The Transcription Factor DPB Confers Antiviral Defence Against Potato Virus X by Modulating MYB-Dependent Signalling.

As a transcriptional co-factor of E2F, DP proteins are typically involved in the regulation of plant cell-cycle-related processes. However, whether DP proteins participate in transcriptional regulation associated with plant antiviral defence remains largely unclear. This study demonstrates that NbDPB, a DP family protein in Nicotiana benthamiana, positively regulates resistance to potato virus X (PVX) by functioning as a transcription factor. Knockout of NbDPB increased plant susceptibility to PVX, while its overexpression significantly suppressed viral accumulation. Furthermore, we identified

Large effect of phosphate-solubilizing bacteria on the growth and gene expression of Salix spp. at low phosphorus levels.

Phosphorus is one of the most important nutrients required for plant growth and development. However, owing to its low availability in the soil, phosphorus is also one of the most difficult elements for plants to acquire. Phosphorus released into the soil from bedrock quickly becomes unavailable to plants, forming poorly soluble complexes. Phosphate-solubilizing bacteria (PSB) can solubilize unavailable phosphorus-containing compounds into forms in which phosphorus is readily available, thus promoting plant growth. In this study, two willow species, Salix dasyclados cv. Loden and Salix schweri

Identification of the DOG1 Gene Family in Cultivated Peanut and Negative Regulation of Ralstonia solanacearum Infection by AhDOG1-L4.

Bacterial wilt, caused by Ralstonia solanacearum, is a devastating soilborne disease often referred to as plant cancer. It threatens hundreds of species of plants and causes losses in yield and a deterioration in the quality of infected plants. Delay of Germination 1 (DOG1) plays vital roles in plant growth, development, and abiotic and biotic stress responses. However, the functions of DOG1 in biotic stress remain unexplored in cultivated peanut (Arachis hypogaea L.). In this study, we systematically identified and characterized the AhDOG1 gene family in peanut using bioinformatics approaches

Genome-wide characterization of MADS-box genes and their roles in axillary bud development in tobacco.

A total of 118 NtMADS-box genes were identified in tobacco, revealing their potential roles in axillary bud development. Preliminary overexpression analysis indicated that NtMADS91 promotes axillary bud development. MADS-box transcription factors are core regulators of plant development, but their functions in axillary bud development in Nicotiana tabacum L. have not been systematically elucidated. In this study, 118 NtMADS-box genes were identified from the tobacco genome. Phylogenetic analysis classified them into type I (comprising the Mα and Mγ subfamilies) and type II (comprising the MIKC

A comparative study of microRNAs in different stages of Eimeria tenella.

Apicomplexan parasites have divergent biogenesis machinery for small RNA generation. Analysis has shown that parasites in Plasmodium and Cryptosporidium as well as many species in Leishmania or Trypanosoma do not have a complete machinery in small RNA biogenesis. Recently, the miRNA-generating system of Toxoplasma has been identified as plant/fungal-like and its miRNAome has been elucidated. However, the microRNA (miRNA) expression profiles and their potential regulatory functions in different stages of Eimeria tenella remain largely unknown. In this study, we characterized the RNA silencing m

C2-mediated decrease in DNA methylation, accumulation of siRNAs, and increase in expression for genes involved in defense pathways in plants infected with beet severe curly top virus.

Cytosine methylation is one of epigenetic information marked on the DNA sequence. In plants, small interfering RNAs (siRNAs) target homologous genomic DNA sequences for cytosine methylation. This process, known as RNA-directed DNA methylation (RdDM), plays an important role in transposon control, regulation of gene expression and virus resistance. In this paper, we demonstrate that the C2 protein encoded by a geminivirus (beet severe curly top virus, BSCTV) mediated a decrease in DNA methylation of repeat regions in the promoters of ACD6, an upstream regulator of the salicylic acid defense pat

Involvement of the small GTPase Rac in the defense responses of tobacco to pathogens.

During the hypersensitive response (HR), plants accumulate reactive oxygen species (ROS) that are likely generated at least in part by an NADPH oxidase similar to that found in mammalian neutrophils. An essential regulator of mammalian NADPH oxidase is the small GTP-binding protein Rac. To investigate whether Rac also regulates the pathogen-induced oxidative burst in plants, a dominant negative form of the rice OsRac1 gene was overexpressed in tobacco carrying the N resistance gene. Following infection with Tobacco mosaic virus (TMV), DN-OsRacl plants developed smaller lesions than wild-type p

Antagonistic Transcriptional and Metabolic Networks Are Associated with Anthocyanin Accumulation in Maize Seedlings.

Background: Anthocyanin accumulation is a developmentally regulated trait shaped by complex interactions between metabolic and transcriptional networks. However, dissecting the regulatory mechanisms underlying anthocyanin biosynthesis is often complicated by confounding variation in plant growth and environmental conditions. Methods: Here, we used a time-resolved multi-omics approach to investigate anthocyanin accumulation in maize seedlings by comparing an anthocyanin-rich inbred line PH19401 with an anthocyanin-deficient line YPX across five developmental stages. Results: Untargeted metabolo

Analysis of gene expression profile of Arabidopsis genes under trichloroethylene stresses with the use of a full-length cDNA microarray.

Trichloroethylene (TCE) is a widespread and persistent environmental contaminant. Plants are able to take up a range of harmful organic compounds, including some of the most abundant environmental pollutants like TCE. In this study, complementary DNA microarrays were constructed to have a better view of transcript expression in Arabidopsis thaliana during TCE-induced stress. The microarray analysis demonstrated the complexity of gene expression patterns resulting from TCE. A total of 1,020 transcripts were differentially up-regulated by TCE. Those genes might specifically contribute to the TCE

Identification of novel pepper genes involved in Bax- or INF1-mediated cell death responses by high-throughput virus-induced gene silencing.

Hot pepper is one of the economically important crops in Asia. A large number of gene sequences, including expressed sequence tag (EST) and genomic sequences are publicly available. However, it is still a daunting task to determine gene function due to difficulties in genetic modification of a pepper plants. Here, we show the application of the virus-induced gene silencing (VIGS) repression for the study of 459 pepper ESTs selected as non-host pathogen-induced cell death responsive genes from pepper microarray experiments in Nicotiana benthamiana. Developmental abnormalities in N. benthamiana

Cross-kingdom comparison of transcriptomic adjustments to low-oxygen stress highlights conserved and plant-specific responses.

High-throughput technology has facilitated genome-scale analyses of transcriptomic adjustments in response to environmental perturbations with an oxygen deprivation component, such as transient hypoxia or anoxia, root waterlogging, or complete submergence. We showed previously that Arabidopsis (Arabidopsis thaliana) seedlings elevate the levels of hundreds of transcripts, including a core group of 49 genes that are prioritized for translation across cell types of both shoots and roots. To recognize low-oxygen responses that are evolutionarily conserved versus species specific, we compared the

Specific patterns of changes in wheat gene expression after treatment with three antifungal compounds.

The two fungicides azoxystrobin and fenpropimorph are used against powdery mildew and rust diseases in wheat (Triticum aestivumL). Azoxystrobin, a strobilurin, inhibits fungal mitochondrial respiration and fenpropimorph, a morpholin, represses biosynthesis of ergosterol, the major sterol of fungal membranes. Although the fungitoxic activity of these compounds is well understood, their effects on plant metabolism remain unclear. In contrast to the fungicides which directly affect pathogen metabolism, benzo(1,2,3) thiadiazole-7-carbothioic acid S-methylester (BTH) induces resistance against whea

A novel approach to enhance resistance to vascular disease by expressing cell-death-inducing fungal elicitors in the xylem tissue.

Constitutive expression of hypersensitive cell-death-inducing effectors secreted by pathogens has proved to be difficult in plants. Herein, xylem-specific promoters (PVND7 or PVND2) were utilized to express three cell-death-inducing proteins (FocSIX1A, FocSIX6 or TvSm1) of fungal origin in banana plants. The regenerated plants were not only healthy but also showed improved resistance to fungal (Foc vcg0124 and Foc vcg0125) infection. These banana plants developed systemic resistance-like symptoms, including elevated jasmonic acid content, callose depositions, increased xylem wall thickness and

Genome-wide identification and characterization of CONSTANS-like gene family in radish (Raphanus sativus).

Floral induction that initiates bolting and flowering is crucial for reproductive fitness in radishes. CONSTANS-like (CO-like, COL) genes play an important role in the circadian clock, which ensures regular development through complicated time-keeping mechanisms. However, the specific biological and functional roles of each COL transcription factor gene in the radish remain unknown. In this study, we performed a genome-wide identification of COL genes in the radish genome of three cultivars including 'Aokubi', 'kazusa' and 'WK10039', and we analyzed their exon-intron structure, gene phylogeny

Histochemical Analyses Reveal That Stronger Intrinsic Defenses in Gossypium barbadense Than in G. hirsutum Are Associated With Resistance to Verticillium dahliae.

Verticillium wilt, caused by Verticillium dahliae Kleb., is a serious threat to cotton (Gossypium spp.) crop production. To enhance our understanding of the plant's complex defensive mechanism, we examined colonization patterns and interactions between V. dahliae and two cotton species, the resistant G. barbadense and the susceptible G. hirsutum. Microscopic examinations and grafting experiments showed that the progression of infection was restricted within G. barbadense. At all pre- and postinoculation sampling times, levels of salicylic acid (SA) were also higher in that species than in G. h
