting to environmental changes, yet the molecular mechanisms behind high-altitude plant adaptations remain elusive. We characterized the dwarf and highly branched traits of the Tibet ecotype of Arabidopsis thaliana (L.) Heynh. Quantitative trait locus (QTL) analysis revealed that GA5 and BRC1, as major genes regulating plant height and branching, respectively, underlie the variation in plant architecture of the Tibet ecotype. Loss-of-function of GA5 disrupts gibberellin biosynthesis, leading to dwarfism, and natural variations in the BRC1 promoter r…
